Abstract:A near-infrared fluorescent probe (IC-V) for detecting viscosity is constructed. The probe has a large Stokes shift (170 nm) and an about 180-fold increase in fluorescence intensity at 700 nm. In addition, IC-V can not only distinguish cancer cells from normal cells, but also monitor viscosity in normal mice and tumor-bearing mice.

Abstract:Lipid droplets (LDs) represent highly dynamic organelles found in almost all organisms and play a pivotal role in regulating the balance of intracellular lipid homeostasis. Viscosity directly affects the metabolism of lipid droplets, and the abnormal change of viscosity will lead to a series of related diseases. Therefore, developing a tool that can monitor changes in intracellular lipid droplet viscosity in real-time is of great significance for disease prevention, diagnosis, and treatment. In this study, a viscosity-sensitive fluorescent probe CA-LD based on an intramolecular twisted charge transfer (TICT) mechanism was developed. The CA-LD probe exhibited strong fluorescence emission at approximately 640 nm and showed a 161-fold increase in fluorescence intensity in 99% glycerol compared to water. Additionally, the probe CA-LD demonstrated the advantages of good anti-interference, excellent biocompatibility and good photostability. Significantly, because of the probe's sensitivity to viscosity, the probe CA-LD successfully distinguished normal cells from cancer cells and has been able to visualize changes in lipid droplets' viscosity induced by oleic acid. In addition, the probe CA-LD was effectively employed to monitor changes in the viscosity of cells and zebrafish induced by different drugs. Therefore, the probe CA-LD can provide an effective means for the detection of lipid droplet viscosity changes and is of great value for physiological diagnosis, pathological analysis and medical research on related diseases.

Abstract:As one of the most significant parameters in cellular microenvironment, viscosity levels could be used to determine the metabolic process of bioactive substances within cells. Abnormal viscosity levels are closely associated with a series of diseases. Therefore, the design and synthesis of fluorescent probes that can monitor changes of intracellular viscosity in real-time is of great significance for the study of disease development process. Here, a new viscosity-recognized NIR fluorescence probe W1 based on quinoline-malonitrile is synthesized, and it is not susceptible to interference substances. Besides, AIE probe W1 shows fast response, excellent photostability, low cytotoxicity, good linear relationship between fluorescence intensity value and viscosity. Based on the above advantages, probe W1 is used to image the change of viscosity level in the cell model induced by nystatin.

Abstract:Viscosity is a crucial indicator of the flow state of proteins, lipids, and polysaccharides in the cell microenvironment and plays a vital role in maintaining normal cellular activities. Abnormal viscosity in any part of the cell constituents can lead to various diseases in the organism. For instance, abnormal mitochondrial viscosity can lead to diseases such as sugar in diabetes and Parkinson's disease. Therefore, real-time monitoring of changes in mitochondrial viscosity in both pathological and physiological environments is relevant. This study describes a water-soluble xylan-based near-infrared fluorescence probe that can detect changes in cellular viscosity. The designed mitochondria-targeting near-infrared fluorophores were introduced into modified xylan to form a viscosity sensing fluorescent probe (NI-XylV). The fluorescence intensity of NI-XylV at 590, 670 nm gradually increases with the increase in viscosity caused by environmental changes, enabling the sensitive detection of viscosity changes in mitochondria within living cells. NI-XylV exhibits good photostability, biocompatibility, excellent mitochondrial targeting, and broad application prospects as a biobased fluorescence probe.

Abstract:Viscosity and hypoxia, as microenvironment parameters, play important roles in maintaining normal biological processes and homeostasis. Therefore, simultaneous and sensitive detection of these elements with simple and effective methods could offer precise information in biology. Here, we report a two-site lysosome-targeting fluorescent probe, NVP, for monitoring viscosity and nitroreductase with dual emission channels (emission shift is 86 nm). The NVP probe has displayed highly sensitive and selective responses towards viscosity and nitroreductase, respectively. Significantly, the fluctuations of viscosity and NTR have been detected in vitro and in vivo. We expect that the dual-responsive fluorescent NVP probe will become a potential molecular tool for the exploration of deeper functions of viscosity and hypoxia.

