Abstract:Controlled drug release has sparked much attention in the field of drug delivery by prolonging the duration of drug effectiveness and reducing drug toxicity. However, the development of controlled-release drug delivery systems remains a significant challenge, primarily due to inadequate drug compatibility, low biocompatibility, and low response sensitivity of the drug carriers. Herein, an innovative strategy for fabricating multifunctional organohydrogels as drug carrier systems is designed. 1-Tetradecanol/water emulsions, stabilized by cellulose acetoacetate (CAA), are initially prepared. The organohydrogels are fabricated by the ternary enzyme-mediated polymerization of the monomer - containing emulsion. The amphipathy of CAA ensures the stability of emulsions during polymerization, and CAA participates in polymerization as an initiator. Through the reversible crystallization-melting transition of 1-tetradecanol, the drug can be encapsulated in/released from the organohydrogel. Moreover, 1-tetradecanol enhances the bioavailability of water-insoluble drugs without any additional cosolvent. Therefore, biofriendly organohydrogels can serve as ideal dressings with adjustable mechanical properties, effective temperature regulation, and temperature-triggered drug release ability.

Abstract:Natural compounds cholic acid and β-cyclodextrin are attached separately as pendant groups in a copolymer with N,N′-dimethylacrylamides. The formation of supramolecular hydrogels is induced by inclusion complex formation between cholic acid and β-cyclodextrin moieties as evidenced by rheological analysis and 1H NMR spectroscopy. Storage modulus of the hydrogel shows a maximum value when the molar ratio of cholic acid to β-cyclodextrin units is adjusted to 1:1. The concentration of the hydrogel can be as low as 5.5 wt %. Both shear-thickening and shear-thinning have been observed when the shear rate gradually increases from 0.01 to 100 s–1. The inclusion complexation renders the gel–sol process reversible under heating and cooling cycles. The self-healing of such hydrogels is observed and confirmed by step-strain rheological measurements. The dynamically reversible host–guest complexation provides reasonably good mechanical properties of the cross-linked polymer network. The natural origin of the constituents may make the hydrogels suitable candidates for biomedical applications pending further tests.

Abstract:Self-healing hydrogels can repair their cracks, and restore their original properties. However, self-healing hydrogels usually face low mechanical strength and poor stability. By the dual crosslinking strategy, a self-healing hyaluronic acid-based hydrogel with enhanced strength was fabricated by dynamic acylhydrazone linkages between aldehyde-modified maleic sodium hyaluronate and 3,3'-dithiobis (propionylhydrazide) and subsequent photopolymerization among maleic groups in the hydrogel network. The hydrogels exhibit fast gelation and excellent self-healing capability due to the dynamic and reversible characteristics of acylhydrazone and disulfide linkages. Furthermore, the dual crosslinking increase the mechanical strength of the hydrogels and prolong their stabilization time. Swelling behaviors, morphology, and mechanical properties could be adjusted by altering the molar ratio of -NH-NH2/-CHO. Besides, the hydrogels displayed interesting pH-responsiveness and cytocompatibility. The hydrogels have potential applications in cell culture, drug delivery, and 3D bioprinting.

Abstract:Self-healing hydrogels with pH-responsiveness could protect loaded drugs from being destroyed till it arrives to the target. The pectin-based hydrogel is a candidate due to the health benefit, anti-inflammation, antineoplastic activity, nontoxicity, and biospecific degradation, et al. However, the abundant existence of water-soluble branched heteropolysaccharide chains influenced its performance resulting in limitation of the potential. In the present study, we prepared a series of self-healing pectin/chitosan hydrogels via the Diels-Alder reaction. Moreover, pectin/chitosan composite hydrogel was prepared as a contrast. By comparison, it can be seen that the Diels-Alder reaction greatly improved the cross-linking density of hydrogels. The self-healing experiments showed excellent self-healing performance. In different swelling mediums, significant transformation in the swelling ratio was shown, indicating well-swelling property, pH- and thermo-responsiveness. The drug loading and release studies presented high loading efficiency and sustained release performance. The cytotoxicity assay that showed a high cell proliferation ratio manifested great cytocompatibility.

