Abstract:Three new eremophilane sesquiterpenes were isolated from the MeOH extract of the roots of Senecio nemorensis. Their structures were identified as 8 alpha-hydroxy-6 ss-(isobutanoyloxy)-1-oxoeremophila-7(11).9-dieno-12.8 ss-lactone (1), 6 ss,8 ss-dimethoxy-1-oxoeremophila-7(11),9-dieno-12,8 alpha-lactone (2) and 10 alpha-hydroxy-6 ss-(isobutanoyloxy)-1-oxocremophila-7(11),8-dieno-12,8-lactone (3), respectively, based on spectroscopic data, including IR. EI-MS, HR-ESI-MS, and 1D- and 2D-NMR data.

Abstract:From the whole plant of Senecio tsoongianus, two pairs of epimers, tsoongianolides A (1) and B (2), and tsoongianolides C (3) and D (4), and two new sesquiterpene lactones, tsoongianolides E (5) and F (6), all of which possess the eremophilane skeleton, were isolated. Two known eremophilane dimers (7) and (8) were also obtained from this species. Their absolute structures were elucidated by 1D- and 2D-NMR techniques and by X-ray diffraction studies, along with chemical evidence.

Abstract:A previously undescribed sesquiterpene lactone, named eupalinolide N (1), was isolated from traditional Chinese medicine "Ye-Ma-Zhui" (Eupatorium lindleyanum) by ethanol reflux extraction, reduced pressure concentration, macroporous resin column chromatography, and C-18 reversed-phase chromatography. Its structure was elucidated by a comprehensive interpretation of spectroscopy evidence as well as ECD calculations. 1 showed anti-inflammatory activity by inhibiting the gene expressions of pro-inflammatory factors including IL-1 beta, TNFa COX-2, and iNOS at the concentration of 7.5 mu M, as well as attenuating the excretion of NO, IL-6, and TNF-alpha in Raw 264.7 macrophages at the concentration of 15 mu M.

Abstract:Two new alkaloids (1,2) and one new enoic acid (3), together with three known piericidins (4-6), were isolated from the liquid fermentation of the salt lake derived Streptomyces sp. QHA10. The structures of 1-3 were elucidated based on extensive spectroscopic data (NMR, HRESIMS) as well as single-crystal X-ray diffraction. Compound 3 showed potential anti-inflammatory activity by inhibiting the production of nitric oxide (NO) in lipopolysaccharide (LPS)-induced RAW 264.7 mouse macrophages with the IC50 value of 24.5 mu M. Abstract:Three unusual sesterterpenoids featuring unprecedented rearranged colquhounane (C25) and tetranorcolquhounane (C21) frameworks, colquhounoids E (1) and F (3) and norcolquhounoid F (2), were isolated from a Lamiaceae medicinal plant Colquhounia coccinea var. mollis. Their structures were elucidated by spectroscopic analysis and quantum chemical calculations. A biomimetic inspired regioselective cyclopropane cleavage was achieved under acidic conditions. The immunosuppressive activities of these new sesterterpenoids were also evaluated.

Abstract:Four new compounds, including three eudesmane sesquiterpenoids (1–3) and a dimeric sesquiterpenoid lactone (4), were isolated from the aerial part of Salvia plebeia R. Br., together with two known eudesmanolides (5–6). Their structures were elucidated by extensive spectroscopic analysis including 1D and 2D NMR, HR-ESI–MS spectra. Antiproliferative activities of isolates against six human myeloid leukemia cell lines were evaluated, salplebeone D (1) and G (4) showed moderate inhibitory activity. In addition, compound 4 was a rare eudesmane sesquiterpenoid dimer obtained from this specie for the first time.

Abstract:The plant genus Eremophila is endemic to Australia and widespread in arid regions. Root bark extract of Eremophila longifolia (R.Br.) F.Muell. (Scrophulariaceae) was investigated by LC-PDA-HRMS, and dereplication suggested the presence of a series of diterpenoids. Using a combination of preparative- and analytical-scale HPLC separation as well as extensive 1D and 2D NMR analysis, the structures of 12 hitherto unreported serrulatane diterpenoids, eremolongine A-L, were established. These structures included serrulatanes with unusual side chain modifications to form hitherto unseen skeletons with, e.g., cyclopentane, oxepane, and bicyclic hexahydro-1H-cyclopenta[c]furan moieties. Serrulatane diterpenoids in Eremophila have recently been shown to originate from a common biosynthetic precursor with conserved stereochemical configuration, and this was used for tentative assignment of the relative and absolute configuration of the isolated compounds. Triple high-resolution α-glucosidase/α-amylase/PTP1B inhibition profiling demonstrated that several of the eremolongines had weak inhibitory activity towards targets important for management of type 2 diabetes.