Abstract:Mitochondria are the powerhouses in cells, providing the energy needed for cellular activities. However, the abnormalities in the mitochondrial microenvironment (e.g., the increased viscosity) can lead to mitochondrial dysfunctions and diseases. Herein, we develop a series of near-infrared (NIR) fluorescence probes for the detection of viscosity. After screening, probe CQ-4 is selected since it shows a great fluorescence enhancement (89-fold) in the NIR window. Its specific response to viscosity is not influenced by pH, polarity and biological species. Under stimulation with monensin or nystatin, CQ-4 can measure the cellular viscosity changes with good biocompatibility. In addition, we can observe an increase of viscosity during starvation. CQ-4 is applied to distinguish cancer cells from normal cells based on the viscosity differences. Furthermore, the probe has been successfully applied to image viscosity in inflamed and tumor-bearing mice in vivo. Therefore, CQ-4 may contribute to the future study about viscosity in the physiological and pathological processes.

Abstract:Blood viscosity changes and blood clots are high-impact diseases, but the pathogenic mechanisms and detection methods are still limited. Due to the complexity of the cellular microenvironment, viscosity is a key factor in regulating the behavior of mitochondria and lysosomes in cells. Conventional fluorescence probes are highly restrictive for complex viscosity detection in live animals. Therefore, we developed two near-infrared fluorescence probes, QL1 and QL2, with dual responses to the pH and viscosity. Notably, QL2 has two maximum fluorescence emissions at 680 and 750 nm, when excitation by 580 and 700 nm, respectively. QL2 exhibited both a pH and viscosity switchable fluorescence response. The two emission peaks exhibited a reverse change trend: the fluorescence at 680 nm decreased by 90%, and the fluorescence at 750 nm increased by about 5-fold with pH from 2 to 10. Meanwhile, both emission peaks show remarkable fluorescence enhancement toward viscosity change, with 185 and 32 times enhancement, respectively. The sensing mechanism and spectral changes are confirmed by DFT calculations. QL2 was further used for viscosity imaging in live cells, zebrafish, and live animals. Most importantly, QL2 is able to successfully track changes in blood clots in live mice and organs, thus enabling the study of blood clots in cerebral strokes and the underlying pathological mechanisms.

Abstract:So many biological functional disorders and diseases, such as atherosclerosis, hypertension, diabetes, Alzheimer's disease, as well as cell malignancy are closely related with the intracellular viscosity. A safe and effective intracellular viscosity detecting method is desired by the biomedical community. Recently, a novel near-infrared fluorescent probe NI-VIS with a twisting intramolecular charge transfer mechanism was developed. The capability of this probe to visualize the viscosity variation in cirrhotic liver tissues and map the micro viscosity in vivo were testified using an experiment. In this work, the twisting intramolecular charge transfer mechanism and fluorescent properties of the probe NI-VIS were studied in detail under quantum mechanical method. The low energy barrier among the different conformations of the probe indicated the occurrence of twisting intramolecular charge transfer due to the rotation of the aryl group in the probe molecule while within the low viscosity environment. The electronic structure analysis on different probe conformations revealed the electron transfer process of the probe under optical excitation. All these theoretical results could provide insights into understand in greater depth the principles and build highly effective fluorescent probe to monitor the viscosity in biological samples.

Abstract:Results are reviewed concerning some effects, at a units's characteristic frequency, of a short-term conditioning stimulus on the responses to perstimulatory and poststimulatory test tones. A phenomenological equation is developed from the poststimulatory results and shown to be consistent with the perstimulatory results. According to the results and equation, the response to a test tone equals the unconditioned or unadapted response minus the decrement produced by adaptation to the conditioning tone. Furthermore, the decrement is proportional to the driven response to the conditioning tone and does not depend on sound intensity per se. The equation has a simple interpretation in terms of two processes in cascade--a static saturating nonlinearity followed by additive adaptation. Results are presented to show that this functional model is sufficient to account for the "physiological masking" produced by wide-band backgrounds. According to this interpretation, a sufficiently intense background produces saturation. Consequently, a superimposed test tone cause no change in response. In addition, when the onset of the background precedes the onset of the test tone, the total firing rate is reduced by adaptation. Evidence is reviewed concerning the possible correspondence between the variables in the model and intracellular events in the auditory periphery.

Abstract:A new two-photon fluorescent probe (MCN) for viscosity imaging was developed based on a 6-substituted quinoline framework. MCN showed an excellent "off-on" fluorescence response (ca. 90-fold enhancement) with viscosity increasing in the glycerol-water viscosity system. MCN showed great sensitivity to viscosity (R-2 = 0.98, x = 0.65), which gave rise to cell imaging for micro-viscosity or real-time cell imaging during apoptosis with low cytotoxicity under two-photon excitation (lambda(ex) = 800 nm). Fluorescence lifetime imaging (FLIM) of living HeLa cells stained with MCN revealed that the intracellular average viscosity value was 73.45 +/- 21.55 cP in cytosol. Imaging in living tissue slices indicated that MCN can work in deep tissue (similar to 130 mu M) under two-photon excitation. Moreover, MCN also showed the capacity for in vivo imaging viscosity in zebrafish with obvious fluorescence emission.

