Abstract:The preparation of a series of 2-pyridinamines and their alkyl derivatives is described.The target compounds are synthesized starting from corresponding 2-cyanopyridines, via incomplete hydrolysis in the presence of hydrogen peroxide in the dilute alkaline mixture of acetone-2% sodium hydroxide solution, and Hoffmann degradation reaction with freshly made NaBrO.This route is of industrial value because of cheap and readily available materials, moderate reaction conditions and convenient operations.

Abstract:In sharp contrast to the well researched and well developed aminocarbonylation of alkenes, the dearomative carbamoylation of abundant arene pi-systems has scarcely been studied despite its great potential to enrich the diversity-oriented synthesis of high-value amides. The formidable challenges associated with such dearomatization include the low reactivity of stable aromatic pi-systems and intricate selectivity issues. Herein, we disclose a general approach toward highly selective dearomative carbamoylations of arene pi-bonds under CO-gas-free strated by tolerating a broad range of nucleophilic partners with high yields and excellent selectivities, thus providing modular access to the divergent synthesis of beta-functionalized primary amides. In addition, diverse downstream derivatizations including a for-mal C-H 1,2-olefination/carbamoylation reaction were conducted, exhibiting great potential in syn-thetic and medicinal chemistry.

Abstract:α-Hydroxy amides are a class of important intermediates in organic synthesis. It plays an important role in the field of science and medicine. However, the preparation methods of α-hydroxy amides are relatively few, and they are usually enzyme-or Lewis acid-catalyzed reactions. These types of methods are not ideal in operation, yield and/or stereoselectivity. We found a new way to synthesize α-hydroxy amides by transforming amino acids into α-hydroxyl acids, and then by amination of the α-hydroxy acids.

Abstract:Diazo carbonyl compounds can be decomposed under various conditions and result in diverse types of chemical transformations.For this reason,their application in organic synthesis have attracted considerable attention.In order to gain deep insight into the mechanism of the synthetically well established Rh(Ⅱ) mediated intramolecular C\|H insertion reaction,the classic physical organic chemistry method,the Hammett linear free energy correlation,has been applied.In the development of new synthetic methodologies of α diazo carbonyl compounds,it was found that Cu(acac)\-2 can effectively promote the diazo decomposition and resulted in selective intramolecular N\|H insertion.On the other hand,the Wolff rearrangement of the α diazo carbonyl compounds catalyzed by Ag(Ⅰ) has been applied to the efficient synthesis of optically pure γ lactams.

Abstract:Transition-metal catalysts have been widely utilized in many organic syntheses. Rhodium acetate was found to be an efficientcatalyst for organic reactions. The purpose of this review is to give an overview of the various reactions of the Rh2 (OAc)4-stabilized carbenoids derived from α-dizao-oxo-compounds. The types of these reactions include cyclication reaction, insertion reaction and the formation of ylides. The results show that the Rh(Ⅱ) carbenoids and the reactive ylide intermediates produced from the Rh(Ⅱ)carbenoids with the heteroatom of the system can undergo many organic translations and give enormous new synthetic methods to prepare compounds containing many function groups in one step.

In the past two decades, the research activities related to diazo compounds have gained increased momentum. Abstract:Diazo compounds had been widely employed as a class of environment friendly and highly efficient coupling reagents in transition-metal-catalyzed C-H bonds insertion reactions.Recently, C-H alkylation of various arenes and heteroarenes has been achieved enormous successes using diazo compounds as the alkylation reagents under a new catalytic model.The recent progress in diazo compounds mediated transition-metal-catalyzed C-H alkylation and tandem C-H alkylation/cyclization is reviewed, including reaction mechanism and synthetic applications.Furthermore, the prospects of this reaction are also discussed.

Abstract:The synthesis of small molecules and complex scaffolds is one of the most important topics in organic synthesis. Aryl methyl ketones, as simple and easily available materials with rich and diverse structures, can generate α-iodoaryl methyl ketones, aryl ketoaldehydes, or sulfur ylide intermediates in the I2–dimethyl sulfoxide (DMSO) system. These molecules can be easily captured by other components in the system in situ to drive various organic unit reactions and continuous cascade transformation to realize the effective construction of C–X bonds, including C–C, C–S, C–N, and C–O bonds. The reaction system of I2–DMSO–aryl methyl ketone has been widely used in the functionalization of the α-C–H bond of aryl methyl ketones, heterocyclic synthesis, total synthesis of natural products, and other reactions. In view of the rich research foundation and great synthesis potential of the I2–DMSO–aryl methyl ketone reaction system, here, we review the latest progress in this field from the aspects of reaction design, the synthesis strategy, and mechanism research, with an emphasis on its synthesis application. We believe that this review will attract the interest of medical and synthetic organic chemists.

Abstract:Diels-Alder reaction is a kind of reaction with high selectivity, high productivity, high reliability and condition tolerability. Ever since its discovery, it has been widely used in the organic synthetic chemistry and become a click chemistry. Recently, the Diels-Alder reaction is also gradually becoming an important linking method in the preparation for polymers with well-defined architectures, due to its broad practicability and orthogonality. Based on the classification of polymer architecture, this paper reviews the applications of Diels-Alder reaction. in preparing these polymers. Moreover, the paper gives the outlook for further prospect of Diels-Alder reaction in the polymer science. The relative key questions include methodology developments, application in the preparation of polymers with more specialized architectures, and broadening the functional usages.

Abstract:The [5+2] cycloaddition is one kind of fascinating and powerful reaction to access seven-membered ring systems existing in complex natural products and pharmaceuticals. In this review, recent developments on the [5+2] cycloadditions and their application in the synthesis of complex natural products are discussed. A focus is given to the way in which the [5+2] cycloadditions have influenced the logical process of the synthesis. Abstract:The class of Double Diels-Alder (DDA) reactions leading the targets concisely where numerous synthetic pursuits have been rewarded with conceptually novel and streamlined method. After the tremendous development and discoveries in various Diels-Alder reactions including its key role applications in many natural products syntheses, DDA reaction is considered as a growing branch of Diels-Alder reaction where two consecutive [4+2] cycloaddition trigger the streamlined construction of polycyclic and macrocyclic architecture. In light of the ever-increasing importance of DDA reactions in chemical sciences, we review the comprehensive study and recent advances of DDA reactions in organic synthesis. This review summarizes key achievements of DDA in preparation of macrocyclic, bicyclic, heterocyclic, and polycyclic structures documented since its revelation in 1980 to 2020. Emphasis is stand on the synthesis of various structural class based on the selectivity of DDA reaction.

Abstract:The nucleophilic addition reaction of diazo-compound derived anions or enolates with electrophilic C=O and C=N bonds have been summarized. The subsequent reactions of the addition products demonstrate diverse reactivities of the diazocarbonyl compounds bearing other functional groups and their potential in organic synthesis. Asymmetric catalysis is also possible with diazocarbonyl compounds as nucleophiles.

However, this year and last year have witnessed unprecedentedly wide applications of this type of compound in many fields well beyond pure organic synthesis, ranging from photo-click reactions, polymer-backbone editing, catalytic Fisher carbene formation, acylsilanes-hydroxylation ligation, etc. The recently-developed quick synthetic method for acylsilanes makes the chemistry even more attractive. The review covers the selected representative examples and focuses on their possible utilities in the future.
