Abstract:Two new eremophilanolides, tsoongianolide E and F, were isolated from Senecio tsoongianus. Their structures were elucidated on the basis of NMR and MS spectra. Both of these two compounds showed in vitro cytotoxicity to cultured KB and A-549 cancer cell lines.

Abstract:From the whole plant of Senecio tsoongianus, two pairs of epimers, tsoongianolides A (1) and B (2), and tsoongianolides C (3) and D (4), and two new sesquiterpene lactones, tsoongianolides E (5) and F (6), all of which possess the eremophilane skeleton, were isolated. Two known eremophilane dimers (7) and (8) were also obtained from this species. Their absolute structures were elucidated by 1D- and 2D-NMR techniques and by X-ray diffraction studies, along with chemical evidence.

Abstract:A new indole alkaloidal glucoside together with three known compounds aurantiamide acetate (2), eleutheroside E (3) and 1-O-caffeoyl-β-D-glucopyranoside (4) has been isolated from ethanol extract of the aerial parts of Clematis terniflora DC. On the basis of their spectroscopic and chemical evidence, the new compound was elucidated as (6-O-β-D-glucopyranosyl-1H-indol-3-yl) carboxylic acid methyl ester (1). Compounds 1 and 3 showed significant cytotoxicity against human ECA-109.

Abstract:A new sesquiterpene lactone (1) was obtained from the cytotoxic fraction of 95% ethanol extract of root barks of Tsoongiodendron odorum Chun together with two known sesquiterpene lactones, costunolide (2) and parthenolide (3). The structure of 1 was elucidated as 5 alpha, 6 alpha, 7 beta, 10 beta - 11 alpha, 13-dihydro-4(15)-eudesmene-12, 6-olide on the basis of chemical and spectral evidence including X-ray diffraction analysis. Costunolide showed cytotoxic activity against human leukemia (HL-60) cell line. Parthenolide showed promising cytotoxic activities in vitro against HCT-8, Bel-7402, SKOV3, KB, HELA and EJ cell lines. Also, the cytotoxic ethyl acetate fraction of ethanol extract of the root barks from which three chemical components were isolated showed promising cytotoxic activities in vitro against KB, BGC-823, Bel-7402, HCT-8, HL-60 cell lines.

Abstract:A new guaiane-type sesquiterpene glycoside, torilolone 8-O-beta-D-glucopyranoside (1), together with a known analogue compound, torilolone 11-O-beta-D-glucopyranoside (2), was isolated from the fruits of Daucus carota L. Their chemical structures were elucidated on the basis of MS, NMR spectroscopic analyses coupled with chemical degradation. The cytotoxic activities of two isolated compounds were evaluated against two human gastric cancer cell lines BGC-823 and AGS using MTT assay.

Abstract:The EtOH extract of the aerial parts of Xanthium sibiricum afforded six sesquiterpene lactones (STLs), including two new eremophilanolides. The structures of the new compounds were established on the basis of spectroscopic methods and the modified Mosher's method to be the C-11 epimers of 2S,4S,5R,7R,8R-2-hydroxy-eremophil-1(10)-en-12,8-olide [11R: 1 (sibiriolide C); 11S: 2 (sibiriolide D)]. The isolated compounds were evaluated for their in vitro cytotoxicities against five human cancer cell lines (Huh-7 hepatocarcinoma, KB nasopharynx carcinoma, Jurkat T cell lymphoblast, BGC-823 and KE-97 gastric carcinoma) using the CellTiter-Glo™ luminescent cell viability assay method. Compounds 4–6, each possessing an α-methylene-γ-lactone moiety, were found to have noteworthy cytotoxic effects with IC50 values ranging from 1.1 to 18.0μM.

Abstract:Three new eremophilane sesquiterpenes were isolated from the MeOH extract of the roots of Senecio nemorensis. Their structures were identified as 8 alpha-hydroxy-6 ss-(isobutanoyloxy)-1-oxoeremophila-7(11).9-dieno-12.8 ss-lactone (1), 6 ss,8 ss-dimethoxy-1-oxoeremophila-7(11),9-dieno-12,8 alpha-lactone (2) and 10 alpha-hydroxy-6 ss-(isobutanoyloxy)-1-oxocremophila-7(11),8-dieno-12,8-lactone (3), respectively, based on spectroscopic data, including IR. Abstract:Chemical investigation of the water extracts from the Senecio cannabifolius Less. led us to find two new compounds (1 and 2), along with 12 known compounds (3–14). The two new compounds were determined as (E, 4R)-4-hydroxy-4,5,5-trimethyl-3-(3-oxobut-1-enyl)cyclohex-2-enone (1) and (E)-4-((1S, 3R, 4R)-1-hydroxy-4,5,5-trimethyl-7-oxabicyclo[4.1.0]heptan-1-yl)but-1-en-3-o-ne (2), respectively. The structures of other compounds were elucidated by extensive analysis of spectral data and in comparison with the literature values. Compounds 1 and 2 were evaluated for inhibitory activity against lipopolysaccharide-induced NO production in RAW 264.7 macrophages, and compound 1 showed potent inhibitory activity with IC50 value of 30.65 μM.

