Abstract:An electrochemical protocol for benzylic C(sp3)-H aminopyridylation via direct C-H/N-H cross-coupling of alkylarenes with N-aminopyridinium triflate has been developed. This method features excellent site-selectivity, broad substrate scope, redox reagent-free and facile scalability. The generated benzylaminopyridiniums can be readily converted to benzylamines via electroreductive N-N bond cleavage.

Abstract:The first aryl iodide catalyzed intramolecular C-H amination of phenylurea has been disclosed for high-efficiency synthesis of benzimidazolone derivatives in excellent yields (up to 97%) by an operationally simple one-step organocatalytic oxidative process. Fluorinated protic alcohols can efficiently accelerate the conversion of this transformation. The straightforward method has good functional group tolerance and can be performed with an inexpensive and readily accessible catalyst with high proficiency.

Abstract:Constructing a C-N bond by merging electrochemistry and nickel catalysis is considered a powerful strategy. Herein, we investigate highly efficient intramolecular amination at room temperature with excellent functional group tolerance. Mechanistic studies suggest that the rapid ligand exchange may lead to the NiI/NiIII catalytic cycle. This method not only provides a new perspective for intramolecular amination but also offers a novel approach for constructing the benzothiazine scaffold.

Abstract:C-H/N-H cross-coupling is an ideal strategy to synthesize various amines but remains challenging owing to the requirement for sacrificial chemical oxidants and the difficulty in controlling the regio- and chemo-selectivity. Herein we report a site-selective electrochemical amination reaction that can convert benzylic C-H bonds into C-N linkages via H-2 evolution without need for external oxidants or metal catalysts. The synthetic strategy involves anodic cleavage of benzylic C-H to form a carbocation intermediate, which is then trapped with an amine nucleophile leading to C-N bond formation. Key to the success is to include HFIP as a co-solvent to modulate the oxidation potentials of the alkylbenzene substrate and the aminated product to avoid overoxidation of the latter.

Abstract:A straightforward strategy for direct benzylic C-H bond amination via an electrochemical Ritter-type reaction is developed. The reaction demonstrates simpler and milder reaction conditions over the existing methods without extra mediator. Moderate to excellent yields up to 94% of the desired amide products were obtained with a broad substrate scope. The removal of the Ac group by a simple step can afford NH-free benzylic amines, providing a suitable approach for the late-stage functionalization of bioactive molecules.

Abstract:Transition-metal-catalyzed oxidative C- H/N-H coupling reaction is an important method to construct C-N bonds. However, stoichiometric oxidants were usually necessary. Here, we report a palladium-catalyzed electro-oxidative intra-molecular C-H/N-H annulation reaction without an external oxidant or additive. A variety of pyrido[1,2-a]benzimidazoles were constructed by a simple, green, and mild path. Experimental results suggested that Pd(0) could be oxidized on the anode to be recycled.

Abstract:An efficient electrochemical dual C(sp(3))-H amination was developed under metal-free and chemical oxidant-free conditions. A series of imidazo[1,5-a]quinazolin-5(4H)-ones and 5-oxo-4,5-dihydroimidazo[1,5-a]quinazoline-3-carbonitriles can be obtained in high yields and the product distribution can be modulated by virtue of this method. The reaction mechanism was investigated and the corresponding intermediates were studied. The reaction features a broad substrate scope, regulation of the product distribution, mild conditions and scalable preparation.

Abstract:A novel and efficient synthesis of pyrido[1,2-Mbenzimidazoles through direct intramolecular aromatic C-H amination of N-aryl-2-aminopyridines has been developed. The reaction, cocatalyzed by Cu(OAc)(2) and Fe(NO3)(3)center dot 9H(2)O, is carried out in DMF under a dioxygen atmosphere. Diversified pyrido[1,2-a]benzimidazoles containing various substitution patterns are obtained in moderate to excellent yields by using this procedure. The results of mechanistic studies suggest that a Cu(III)-catalyzed electrophilic aromatic substitution (SEAr) pathway is operating in this process. The unique role of iron(III) is believed to lie in its ability to facilitate formation of the more electrophilic Cu(III) species. In the absence of iron(III), a much less efficient and reversible Cu(II)-mediated SEAr process takes place.

Abstract:The direct α-C(sp3)-H functionalization of widely available tertiary amines holds promise for the rapid construction of complex amine architectures. The activation of C(sp3)-H bonds through electron transfer and proton transfer by oxidants, photoredox catalysis and electrochemical oxidation have received wide attention recently. In these reactions, the direct capture and identification of the key reactive radical intermediates are technically difficult due to their short life-time. Herein, an online electrochemical mass spectrometry (MS) methodology was utilized to probe the short-lived intermediates in the electrochemical oxidative α-C(sp3)-H functionalization of tertiary amines. The resulting electrochemical oxidation intermediates, α-amino radical cation and iminium cation were successfully detected. Further, the α-amino C(sp3) radical added to the double bond of a phenyl trans-styryl sulfone, yielding another C(sp3) radical that leads to the final vinylation. Based on the mass spectrometric elucidation of the reactivity of the α-amino radical, a scale-up electrochemical radical vinylation methodology was established, with which a large variety of allylic amines with broad functional group tolerance were synthesized.

Abstract:The iodine(III) compound promoted CH amination and tandem CH amination/acetoxylation of guanidines are achieved for the first time to provide efficiently 2-aminobenzimidazoles and acetoxyl-substituted 2-aminobenzimidazoles, respectively. The amount and type of iodine(III) compounds control the selective syntheses of two types of 2-aminobenzimidazoles. This reaction shows good regioselectivity when unsymmetrical substrates are used.

Abstract:The intramolecular Csp(3)-H and/or C-C bond amination is very important in modern organic synthesis due to its efficiency in the construction of diversified N-heterocycles. Herein, we report a novel intramolecular cyclization of alkyl azides for the synthesis of cyclic imines and tertiary amines through selective Csp(3)-H and/or C-C bond cleavage. Two C-N single bonds or a C=N double bond are efficiently constructed in these transformations. The carbocation mechanism differs from the reported metal nitrene intermediates and therefore enables metal-free and new transformation.
