Abstract:3-Hydroxybenzylhydrazine dihydrochloride was synthesized from 3-hydroxybenzaldehyde and methoxycarbonylhydrazine via condensation, hydrogenation, decarboxylation and salt formation in a total yield of 77.1%. The yield of condensation was 94.6% and of hydrogenation was 99.8% by using Pd/C (5%) which can be reused 3 times at the optimal quantity of 3% (W/W). The yield of salt formation was 81.7%. This route is easy to industrialize for its simple operation, mild reacting condition and low cost.

Abstract:An improved process was reported to solve the problems such as low yield and high costs in the synthesis of 3-n-propyl-2, 4-dihydroxyacetophenone, which is a key intermediate of Leukotriene receptor antagonist. The target compound was synthesized starting from m-dihydroxybenzene, via acylation, allylation, Claisen rearrangement and hydrogenation reduction. The optimal reaction conditions were as follows. The acylation reaction which used acetic acid as acylation agent and ZnCl 2 as catalyst was carried out under reflux for 5 h, and 77.9% yield was obtained. The allylation reaction using allyl chloride as allylation reagent and NaBr, KI as catalyst was carried out at 40°C for 2 h and continued at 70°C for 14 h, to give 98.6% yield. In Claisen rearrangement carried out at 205°C for 4.5 h, diphenyl ether was used as reaction solvent and NaCl was used as catalyst, and the yield was improved to 67.4%. In the final step, the hydrogenated reduction with Pd/C (w(Pd) is 5%) as catalyst was carried out at 45°C for 6 h, and 88.6% yield was obtained. The process is much of industrial value because of high yield, cheap and available materials, and convenient operations. The structure of the main compounds was confirmed by 1H-NMR, GC-MS and IR.

Abstract:A new route for the synthesis of 6-bromo-4,4-dimethylthiochroman was proposed and the optimized conditions for each step were obtained.Bromobenzene was selected as starting material and 6-bromo-4,4-dimethylthiochroman was prepared via chlorosulfonation,reduction,thioetherification and cyclization with the yield of 88.5%.Except the reduction,which had to be performed at a higher temperature,the other three steps proceeded at temperatures lower than 50oC.Thus both the energy cost and byproducts were decreased.Compared to previous routes,the proposed one in this work is advanced for its lower cost and less contamination,so it is suitable to be industrially amplified.

Abstract:1-Chloro-2,5-dimethoxybenzene was prepared from p-benzoquinone through chlorination and methylation with total yield of 78%. p-Benzoquinone was firstly converted catalytically to 1-chloro-2,5-dihydroxybenzene in the binary system of dichloroethane and hydrochloric acid at 35℃ with yield of 89%, then the final product was synthesized by methylation of obtained 1-chloro-2,5-dihydroxybenzene with dimethyl sulfate in the presence of sodium hydroxide. The intermediate 1-chloro-2,5-dihydroxybenzene and the obtained title compound 1-chloro-2,5-dimethoxybenzene were characterized by IR and GC-MS respectively. The results showed that their chemical structures were correct and the purity of title compound was over 98%.

Abstract:A one-pot two-step reaction of 4-aminocoumarin with arylglyoxal monohydrates and p-toluenesulfonates has been developed, which offers 3-alkoxy-substituted chromeno[4,3-b]pyrrol-4(1H)-ones in moderate to good yields. At first, arylglyoxal monohydrates and 4-aminocoumarin were converted into 3-hydroxychromeno[4,3-b]pyrrol-4(1H)-one intermediates in AcOH under reflux. Then the title compounds in an in situ reaction of the intermediates with p-toluenesulfonates catalyzed by DBU in refluxing toluene were obtained.

Abstract:Methyl 3-oxo-2,6,6-trimethylcyclohex-1-ene-1-carboxylate was synthesised over eight steps from the starting material of citral instead of α-ionone. KMnO4, HC(OEt)3 and O2/TEMPO/CuCl were substituted for NaIO4, CH3I and pyridinium chlorochromate, respectively. Every step was simplified and gave a 48% overall yield. The procedure is suitable for large scale production.

Abstract:p-trifluoromethylbenzonitrile was prepared from p-(trifluoromethyl)chlorobenzene and potussium cyanide catalyzed by tris(triphenylphosphine)nickle(0), which was prepared from anhydrous NiCl2 and triphenylphosphine in the presence of zinc.And then, the produced intermideate, p-trifluoromethylbenzonitrile was treated with hydrogen sulfide to give p-(trifluoromethyl)thiobenzamide and the yield of 90% with the purity of 99.7% was obtained for the objective product.The structure is correct through IR and GC-MS testing.The operation process is simple and reaction condition is moderate.

Abstract:2, 4-Dimethoxybenzaldehyde was prepared using 1, 3-dimethoxybenzene as raw material through Vilsmeier formylation with DMF directly. The compound was characterized by IR and NMR as a qualified product. Under the optimal conditions the product obtained from the orthogonal experiments could be given in 94.2% yield with purity of 98.64%, after purification the purity of final compound could be reached 99.6%.

Abstract:The synthesis 2-tert-butyl-4-methylphenol is of great significance because of its wide application in industry, and the development of a highly efficient catalyst is necessary for the alkylation of p-cresol and tert-butyl alcohol. Here, an efficient and mild method was established. Caprolactam was chosen as the hydrogen-bonding acceptor; p-toluenesulfonic acid was employed as the hydrogen-bonding donor, and a deep eutectic solvent (DES) was prepared to catalyze the alkylation reaction. The structure of the deep eutectic solvent catalyst was characterized by 1H NMR spectra, thermogravimetric analysis, and Fourier transform infrared spectra (FT-IR). In addition, response surface design based on the Box–Behnken method was employed to optimize the alkylation reaction process parameters, and the study of reaction kinetics was also carried out subsequently. The recycle performance of the catalyst was evaluated by recovery experiments, and a good result was obtained. By drawing comparisons with the literature reported, we provide a mild method for the synthesis of 2-tert-butyl-4-methylphenol.