Abstract:Therapeutic drugs for human cardiovascular disease require specific carriers. Hydrogels have a porous structure, plasticity, and biocompatibility, and are ideal carriers for cardiovascular drugs. Moreover, for their intended functions, hydrogels also need to have high mechanical properties, self-healing properties, good pH-responsive performance, and other properties. Here, based on the interactions between multiple dynamic covalent bonds, we effectively synthesized hybrid hydrogels of carboxymethyl chitosan (C-CS), aldehyde-modified hyaluronic acid (OHA), polyvinyl alcohol (PVA), and boric acid (BA). The multiple covalent bonds reinforce each other, endowing the hydrogels with high mechanical properties, self-healing properties and pH responsiveness. In addition, the whole preparation process does not require crosslinking agents and the operation process is simple. The results show that the 5% PVA+C-CS+OHA+BA hydrogel has high mechanical properties (tensile stress 40.07 KPa; compressive stress 553.38 KPa), good self-healing performance (up to 92%) and excellent pH responsive performance. In addition, the hydrogel has high drug-loading capacity (117.54 mg/g) and sustained-release performance. In conclusion, this hybrid hydrogel can potentially be used as a drug carrier for clinical treatment.

Abstract:Abstract A new double-layer, pH-sensitive, composite hydrogel sustained-release system based on polysaccharides and synthetic polymers with combined functions of different inner/outer hydrogels was prepared. The polysaccharides inner core based on sodium alginate (SA) and carboxymethyl cellulose (CMC), was formed by physical crosslinking with pH-sensitive property. The synthetic polymer out-layer with enhanced stability was introduced by chemical crosslinking to eliminate the expansion of inner core and the diffusion of inner content. The physicochemical structure of the double-layer hydrogels was characterized. The drug-release results demonstrated that the sustained-release effect of the hydrogels for different model drugs could be regulated by changing the composition or thickness of the hydrogel layer. The significant sustained-release effect for BSA and indomethacin indicated that the bilayer hydrogel can be developed into a novel sustained delivery system for bioactive substance or drugs with potential applications in drugs and functional foods.

Abstract:Self-healing hydrogels which have magical self-healing properties have attracted much attention in recent years. Here, we prepared a self-healing hydrogel with biocompatibility and NaHCO3 degradability. The hydrogel is constructed from cross-linking of poly(N,N-dimethyl acrylamide-stat-4-formylphenyl acrylate) P(DMA-stat-FPA) by pectin achlydrazide (pectin-AH). This hydrogel has base labile phenolic bonds connection and acylhydrazone bonds, which made the hydrogel degradable in extremely mild base of NaHCO3 solution and showed self-healing property. At the same time, the pectin endowed the hydrogels with good biocompatibility and biodegradabilty. In addition, hydrogel has micro-porous structure and showed controlled drug release behavior. These excellent properties made this hydrogel very useful in biomedical fields such as tissue engineering, drug carriers and biosensors.

Abstract:A novel crosslinked hydrogel was prepared from sodium carboxymethyl cellulose (CMC) and hydroxyethyl cellulose (HEC) using ammonium persulfate as an initiator and methylenebisacrylamide as a crosslinker for drug delivery. The chemical structure of the copolymer was characterized by Fourier transform infrared spectroscopy and X-ray diffraction, and the morphology was observed under scanning electron microscopy. The swelling behavior of the hydrogels confirmed the pH-and ionic strength-sensitivity. The reversibility of the hydrogels and the on-off switching behavior were also investigated, providing the potential for drug delivery. The release of bovine serum albumin (BSA) from drug-loaded hydrogels was studied at different pH conditions to simulate gastrointestinal conditions. The amount of BSA released from the hydrogels at pH 1.2 was relatively low (17.8%), while 85.2% was released at pH 7.4. According to the results, the CMC/HEC hydrogel has the potential for use in the controlled release of oral medication.

Abstract:An interpenetrating double network hydrogel based on cellulose backbone and poly-dopamine dendritic structure was successfully prepared with the advantages of controlled hydrophobic drug loading, pH-responsive ability, and long-term bacteriostatic property. 13C-NMR was used to process the structural characterization and explain the mechanism of hydrogel system’s construction. Hydrophobic drug of ciprofloxacin with carbonyl group can evenly carry in the hydrogel system, and the highest loaded ratio of ciprofloxacin was reach to 80.56%. Besides, the swelling property and the controlled drug loading ability of our prepared hydrogel were carefully studied, which showed a close relation with the hydrogel design. Moreover, the in vitro drug release experiments demonstrated that the hydrogel had an obvious pH-responsive release property of ciprofloxacin, and the result of bacteriostatic experiment also showed a long-term bacteriostatic effect on Escherichia coli, which showed potential application on biological and medical fields, including wound treatment.Graphical abstract.

