Abstract:A near-infrared fluorescent probe (IC-V) for detecting viscosity is constructed. The probe has a large Stokes shift (170 nm) and an about 180-fold increase in fluorescence intensity at 700 nm. Abstract:Viscosity is a key factor that determines the diffusion-controlled processes in biological systems. The matrix of mitochondria contains various enzymes and other proteins with high density, and the diffusion of which are severely restricted by the cristae, making it the most crowded place in the cells. Herein, we reported a new near-infrared probe NV-1 with increased Stokes shift for monitoring viscosity changes in mitochondria. A remarkable increase of the fluorescence was observed in glycerol compared with which was observed in methanol at 744 nm. The probe was applied for measuring viscosity changes not only in mitochondria, but also in vivo (in zebra fishes and mice).

Abstract:The detection of viscosity is of great significance for medical research. Herein, we have developed a two-photon fluorescent probe CB-V for monitoring micro-viscosity changes. The fluorescence emission intensity of CB-V increased 9.6-fold from methanol to glycerol exhibiting an excellent fluorescence response. With excellent properties of CB-V, monitoring the viscosity variations has been achieved not only in living cells but also in zebra fish and mice.

Abstract:Formaldehyde (FA) and viscosity play multiple roles in human health and diseases, and viscosity has great regional differences due to the diversity of subcellular organelles. However, it is challenging to achieve dual detection of viscosity and FA in subcellular organelles. Herein, we developed a near infrared (NIR) fluorescent probe FA-Cy, which can simultaneously monitor the viscosity and FA concentration of mitochondria in living cells. Abstract:The status of living cells can be reflected by the changes of intracellular viscosity. Small molecular fluorescent probes are robust tools for measuring viscosity changes in living cells. Therefore, we constructed a near-infrared viscosity sensitive probe ZF1, which had a large Stokes shift, in methanol the Stokes reached to 148 nm and in glycerol it reached to 126 nm in viscous media, a significant enhancement of fluorescence at around 740 nm was observed. The probe ZF1 exhibited an 18.43-fold increasement of fluorescence intensity in glycerol. Moreover, in living cells, the probe ZF1 was applicable to detection of the viscosity changes.

Abstract:Monitoring the intracellular viscosity changes is crucial for better understanding diffusion-controlled cellular processes. Herein, we synthesized a novel phenyl-substituted imidazole-fused rhodamine analogue RV-1 with a long absorption wavelength at 573 nm and a large Stokes shift of about 82 nm for monitoring the changes in cellular viscosity. The new probe RV-1 showed a very strong fluorescence emission at around 655 nm, with a 48.5-fold enhancement of fluorescence intensity from methanol to 99% glycerol. Significantly, the innovative probe RV-1 was successfully applied for the detection of viscosity changes not only in living cells, but also in zebra fish and living mice.

Abstract:Viscosity and hypoxia, as microenvironment parameters, play important roles in maintaining normal biological processes and homeostasis. Therefore, simultaneous and sensitive detection of these elements with simple and effective methods could offer precise information in biology. Here, we report a two-site lysosome-targeting fluorescent probe, NVP, for monitoring viscosity and nitroreductase with dual emission channels (emission shift is 86 nm). The NVP probe has displayed highly sensitive and selective responses towards viscosity and nitroreductase, respectively. Significantly, the fluctuations of viscosity and NTR have been detected in vitro and in vivo. We expect that the dual-responsive fluorescent NVP probe will become a potential molecular tool for the exploration of deeper functions of viscosity and hypoxia.

Abstract:Carbon monoxide (CO) is an important gas transporter and is closely associated with a series of physiological and pathological processes. So far, it is clear that plenty of fluorescent probes have been prepared, but most of them still have some disadvantages and is unsuitable for imaging in vivo. Herein, a novel fluorecent probe named HPQ-BI-CO is synthesized and used to detect CO. In this probe, the benindole group is introduced to extend the wavelength. This probe possess a variety of advantages such as long emission wavelength, high sensitivity and selectivity, and rapid response capability. Moreover, the probe possesses a low cytotoxicity, which make it apply in the imaging of exogenous and endogenous CO in living 4T1 cells. Furthermore, this probe can be utilized as a promising tool for fluorescent bioimaging of CO in vivo like zebrafish.

