Abstract:Ascorbic acid (AA) is a powerful antioxidant in food safety and disease treatment. It is of great significance to develop a low-cost, high-stability, and easy-to-operate colorimetric method for quantitative detection of AA in food or human body. Although various nanozymes have been developed for the colorimetric detection of AA, the size regulation of the catalytic center of nanozymes remains a challenge. In this work, we propose a combined strategy of flow chemistry synthesis and pyrolysis to realize the controllable adjustment of the catalytic center size of nanozymes. Zinc-cobalt zeolitic imidazole frameworks (ZnCo-ZIFs) with different sizes are synthesized by flow chemistry. Nitrogen-doped carbon materials with different Co catalytic centers (80 nm-10 nm) are then obtained by pyrolysis of ZnCo-ZIFs precursors. Among them, cobalt quantum dot embedded nitrogen-doped carbon (Co QDs/N-C) exhibits excellent oxidase activity, with Vmax and Km of 4.19 × 10-7 M s-1 and 0.12 mM. Therefore, a simple, low-cost, and stable colorimetric method for the detection of AA is established with a good linear relationship (3-500 μM) and low detection limit (0.40 μM). This work has certain guiding significance for the size regulation of catalytic center of nanozyme, and the detection method has broad application prospects in biochemical sensing field.
N-Doped Carbon Dots as Fluorescent “Turn-Off” Nanosensors for Ascorbic Acid and Fe 3+ Detection

Abstract:Carbon-based sensors for the detection of ascorbic acid (AA) and ferric ions have drawn a great deal of attention recently, owing to their excellent optical properties and good biocompatibility for biological applications. In this work, a series of nitrogen-doped carbon dots (NCDs) were fabricated by a microwave-assistant approach with a combination of dl-malic acid and urea as precursors. Under optimal experimental conditions, NCDs with an average size of 4.52 ± 0.05 nm were prepared, containing amino N and C–N functional groups on the surface of carbon cores. Optical analysis showed that the NCDs exhibited excitation-dependent and concentration-dependent emission properties. A single emission at 450 nm was observed with two luminescent centers at 280 and 370 nm for concentrations ranging from 0.02 to 0.08 mg/mL. Moreover, the NCDs were further used as a fluorescence sensor to detect AA and Fe3+ in solution. From the metal ion sensing research, Fe3+ demonstrated significant quenching abilities on NCDs with a detection limit of 1.9 μM. More importantly, the NCDs also showed an excellent quenching response by AA through the static quenching mechanism and inner filter effect with a detection limit of 2.6 μM. Additionally, the low cell toxicity against MA104 and 293T cells from both monkeys and humans were affirmed, respectively. Therefore, the NCDs developed in the present work provide a “turn-off” strategy for the highly sensitive detection of AA and Fe3+ ions and can be potentially applied in both environmental and biological systems.

Abstract:Carbon dots (CDs) have caught enormous attention owing to their distinctive properties, such as their high water solubility, tunable optical properties, and easy surface modification, which can be generally used for the detection of heavy metals and organic pollutants. Herein, nitrogen and fluorine co-doped carbon dots (NFCDs) were designed via a rapid, low-cost, and one-step microwave-assisted technique using DL-malic acid and levofloxacin. The NFCDs emitted intense green fluorescence under UV lighting, and the optical emission peak at 490 nm was observed upon a 280 nm excitation, with a high quantum yield of 21.03%. Interestingly, the spectral measurements illustrated excitation-independent and concentration-independent single-color fluorescence owing to the presence of nitrogen and fluorine elements in the surface functional groups. Additionally, the NFCDs were applied for the selective detection of Fe3+ and ascorbic acid based on the “turn-off” mode. The detection limits were determined as 1.03 and 4.22 μM, respectively. The quenching mechanisms were explored using the static quenching mechanism and the inner filter effect. Therefore, a NFCDs fluorescent probe with single color emission was successfully developed for the convenient and rapid detection of Fe3+ and ascorbic acid in environments.