Abstract:This study introduces an innovative approach in the field of biomaterials through the development of a multifunctional, printable scaffold designed for the advanced delivery of polyphenols. Addressing challenges of burst-release, poor compounds solubility and bioavailability, we engineered a supramolecular-assembled dual crosslinked hydrogel combining fucoidan (F) and chitosan (CS) encapsulating nanoliposomes. The novel integration of dibenzaldehyde-terminated PEG (DB) within the hydrogel, created a synergistic crosslinking through the formation of the covalent Schiff base bonds and physical interactions which enhanced the hydrogel’s overall characteristics. Tailoring F content (DBCS-F), allowed the crosslinking density modulation, yielding a robust gel network structure with enhanced mechanical properties, evidenced by a remarkable compressive stress resistance of 1.73 MPa at 60% strain, coupled with high compressive resilience (79.31% stress recovery after 6 cycles), surpassing single crosslinked hydrogel (DBCS). The rheological analysis further confirmed the hydrogel’s improved viscoelasticity, stable network structure, and shear-thinning behavior, affirming its excellent injectability and suitability for 3D printing applications. Most notably, the incorporation of catechin/juglone-loaded nanoliposomes contributed to a pH-responsive sustained release of polyphenols in simulated gastrointestinal conditions. The developed dual crosslinked hydrogel, integrating injectability, self-healing, and pH-responsiveness presents a significant advancement in biomaterials and holds a promising approach for the fabrication of personalized 3D printed scaffolds aimed for sustained and targeted polyphenols release.

Abstract:Most structural self-healing materials were developed based on either reversible supramolecular interaction or dynamic covalent bonding. It seems a good idea to incorporate self-healing properties into high-performance materials. In this study, we fabricated the alginate-based cyclodextrin and polyacrylamide azobenzene highly stretchable and tough interpenetrating composite hydrogel with self-repairing behavior under light irradiation. Initially, the alginate-based cyclodextrin and polyacrylamide azobenzene were designed and synthesized. The corresponding structural, thermal, and morphological properties of hydrogels were characterized. The reversible transformation of the sol-gel can be achieved by the irradiation upon ultraviolet light and visible light. The self-healing behavior of this composited gel is based on the host-guest interaction between cyclodextrin and azobenzene. The recovery gel elongation at 48 h healing in the dark condition was is 0.04 MPa, with an elongation of 1140 %. Therefore, this gel can achieve self-healing ability while maintaining highly stretchable and tough performance.

Abstract:Stimuli-responsive polymeric hydrogels based on host-guest interactions have demonstrated potential as smart drug delivery platforms. However, host-guest interactions alone have been repeatedly highlighted to provide insufficient mechanical strength for the formed hydrogels likely due to the generally low grafting degrees of functional groups and steric hindrance of polymer backbone and grafted functions. Herein, a combination strategy of reversible addition-fragmentation chain-transfer polymerization (RAFT) and dual crosslinking was employed to fabricate thermo-and pH-dual responsive hydrogels with enhanced mechanical properties. Spe-cifically, two kinds of poly (N-isopropyl acrylamide) (PNIPAM)-based well-defined thermo-sensitive polymers, denoted as PH and PG, consisting of beta-cyclodextrin (beta-CD) and adamantane (Ad) groups were synthesized via RAFT copolymerization. A facile mixing of PH and PG with commercially available adipic dihydrazide led to the construction of hydrogels via host-guest interactions and dynamic acylhydrazone bonding. The resulting hydrogels not only exhibited pH-switchable sol-gel transitions and temperature-dependent swelling behaviors, but also possessed good stretchability (fracture stress up to 39.97 kPa) and self-healing property (self-healing efficiency above 90%). Notably, the resulting hydrogels showed pH-responsive controlled spatiotemporal release administration of rhodamine B due to the presence of dynamic acylhydrazone links. Together with good cyto-compatibility, the hydrogels developed herein represent promising candidates for potential transdermal drug delivery.

