Abstract:A rapid and efficient method for synthesis of α, β-conjugated acetylenic ketones involves reaction of phenylacetylene with aroyl chlorides in the presence of catalytic amount CuI under microwave irradiation to give the corresponding α, β-conjugated acetylenic ketones 3 a–i in 76–93% yields.

Abstract:A microwave (MW)-assisted three-component coupling of amino alcohols, aldehydes, and alkynes is developed under catalysis by CuCl. Compared with thermal conditions, MW irradiation greatly increases the reaction efficiency. The reactions of various primary N-alkyl/arylamino alcohols, aliphatic/aromatic aldehydes, and alkynes are systematically investigated, affording the desired products in moderate to good yields. Notably, acetylene is also an effective reactant under the current MW-assisted conditions.

Abstract:Methods toward the dimerization and homocoupling of terminal alkynes have been established that employ Co complexes along with tBuOK under different atmosphere. A variety of Z-1,3-enynes were furnished with a high Z:E (up to 93:7) ratio in moderate to very good yields under inert atmosphere. Alternatively, the Glaser homocoupling reaction of terminal alkynes was also accomplished with a Co catalyst, successfully affording various 1,3-diynes in excellent yields (up to 93%) under oxygen.

Abstract:Biaryls and bis-heterocycles can be synthesized conveniently,efficiently,and generally via palladium(0)-catalyzed homocoupling of aryl bromides with hexabutylditin as reductant.This method delivers biaryls in good to excellent yields in 15 min under controlled microwave irradiation.Various bromo-derivatives of electron-rich or electron-poor arenes and aromatic heterocycles including benzene,thiophene,furan,pyridine,thiazole,and pyrimidine are easily coupled.This homocoupling method is also applicable to the bromides of fused-ring arenes such as naphthalene,quinoline,benzothiophene,and indole.The procedure is compatible with various reactive functionalities such as hydroxy,amino,carboxyl,formyl,acyl,alkoxycarbonyl,nitro,and cyano groups.The reaction appears to proceed through in situ formation of an aryl tin,followed by a Stille coupling to the aryl bromide.

Abstract:Despite the success of Sonogashira coupling for the synthesis of arylalkynes and conjugated enynes, the engagement of unactivated alkyl halides in such reactions remains historically challenging. We report herein a strategy that merges Cu-catalyzed alkyne transfer with the aryl radical activation of carbon–halide bonds to enable a general approach for the coupling of alkyl iodides with terminal alkynes. This unprecedented Sonogashira-type cross-coupling reaction tolerates a broad range of functional groups and has been applied to the late-stage cross-coupling of densely functionalized pharmaceutical agents as well as the synthesis of positron emission tomography tracers.

Abstract:An efficient and effective microwave-assisted cross-coupling of terminal alkynes with various aryl chlorides including sterically hindered, electron-rich, electron-neutral, and electron-deficient aryl chloride is developed. It proceeds faster and generally gives good to excellent yields and also can be extended successfully to the Suzuki coupling and Buchwald-Hartwig amination, as well as the Heck coupling with inert aryl chlorides. The short reaction times and simple reaction conditions coupling with a broad substrate scope render this method particularly attractive for the efficient preparation of biologically and medicinally interesting molecules.

Abstract:Microwave irradiation promoted the intramolecular cycloaddition of 2-azidoacetamides derived from a-chiral propargylic amines, affording 1,4,5-trisubstituted triazoles 4 bearing a chiral aminomethyl side chain at C5. In contrast, for the same substrates 3a-k, Cu(I)-NHC complexes catalyzed the intermolecular cycloaddition in an unexpected desilylative fashion, leading to 1,4-disubstituted triazoles 5. This demonstrates that 1-silyl alkynes can be employed as substrates for CuAAC with a suitable coupling partner.

Abstract:An efficient copper-catalyzed method has been developed for Sonogashira couplings of aryl halides with terminal alkynes in water. The protocol uses inexpensive CuBr as the catalyst, 1,10-phenanthroline as the ligand, tetrabutylammonium bromide (TBAB) as the phase-transfer catalyst, environmentally friendly water as the solvent, and various internal alkynes were synthesized in good to excellent yields.

Abstract:We report a microwave-assisted synthetic protocol for the C-S cross-coupling reaction of aryl iodides and 8-mercaptoadenine using CuI and n-Bu4NBr with NaOt-Bu in DMF. The method is rapid, simple, and cost efficient, and leads in high yields to biologically relevant aryl sulfides.

Abstract:A highly efficient method for the synthesis of tri-aryl-substituted allenes has been developed through Cu(I)-catalyzed coupling of diaryldiazomethanes with terminal alkynes. The reaction is under mild conditions and uses simple and inexpensive CuI as the catalyst. Mechanistically, the reaction follows a pathway involving Cu(I) carbene migratory insertion.
