Abstract:ABSTRACTChitosan‐based single network hydrogels with imine bonds have excellent self‐healing capability, while poor mechanical properties limit their applications. Here, chitosan‐polyacrylamide‐based double network hydrogels were prepared via in situ free‐radical polymerization of acrylamide in the presence of N‐carboxyethyl chitosan (CEC) and dibenzaldehyde‐terminated telechelic poly(ethylene glycol), which had excellent mechanical properties, self‐healing, and dual‐responsive shape memory abilities. The maximum tensile strength and elongation at break could reach 460 kPa and 4600%, respectively. Meanwhile, owing to the reversibility of imine bonds, elongation and strength at break of hydrogels could heal by 84.2 and 93.2% under alkali stimulation at 35 °C, respectively. Furthermore, the hydrogels also had good shape memory abilities for pH‐stimuli responsiveness of the imine bonds and metal ions stimuli responsiveness of CEC. Abstract:A continuously stable moist healing environment is immensely beneficial for wound healing, which can be availably achieved by providing an in situ hydrogel with enough strength resembling skin tissue and self-healing ability. Herein, through a dual-crosslinking strategy, hyaluronic acid-based hydrogels with excellent self-healing capacity and enhanced mechanical properties are fabricated via the acylhydrazone linkages and subsequent photocrosslinking based on hydrazide-modified sodium hyaluronate and aldehyde-modified maleic sodium hyaluronate. The hydrogels demonstrate the fast gelation process (< 1 min), the controlled swelling behaviors, and the good biocompatibility. Notably, they possess enhanced mechanical strength similar to the human dermis (∼ 2.2 kPa). Also, they can self-heal rapidly with a self-healing efficiency of ∼90 % at 6 h. Based on this, the hyaluronic acid-based hydrogels, without any biological factors involved, can facilitate the full-thickness skin wound reconstruction process by accelerating the three phases of the wound repair, including reducing wound inflammation in the inflammatory phase, promoting angiogenesis in the proliferative phase, and promoting the deposition and reconstruction of collagen in the remodeling phase. The produced hyaluronic acid hydrogel can serve as an ideal candidate for wound healing.

Abstract:We report a dual ionic cross-linking approach for the preparation of double-network hydrogels with robustness, high strength, and toughness, sodium alginate/poly(acrylamide-co-acrylic acid)/Fe3+ (SA/P(AAm-co-AAc)/Fe3+), in a facile "one-step" dual ionic cross-linking method. We take advantage of the abundant carboxyl groups on alginate molecules and the copolymer chains and their high coordination capacity with multivalent metal ions to obtain hydrogels with high strength and toughness. The optimal SA/P(AAm-co-AAc)/Fe3+ (SA 2 wt % and AAc S mol %) hydrogels showed a remarkable mechanical performance with 3.24 MPa tensile strength and 1228% strain, both of which remained stable with 76% water content and were highly swelling resistant in an aqueous environment. The hydrogels possessed high fatigue resistance, self recovery, pH-triggered healing capability, shape memory, and reversible gel sol transition facilitated by pH regulation. Moreover, they show three-dimensional (3D) printing processability by properly adjusting the solution viscosity. The approach may provide a convenient way of obtaining hydrogels having high strength and toughness with a number of desirable properties for a broad range of biomedical applications.

Abstract:<p>Natural hydrogels are widely investigated for biomedical applications because of their structures similar to extracellular matrix of native tissues, possessing excellent biocompatibility and biodegradability. However, they are often susceptible to mechanical disruption. In this study, self-healing hyaluronic acid (HA) hydrogels are fabricated through a facile dynamic covalent Schiff base reaction. Dialdehyde-modified HA (AHA) precursor was synthesized, and then the AHA/cystamine dihydrochloride (AHA/Cys) hydrogels were formed by blending AHA and Cys at acidic pH levels. By varying Cys to AHA ratio, the hydrogel morphology, swelling and kinetics of gelation could be controlled. Gelation occurred fast, which was predominantly attributed to Schiff base reaction between the dialdehyde groups on AHA and amimo groups on Cys. The hydrogel exhibited improved mechanical properties with increase in Cys content. Furthermore, due to dynamic imine bonds, this hydrogel demonstrated excellent self-healing ability based on the stress after mechanical disruption. Also, it was found to be pH-responsive and injectable. Taken together, this kind of hyaluronic acid hydrogel can provide promising future for various biomedical applications in drug delivery, bioprinting, smart robots and tissue regeneration.</p>

