Abstract:A cobalt(III)-catalyzed C-2 selective C–H alkynylation of indoles using hypervalent iodine-alkyne reagents is described. A broad range of synthetically useful functional groups (−F, −Cl, −Br, −CO2Me, −CN) were tolerated, providing an efficient and robust protocol for the synthesis of C-2 alkynylated indoles. The pyrimidyl and silyl protecting groups could be easily removed to give the corresponding 2-ethynyl-1H-indole.

Abstract:A Rh(I)‐catalyzed chelation‐assisted C2‐selective C−H decarbonylative alkylation of indoles with readily available, cheap, safe and structurally diverse alkyl carboxylic acids or anhydrides has been developed. A wide variety of primary and secondary alkyl carboxylic acids and differently substituted indoles are compatible with this transformation, allowing facile synthesis of various C2‐alkylated indoles with high efficiency and broad tolerance of diverse functional groups. The reaction proceeds in the absence of any external oxidant, and the presence of readily installable and removable N‐pyrimidyl directing group is critical for catalysis. The process is convenient and scalable, and avoids the use of a dried solvent and an inert atmosphere. Moreover, selective C7‐alkylation and C2, C7‐dialkylation have also been achieved by slightly modifying the reaction conditions.

Abstract:A Co(III)-catalyzed, internal and terminal alkyne-compatible indole synthesis protocol is reported herein. The N-amino (hydrazine) group imparts distinct, diverse reactivity patterns for directed C-H functionalization/cyclization reactions. Notable synthetic features include regioselectivity for a meta-substituted arylhydrazine, regioselectivity for a chain-branched terminal alkyne, formal incorporation of an acetylenic unit through C2-desilylation on a C2-silylated indole derivative, formal inversion of regioselectivity through consecutive C3-derivatization and C2-desilylation processes, and formal bond migration for a linear-chain terminal alkyne.

Abstract:A highly chemo-, regio- and stereoselective cobalt-catalyzed Markovnikov hydrosilylation of alkynes was developed. Various functionalized groups, such as halides, free alcohols, free aniline, ketones, esters, amides, and nitriles are tolerated, which may lead to further applications and late-stage derivatizations. To date, this is the most efficient cobalt catalytic system (TOF=65520h(-1); TOF=turnover frequency) for hydrosilylation of alkynes. The Hiyama-Denmark cross-coupling reactions of vinylsilanes with aryl iodides underwent smoothly to afford 1,1-diarylethenes. A unique regioselectivity-controllable hydrosilylation/hydroboration reaction of alkynes was also described.

Abstract:A cobalt(II)-catalyzed C2-H carbonylation of indoles with amines toward indole-2-carboxamides has been developed. By employing Co(OAc)(2)·4H(2)O as an inexpensive catalyst and using benzene-1,3,5-triyl triformate (TFBen) as the CO surrogate, a variety of indole-2-carboxamide derivatives were produced in moderate to high yields. Additionally, several bioactive-molecule-related compounds can be applied as substrates, as well.

Abstract:<p>A new Rh(III)-catalyzed direct C-H alkenylation of 2,3-unsubstitued indoles with alkenyl borates through C-H activation followed by transmetallation was described. This protocol provides an efficient method for the synthesis of terminal C2-alkenylindoles under mild conditions, and also shows broad substrate scope and high functional group tolerance with respect to both components. Furthermore, the C2-alkenylated indole can be easily transformed into carboline derivative, which is an important polycyclic indole moiety in natural products and drug molecules.</p><p>This article is protected by copyright. Abstract:A mild Co(III)-catalyzed oxidative annulation of N-arylureas and internal alkynes has been developed. The use of less electrophilic ureas other than acetamides as directing groups is crucial for the reaction. Abstract:With alanine as a transient directing group, Pd-catalyzed regioselective alkynylation at the indole C4-position was successfully established in a good yield. The total synthesis of the PAF antagonist demonstrated the synthetic utility of this protocol. The regioselectivity was explicitly proven by the prepared C4-selective palladacycle intermediate in the catalytic process and the DFT calculation of the energy barriers of C4- and C2-site-selective C-H activation of indole.

Abstract:Herein, we report a straightforward approach for synthesizing C3 -alkylated indoles selectively via an iron-catalyzed alkylation of indoles using alcohols as the alkylating agents. A well-defined, air-stable, and easy-to-prepare Fe(II) catalyst of a redox-active tridentate arylazo scaffold was used as a catalyst. Various C3 -alkylated indoles were prepared selectively in moderate to good isolated yields by coupling indoles with different substituted alcohols. The methodology is compatible with the gram-scale synthesis. Control experiments were performed to unveil the mechanism, which revealed that the alkylation reaction proceeds via borrowing-hydrogen pathway where the coordinated azo-aromatic ligand actively participates during catalysis, acting as an electron and hydrogen reservoir. Abstract:An air‐stable, inexpensive, and isolable cobalt(II) complex (C1) of N‐((1‐methyl‐1H‐imidazol‐2‐yl)methyl)‐2‐(phenylselanyl)ethan amine (L1) was synthesized and characterized. The complex was used to catalyze a one‐pot cascade reaction between 2‐(2‐aminophenyl)ethanols and benzyl alcohol derivatives. Interestingly, 2‐aryl‐3‐formylindole derivatives were formed instead of N‐alkylated or C‐3 alkylated indoles. A broad substrate scope can be activated using this protocol with only 5.0 mol% catalyst loading to achieve up to 87% yield of 2‐aryl‐3‐formylindole derivatives. The mechanistic studies suggested that the reaction proceeds through tandem imine formation followed by cyclization.