Abstract:In this work, a versatile and simple strategy to build an injectable interpenetrating polymer network (IPN) hydrogel with tunable mechanical properties and self-healing ability is reported. The gelation time and mechanical properties can be tuned by pH or the solid content. Due to the exchange of acylhydrazone and boronate ester bonds formed between functionalized prepolymers, the hydrogels can self-heal at either acidic or basic condition. Furthermore, the pH-induced IPN to semi-IPN reversible transition endows the hydrogels with tunable pore structure and mechanical properties. It is hoped that this novel strategy will provide new opportunities in design and preparation of 3D-printable IPN hydrogels, fostering their use in a wide range of applications such as controlled drug release and tissue engineering.

Abstract:Tumor local chemotherapy employing injectable hydrogel reservoirs is a promising platform to achieve precise drug administration. However, balanced injectability, pH-responsiveness and long-term hydrolysis resistance of self-healing hydrogels remain appealing challenges. Herein, a modular preassembly strategy combining host-guest interactions with dynamic acylhydrazone bonds, was exploited to fabricate injectable cellulose-based hydrogels (CAAs) dressed with self-healing properties, pH-responsiveness and hydrolytic degradation resistance. Attributed to the host-guest interaction between β-cyclodextrin (CD) and 1-adamantane (AD), the hydrogels exhibited injectability, self-healing properties (healing efficiency of 97.5%) and rapid recovery (< 10 min) without external stimuli in physiological environment. Moreover, the hydrogels equipped with dynamic acylhydrazone linkages underwent slow hydrolytic degradation (> 30 days) and pH-responsive behavior, endowing the hydrogels with precise spatiotemporal drug release administration. The in vivo application of CAA as a carrier was studied using doxorubicin (DOX) model drug, and the results shows that using CAA as DOX carrier not only greatly enhances the anti-tumor efficacy of DOX, but also reduced the side effects of DOX. STATEMENT OF SIGNIFICANCE: With the preassemble approach combining host-guest interactions with dynamic acylhydrazone bonds, this work demonstrated a multi-functional self-healing hydrogel as drug carrier developed by using natural polysaccharides, which offers a new avenue for the high-value utilization of biomass. The strategy demonstrated in the present work may also supply a pathway for the preparation and regulation of hydrogels as intelligent biomedicine materials.

Abstract:Cellulose hydrogels have gained attention in the field of wound healing due to their biodegradability, biocompatibility, and the capacity to sustain a humid environment that promotes healing. Conventional cellulose hydrogels were usually lacked responsiveness to changing wound conditions, and limited capacity for controlled release of active substances. The composite hydrogels with Berberine (BBR) loading were prepared from bamboo parenchymal cellulose and in situ crosslinking carboxylated-β-cyclodextrin (BPCH-B) via dissolution. The inclusion of BBR enhanced the antibacterial properties of cellulose hydrogel while maintaining biocompatibility and drug delivery capabilities. The dual-responsive dressing was demonstrated to modulate drug release kinetics in accordance with the pH and temperature conditions prevailing within the wound site. Specifically, study exhibited a significant increase in drug release (over 70 %) under alkaline pH (7.6) and temperature (40 °C) conditions. Full-thickness wound healing experiments indicated that BPCH-B had better healing ability, and the wound healing area of BPCH-B treated was 80 % within 12 days, while the control group was only 50 %. This strategy for generating functional wound healing can be further control release of drug compounds for treatment of wounds, enabling development of practical wound care materials.

Abstract:In this study, we reported a pH/near-infrared (NIR)-responsive hydrogel for on-demand drug delivery and wound healing. Cellulose nanofibrils (TOCNFs) were produced by 2, 2, 6, 6-tetramethylpiperidine-1-oxyl (TEMPO)-mediated oxidation method. Polydopamine (PDA) was introduced into TOCNFs network to fabricate a PDA/TOCNFs hydrogel through ion-crosslinking with calcium ion as a crosslinker. Then, tetracycline hydrochloride (TH) which loaded on PDA could be released from the prepared hydrogels in an on-demand fashion under NIR exposure or at lower pH conditions. In addition, the in vivo skin defect experiments illustrated that the resultant composite hydrogel had a synergistic effect on promoting wound healing. Therefore, owing to the unique pH/NIR responsiveness, long time period drug releasing property, antibacterial and mechanical properties, together with their biodegradability and biocompatibility, the prepared PDA/TOCNFs hydrogel is highly promising as a new drug delivery vehicle, offering simple and safe alternatives to the conventional systems based on the petroleum-based polymers.