Abstract:The self-healing hydrogel offering intrinsic antibacterial activity is often required for the treatment of wounds because it can provide effective wound protection and prevent wound infection. Herein, antibacterial hyaluronic acid hydrogels with enhanced self-healing performances are prepared by multiple dynamic-bond crosslinking between aldehyde hyaluronic acid, 3, 3′- dithiobis (propionyl hydrazide) and fungal-sourced quaternized chitosan. Due to the formation of these different types of reversible interactions e.g. hydrazone bonds, disulfide bonds, and electrostatic interactions, the hyaluronic acid hydrogels can gel rapidly and exhibit excellent self-healing ability, which can heal completely within 1 h. Furthermore, the hydrogels show good antibacterial activity against E. coli and S. aureus with an inhibition ratio of ~100 % and above 75 %, respectively. Additionally, the hydrogels are cytocompatible, which makes them the potential for biomedical applications e.g. cell culture, tissue engineering, and wound dressing.

Abstract:The wide development of hydrogels had been used in many filed due to the high water-containing and tough three-dimensional structure, however, the poor mechanical and multi-functional properties of hydrogel can be limited in its applications deeply. Herein, the dual responsive self-healing hydrogels with tough mechanical properties were manufactured by dual-physical cross-linking based on biodegradable aliphatic polycarbonate. Choosing the soft and hard segments to design the polymeric hydrogel not only can facilitate the dual-dynamic bonding interactions but also the resilient hydrogels possess robust and controllable mechanical strength (6.51 MPa). Furthermore, the results of swelling and stability tests of the materials indicated that the swelling ability of the biodegradable hydrogels can be regulated by the hydrophilic group, and the maximal swelling ratio in water and the equilibrium water content is 66% and 40%, respectively. It is worth mentioning that the tough hydrogels embrace dual-responsive high efficiency of self-healing ability, and the self-healing time is 2 h at 50 °C or 10 h under pH = 5, suggesting that the obtained hydrogels can respond to temperature and pH value to drive the fracture interface for fast self-healing, which will offer new opportunities for stimuli-responsive materials and wound healing.

Abstract:Conventional wound healing materials suffer from low efficiency, poor mechanical strength, and nontunable properties. Responsive hydrogels are appealing candidates for tissue engineering. Herein, we developed a double-cross-linked hydrogel system composed of hyperbranched PEG-based polymer, comprising pre-cross-linked acetal structure and numerous terminal acrylate groups, which can form hydrogels in situ and can be further strengthened by UV irradiation. The hyperbranched glycidyl methacrylate-co-poly(ethylene glycol) diacrylate polymers (HB-GMA-PEGs) were first synthesized via in situ deactivation enhanced atom transfer radical polymerization (DE-ATRP). A series of pre-cross-linked materials were achieved after postfunctionalization. The material can absorb a high amount of water to form hydrogels, and the gel stiffness was evaluated in detail before and after UV irradiation. The in vitro cytotoxicity experiments were conducted with the resulting materials and have demonstrated their good biocompatibility. The results indicate that this type of hydrogel with high water uptake capacity has appealing potential as a responsive biomaterial for wound closure.
Tough Supramolecular Hydrogels with Excellent Self-Recovery Behavior Mediated by Metal-Coordination Interaction

Abstract:Based on the strategy of dynamic metal-coordination for improving mechanical properties, a further ideal was taken to prepare the tough hydrogels with the combination of self-crosslinking monomer and metal-coordination complexes. Herein, a series of hydrogels of poly(acrylamide-co-acrylic acid-co-N-hydroxymethyl acrylamide) (P(AM-co-AAc-co-NMAM)) consisting the chemical crosslinking induced by NMAM units and the physical crosslinking derived from the coordination complexes of carboxyl-Fe3+ were prepared. The molecular structure was investigated with the ATR-FTIR, Raman and UV-vis spectra. These hydrogels with different water content of 57-93% possess good mechanical performances. The optimal hydrogels possess high tensile strength (8.56 MPa), prominent modulus (15.5 MPa), remarkable toughness (37.85 MJ/m(3)) and superb tearing energy (7062 J/m(2)). The tough hydrogels also display excellent self-recovery (95% toughness recovery within 50 min), pH-triggered healing, shape memory and plasticity abilities. These hydrogels having high strength and toughness may broaden range of potential applications in load-bearing soft actuators, flexible electronics, etc.