Abstract:Cellular viscosity is a prominent micro-environmental parameter and peroxynitrite is an essential reactive oxygen special, both of which are involved in various pathological and physiological processes. When the intracellular viscosity is abnormal or the ONOO- concentration is irregular, the normal function of cells will be disturbed. Herein, we rationally designed and synthesized a novel multichannel fluorescent probe (probe 1) for multichannel imaging of viscosity and peroxynitrite. Probe 1 displayed about 108-fold enhancement as the viscosity increased from 1.005 cP to 1090 cP. Moreover, the fluorescence inten-sity at 540 nm was quickly increased after adding ONOO-. It should be noted that probe 1 has high sen-sitivity, selectivity and low cytotoxicity, which can be successfully employed for the visualization of exogenous and endogenous ONOO- and imaging viscosity changes in HeLa cells by different fluorescent signals. Furthermore, probe 1 could monitor the change of ONOO- induced by LPS (lipopolysaccharide) and IFN-gamma (interferon-gamma) in zebrafish. This result reveals that probe 1 may inspire more diagnostic and therapeutic programs for viscosity-peroxynitrite related diseases shortly. (C) 2021 Elsevier B.V. All rights reserved.

Abstract:As an important cellular microenvironmental parameter, viscosity could reflect the status of living cells. Small molecular fluorescent probes are a vital tool to measure the change of viscosity in living cells. A novel fluorescence probe ZL-1 with a large Stokes shift (in methanol it reached to 153 nm and in glycerol it reached to 125 nm) and excellent sensitivity toward viscosity was developed. The sharp enhancement of the emission intensity for the probe ZL-1 from low viscous methanol to high viscous glycerol indicated that the probe ZL-1 could respond to the viscosity variations. Moreover, the probe ZL-1 has been successfully utilized to detect of the viscosity variations in living cells.

Abstract:Viscosity is a crucial indicator of the flow state of proteins, lipids, and polysaccharides in the cell microenvironment and plays a vital role in maintaining normal cellular activities. Abnormal viscosity in any part of the cell constituents can lead to various diseases in the organism. For instance, abnormal mitochondrial viscosity can lead to diseases such as sugar in diabetes and Parkinson's disease. Therefore, real-time monitoring of changes in mitochondrial viscosity in both pathological and physiological environments is relevant. This study describes a water-soluble xylan-based near-infrared fluorescence probe that can detect changes in cellular viscosity. The designed mitochondria-targeting near-infrared fluorophores were introduced into modified xylan to form a viscosity sensing fluorescent probe (NI-XylV). The fluorescence intensity of NI-XylV at 590, 670 nm gradually increases with the increase in viscosity caused by environmental changes, enabling the sensitive detection of viscosity changes in mitochondria within living cells. NI-XylV exhibits good photostability, biocompatibility, excellent mitochondrial targeting, and broad application prospects as a biobased fluorescence probe.

Abstract:Herein, seven viscosity-sensitive probes were developed via simple structural modification of dicyanoisophorone (DCO)-derived dyes. Among them, DCO-5 significantly enhances (180-fold) the response signal in highly viscous aqueous media while showing insensitivity to polarity changes or pH variations, and enables the successful detection of viscosity changes in nystatin-treated HepG2 cells, PC 12 cells and zebrafish.

Abstract:As one of the most significant parameters in cellular microenvironment, viscosity levels could be used to determine the metabolic process of bioactive substances within cells. Abnormal viscosity levels are closely associated with a series of diseases. Therefore, the design and synthesis of fluorescent probes that can monitor changes of intracellular viscosity in real-time is of great significance for the study of disease development process. Here, a new viscosity-recognized NIR fluorescence probe W1 based on quinoline-malonitrile is synthesized, and it is not susceptible to interference substances. Besides, AIE probe W1 shows fast response, excellent photostability, low cytotoxicity, good linear relationship between fluorescence intensity value and viscosity. Based on the above advantages, probe W1 is used to image the change of viscosity level in the cell model induced by nystatin.

