Abstract:Background: Cirrhosis represents the terminal stage of liver disease progression and timely intervention in a diseased liver can enhance the likelihood of recovery. Viscosity, a crucial parameter of the cellular microenvironment, is intricately linked to the advancement of cirrhosis. However, viscosity monitoring still faces significant challenges in achieving non-invasive and rapid early diagnosis of cirrhosis. Near-infrared (NIR) fluorescence imaging has the advantages of high sensitivity, non-destructive detection, and ignoring background fluorescence interference, plays an important role in diagnosing and treating various biological diseases. Hence, monitoring cellular viscosity changes with NIR fluorescence probe holds great significance in the early diagnosis of cirrhosis. Results: In this study, the NIR fluorescence probe based on the intramolecular charge transfer (TICT) mechanism was developed for imaging applications in mouse model of liver cirrhosis. A molecular rotor-type viscosity-responsive probe was synthesized by linking dioxanthracene groups via carbon-carbon double bonds. The probe demonstrated remarkable sensitivity, high selectivity and photostability, with its responsiveness to viscosity largely unaffected by factors such as polarity, pH, and interfering ions. The probe could effectively detect various drug-induced changes in cellular viscosity, enabling the differentiation between normal cells and cancerous cells. Furthermore, the enhanced tissue penetration capabilities of probe facilitated its successful application in mouse model of liver cirrhosis, allowing for the assessment of liver disease severity based on fluorescence intensity and providing a powerful tool for early diagnosis of cirrhosis. Significance: A NIR viscosity-sensitive fluorescent probe was specifically designed to effectively monitor alterations in cellular and organ viscosity, which could advance the understanding of the biological characteristics of cancer and provide theoretical support for the early diagnosis of cirrhosis. Overall, this probe held immense potential in monitoring viscosity-related conditions, expanding the range of biomedical tools available.

Abstract:Fluorescent labeling technology has been prevailing in the field of biotechnology, but in the more complex pathology, such as diabetes, tracking lipid droplets (LDs) viscosity remains a vacancy. To fill this gap, a new fluorescent probe (ABI-V), was herein synthesized for detecting the viscosity change of LDs in complicated biological systems. This probe displayed a larger stokes shift (195 nm), near-infrared emission and high sensitivity and selectivity to viscosity, exhibiting a 95-fold fluorescence enhancement from methanol to glycerol. In light of its distinguishable optical signals toward viscosity, the blood viscosity of diabetic mice was measured and a stronger fluorescence was obtained in the blood of diabetic mice with 2.5 times higher than normal mice. Importantly, with the assistance of ABI-V, the LDs viscosity changes in diabetic liver injury was successfully monitored at the organ and tissue levels and the results indicate that the viscosity of LDs in the liver increased under diabetic pathological conditions. Moreover, the LDs viscosity in metformin-treated diabetic liver injury was also detected accompanied by decreased fluorescence intensity comparing with the pure diabetic model, suggesting that the proposed probe ABI-V can achieve tracking the efficacy of diabetes treatment by monitoring the viscosity changes. This work provides a rewarding perspective to elucidate the role of LDs viscosity in the pathology of diabetes.

Abstract:As important organs of the human body, the liver and kidneys ensure the stability of the internal environment of the body and allow the body to carry out normal metabolism. The lack of effective methods for early diagnosis of liver and kidneys diseases is an important reason for delaying the treatment of kidney diseases. Therefore, the development of a simple, non-invasive and effective test is essential for the early diagnosis of liver and kidneys disease. Herein, a new viscosity fluorescent probe, Mito-ND, is rationally designed and synthesized in order to solve the practical problem of realizing the diagnosis of liver and kidneys diseases. The constructed fluorescent probe Mito-ND has the advantages of near-infrared emission, mitochondrial localization, high chemical and photo-stable properties, and sensitive viscosity detection performance, etc. Using this fluorescent probe, besides in vitro cell viscosity fluorescence imaging, the viscosity fluorescence imaging of mouse liver and kidney injury are also achieved for the first time. Mito-ND provides more accurate tools for the non-invasive diagnosis of viscosity-related diseases such as liver injury and kidney injury. Therefore, Mito-ND provides more accurate detection techniques for the early diagnosis of liver and kidney diseases such as liver injury and kidney injury.