Abstract:Cell therapies have great potential for the treatment of many different diseases, while the direct application of cells to the targeted location leads to limited therapeutic outcomes due to the low cell engraftment and cell survival rate. Injectable hydrogels have been developed to facilitate cell delivery; however, those currently developed hydrogel systems still face the limited cell survival rate. Here, an injectable and self-healable hydrogel is reported through the combination of hyperbranched PEG-based multi-hydrazide macro-crosslinker (HB-PEG-HDZ) and aldehyde-functionalized hyaluronic acid (HA-CHO), with gelatin added to increase the crosslinking density and cell activity. The hydrogels can be formed only in 7 s due to the relatively high content of the functional end groups. The reversible crosslinking mechanism between the hydrazide and aldehyde groups endows the hydrogel with shear-thinning and self-healing properties. The hydrogels with gelatin exhibit relatively better mechanical properties and cell activity. The hydrogels can improve the survival, attachment, and engraftment of injected cells due to the rapid sol-gel transition, which can promote an enhanced regenerative response.
Dual Physically Cross-Linked κ-Carrageenan-Based Double Network Hydrogels with Superior Self-Healing Performance for Biomedical Application

Abstract:Chemically linked double network (DN) hydrogels display extraordinary mechanical attributes but mostly suffer from poor self-healing property and unsatisfactory biocompatibility due to the irreversible breaks in their chemical-linked networks and the use of toxic chemical cross-linking agents. To address these limitations, we developed a novel κ-carrageenan/polyacrylamide (KC/PAM) DN hydrogel through a dual physical-cross-linking strategy, with the ductile, hydrophobically associated PAM being the first network, and the rigid potassium ion (K+) cross-linked KC being the second network. The dual physically cross-linked DN (DPC-DN) hydrogels with optimized KC concentration exhibit excellent fracture tensile stress (1320 ± 46 kPa) and toughness (fracture energy: 6900 ± 280 kJ/m3), comparable to those fully chemically linked DN hydrogels and physically chemically cross-linked hybrid DN hydrogels. Moreover, because of their unique dual physical-cross-linking structures, the KC/PAM hydrogels also demonstrated rapid self-recovery, remarkable notch-insensitivity, self-healing capability, as well as excellent cytocompatibility toward stem cells. Accordingly, this work presents a new strategy toward fabricating self-repairing DPC-DN hydrogels with outstanding mechanical behaviors and biocompatibility. The new type of DN hydrogels demonstrates strong potentiality in many challenging biomedical applications such as artificial diaphragm, tendon, and cartilage.
Highly Efficient Self‐Healable and Dual Responsive Cellulose‐Based Hydrogels for Controlled Release and 3D Cell Culture

Abstract:To face the increasing demand of self‐healing hydrogels with biocompatibility and high performances, a new class of cellulose‐based self‐healing hydrogels are constructed through dynamic covalent acylhydrazone linkages. The carboxyethyl cellulose‐graft‐dithiodipropionate dihydrazide and dibenzaldehyde‐terminated poly(ethylene glycol) are synthesized, and then the hydrogels are formed from their mixed solutions under 4‐amino‐DL‐phenylalanine (4a‐Phe) catalysis. The chemical structure, as well as microscopic morphologies, gelation times, mechanical and self‐healing performances of the hydrogels are investigated with 1H NMR, Fourier transform infrared spectroscopy, atomic force microscopy, rheological and compression measurements. Their gelation times can be controlled by varying the total polymer concentration or 4a‐Phe content. The resulted hydrogels exhibit excellent self‐healing ability with a high healing efficiency (≈96%) and good mechanical properties. Moreover, the hydrogels display pH/redox dual responsive sol‐gel transition behaviors, and are applied successfully to the controlled release of doxorubicin. Importantly, benefitting from the excellent biocompatibility and the reversibly cross‐linked networks, the hydrogels can function as suitable 3D culture scaffolds for L929 cells, leading to the encapsulated cells maintaining a high viability and proliferative capacity. Therefore, the cellulose‐based self‐healing hydrogels show potential applications in drug delivery and 3D cell culture for tissue engineering.

