Abstract:α-Hydroxy amides are a class of important intermediates in organic synthesis. It plays an important role in the field of science and medicine. However, the preparation methods of α-hydroxy amides are relatively few, and they are usually enzyme-or Lewis acid-catalyzed reactions. These types of methods are not ideal in operation, yield and/or stereoselectivity. We found a new way to synthesize α-hydroxy amides by transforming amino acids into α-hydroxyl acids, and then by amination of the α-hydroxy acids.

Abstract:In order to gain deep insight into the mechanism of the synthetically well established Rh(II)-mediated intramolecular C-H insertion reaction, the classic physical organic chemistry method, the Hammett linear free energy correlation, has been applied. In the development of new synthetic methodologies of alpha-diazo carbonyl compounds, it was found that Cu(acac)(2) can effectively promote the diazo, decomposition and resulted in selective intramolecular N-H insertion. On the other hand, the Wolff rearrangement of the alpha-diazo carbonyl compounds catalyzed by Ag(I) has been applied to the efficient synthesis of optically pure alpha-lactams.

Abstract:Diazo compounds had been widely employed as a class of environment friendly and highly efficient coupling reagents in transition-metal-catalyzed C-H bonds insertion reactions.Recently, C-H alkylation of various arenes and heteroarenes has been achieved enormous successes using diazo compounds as the alkylation reagents under a new catalytic model.The recent progress in diazo compounds mediated transition-metal-catalyzed C-H alkylation and tandem C-H alkylation/cyclization is reviewed, including reaction mechanism and synthetic applications.Furthermore, the prospects of this reaction are also discussed.
Selective alpha-Bromination of Functionalized Ketene Dithioacetals

Abstract:Functionalized ketene dithioacetals are important intermediates in organic synthesis. alpha-Functionalization reactions based on them provide potential precursors for the construction of usefully carbo- and hetero-cyclic compounds. In this paper, a versatile alpha-bromination of functionalized ketene dithioacetals was developed. In the presence of H2O, the reactions of functionalized ketene dithioacetals(1) with CuBr2 in THF at room temperature gave various alpha-bromo ketene dithioacetals (2) in excellent yields with high regioselectivities. The mechanism of this alpha-bromination is proposed to undergo an addition of bromine formed in situ to compound 1 and sequontial elimination of the alpha-proton. The simplicity of execution, mild conditions, inexpensive, easily usable brominating agent and high yields, make this synthetic method attractive for practical applications.

Abstract:The nucleophilic addition reaction of diazo-compound derived anions or enolates with electrophilic C=O and C=N bonds have been summarized. The subsequent reactions of the addition products demonstrate diverse reactivities of the diazocarbonyl compounds bearing other functional groups and their potential in organic synthesis. Asymmetric catalysis is also possible with diazocarbonyl compounds as nucleophiles.

Abstract:The synthesis of small molecules and complex scaffolds is one of the most important topics in organic synthesis. Aryl methyl ketones, as simple and easily available materials with rich and diverse structures, can generate α-iodoaryl methyl ketones, aryl ketoaldehydes, or sulfur ylide intermediates in the I2–dimethyl sulfoxide (DMSO) system. These molecules can be easily captured by other components in the system in situ to drive various organic unit reactions and continuous cascade transformation to realize the effective construction of C–X bonds, including C–C, C–S, C–N, and C–O bonds. The reaction system of I2–DMSO–aryl methyl ketone has been widely used in the functionalization of the α-C–H bond of aryl methyl ketones, heterocyclic synthesis, total synthesis of natural products, and other reactions. In view of the rich research foundation and great synthesis potential of the I2–DMSO–aryl methyl ketone reaction system, here, we review the latest progress in this field from the aspects of reaction design, the synthesis strategy, and mechanism research, with an emphasis on its synthesis application. We believe that this review will attract the interest of medical and synthetic organic chemists.

Abstract:Cyclooctyne molecules have found wide applications in the strain-promoted azide–alkyne cycloaddition (SPAAC) reactions, which avoid the biotoxicity caused by the use of Cu(I) catalysts. Among the various cyclooctyne systems, dibenzocyclooctyne (DBCO) series have displayed the highest reaction activity. However, the synthesis processes of such structures are time-consuming, which to some extent limit their large-scale development and application. This review has summarized current synthesis routes of two DBCO molecules, aza-dibenzocyclooctyne (DIBAC) and biarylazacyclooctynone (BARAC).

Abstract:The class of Double Diels-Alder (DDA) reactions leading the targets concisely where numerous synthetic pursuits have been rewarded with conceptually novel and streamlined method. After the tremendous development and discoveries in various Diels-Alder reactions including its key role applications in many natural products syntheses, DDA reaction is considered as a growing branch of Diels-Alder reaction where two consecutive [4+2] cycloaddition trigger the streamlined construction of polycyclic and macrocyclic architecture. In light of the ever-increasing importance of DDA reactions in chemical sciences, we review the comprehensive study and recent advances of DDA reactions in organic synthesis. This review summarizes key achievements of DDA in preparation of macrocyclic, bicyclic, heterocyclic, and polycyclic structures documented since its revelation in 1980 to 2020. Emphasis is stand on the synthesis of various structural class based on the selectivity of DDA reaction.

Abstract:We have witnessed the striking advancement of CH functionalization in organic synthesis over the past decade. This short review spotlights the very recent applications of CH functionalization in natural product synthesis and drug synthesis. Some representative examples of natural product total synthesis facilitated by CH functionalization are classified by CO, CC, CN or C–X bond formation. Three different total syntheses are highlighted in details, in which iterative CH functionalization strategy is involved. Another example of Merck's synthesis of anacetrapib is also discussed to briefly demonstrate the broad application of CH functionalization strategy in process chemistry of pharmaceutical industry.