Abstract:In order to prepare p-hydroxybenzaldehyde by heterogeneous catalytic oxidation of p-cresol,catalyst of Co/Cu/γ-Al2O3 was prepared by impregnation with carrier γ-Al2O3,p-cresol was synthesized from p-hydroxybenzaldehyde.The effect of various reaction parameters on catalytic activity was discussed.The catalyst was characterized by TPR and the product was characterized by IR.It was found that when the catalyst calcined at 400 ℃ the catalytic activity was the best due to Co3+ → Co2+ reduction more easily occurs.Under suitable conditions(catalyst/p-cresol=3%~4%,NaOH/p-cresol =3/1,temperature=65 ℃)the highest yield of p-hydroxybenzaldehyde reached 45% in 10 h.

Abstract:Some synthetic methods of 1-bromo-3,4,5-trimethoxybenzene BTMOB are compared. One can obtain good yield by Sandmeyer reaction after 3,4,5-trimethoxyaniline is diazotized. The optimum reaction conditions are determined: n (ArNH2):n(NaNO2):n(NaBr) = 1.0:1.2:1.5, temperature of diazotization below 5°C and the complex of diazonium salt refluxed for 1 h during decomposition. Yield of BTMOB is about 87.5%.

Abstract:2,4,6-Trimethylbenzaldehyde(3) was synthesized from mesitylene via bromination,Grignard reaction and oxidation in an overall yield of about 64.2% with a purity of 97%.1,3,5Mesitylene reacted with bromine in water gaving 90% 1-bromo-2,4,6-trimethylbenzene(1),which was made into Grignard reagent and then reacted with formaldehyde to give 79% 2,4,6-trimethylbenzyl alcohol(2).2 was oxidated by chromic acid.The product was distilled giving 3 in yield of 90.3%.The structures of 2 and 3 were confirmed by 1H NMR and MS.

Abstract:A multistep conversion system composed of phenol hydroxylase (PHIND) and 2,3-dihydroxy-biphenyl 1,2-dioxygenase (BphCLA-4) was used to synthesize methylcatechols and semialdehydes from o- and m-cresol for the first time. Docking studies displayed by PyMOL predicted that cresols and methylcatechols could be theoretically transformed by this multistep conversion system. High performance liquid chromatography mass spectrometry (HPLC-MS) analysis also indicated that the products formed from multistep conversion were the corresponding 3-methylcatechol, 4-methylcatechol, 2-hydroxy-3-methyl-6-oxohexa-2,4-dienoic acid (2-hydroxy-3-methyl-ODA) and 2-hydroxy-5-methyl-6-oxohexa-2,4-dienoic acid (2-hydroxy-5-methyl-ODA). The optimal cell concentrations of the recombinant E. coli strain BL21 (DE3) expressing phenol hydroxylase (PHIND) and 2,3-dihydroxy-biphenyl 1,2-dioxygenase (BphCLA-4) and pH for the multistep conversion of o- and m-cresol were 4.0 (g·L−1 cell dry weight) and pH 8.0, respectively. For the first step conversion, the formation rate of 3-methylcatechol (0.29 μmol·L−1·min−1·mg−1 cell dry weight) from o-cresol was similarly with that of methylcatechols (0.28 μmol·L−1·min−1·mg−1 cell dry weight) from m-cresol by strain PHIND. For the second step conversion, strain BphCLA-4 showed higher formation rate (0.83 μmol ·L−1·min−1·mg−1 cell dry weight) for 2-hydroxy-3-methyl-ODA and 2-hydroxy-5-methyl-ODA from m-cresol, which was 1.1-fold higher than that for 2-hydroxy-3-methyl-ODA (0.77 μmol·L−1·min−1·mg−1cell dry weight) from o-cresol. The present study suggested the potential application of the multistep conversion system for the production of chemical synthons and high-value products.

Abstract:3,4-Dimethoxybenzaldehyde was synthesized from catechol through etherification and Vilsmeier reaction,and the yield was 87.8%.The second step,Vilsmeier reaction,was improved with lower N and P waste and much more environment friendly.The yield increased from 86.5% to 91.2%,and the amount of Vilsmeier decreased from 2.5 times to 1.5 times.

Abstract:Using prenal and prenol as precursors,3-methyl-2-butenal diprenyl acetal was obtained via an acetalization reaction using senecioic acid as catalyst.Then an elimination of 3-methyl-2-butenal diprenyl acetal catalyzed by ammonium dihydrogen phosphate yielded the cis/trans-prenyl-3-methyl-butadienyl ether.Further rearrangement of the elimination product through heating produced to the target citral.The conditions of acetalization reaction and elimination reaction were optimized.It was found that a conversion yield of 63%～64% for prenal could be achieved at 70～75 ℃ for 8 h with 0.3% senecioic acid as catalyst.After azeotropism treatment,the purity and yield of 3-methyl-2-butenal diprenyl acetal are 97.5% and 96.8%,respectively.Using 0.2%～0.5% ammonium dihydrogen phosphate as catalyst,the elimination reaction was carried out at 100～130 ℃ and 2.66 kPa.The cis/trans-prenyl-3-methylbutadienyl ether was distilled from the reaction batch and its purity and yield are 95.9% and 97.0%,respectively.The rearrangement reaction temperature was 120～130 ℃,the purity and yield of citral are 97.5% and 90.2%,respectively.
