Abstract:The rapid development of industrialization is causing several fundamental problems in plants due to the interaction between plants and soil contaminated with metallic nanoparticles (NPs). Numerous investigations have been conducted to address the severe toxic effects caused by nanoparticles in the past few decades. Based on the composition, size, concentration, physical and chemical characteristics of metallic NPs, and plant types, it enhances or lessens the plant growth at various developmental stages. Metallic NPs are uptaken by plant roots and translocated toward shoots via vascular system based on composition, size, shape as well as plant anatomy and cause austere phytotoxicity. Herein, we tried to summarize the toxicity induced by the uptake and accumulation of NPs in plants and also we explored the detoxification mechanism of metallic NPs adopted by plants via using different phytohormones, signaling molecules, and phytochelatins. This study was intended to be an unambiguous assessment including current knowledge on NPs uptake, accumulation, and translocation in higher plants. Furthermore, it will also provide sufficient knowledge to the scientific community to understand the metallic NPs-induced inhibitory effects and mechanisms involved within plants.

Abstract:Silver nanoparticles (AgNPs) are extensively utilized in industrial, biotechnological and medical fields because of their specific physical and chemical properties, which shown small size effects, surface and interface effects, quantum scale effects. Inevitably, wastewater containing AgNPs is leached into the soil. AgNPs in soil may be absorbed by plant roots and they affect plant growth. Understanding the complex interactions between AgNPs and plants is of crucial significance since plants are essential to the ecological environment as key players in ecosystems. Most previous reports have focused on AgNP phytotoxicity rather than positive effects in the plant field. This article reviews recent studies on the important aspects about nanoparticles including the green synthesis, absorption, migration, accumulation, biotransformation, biological effects, and the underlying molecular mechanisms of AgNPs affecting plants. We provide insights into the interactions between AgNPs and plants, helping to further understand AgNP-related effects and biosafety in agricultural production, as well as the practical application of various nanomaterials in agriculture.
Uptake Kinetics of Silver Nanoparticles by Plant: Relative Importance of Particles and Dissolved Ions

Abstract:Quantifying the relative bioavailability of particles versus ions is a key step toward understanding the mechanisms of bioaccumulation and toxicity of silver nanoparticles (AgNPs). Here we investigated AgNP uptake kinetics by wheat Triticum aestivum L. at different ages (i.e. 15- and 30-day after germination) in hydroponics. The concentration-dependent accumulation of AgNPs under the experimental conditions, in which AgNP dissolution in bulk suspension and at the biological interface was ruled out, confirmed the direct uptake of particles. This was further validated by the detection of Ag-containing particles within plants by single particle inductively coupled plasma mass spectrometry. Plants differentiated particles and dissolved ions, with uptake rate constants of particles and dissolved ions 4.3 +/- 0.6 to 5.2 +/- 0.6 and 288.6 +/- 13.4 to 450.6 +/- 63.4 L kg(-1) h(-1), respectively. Plant age appeared to have a negligible influence on uptake rate constant ratio of particles to ions. As a result, the relative contribution of particulate uptake to overall AgNP accumulation varied as a function of AgNP dissolution in nature. Particulate uptake was dominant when the dissolution was less than 1.2%. These findings help clearing up the inconsistency of uptake mechanisms of AgNPs in literature and could be used to predict their environmental impact and significance.
Nonselective Uptake of Silver and Gold Nanoparticles by Wheat.

Abstract:Metallic nanoparticles (NPs) show unique reactivity to crop plants, but the uptake mechanisms remain unclear. We quantitatively evaluated the phytoavailability of particles to wheat (Triticum aestivum L.) in hydroponics upon exposure to AgNPs (15 nm) or AuNPs (13 and 33 nm). Particles were physically separated from the released Ag ions by a dialysis membrane, under which particle-specific uptake of AgNPs could be discerned. Plants did not differentiate AgNPs and AuNPs during particle uptake, with uptake rate constants of 1.1 +/- 0.1, 1.2 +/- 0.3, and 1.2 +/- 0.1 L kg(-1) h(-1) for AgNPs, AuNPs (13 nm), and AuNPs (33 nm), respectively. We found little effect of particle size (13 or 33 nm AuNPs) or core composition (Ag or Au) on particle bioavailability. Plants stimulated the subsequent uptake of Evans blue stain and showed cell damage in root tips. These results imply similar physiological processes involved in particle-specific uptake of AgNPs and AuNPs. The internalization of particles was further confirmed by single particle inductively coupled plasma mass spectrometry (spICP-MS) and transmission electron microscope-energy dispersive spectrometer (TEM-EDS) analysis. The work here builds the knowledge base for the nature of particle-specific uptake of different NP types by crop plants.
Discerning the Sources of Silver Nanoparticle in a Terrestrial Food Chain by Stable Isotope Tracer Technique