Abstract:Ascorbic acid (AA) is involved in many physiological activities of the body and plays an important role in maintaining and promoting human health. It is also present in many natural and artificial foods. Therefore, the development of highly sensitive and accurate AA sensors is highly desirable for human health monitoring, as well as other commercial application fields. Herein, an ultrasensitive and selective electrochemical sensor based on an aptamer was developed for the determination of AA for the first time. The aptasensor was fabricated by modifying a composite made of polyaniline (PANI) and gold nanoparticles (AuNPs) on a glassy carbon electrode. The morphologies and electrochemical properties of the resulting electrodes were characterized by various analytical methods. The results indicated relatively good electrical conduction properties of PANI for accelerated electron transfer. The modification with AuNPs provided signal amplification, suitable for applications as novel platforms for the sensitive sensing of AA. Under optimized conditions, the proposed aptasensor displayed a wide linear response toward the detection of AA from 1.0 to 1.0 x 10(5) ng L-1 coupled with a low detection limit of 0.10 ng L-1. The sensor also exhibited excellent selectivity and high stability, with at least 2000-fold higher sensitivity than similar previously reported methods. Importantly, the aptasensor exhibited promising properties for the determination of AA in real fruits, vegetables, and infant milk powder, thereby showing potential for food analysis.

Abstract:Herein, we presented an ingenious method for the detection of trace ascorbic acid (AA) using fluorescent sensing system, which was composed of periodate (IO 4 − ) and nitrogen-boron co-doped carbon dots (N, B-CDs). Highly photoluminescent carbon dots with a quantum yield of 84.5% were synthesized by a one-step hydrothermal method using citric acid, ethylenediamine and boric acid as raw materials. The fluorescence of N, B-CDs could be statically quenched by IO 4 − , and non-fluorescent complexes were formed on the surface of carbon dots at the same time. With the pre-addition of ascorbic acid, the process of fluorescence quenching by IO 4 − was inhibited due to the preferential redox reaction between AA and IO 4 − . Based on the principle mentioned above, we designed the "inhibition" fluorescent sensor that can easily realize the detection of AA, and an extremely low limit of detection of 0.035 nM with a linear interval of 0–1600 pM was obtained. In addition, the sensing system has been applied to the detection of AA in human urine and obtained satisfactory results. The developed "inhibition" fluorescent sensor with high sensitivity and selectivity, has shown great potential and application prospects in the field of biosensing.

Abstract:Nitrogen-doped carbon dots (N-CDs) were prepared by self-exothermic procedure using grasshopper powder as a single precursor. The prepared N-CDs not only have excellent fluorescence properties, but also can catalyze and enhance the ultra-weak chemiluminescence of NaHCO3-H2O2. The reaction conditions of NaHCO3-H2O2-N-CDs CL were optimized. Under the optimal experimental conditions, when AA was added to the NaHCO3-H2O2-N-CDs CL system, AA had a significant inhibitory effect on the CL intensity of NaHCO3-H2O2-N-CDs. There was a good linear relationship between the calculated lg(I0/I) and the concentration of AA (C), and the calibration curve equation was lg(I0/I) = 0.03667 C-0.00708 (µM). The established CL analysis method has a detection limit of 0.12 µM for AA and a linear range of 0-50 µM. The selectivity of CL method was evaluated, and the method was successfully applied to the determination of AA in vegetable and fruit samples. The spiked recoveries were between 88.9% and 118.9%, which indicated that the method was simple, rapid, and sensitive, and had great potential in the determination of AA in foods.

Abstract:Label-free detection of ascorbic acid (AA) with high sensitivity and specificity based on the effects of AA on fluorescence quenching of carbon nitride quantum dots (CNQDs) is described. The CNQDs with a high fluorescence quantum yield of 21% and good dispersibility in water are prepared by reacting g-C3N4 sheets with ethylenediamine (EN). Fluorescence at 510 nm from the CNQDs is quenched gradually by addition of AA. As the AA concentration increases, the activity of the lone pair (LP) state of the CNQDs diminishes resulting in reduced fluorescence from the CNQDs and the mechanism of the static quenching effect is discussed. Since the CNQDs have a large specific surface area and abundant amino groups and AA exists in the anionic form at the physiological pH, the electrostatic interaction between CNQDs and AA inhibits excitation and emission of the LP states in the CNQDs. Owing to steric effects and hydrogen bonding, the CNQDs constitute a sensitive and selective detection platform for AA in a wide range from 0.5 to 200 μM with a detection limit of 150 nM (signal-to-noise ratio of 3). More importantly, the strategy can successfully be applied to the detection of AA concentrations in serum samples. This simple method which provides quantitative AA determination has large potential in clinical and health-related applications and the mechanism provides insights into intracellular AA monitoring.