Abstract:Salvia przewalskii Maxim is a perennial plant from the genus Salvia (family Lamiaceae). The roots of S. przewalskii were long used as a traditional herb to treat blood circulation related illnesses in China. As part of our continuing interest in polycyclic natural products from medicinal plants, two unprecedented adducts comprised of a dinor-diterpenoid and a 9′-nor-rosmarinic acid derivative, linked by a 1,4-benzodioxane motif (1 and 2), were isolated from the roots of S. przewalskii. Their structures were established by extensive spectroscopic approaches including 1D, 2D NMR, and HRFABMS. Their cytotoxic activities against five human tumor cell lines were evaluated.

Abstract:Salinopyridins A and B, two novel polyethers containing a rare pyridine moiety, were isolated from an earwig symbiotic Streptomyces sp. NA4227. Their structures were determined by extensive spectroscopic analysis. Based on the structure, bioinformatics analysis and gene mutation experiments, we proposed that salinopyridins share the same biosynthetic pathway with salinomycin, and the formation of pyridine moiety is assembled through a nonenzymatic process. Salinopyridins A and B exhibited potent activity against several Gram-positive bacteria.

Abstract:Two new stilbenoids, stemobenoids A (1) and B (2), together with three known compounds were obtained from the roots of Stemona tuberosa. The structures of the new compounds were established by extensive spectroscopic analysis, including HRMS, 1D and 2D NMR data. Compounds 1 and 2 displayed potent quinone reductase inducing activity in Hepa 1c1c7 cells.

Abstract:(±)-Salvicatone A (1), a C27-meroterpenoid featuring a unique 6/6/6/6/6-pentacyclic carbon skeleton with a 7,8,8a,9,10,10a-hexahydropyren-1 (6H)-one motif, was isolated from the roots and rhizomes of Salvia castanea Diels f. tomentosa Stib. Its structure was characterized by comprehensive spectroscopic analyses along with computer-assisted structure elucidation, including ACD/structure elucidator and quantum chemical calculations with 1H/13C NMR and electronic circular dichroism. Biogenetically, compound 1 was constructed from decarboxylation following [4 + 2] Diels-Alder cycloaddition reaction between caffeic acid and miltirone analogue. Bioassays showed that (-)-1 and (+)-1 inhibited nitric oxide production in lipopolysaccharide-induced RAW264.7 macrophage cells with an IC50 value of 6.48 ± 1.25 and 15.76 ± 5.55 μM, respectively. The structure-based virtual screening based on the pharmacophores in ePharmaLib, as well as the molecular docking and molecular dynamics simulations study, implied that (-)-1 and (+)-1 may potentially bind to retinoic acid receptor-related orphan receptor C to exert anti-inflammatory activities.

Abstract:A 6-eremophilene derivative 2 with an unusual pendant was produced as phytoalexin in the fresh leaves of Chloranthus anhuiensis K. F. Wu in response to abiotic stress elicitation by CuCl2. The phytoalexin was characterized as a sesquiterpene with a (2,6-dimethoxy-4-hydroxy)benzyl moiety pendant. Two new sesquiterpenes, (3R,4S,5R,10S,11S)-3-hydroxy-8-oxo-6-eremophilen-12-oic acid (1) and (3R,4S,5R,6R,8R,10S)-3,6,8-trihydroxy-7(11)-eremophilen-12,8-olide (3), one new sesquiterpene glycoside, anhuienoside A (4), one new caffeoyl phenylethanoid diglycoside with an unusual cyclic structure, anhuienoside B (5), and one new tyramine derivative, N-acetyltyramine 1-O-β-d-glucoside (6), and three known compounds were also isolated. Their structures and relative configurations were established by spectroscopic means. The absolute configurations of 1−3 were defined mainly by comparison of quantum chemical TDDFT calculated and experimental ECD spectra.