Abstract:The growing list of potential applications of soft materials increases the demand for, among others, new multifunctional hydrogels. We propose a hybrid double-network hydrogel based on synthetic poly(acrylic acid) and alginate, a natural polymer. These polymer networks were double crosslinked: chemically and physically. The ferrous ion was used to catalyze the decomposition of the initiator and later, after its oxidation by the initiator to the ferric ion, was coordinated by the carboxylic groups of the polymer networks and acted as a physicochemical crosslinker. The composition of the hydrogel was optimized to get improved mechanical and electrical properties, and the self-healing ability. The introduction of the coordination bond between Fe3+ and the carboxylic groups of AA and alginate molecules created the ability of self-healing of the hydrogels; they could regenerate without any external stimulus and regain their electrical conductivity and mechanical properties.

Abstract:Hyaluronic acid hydrogels are promising materials for diverse applications, yet their potential is hampered by limitations such as low self‐healing efficiency and insufficient mechanical strength. Inspired by the heterogeneous structures of spider silk, we introduce a novel dual dynamically crosslinked network hydrogel. This hydrogel comprises an acylhydrazone‐crosslinked network, utilizing aldehyde hyaluronic acid (AHA) and 3,3'‐dithiobis (propionohydrazide) (DTP) as a first network, and a secondary network formed by hydrogen bonds‐crosslinked network between tannic acid (TA) and silk fibroin (SF) with β‐sheet formation. The hydrogel exhibits exceptional self‐healing ability due to the dynamic and reversible nature of Schiff base bonds, disulfide bonds, and hydrogen bonds, achieving complete healing within 5 minutes. Additionally, the spider silk‐inspired heterogeneous structures enhance mechanical properties. Furthermore, the incorporation of TA provides enhanced adhesion, as well as remarkable antibacterial and antioxidant properties. This innovative hyaluronic acid‐based hydrogel, inspired by spider silk, offers a promising avenue to fortify both the mechanical strength and self‐healing capabilities of hydrogels, thus expanding opportunities for applications in tissue engineering and biomedicine.

Abstract:<p>Most structural self-healing materials were developed based on either reversible supramolecular interaction or dynamic covalent bonding. It seems a good idea to incorporate self-healing properties into high-performance materials. In this study, we fabricated the alginate-based cyclodextrin and polyacrylamide azobenzene highly stretchable and tough interpenetrating composite hydrogel with self-repairing behavior under light irradiation. Initially, the alginate-based cyclodextrin and polyacrylamide azobenzene were designed and synthesized. The corresponding structural, thermal, and morphological properties of hydrogels were characterized. The reversible transformation of the sol-gel can be achieved by the irradiation upon ultraviolet light and visible light. The self-healing behavior of this composited gel is based on the host-guest interaction between cyclodextrin and azobenzene. The recovery gel elongation at 48 h healing in the dark condition was is 0.04 MPa, with an elongation of 1140 %. Therefore, this gel can achieve self-healing ability while maintaining highly stretchable and tough performance.</p>

Abstract:Self-healing hydrogel systems usually suffer from poor mechanical performance stemmed from weaker and reversible non-covalent interactions or dynamic chemical bonds, which hamper their practical applications. This issue is addressed by adopting a double-crosslinking design via employing both dynamic Schiff base bonds and non-dynamic photo-induced crosslinking. This leads to the formation of a special topological structure which simultaneously provide good self-healing capability (healing efficiency of 73.5%) and good mechanical performance (good elastic recovery and tensile modulus of 157.4 kPa, close to modulus of native skin). The quaternary ammonium and protonated amino groups can provide superior antibacterial capability, while Schiff base formation between residual aldehyde groups and amino groups on tissue surface contribute to hydrogel’s good adhesion to tissues (5.9 kPa). Furthermore, the multifunctional hydrogel with desirable mechanical performance, self-healing capability, superior antibacterial capability and good tissue adhesion can significantly promote healing of infectious cutaneous wound, tissue remodeling and regeneration.

Abstract:Superior flexibility and toughness can be achieved in bioactive hydrogels by the use of a double polymer network with complementary properties. Inspired by this design principle, we here combine polyacrylic acid (PAA) and sodium alginate (SA) to obtain a dual-reinforced double interpenetrating network (d-DIPN) hydrogel. The dual reinforcement involves ionic cross-linking and introduction of SiO2 nanoparticles, which leads to extraordinary improvements in strength and toughness. Compared with the standard PAA hydrogel that offers an elongation of 240% and a breakage stress of 0.03 MPa, the prepared SA(Ca2+)-PAA-SiO2 hydrogel shows an elongation above 1000% and a breakage stress of 1.62 MPa. Moreover, the combination of strong covalent cross-links and weak reversible interactions provides the d-DIPN hydrogel with swelling resistance and self-healing behavior, adhesive abilities, and shape memory performance. Furthermore, we show that the biocompatibility and bone cell proliferation ability of the hydrogels can be improved through a mineralization process despite an observed reduction in breakage strain and stress. Taken as a whole, our work paves the way for the design of strong and tough hydrogels, with potential applications within biomedicine and particularly tissue engineering.