Abstract:Synthesis of nitrogen-doped carbon dots (N-CDs) using urea and ascorbic acid as carbon sources is addressed in this study using a microwave-assisted hydrothermal technique. The samples were characterized by ultraviolet, photoluminescence (PL) and transmission electron microscopy analyses, which confirmed the formation of CDs. Besides PL properties, the peroxidase-like catalytic activity of the CDs and N-CDs was also compared. Interestingly, the N-CDs exhibit higher peroxidase-like catalytic activity than pure CDs and can be used to detect H 2 O 2 . Additionally, we synthesized several N-CD ratios and evaluated their peroxidase-like catalytic activity.

Abstract:Ascorbic acid (AA) is essential for human body. Thus, its selective, sensitive, and accurate sensing and imaging are greatly important for clinical chemistry and diagnostics. Here, carbon dots (CDs) with two emissions at 520 and 668 nm under 415 nm excitation were effectively fabricated via solvothermal method using black pepper as raw materials. Formation of nonfluorescent CDs-Fe3+ complex led to the main quenching of near-infrared (NIR) emission at 668 nm. Then redox reaction between Fe3+ and AA recovered the fluorescence. Thus, the resultant CDs were developed as a NIR ratiometric nanoprobe for AA determination based on off-on mode with excellent selectivity and high sensitivity. In addition, the good photostability, low toxicity and excellent biocompatibility made as-prepared CDs a biosensing platform for intracellular AA imaging. As a novel member of dual-emission CDs, the proposed strategy has tremendous potential to broaden their applications in biological fields.

Abstract:A fluorescent carbon dots probe for the detection of aqueous nitrite was fabricated by a one-pot hydrothermal method, and the transmission electron microscope, X-ray diffractometer, UV–Vis absorption spectrometer and fluorescence spectrophotometer were used to study the property of carbon dots. The fluorescent property of carbon dots influenced by the concentration of aqueous nitrite was studied. The interaction between the electron-donating functional groups and the electron-accepting nitrous acid could account for the quenching effect on carbon dots by adding aqueous nitrite. The products of the hydrolysis of aqueous nitrite performed a stronger quenching effect at lower pH. The relationship between the relative fluorescence intensity of carbon dots and the concentration of nitrite was described by the Stern–Volmer equation ( I 0 / I − 1 = 0.046[ Q ]) with a fine linearity ( R 2 = 0.99). The carbon dots-based probe provides a convenient method for the detection of nitrite concentration.

Abstract:The BNQDs are synthesized by simple one‐step hydrothermal method, using amino acids as nitrogen source and boric acid as boron source. The BNQDs prepared by different amino acids emit strong blue fluorescence with the excitation wavelength from 280 to 400 nm. An "on‐off‐on" BNQDs fluorescent sensor is designed to detect ascorbic acid with Fe3+ as medium, extending the application in sensing. The fluorescence of BNQDs is quenched by Fe3+ on basis of the inner filter effect. With increasing of ascorbic acid, the fluorescence of BNQDs is recovered due to the oxidation‐reduction between Fe3+ and ascorbic acid. The assay is capable of recognizing ascorbic acid in the concentration range of 1–100 μM with a detection limit of 0.0833 μM. In addition, the method has the potential to determine Fe3+ and ascorbic acid in real water samples and human serum.