Abstract:The increasing use of silver-containing nanoparticles (NPs) in commercial products has led to NP accumulation in the environment and potentially in food webs. Identifying the uptake pathways of different chemical species of NPs, such as Ag2S-NP and metallic AgNPs, into plants is important to understanding their entry into food chains. In this study, soybean Glycine max L. was hydroponically exposed to Ag2S-NPs via their roots (10-50 mg L-1) and stable-isotope-enriched 109AgNPs via their leaves [7.9 μg (g fresh weight)-1]. Less than 29% of Ag in treated leaves (in direct contact with 109AgNP) was accumulated from root uptake of Ag2S-NPs, whereas almost all of the Ag in soybean roots and untreated leaves sourced from Ag2S-NPs. Therefore, Ag2S-NPs are phytoavailable and translocate upward. During trophic transfer the Ag isotope signature was preserved, indicating that accumulated Ag in snails most likely originated from Ag2S-NPs. On average, 78% of the Ag in the untreated leaves was assimilated by snails, reinforcing the considerable trophic availability of Ag2S-NPs via root uptake. By highlighting the importance of root uptake of Ag2S-NPs in plant uptake and trophic transfer to herbivores, our study advances current understanding of the biogeochemical fate of Ag-containing NPs in the terrestrial environment.
Uptake of Silver-Containing Nanoparticles in an Estuarine Plant: Speciation and Bioaccumulation

Abstract:Understanding the bioaccumulation of silver-containing nanoparticles (Ag-NPs) with different species, concentrations, and sizes in estuarine plants is critical to their related environmental risk. Herein, the distribution of Ag-NPs in tidewater, sediments, and plants (Scirpus triqueter) of field-constructed mesocosm was investigated, where tidewater was exposed to Ag0-NPs and Ag+ at environmentally relevant concentrations. Particle number concentrations (PNCs) and sizes of Ag-NPs with various species were analyzed using a multistep selective dissolution method followed by the single-particle- inductively coupled plasma mass spectrometry technique. After 30 days of exposure, more than half of Ag0-NPs were dissolved to Ag+ and about 1/4 of Ag+ were transformed into Ag0-/AgCl-NPs in tidewater. Ag-NPs in stems exposed to Ag0-NPs were found to be dominated by metallic Ag, while Ag+ exposure led to more Ag2S-NPs in stems. In roots, 71% and 51% of Ag-NPs were found as Ag2S-NPs for Ag0-NPs and Ag+ treatment groups, respectively. Plant stems had a significantly higher enrichment of Ag-NPs than roots. Based on both random forests and structure equation models, it is suggested that salinity of tidewater can regulate Ag0-NPs in tidewater indirectly by influencing AgCl-NPs in tidewater and further affect the total PNCs of Ag-NPs in plant stems. Moreover, elevated sulfate-reducing bacteria (SRB) result in more Ag2S-NPs in rhizosphere sediments, thereby enhancing the bioaccumulation of Ag-NPs by roots.
Effects of Exposure Pathways on the Accumulation and Phytotoxicity of Silver Nanoparticles in Soybean and Rice

Abstract:Abstract The widespread use of silver nanoparticles (AgNPs) raises concerns both about their accumulation in crops and human exposure via crop consumption. Plants take up AgNPs through their leaves and roots, but foliar uptake has been largely ignored. To better understand AgNPs–plant interactions, we compared the uptake, phytotoxicity and size distribution of AgNPs in soybean and rice following root versus foliar exposure. At similar AgNP application levels, foliar exposure led to 17–200 times more Ag bioaccumulation than root exposure. Root but not foliar exposure significantly reduced plant biomass, while root exposure increased the malondialdehyde and H2O2 contents of leaves to a larger extent than did foliar exposure. Following either root or foliar exposure, Ag-containing NPs larger (36.0–48.9 nm) than the originally dosed NPs (17–18 nm) were detected within leaves. These particles were detected using a newly developed macerozyme R-10 tissue extraction method followed by single-particle inductively coupled plasma mass spectrometry. In response to foliar exposure, these NPs were stored in the cell wall and plamalemma of leaves. NPs were also detected in planta following Ag ion exposure, indicating their in vivo formation. Leaf-to-leaf and root-to-leaf translocation of NPs in planta was observed but the former did not alter the size distribution of the NPs. Our observations point to the possibility that fruits, seeds and other edible parts may become contaminated by translocation processes in plants exposed to AgNPs. These results are an important contribution to improve the risk assessment of NPs under environmental exposure scenarios.