Abstract:Mitochondria, as an important organelle in cells, play an important role in the production of adenosine triphosphate (ATP) and cell signal transduction, as well as control of the cell cycle and growth. Abnormal viscosity is a pivotal factor for indicating mitochondrial dysfunction. In addition, abnormal changes in mitochondrial viscosity have been confirmed to be closely related to many diseases and malfunctions, such as cell malignancy, atherosclerosis, diabetes, and Alzheimer's disease. Therefore, the development of mitochondrial viscosity response probe and its application in mitochondrial viscosity measurement are considered as a new tool to understand diseases. So far, most of the fluorescent probes used to detect mitochondrial viscosity have emission wavelengths in the visible region, making them less penetrable to tissue. In this work, we report a novel near-infrared fluorescent probe (MN-V) for the detection of mitochondrial viscosity in live cells, which exhibited near infrared emission (795 nm) and high sensitivity towards viscosity changes. The fluorescence intensity of the probe at 795 nm has a 37 times increase from pure PBS solution to glycerol. There was a linear relationship between the fluorescence intensity and the media viscosity. The probe MN-V was applicable to detection of the viscosity changes in living cells and zebrafish. Most importantly, with the probe we found that during cancer cells ferroptosis process was accompanied by an increase in mitochondrial viscosity.

Abstract:Carbon monoxide (CO) is an important gas transporter and is closely associated with a series of physiological and pathological processes. So far, it is clear that plenty of fluorescent probes have been prepared, but most of them still have some disadvantages and is unsuitable for imaging in vivo. Herein, a novel fluorecent probe named HPQ-BI-CO is synthesized and used to detect CO. In this probe, the benindole group is introduced to extend the wavelength. This probe possess a variety of advantages such as long emission wavelength, high sensitivity and selectivity, and rapid response capability. Moreover, the probe possesses a low cytotoxicity, which make it apply in the imaging of exogenous and endogenous CO in living 4T1 cells. Furthermore, this probe can be utilized as a promising tool for fluorescent bioimaging of CO in vivo like zebrafish.

Abstract:Viscosity and pH are two crucial microenvironment parameters, and their abnormal fluctuations at the cellular level can act as key indicators of various diseases diagnosis. Considering that the traditional viscosity/pH meters cannot work in the cellular environment, developing some viscosity/pH-sensitive fluorescent probes is urgently demanded. Herein, a novel dual-functional camphor-based fluorescent probe PN-SP for viscosity and pH detection was constructed based on spiropyran open/closed-ring switching strategy. PN-SP exhibited an intense green emission in response to alkaline pH while a bright red fluorescence was observed in high-viscosity media, which could avoid some problems caused by the use of multiple probes such as cross-talk spectrum and different localizations. The sensing performances of PN-SP were also demonstrated for monitoring viscosity and pH fluctuations in living HeLa cells by means of different emisson channels. Therefore, we believed that this work will provide guidance for designing some dual-functional fluorescent probes.

Abstract:As a basic parameter of the intracellular microenvironment, viscosity is closely related to the development of cancer. Thus, it is necessary to utilize a sensitive tool to visualize the viscosity in tumor cells and mice, which is helpful for the diagnosis of cancer. Herein, a novel dual-modal probe (IX-V) that has a near-infrared fluorescence (NIRF) and photoacoustic (PA) response to viscosity is synthesized. In low viscosity media, the probe has no fluorescence. With the increase of viscosity, the fluorescence is produced in the near-infrared region due to the inhibition of the TICT process. At the same time, the probe shows different photoacoustic (PA) signals in different viscosity media. Most notably, the viscosity in tumor cells has been imaged successfully by the application of IX-V, and the probe can effectively distinguish cancer cells from normal cells co-cultured in one dish by the difference of fluorescence intensity. In addition, the probe has been used for dual-modal imaging (NIRF and PA) of viscosity in tumor mice, which provides a tool for exploring the relationship between viscosity and diseases. That is to say, IX-V can achieve complementary imaging effects and has great application prospects in the tumor diagnosis.