Abstract:As the primary organ for drug metabolism and detoxification, the liver is susceptible to damage and seriously impaired function. In situ diagnosing and real-time monitoring of liver damage are thus of great significance but remain limited owing to the lack of reliable in vivo visualization protocols with minimal invasion. Herein, we reported for the first time an aggregation-induced emission (AIE) probe, namely DPXBI, emitting light in the second near-infrared window (NIR-II) for early diagnosis liver injury. DPXBI featured by strong intramolecular rotations, excellent aqueous solubility and robust chemical stability, is powerfully sensitive to viscosity alteration affording rapid response and high selectivity, through NIR-II fluorescence intensity changes. The prominent viscosity-responsive performance enables DPXBI to accurately monitor both drug-induced liver injury (DILI) and hepatic ischemia-reperfusion injury (HIRI) with excellent image contrast to the background. By using the presented strategy, the detection of liver injury in mouse model can be achieved at least several hours earlier than typical clinical assays. Moreover, DPXBI is able to dynamically track the liver improvement process in vivo in the case of DILI when the hepatotoxicity is alleviated by using hepatoprotective medication. All these results demonstrate that DPXBI is a promising probe for investigating viscosity-associated pathological and physiological processes.

Abstract:This study introduces a novel near-infrared fluorescent probe named VSP, designed to effectively and sensitively monitor viscosity changes both internally and externally. Through an enhanced conjugation structure of the fluorescent molecule and the incorporation of hydrophilic groups, the VSP probe showcases remarkable sensitivity, selectivity, and stability in its ability to detect viscosity alterations. Notably, the linear correlation between its fluorescence intensity and glycerol content presents the possibility of quantitative viscosity detection, holding significant promise for applications in biomedical research and clinical diagnostics. Moreover, its biocompatibility supports its use for monitoring viscosity within cellular environments. An exceptional attribute is its ability to accurately distinguish between hyperlipidemic model mice and normal mice in animal imaging as well as serum samples, underscoring its potential for early-stage diagnosis of elevated lipid levels and risk assessment for cardiovascular diseases. VSP's potential was demonstrated in cell imaging and differentiation between high-fat and normal mice based on blood serum viscosity. This promising tool holds significant potential for biomedical research and clinical applications in monitoring and analyzing changes in biological systems.

Abstract:Elucidating the intrinsic correlation between non-alcoholic fatty liver (NAFL) and lysosomal viscosity is a challenging work owing to the disadvantages of state-of-the-art technologies for NAFL identification. Herein, we constructed a red emission imaging fluorophore L -TPQ based on D-π-A (donor–π–bridge-acceptor) structure for identifying NAFL tissues by tracking the change in lysosomal viscosity. Specifically, L -TPQ is ultrasensitive to viscosity with a remarkable enhancement in fluorescence intensity (about 910 fold) and fluorescence quantum yield (about 221 fold) through a twisted intra-molecular charge transfer (TICT) mechanism. In addition, L -TPQ shows a single selectivity toward viscosity with an excellent pH stability and insignificant interference from environmental polarity or biomolecules. Moreover, probe L -TPQ is lysosome-specific, which is capable of monitoring the lysosome movement, lysosomal viscosity change, and mitophagy. Importantly, by means of this probe, we have successfully observed an increased viscosity in the pathological model of NAFL mice. Given the comprehensive property of probe L -TPQ , it is expected to serve as a convenient and effective tool for better understanding the pathophysiology of NAFL and the early diagnosis of this disease.