Abstract:Viscosity, an essential parameter of the cellular microenvironment, has the ability to indicate the condition of living cells. It is closely linked to numerous diseases like Alzheimer's disease, diabetes, and cardiovascular disorders. Therefore, it is necessary to design tools to effectively monitor viscosity changes, which could provide promising avenues for therapeutic interventions in these diseases. Herein, we report a novel mitochondria-targeting fluorescent probe GX-VS which was suitable for the detection of viscosity changes in vivo and in vitro. The probe GX-VS had many advantages such as long emission wavelength (650 nm), large Stokes shift (105 nm), significant fluorescence enhancement (59-fold), high sensitivity, good biocompatibility and so on. Biological experiments showed that the probe could target mitochondria and detect viscosity alterations in HeLa cells. Moreover, it has been successfully utilized to monitor viscosity changes induced by lipopolysaccharides (LPS) in inflammatory zebrafishes and living mice, which further underscored the capacity of GX-VS to explore fluctuations in viscosity within living organisms.

Abstract:Rational design of specific ratiometric viscosity probes with small molecular weight is a challenge in practical biotechnology applications. Herein two novel water-soluble, small-molecular ratiometric probes, bearing N-methyl benzothiazolium moiety (DSF and DBF), are designed for two-photon fluorescent imaging as a functional of local viscosity. The dye DSF, a light-up fluorescent probe, is sensitive to local viscosity and selectively stains nuclear DNA, which can be used to inspect asynchronous cells under confocal microscopy. While the dye DBF as a molecular rotor displays strong fluorescence enhancement in viscous media or binding to RNA. It exhibits dual absorption and emission as well, and only the red emission is markedly sensitive to viscosity changes, providing a ratiometric response and selectively imaging nucleolic and cytosolic RNA. Interestingly it is shown, for the first time, that the intracellular targeting and localization (DNA and RNA) of the two dyes are entirely realized simply by modifying the substituent attached to the benzothiazolium.

Abstract:Developing near-infrared (NIR) fluorescence probes for detection of intracellular viscosity is still sufficiently challenging. In this work, three kinds of D-A-D type naphthyl and 2,1,3‑benzoxadiazol hybrid NIR dyes functionalized with amino (NY1), N‑methylamino (NY2) and N,N‑dimethylamino (NY3) groups for intracellular micro-viscosity detection were designed and synthesized. All the probes exhibited very weak NIR emission in low viscosity environment and obvious fluorescence enhancement with the increased viscosity. Different substituent groups had a high impact on the photophysical properties and response sensitive of the probes to viscosity. The structure-property relationships were systematic investigated. The results showed that stronger electron-donating ability and larger steric effect of N,N‑dimethylamino led to a narrower energy gap and more sensitive to viscosity environment. Therefore, NY3 exhibited higher signal noise ratio for viscosity detection and was successfully applied for imaging the changes of intracellular micro-viscosity. This work provides an efficient way to design powerful NIR fluorescence probes for viscosity detection.

Abstract:A new two-photon fluorescent probe (MCN) for viscosity imaging was developed based on a 6-substituted quinoline framework. MCN showed an excellent "off-on" fluorescence response (ca. 90-fold enhancement) with viscosity increasing in the glycerol-water viscosity system. MCN showed great sensitivity to viscosity (R-2 = 0.98, x = 0.65), which gave rise to cell imaging for micro-viscosity or real-time cell imaging during apoptosis with low cytotoxicity under two-photon excitation (lambda(ex) = 800 nm). Fluorescence lifetime imaging (FLIM) of living HeLa cells stained with MCN revealed that the intracellular average viscosity value was 73.45 +/- 21.55 cP in cytosol. Imaging in living tissue slices indicated that MCN can work in deep tissue (similar to 130 mu M) under two-photon excitation. Moreover, MCN also showed the capacity for in vivo imaging viscosity in zebrafish with obvious fluorescence emission.

Abstract:Rational design of water-soluble probes for mitochondrial viscosity in practical biological applications remains a challenge. Herein, we described a novel hydro soluble benzothiazole salt derivative MitoSN, which exhibits specifically response and singular sensitivity to the mitochondria viscosity in living Hela cells. MitoSN displays an excellent fluorescence enhancement (ca. 35-fold) with the increase of the viscosity in the water-glycerol system. Moreover, confocal microscopy indicates that MitoSN is sensitive to the local viscosity and selectively stains mitochondria, the body of zebrafish as well. Importantly, MitoSN is capable to identify the viscosity difference of mitochondria in normal and nystatin treated Hela cells. The work provides a useful tool to monitor the changes of viscosity in the mitochondrial microenvironment.