The introduction of normal human dermal fibroblast (NHDF) cells enables the formation of dually crosslinked hydrogels through hydrogen interactions between cell secretions ( mainly the secretory proteins) and polymer chains. This allows an enhancement of the self-healing efficiency from 73% to 92% and an acceleration of the self-healing rate (12 times). Based on the excellent biocompatibility, antibacterial property and low toxicity, we extended the use of dually crosslinked hydrogels as wound dressings to expedite wound healing. This cell-loading concept for constructing dually crosslinked hydrogels offers an available pathway to design smart materials useful for biomedical applications.

Abstract:Endowing polymer hydrogels with good self-healing ability can autonomously repair damage with improved reliability. In this work, the benzaldehyde group was first grafted onto a biocompatible poly(vinyl alcohol) (PVA) molecular chain by esterification to obtain aldehyde-functionalized PVA (APVA), and the reversible imine bonds were further formed by reacting with amine groups on a quaternized chitosan (HTCC) chain. And thus, the self-healing APVA/HTCC hydrogel was fabricated with such imine bonds as crosslinking points together with hydrogen bonds. Many more imine bonds of hydrogels formed with increasing aldehyde content, resulting in increasing crosslinking density, decreasing average pore diameter and formation of a compact dynamic network, imparting certain mechanical strength and toughness with hydrogels. Furthermore, the healing efficiency of the hydrogel reached as high as 91.7% by self-healing without any external stimulus and its microstructure could be reconstructed after damage, exhibiting rapid recovery and dynamic features. Biocompatible self-healing PVA-based hydrogels exhibited great potential application in biomedical fields, like smart infill biomaterials, tissue engineering scaffolds, etc.

Abstract:A gamma-glutamyl kinase (ATP-L-glutamate 5-phosphotransferase) was purified about 85-fold from crude extracts of Pseudomonas aeruginosa strain PAO 1 by (NH4)2SO4 precipitation, molecular-sieving by Sephadex G-150 and DEAE-cellulose chromatography. The molecular weight of this enzyme was 84,000. The preparation catalysed formation of gamma-glutamyl hydroxamate from L-glutamate, ATP and Mg2+ or Mn2+ with concomitant hydrolysis of ATP to ADP + Pi. L-Proline inhibited the gamma-glutamyl kinase activity by 50% at 5 mM and almost completely at 30 mM. The inhibition of L-proline was non-competitive, wherease L-methionine-DL-sulphoximine inhibited the enzyme competitively. Proline was found to inhibit the gamma-glutamyl kinase activity of the wild-type strain and of representatives of two of the three transductional classes of proline-auxotrophic mutants. Strain PAO 879, a mutant representing the third transductional class of proline auxotrophs, lacked proline-inhibitible gamma-glutamyl kinase. Thiol-blocking reagents inhibited the gamma-glutamyl kinase and this effect was prevented by dithiothreitol.

Abstract:A novel biocompatible polysaccharide‐based self‐healing hydrogel, CEC‐l‐OSA‐l‐ADH hydrogel (“l” means “linked‐by”), is developed by exploiting the dynamic reaction of N‐carboxyethyl chitosan (CEC) and adipic acid dihydrazide (ADH) with oxidized sodium alginate (OSA). The self‐healing ability, as demonstrated by rheological recovery, macroscopic observation, and beam‐shaped strain compression measurement, is attributed to the coexistence of dynamic imine and acylhydrazone bonds in the hydrogel networks. The CEC‐l‐OSA‐l‐ADH hydrogel shows excellent self‐healing ability under physiological conditions with a high healing efficiency (up to 95%) without need for any external stimuli. In addition, the CEC‐l‐OSA‐l‐ADH hydrogel exhibits good cytocompatibility and cell release as demonstrated by three‐dimensional cell encapsulation. With these superior properties, the developed hydrogel holds great potential for applications in various biomedical fields, e.g., as cell or drug delivery carriers.