Abstract:As a basic parameter of the intracellular microenvironment, viscosity is closely related to the development of cancer. Thus, it is necessary to utilize a sensitive tool to visualize the viscosity in tumor cells and mice, which is helpful for the diagnosis of cancer. Herein, a novel dual-modal probe (IX-V) that has a near-infrared fluorescence (NIRF) and photoacoustic (PA) response to viscosity is synthesized. In low viscosity media, the probe has no fluorescence. With the increase of viscosity, the fluorescence is produced in the near-infrared region due to the inhibition of the TICT process. At the same time, the probe shows different photoacoustic (PA) signals in different viscosity media. Most notably, the viscosity in tumor cells has been imaged successfully by the application of IX-V, and the probe can effectively distinguish cancer cells from normal cells co-cultured in one dish by the difference of fluorescence intensity. In addition, the probe has been used for dual-modal imaging (NIRF and PA) of viscosity in tumor mice, which provides a tool for exploring the relationship between viscosity and diseases. That is to say, IX-V can achieve complementary imaging effects and has great application prospects in the tumor diagnosis.

Abstract:Mitochondria are the powerhouses in cells, providing the energy needed for cellular activities. However, the abnormalities in the mitochondrial microenvironment (e.g., the increased viscosity) can lead to mitochondrial dysfunctions and diseases. Herein, we develop a series of near-infrared (NIR) fluorescence probes for the detection of viscosity. After screening, probe CQ-4 is selected since it shows a great fluorescence enhancement (89-fold) in the NIR window. Its specific response to viscosity is not influenced by pH, polarity and biological species. Under stimulation with monensin or nystatin, CQ-4 can measure the cellular viscosity changes with good biocompatibility. In addition, we can observe an increase of viscosity during starvation. CQ-4 is applied to distinguish cancer cells from normal cells based on the viscosity differences. Furthermore, the probe has been successfully applied to image viscosity in inflamed and tumor-bearing mice in vivo. Therefore, CQ-4 may contribute to the future study about viscosity in the physiological and pathological processes.

Abstract:Exploring the connection between blood viscosity and liver in it have great significance for the detection and treatment of hyperlipidemic. Therefore, we designed a new near-infrared viscosity sensitive fluorescent probe Vis-LW based on the twisted intramolecular charge transfer mechanism. The probe exhibited a strong dependence on viscosity as well as good anti-interference performance. With the aid of Vis-LW, we successfully detected the viscosity changes in chloroquine induced cells and realized the diagnosis of diseases in hyperlipidemic mice by utilizing blood viscosity as a diagnostic indicator for hyperlipidemia. Not only that, the increased liver viscosity of hyperlipidemic mice was found for the first time, the reduced blood and liver viscosity after treatment with lipid-lowering drug fenofibrate was also tracked. This work provides a new method for the diagnosis of hyperlipidemia and offered a new idea on the research and treatment of hyperlipidemia in the future.

Abstract:Liver fibrosis is a major stage in the development of liver disease, and it is important to investigate its pathogenesis for early intervention or even reversal. Recent studies have found that intestinal disease can aggravate liver fibrosis through the role of the "gut-liver axis". Hypoxia is considered to be a typical characteristic of many diseases including ulcerative colitis and liver fibrosis. However, there is no fluorescent probe for hypoxia detection to investigate the "gut-liver axis". Herein we design and synthesize a turn-on fluorescent probe termed Cy-AP, which displays high sensitivity and selectivity to hypoxia given by sodium dithionite (Na2S2O4) in vitro with near-infrared (NIR) emission (725 nm). The possible response mechanism of Cy-AP toward hypoxia is given and proved though HPLC, MS, and theory calculation. Moreover, on the basis of low cell cytotoxicity, the probe is used in visualizing hypoxia in four cell lines (HepG2, HCT116, HeLa, and MCF-7 cells) by fluorescence imaging, flow cytometry, and 3D imaging. Furthermore, due to its NIR emission, Cy-AP can monitor the hypoxia condition in vivo such as in liver fibrosis mice and ulcerative colitis mice models. In particular, the probe can validate the existence and mechanism of the "gut-liver axis" in vivo by monitoring hypoxia. To the best of our knowledge, this is the first work to give a strategy for studying the "gut-liver axis" by a NIR hypoxia-sensitive fluorescent probe, which will provide some powerful support for delaying the progression of liver fibrosis and thus promoting the treatment of liver disease.