Abstract:Two new eremophilane-type sesquiterpenoids eremophil-6-en-11-ol (1) and (7 alpha,9 alpha,10 alpha)-9,10-epoxy-eremophilan-11-ol (2), together with a known eremophilane-type (6 alpha,8 alpha)-6,8-dihydroxyeremophil-7(11)-en-12-oic acid 12,8-lactone (3) were isolated from the rhizomes of Ligularia veitchiana. The structures of 1 and 2 were established by spectral analysis including H-1- and C-13-NMR, HSQC, HMBC, and HR-ESI-MS data. The compounds 1 and 3 were assessed against lung-cancer (A549) and stomach-cancer (BCG823) cell lines by the MTT method. The results showed that I exhibited significant inhibiting activities on the growth of these cancer cells with IC50 values between 1 - 100 mu g/ml, whereas compound 3 had no effect on the same cell lines.

Abstract:Five undescribed sesquiterpene coumarins, one undescribed coumarin derivative, and twenty-five known analogues, were isolated from the resin of Ferula sinkiangensis K.M.Shen. The planar structures and relative configurations of the undescribed compounds were determined by NMR experiment and HRESIMS data. The absolute configurations were established by Electrostatic Circular Dichroism method. Among these analogues, Sinkiangenol E showed the best cytotoxic activity against HeLa cervical cancer cells. Annexin V-FITC/PI staining indicated that Sinkiangenol E induced apoptosis in HeLa cells. Cell cycle analysis showed Sinkiangenol E arrested cell cycle at G0/G1 phase. Western blot results proved that Sinkiangenol E affected apoptosis-related and cell cycle regulation-related protein expression by activating the MAPK pathway.

Abstract:Erythrosides A-G, seven undescribed oleanane-type triterpenoid saponins, were isolated from the seeds of Erythrophleum fordii. Their structures with absolute configurations were determined by extensive spectroscopic analysis including one-dimensional [1D] and two-dimensional [2D] nuclear magnetic resonance [NMR], high-resolution electrospray ionization mass spectroscopy [HR-ESI-MS] analysis, and chemical methods. Erythrosides A-G featured a diverse oligosaccharide chain containing 4–6 pentoses or hexoses at C-3 and a monoterpenic acid or a (E)-cinnamic acid unit at the C-21 position. Of particular interest, erythrosides A-C, E and F contained a rare alpha xylose in their sugar chains. The bioassay results indicated that erythrosides A-C showed moderate cytotoxic activities against human lung cancer cell line PC9 with IC50 values of 13.14, 16.67 and 17.59 μM, respectively (the positive control, Taxol, IC50 = 0.60 nM).

Abstract:Two new tigliane diterpenes, named eupneonoids A (1) and B (2), together with four known analogues (3-6) were isolated from the whole plant of Euphorbia neorubella Bruyns. Their structures were deduced based on the detailed spectroscopic analysis. All the isolates displayed cytotoxic effects against A549 human tumor cell lines with IC50 values ranging from 1.318 to 7.042 mu M.

Abstract:A phytochemical investigation guided by H-1 NMR and LC-MS data on the ethanol extract of Syringa pinnatifolia stems led to the isolation of 11 new dimeric eremophilane sesquiterpenoids, namely, syringenes A-K (1-11) and one known analog (12, syringene L). These structures were elucidated by extensive analysis of spectroscopic data, single-crystal X-ray diffraction, and computational methods. Biological assays revealed that 1-12 exhibited different degrees of anti-inflammatory effects, and 5 and 6 showed significant cytotoxicity against human hepatoma HepG2 cells with IC50 values of 12.3 and 12.9 mu M, respectively. Furthermore, flow cytometry assays and western blot analysis revealed that 5 and 6 promoted the apoptosis of HepG2 cells by activating ERK. Finally, the molecular docking analysis implied that the carbonyl and hydroxy groups at the C-11/C-6' of 5 and 6 had a good binding affinity with ERK.

Abstract:Three new ent-kaurane diterpenoids, silvaticusins A-C (1-3), along with a new ent-kaurane dimer silvaticusin D (4) were isolated from the aerial parts of Isodon silvaticus. The structures of these new compounds were established mainly by comprehensive analysis of their NMR and MS data. The absolute configuration of compounds 1 and 4 were determined using a single-crystal X-ray diffraction and computational methods, respectively. Compounds 2 and 3 were found to exhibit remarkable cytotoxic effects against five human tumor cell lines (HL-60, A-549, SMMC-7721, MDA-MB-231, and SW-480), with IC50 values spanning from 1.27 ± 0.08 to 7.52 ± 0.33 μM.

Abstract:Two new monoterpenoid indole alkaloids, gelselegandines F (1) and G (2), were isolated from the aerial parts of Gelsemium elegans. Their structures were elucidated by means of spectroscopic techniques and quantum chemical calculations. The ECD calculations were conducted at the B3LYP/6-311G(d,p) level and NMR calculations were carried out using the Gauge-Including Atomic Orbitals (GIAO) method. Structurally, the two new compounds possessed rare, cage-like, monoterpenoid indole skeletons. All isolated compounds and the total alkaloids extract were tested for cytotoxicity against four different tumor cell lines. The total alkaloids extract of G. elegans exhibited significant antitumor activity with IC50 values ranging from 32.63 to 82.24 ug/mL. In order to discover anticancer leads from the active extraction, both new indole compounds (1–2) were then screened for cytotoxicity. Interestingly, compound 2 showed moderate cytotoxicity against K562 leukemia cells with an IC50 value of 57.02 uM.