Abstract:A versatile probe with mitochondria targeting, PN, for detection of viscosity alteration in HIRI mouse model was fabricated. PN had high sensitivity and selectivity, and with a large Stokes shift (∼190 nm) for viscosity. PN is expected to be used for early diagnosis and treatment assessment of HIRI disease, and also provides diagnostic basis for other related diseases.

Abstract:The abnormal microenvironment parameter, viscosity, is closely connected with various diffusion processes, signal transduction, molecule interactions, and various diseases. It is greatly significant to design viscosity-dependent near-infrared (NIR) small molecule fluorescence probes for visualizing biological processes or diagnosing diseases. Herein, through the stepwise modulating structure of the silicon–rhodamine fluorophore (SR), we report three viscosity probes with allyl or methyl group as rotors, named SR-T-Al, SR-S-Al, and SR-T-Me. Among them, SR-T-Al demonstrates better viscosity responsibility from 1.0 to 1410.4 cP of viscosity. Therefore, the probe of SR-T-Al is successfully applied to sensitively monitor lysosome microscopic viscosity changes of living cells induced by oxygen stress. What’s more, based on its advantages in NIR emission (669 nm) and large Stokes shift (201 nm), we also use it to image variations of viscosity in an acute hepatitis mouse induced by carbon tetrachloride. Both time and concentration-dependent induction models display the great ability of SR-T-Al to detect viscosity alteration. All the experimental results indicated that this allyl-rotor-based NIR viscosity probe could provide a general platform to monitor abnormal physiological processes and diseases relating to viscosity.

Abstract:Background: Cirrhosis represents the terminal stage of liver disease progression and timely intervention in a diseased liver can enhance the likelihood of recovery. Viscosity, a crucial parameter of the cellular microenvironment, is intricately linked to the advancement of cirrhosis. However, viscosity monitoring still faces significant challenges in achieving non-invasive and rapid early diagnosis of cirrhosis. Near-infrared (NIR) fluorescence imaging has the advantages of high sensitivity, non-destructive detection, and ignoring background fluorescence interference, plays an important role in diagnosing and treating various biological diseases. Hence, monitoring cellular viscosity changes with NIR fluorescence probe holds great significance in the early diagnosis of cirrhosis. Results: In this study, the NIR fluorescence probe based on the intramolecular charge transfer (TICT) mechanism was developed for imaging applications in mouse model of liver cirrhosis. A molecular rotor-type viscosity-responsive probe was synthesized by linking dioxanthracene groups via carbon-carbon double bonds. The probe demonstrated remarkable sensitivity, high selectivity and photostability, with its responsiveness to viscosity largely unaffected by factors such as polarity, pH, and interfering ions. The probe could effectively detect various drug-induced changes in cellular viscosity, enabling the differentiation between normal cells and cancerous cells. Furthermore, the enhanced tissue penetration capabilities of probe facilitated its successful application in mouse model of liver cirrhosis, allowing for the assessment of liver disease severity based on fluorescence intensity and providing a powerful tool for early diagnosis of cirrhosis. Significance: A NIR viscosity-sensitive fluorescent probe was specifically designed to effectively monitor alterations in cellular and organ viscosity, which could advance the understanding of the biological characteristics of cancer and provide theoretical support for the early diagnosis of cirrhosis. Overall, this probe held immense potential in monitoring viscosity-related conditions, expanding the range of biomedical tools available.

Abstract:We have constructed a novel fluorescent probe PPF‐1 for detecting viscosity in vitro and in vivo. Lipid droplet, an intracellular lipid reservoir, is vital for energy metabolism and signal transmission in cells. The viscosity directly affects the metabolism of lipid droplets, and the abnormal viscosity is associated with the occurrence and development of various diseases. Therefore, it is indispensable to develop techniques that can detect viscosity changes in intracellular lipid droplets. Based on twisted intramolecular charge transfer (TICT) mechanism, a novel small‐molecule lipid droplet‐targeted viscosity fluorescence probe PPF‐1 was designed. The probe was easy to synthesize, it had a large Stokes shift, stable optical properties, and low bio‐toxicity. Compared to being in methanol solution, the fluorescence intensity of PPF‐1 in glycerol solution was increased 26.7‐fold, and PPF‐1 showed excellent ability to target lipid droplets. Thus, the probe PPF‐1 could provide an effective means of detecting viscosity changes of lipid droplets and was of great value for physiological diagnosis of related diseases, pathological analysis, and medical research.