Abstract:A fluorescent probe for the determination of nitrite (NO2-) was fabricated by using green fluorescent nitrogen doped carbon dots (NCDs). The NCDs were synthesized via a one-pot hydrothermal carbonization of citric acid in the presence of p-phenylenediamine as the nitrogen source. The N content of the NCDs was high to 17.09% and consisted of a variety of functional groups on the NCDs surface, including sp2-hybridized CN, porphyrin C-N-C and amino N in N-(C) 3 or H-N-(C) 2 et al. N atoms were also doped within the framework of the NCDs. The almost monodisperse NCDs (average particle diameter = 3.67 nm) exhibited green photoluminescence (PL) with excitation/emission maxima of 360/505 nm. The PL of the NCDs was dependent on both excitation wavelength and solution pH. The NCDs showed a strong PL quenching response to NO2- under acidic conditions (pH = 2.5). The PL intensity of the NCDs was inversely proportional to the concentration of NO2- between 0.02 and 40 μM (R2 = 0.992), with a detection limit of 21.2 nM. The practical use of the nanoprobe for NO2- determination in food samples was also demonstrated, successfully. NCD-nitroso compounds formed because of reaction between the abundant amide groups on the surface of NCDs with the NO2-, which caused an inner filter effect and static PL quenching. Importantly, the NCDs had low cellular toxicity and were successfully used as a multicolor cellular imaging agent for Hepg2 cells.
N, P Co-Doped Carbon Dots as Multifunctional Fluorescence Nano-Sensor for Sensitive and Selective Detection of Cr(VI) and Ascorbic Acid

Abstract:It is particularly important to monitor Cr(VI) for its high toxicity. In this paper, a novel, simple, low-cost and “on-off-on” fluorescence sensor of carbon dots doped with nitrogen and phosphorus (N, P-CDs) was developed via one-step hydrothermal method for highly sensitive and good selective detection of Cr(VI) and ascorbic acid (AA). The prepared N, P-CDs exhibited the ability for detection of Cr(VI) based on the inner filter effect (IFE) and static quenching. Under optimized conditions, the fluorescence quenching efficiency of N, P-CDs showed a good linear correlation with Cr(VI) concentration ranged from 0.68 to 87.38 μmol/L (R2 = 0.9946). The limit of detection (LOD) was 0.18 μmol/L, which was acceptable compared with the maximum Cr(VI) concentration of 0.96 μmol/L in drinking water prescribed by WHO. In addition, the N, P-CDs/Cr(VI) hybrid were also used as “turn–on” fluorescent transducers for detecting AA with a wide linear region ranged from 0.02 to 933.33 μmol/L (R2 = 0.9972). Furthermore, the N, P-CDs fluorescence sensor had admissible applicability for Cr(VI) and AA detection in actual water samples with acceptable recovery rate, indicating that the fluorescence sensor had great application potential in environmental monitoring and food field.

Abstract:In this work, nitrogen-doped carbon quantum dots (N-CQDs) with high fluorescence quantum yield (46.2%) were successfully synthesized through one-pot hydrothermal carbonization approach assisted by ethylenediamine as surface passivation agent, wherein alanine acted as a single carbon source, ethylenediamine as the surface passivation agent. The obtained biocompatible N-CQDs could effectively label both the MCF-7 cell membranes and the nucleus, suggesting the potential application in biolabeling and bioimaging. Furthermore, the addition of dihydronicotinamide adenine dinucleotide (NADH) could dramatically quench the fluorescence of N-CQDs. Therefore, a novel protocol for NADH determination was established and its mechanism was discussed. This proposed fluorescence sensor displayed an enhanced performance for NADH determination with the linear range up to 80 mu M and the detection limit down to 25.1 nM. (C) 2015 Elsevier B.V. Abstract:The synthesis of water-soluble nitrogen-doped carbon dots has received great attention, due to their wide applications in oxygen reduction reaction, cell imaging, sensors, and drug delivery. Herein, nitrogen-doped, carbon-rich, highly photoluminescent carbon dots have been synthesized for the first time from ammonium citrate under hydrothermal conditions. The obtained nitrogen-doped carbon dots possess bright blue luminescence, short fluorescence lifetime, pH-sensitivity and excellent stability at a high salt concentration. They have potential to be used for pH sensors, cell imaging, solar cells, and photocatalysis.