Abstract:Despite the tremendous interest in silver nanoparticles (AgNPs) based on their wide range of applications, several key questions remain unanswered, especially regarding the trophic transfer and toxicological consequences of AgNPs along the food chain. A detailed understanding of the mechanisms that drive AgNP trophic transfer is currently limited due to the lack of information regarding the steady-state accumulation of AgNPs and their in vivo fate in the food chain. This results in variable patterns of AgNP trophic transfer among various food chains. However, biodynamic approaches in combination with the new tools emerging from methodological advances may provide unique perspectives on the accumulation of AgNPs, their in vivo fate and the molecular features driving toxicity in the food chain. The successful commercialization and use of AgNPs in sustainable development and the avoidance of unintended consequences for the environment and human health will also require a determination of the biogeochemical factors that influence AgNP trophic transfer. This review focuses on the current state of knowledge regarding (i) the trophic availability of AgNPs in the food chain, evaluated using conventional and biodynamic approaches, (ii) the processes that control AgNP trophic transfer, and (iii) the toxicological consequences of AgNP trophic transfer.
Migration of silver nanoparticles in an aquatic system with Eichhornia crassipes

Abstract:The migration and dissolution of AgNPs in an aquatic system with plants was investigated. By using a hydroponic system with Eichhornia crassipes, the absorption and transportation processes of silver nanoparticles were investigated. The results show that AgNPs concentrations in the water phase declined with the increase in time, and the reduction degree was dependent on the initial concentrations of AgNPs. The silver concentrations in the roots ( r=0. 98, p＜0. 05) , stems and leaves ( r=1, p＜0. 001) were significantly positively correlated with the initial concentrations of AgNPs. Silver nanoparticles accumulated in plant roots more than stems and leaves. Compared with the addition of AgNO3at identical concentrations, lower removal rates of silver and plant uptake were observed in the AgNPs stress systems. A significant positive correlation was also found between the initial AgNPs concentrations and the removed amount of silver ( r=0. 99, p ＜0. 001 ) . For AgNPs, the primary removal mechanisms in these aquatic systems were agglomeration and sedimentation, while the absorption by plants had a relatively weak contribution.

Abstract:Silver nanoparticles (NPs) are used in more consumer products than any other nanomaterial and their release into the environment is unavoidable. Of primary concern is the wastewater stream in which most silver NPs are transformed to silver sulfide NPs (Ag2S-NPs) before being applied to agricultural soils within biosolids. While Ag2S-NPs are assumed to be biologically inert, nothing is known of their effects on terrestrial plants. The phytotoxicity of Ag and its accumulation was examined in short-term (24 h) and longer-term (2-week) solution culture experiments with cowpea (Vigna unguiculata L. Walp.) and wheat (Triticum aestivum L.) exposed to Ag2S-NPs (0-20 mg Ag L-1), metallic Ag-NPs (0-1.6 mg Ag L-1), or ionic Ag (AgNO3; 0-0.086 mg Ag L-1). Although not inducing any effects during 24-h exposure, Ag2S-NPs reduced growth by up to 52% over a 2-week period. This toxicity did not result from their dissolution and release of toxic Ag+ in the rooting medium, with soluble Ag concentrations remaining below 0.001 mg Ag L-1. Rather, Ag accumulated as Ag2S in the root and shoot tissues when plants were exposed to Ag2S-NPs, consistent with their direct uptake. Importantly, this differed from the form of Ag present in tissues of plants exposed to AgNO3. For the first time, our findings have shown that Ag2S-NPs exert toxic effects through their direct accumulation in terrestrial plant tissues. These findings need to be considered to ensure high yield of food crops, and to avoid increasing Ag in the food chain.
Attenuation of photosynthesis in nanosilver-treated Arabidopsis thaliana is inherently linked to the particulate nature of silver