Abstract:Smart hydrogels have shown great potential applications in disease treatment due to their controlled and local drug-release ability. Herein, a smart hydrogel with pH-responsive, injectable, and self-healing properties for controlled release of taxifolin (TFL) was prepared by freezing-thawing and photo-crosslinking methods. The crosslinking network of hydrogels (CS-CA hydrogels) was constructed by the hydrogen bonds , Schiff base bonds, and cyclobutane rings using chitosan (CS) and coumarin (CA) as raw materials. The CS-CA hydrogel demonstrated a compressive strength of 1.04 MPa, a self-healing efficiency of 99.9 %, and could maintain structural and functional integrity after injection. In addition, the drug release rate and shape of the CS-CA hydrogels were tunable due to its pH sensitivity. The TFL cumulative release reached 60 % within 12 h at pH = 4, and after equilibration, the cumulative release of TFL at pH = 4 (80 %) was significantly higher than at pH = 9.2 (50 %). The CCK8 experiment showed that the resulting hydrogel had no cytotoxicity. Meanwhile, subcutaneous implantation experiments in mice showed that the CS-CA hydrogels had favorable biodegradability and compatibility.

Abstract:In this study, we reported the development of a new drug encapsulation strategy and a robust hybrid hydrogel platform for controlled and sustained release of small and large molecule ionic drugs. A biodegradable, biocompatible and temperature stimuli responsive hybrid hydrogel platform was fabricated from arginine based unsaturated poly(ester amides) (Arg-UPEAs), Pluronic diacrylate (Pluronic-DA) and alginate by the UV photo-crosslinking method, combining the favorable properties of a hydrogel and a polyelectrolyte complex. The hydrogels were systematically characterized, including the swelling mechanics, mechanical properties, biodegradation and interior morphologies. In vitro biocompatibility study showed that the hydrogels could support the cell attachment and proliferation well. Some model drugs, such as hydrochloride salts of hydralazine, insulin and interleukin-12, were encapsulated into the hydrogels and the drug release behavior was investigated using HPLC, LC-MS, BCA assay and ELISA assay. The obtained release profiles indicated that the Pluronic/Arg-UPEA/alginate hybrid hydrogels could release ionic drugs over weeks in vitro via a sustained manner. The structure-function study of hydrogels indicated that the polymer structure, hydrogel composition and environmental temperature had strong effects on the hydrogel properties and their drug release profiles.
Self‐healing injectable stimuli responsive polysaccharide hydrogel constructed with dynamic covalent bonds

Abstract:The self‐healing injectable hydrogels are co‐crosslinked with imine borate and borate ester bonds. The hydrogels are responsive to both pH values and oxidative environments. Hydrogels based on dynamic covalent bonds have unique properties and are competent in many biomedical fields. However, critical issues such as biocompatibility, mechanical properties, and stimulus responsiveness are required to be solved. It is also expected to develop a simple preparation method with mild conditions. Based on these objectives, chitosan (CS), sodium alginate (SA) and 2‐formylphenylboronic acid (2‐FPBA) are used to prepare the dynamic covalent bond hydrogels within minutes neither chemical medications nor severe conditions. Benefiting from the imine borate and boronic ester bonds both formed in the hydrogels, the hydrogels are stable, injectable, and of efficient self‐healing properties. The damage‐recovery process can be repeated for more than 6 cycles. The microstructure of the hydrogels recoded by atomic force microscopy (AFM) is consistent with the mechanical properties and both of them could be affected by the concentrations of SA and 2‐FPBA. Moreover, the dynamic hydrogels are responsive to pH values and oxidative environment. When the model drug 5‐fluorouracil (5‐FU) is loaded in the dynamic hydrogels, the release profiles are sustainable and present significant differences with various stimuli. These dynamic covalent bond hydrogels have a promising prospect in tissue engineering and drug delivery.