Abstract:A nitrogen and sulfur co-doped graphene quantum dots (N/S-GQDs) and permanganate was employed to prepare a N/S-GQDs/KMnO 4 dual-signal readout colorimetric and fluorescent probe for sensitive detection of ascorbic acid (AA). In the detection system, the addition of KMnO 4 quenched significantly the fluorescence (FL) of N/S-GQDs, meanwhile, the transparent aqueous dispersion of N/S-GQDs changed into yellow–brown with new absorption peak (maximum absorption at 338 nm). Interestingly, introduction of AA into the N/S-GQDs/KMnO 4 system could recover the quenched fluorescence and result in a decrease in the absorbance (at 338 nm) of the test solution in a concentration dependent manner, and hence demonstrated the potential practical use of the sensing system for AA detection. Based on various spectroscopic studies, it has been concluded that the static as well as inner filter effect was the main quenching mechanisms in the detection system. The dual-response sensor showed linear response in the concentration range between 0.01 and 30 μM with the limit of detection (LOD) of 8.3 nM and 100 nM, respectively, for the fluorescent and colorimetric detections. In addition, the proposed detection system has been successfully applied to the detection of AA in vitamin C tablets. Moreover, the proposed detection strategy is convenient and fast, prevents complex and tedious probe preparation procedure, and capable of visual detection of AA.

Ultrathin g-C3N4 nanosheet-CoOOH nanocomposite for fluorescence imaging of ascorbic acid in living cells

Abstract:Ascorbic acid (AA), a critical cellular metabolite involved in many biochemical pathways, is an important antioxidant in human body. Therefore, it is of great significance to monitor AA in living cells. Nowadays, there are various technologies developed for the detection of AA, but few methods could sensitively and selectively detect the intracellular AA. Here, we reported a highly efficient biosensor (g-C3N4-CoOOH nanocomposite) based on ultrathin graphitic carbon nitride (g-C3N4) nanosheets and CoOOH nanoflakes, for sensitive detection and fluorescence imaging of AA in living cell. The g-C3N4 used here as fluorescence donor is a promising bioimaging nanomaterial because of their high fluorescence quantum yield, good biocompatibility and low toxicity. In addition, the CoOOH was used to be perfect fluorescence quencher. Herein, we enabled the CoOOH in situ to form a layer on the surface of g-C3N4, resulting in fluorescence quench of the g-C3N4. Upon the addition of AA, the CoOOH nanoflakes were reduced to Co2+, and the system gave a "turn on" fluorescence signal. It developed as an efficient sensing platform for AA, and the linear range was from 5 to 50 μM with a 1.6 μM detection limit. This novel biosensor, g-C3N4-CoOOH nanocomposite exhibited highly selective response toward AA relative to other biomolecules. Furthermore, this biosensor was used successfully to visualize and monitor AA in living cells. Hopefully, we believe that this biosensor would provide a low-cost and highly sensitive platform for AA detection and bioimaging. Abstract:<h3 class="a-plus-plus">Abstract</h3> Ascorbic acid (AA) detection in biological sample and food sample is critical for human health. Herein, a MnO2 nanosheet (MnO2-NS)-based ratiometric fluorescent nanosensor has been developed for high sensitive and specific detection of AA. The MnO2-NS presents peroxidase-like activity and can oxidize non-fluorescent substrate of o-phenylenediamine (OPDA) into fluorescent substrate, presenting maximum fluorescence at 568 nm (F568). If MnO2-NS is premixed with AA, the MnO2-NS is then decomposed as Mn2+ by AA, decreasing the fluorescent intensity of F568. Meantime, AA is oxidized as dehydroascorbic acid (DHAA), which can react with OPDA to generate fluorescent substrate. A new fluorescence response is found at 425 nm (F425). The dual fluorescent responses can be excited with a universal excitation wavelength, simplifying the detection procedure. With F425/F568 as readout, limit of detection for AA reaches as low as 10.0 nM. Satisfactory recoveries are found for AA detection in serum and diverse beverages. The ratiometric strategy significantly eliminates false-negative and false-positive results, providing a cost-effective, rapid, and reliable way for AA detection in real sample.