Abstract:Silver nanoparticles (AgNPs) are known to affect the physiology and morphology of plants in various ways, but the exact mechanism by which they interact with plant cells remains to be elucidated. An unresolved question of silver nanotoxicology is whether the interaction is triggered by the physical features of the particles, or by silver ions leached from their surface. In this study, we germinated and grew Arabidopsis thaliana seedlings in synthetic medium supplemented with sub-morbid concentrations (4 μg/mL) of AgNPs and silver nitrate (AgNO3). This treatment led to in planta accumulation of 106 μg/g and 97 μg/g of silver in the AgNO3- and AgNP-exposed seedlings, respectively. Despite the statistically indistinguishable silver accumulation, RNA sequencing data demonstrated distinct changes in the transcriptome of the AgNP-exposed, but not in the AgNO3-exposed plants. AgNP exposure induced changes in the expression of genes involved in immune response, cell wall organization, photosynthesis and cellular defense against reactive oxygen species. AgNO3 exposure, on the other hand, caused the differential expression of only two genes, neither of which belonged to any AgNP-enriched gene ontology categories. Moreover, AgNP exposure led to a 39% reduction (p < 0.001) in total chlorophyll concentration relative to untreated plants which was associated with a 56.9% and 56.2% drop (p < 0.05) in carbon assimilation rate at ambient and saturating light, respectively. Stomatal conductance was not significantly affected by AgNP exposure, and limitations to carbon assimilation, as determined through analysis of light and carbon dioxide (A/Ci) curves, were attributed to rates of electron transport, maximum carboxylation rates and triose phosphate use. AgNO3-exposure, on the other hand, did not lead to significant reduction either in chlorophyll concentration or in carbon assimilation rate. Given these data, we propose that the impact of AgNPs cannot be simply attributed to the presence of the metal in plants, but is innate to the particulate nature of nanosilver.
Trophic transfer of silver nanoparticles shifts metabolism in snails and reduce food safety

Abstract:Food security and sustainable development of agriculture has been a key challenge for decades. To support this, nanotechnology in the agricultural sectors increases productivity and food security, while leaving complex environmental negative impacts including pollution of the human food chains by nanoparticles. Here we model the effects of silver nanoparticles (Ag-NPs) in a food chain consisting of soil-grown lettuce Lactuca sativa and snail Achatina fulica . Soil-grown lettuce were exposed to sulfurized Ag-NPs via root or metallic Ag-NPs via leaves before fed to snails. We discover an important biomagnification of silver in snails sourced from plant root uptake, with trophic transfer factors of 2.0–5.9 in soft tissues. NPs shifted from original size (55–68 nm) toward much smaller size (17–26 nm) in snails. Trophic transfer of Ag-NPs reprograms the global metabolic profile by down-regulating or up-regulating metabolites for up to 0.25- or 4.20- fold, respectively, relative to the control. These metabolites control osmoregulation, phospholipid, energy, and amino acid metabolism in snails. Consumption of these Ag-NP contaminated snails causes non-carcinogenic effects on human health. Global public health risks decrease by 72% under foliar Ag-NP application in agriculture or through a reduction in the consumption of snails sourced from root application. The latter strategy is at the expense of domestic economic losses in food security of 58.3 million annually for countries such as Nigeria and Cameroon. Foliar Ag-NP application in nano-agriculture has lower hazard quotient risks on public health than root application to ensure global food safety, as brought forward by the United Nations Sustainable Development Goals.

Abstract:Due to their unique characteristics, nanoparticles are increasingly used in agricultural production through foliage spraying and soil application. The use of nanoparticles can improve the efficiency of agricultural chemicals and reduce the pollution caused by the use of agricultural chemicals. However, introducing nanoparticles into agricultural production may pose risks to the environment, food and even human health. Therefore, it is crucial to pay attention to the absorption migration, and transformation in crops, and to the interaction with higher plants and plant toxicity of nanoparticles in agriculture. Research shows that nanoparticles can be absorbed by plants and have an impact on plant physiological activities, but the absorption and transport mechanism of nanoparticles is still unclear. This paper summarizes the research progress of the absorption and transportation of nanoparticles in plants, especially the effect of size, surface charge and chemical composition of nanoparticle on the absorption and transportation in leaf and root through different ways. This paper also reviews the impact of nanoparticles on plant physiological activity. The content of the paper is helpful to guide the rational application of nanoparticles in agricultural production and ensure the sustainability of nanoparticles in agricultural production.

Abstract:Nanoparticles (NPs), especially biosynthesised in living plants by absorbing soluble salts and reducing metal ions, are extensively used in various fields. This work aimed at investigating the in vivo biosynthesis of silver NPs (Ag-NPs) in maize and the spatial distribution of the NPs and some important nutrient elements in the plant. The content of silver in plant was examined by inductively coupled plasma-atomic emission spectrometer showing that Ag can be absorbed by plant as soluble salts. The NPs in different parts of maize plant were detected and analysed by transmission electron microscopy, demonstrating the synthesis of NPs and their transport from the root to the shoots. Two-dimensional proton induced X-ray emission of silver, chlorine and several nutrient elements elucidated the possible relationship between synthesis of NPs and several nutrient elements in plant tissues. To their knowledge, this is the first report of possibility of synthesis of Ag-NPs in living plants maize (Zea mays L.). This study presents direct evidence for synthesis of NPs and distribution of related nutrient elements in maize, which has great significance for studying synthetic application of NPs in crop plants.

Abstract:The abundance of silver nanoparticles (AgNPs) in consumer products has led to their environmental release and therefore to concern about their impact on human health. Given the limited information on oral bioaccessibility of soil Ag, we used a physiologically based extraction test (PBET) to evaluate the bioaccessibility of AgNPs and AgNO3 from soil digestion. The AgNPs underwent several biochemical transformations, including their simultaneous dissolution and agglomeration in gastric fluid followed by the disintegration in the intestinal fluid of the agglomerates into NPs containing silver and chlorine. Therefore, Ag-containing soil exposed the intestine to nanoparticulate Ag in forms that were structurally different from the original forms. The bioaccessibility of AgNPs (0.5 +/- 0.05% 10.9 +/- 0.7%) was significantly lower than that of AgNO3 (4.7 +/- 0.6%-14.4 +/- 0.1%), as a result of the lower adsorption of nanoparticles to soil residues during the digestive process. For the soils tested, the bioaccessibility of AgNPs increased with decreasing clay contents and lower pH. By identifying the soil properties that control AgNP bioaccessibility, a more efficient and accurate screening can be performed of soil types that pose the greatest health risk associated with AgNP exposure. Trophic Transfer of Silver Nanoparticles Shifts Metabolism in Snails and Reduces Food Safety.

Abstract:Food security and sustainable development of agriculture has been a key challenge for decades. To support this, nanotechnology in the agricultural sectors increases productivity and food security, while leaving complex environmental negative impacts including pollution of the human food chains by nanoparticles. Here we model the effects of silver nanoparticles (Ag-NPs) in a food chain consisting of soil-grown lettuce Lactuca sativa and snail Achatina fulica. Soil-grown lettuce were exposed to sulfurized Ag-NPs via root or metallic Ag-NPs via leaves before fed to snails. We discover an important biomagnification of silver in snails sourced from plant root uptake, with trophic transfer factors of 2.0-5.9 in soft tissues. NPs shifts from original size (55-68 nm) toward much smaller size (17-26 nm) in snails. Trophic transfer of Ag-NPs reprograms the global metabolic profile by down -regulating or up-regulating metabolites for up to 0.25-or 4.20-fold, respectively, relative to the control. These metabolites control osmoregulation, phospholipid, energy, and amino acid metabolism in snails, reflecting molecular pathways of biomagnification and pontential adverse biological effects on lower trophic levels. Consumption of these Ag-NP contaminated snails causes non-carcinogenic effects on human health. Global public health risks decrease by 72% under foliar Ag-NP application in agriculture or through a reduction in the con-sumption of snails sourced from root application. The latter strategy is at the expense of domestic economic losses in food security of $177.3 and $58.3 million annually for countries such as Nigeria and Cameroon. Foliar Ag-NP application in nano-agriculture has lower hazard quotient risks on public health than root application to ensure global food safety, as brought forward by the United Nations Sustainable Development Goals.

The retention propensities of AgNPs in soils dictate their fate, transport, and transformation in the subsurface environments.