Abstract:As a pivotal parameter of microenvironment, viscosity is closely related to many diseases and dysfunction in vivo. Lipid droplets, as organelles that store and metabolize lipids, are involved in various physiological processes related to metabolism in cells. Diabetes is one of the metabolic diseases that may lead to dysfunction of organs, which is associated with abnormal viscosity changes. It is of great significance for the study of related diseases to clarify the relationship between viscosity variations in vivo and monitor the changes of lipid droplets. Here, a near-infrared fluorescent probe DPX ((E)− 6-(dimethylamino)− 4-(4-(diphenylamino)benzylidene)− 9-methyl-1,2,3,4-tetrahydroxanthylium) with AIE characteristics was designed, which shows an "off-on" fluorescence response to viscosity based on twisting intramolecular charge transfer (TICT) mechanism. The probe DPX features excellent mitochondria and lipid droplets targeting, and sensitively monitor accumulation of LDs induced by oleic acid and mitochondrial viscosity variations in HeLa cells. More importantly, DPX has been successfully applied to visualize viscosity changes of diabetic liver injury in vivo, which may be benefit for development of diabetes related clinical research.

Abstract:So many biological functional disorders and diseases, such as atherosclerosis, hypertension, diabetes, Alzheimer's disease, as well as cell malignancy are closely related with the intracellular viscosity. A safe and effective intracellular viscosity detecting method is desired by the biomedical community. Recently, a novel near-infrared fluorescent probe NI-VIS with a twisting intramolecular charge transfer mechanism was developed. The capability of this probe to visualize the viscosity variation in cirrhotic liver tissues and map the micro viscosity in vivo were testified using an experiment. In this work, the twisting intramolecular charge transfer mechanism and fluorescent properties of the probe NI-VIS were studied in detail under quantum mechanical method. The low energy barrier among the different conformations of the probe indicated the occurrence of twisting intramolecular charge transfer due to the rotation of the aryl group in the probe molecule while within the low viscosity environment. The electronic structure analysis on different probe conformations revealed the electron transfer process of the probe under optical excitation. All these theoretical results could provide insights into understand in greater depth the principles and build highly effective fluorescent probe to monitor the viscosity in biological samples.

Abstract:Mitochondria, as an important organelle in cells, play an important role in the production of adenosine triphosphate (ATP) and cell signal transduction, as well as control of the cell cycle and growth. Abnormal viscosity is a pivotal factor for indicating mitochondrial dysfunction. In addition, abnormal changes in mitochondrial viscosity have been confirmed to be closely related to many diseases and malfunctions, such as cell malignancy, atherosclerosis, diabetes, and Alzheimer's disease. Therefore, the development of mitochondrial viscosity response probe and its application in mitochondrial viscosity measurement are considered as a new tool to understand diseases. So far, most of the fluorescent probes used to detect mitochondrial viscosity have emission wavelengths in the visible region, making them less penetrable to tissue. In this work, we report a novel near-infrared fluorescent probe (MN-V) for the detection of mitochondrial viscosity in live cells, which exhibited near infrared emission (795 nm) and high sensitivity towards viscosity changes. The fluorescence intensity of the probe at 795 nm has a 37 times increase from pure PBS solution to glycerol. There was a linear relationship between the fluorescence intensity and the media viscosity. The probe MN-V was applicable to detection of the viscosity changes in living cells and zebrafish. Most importantly, with the probe we found that during cancer cells ferroptosis process was accompanied by an increase in mitochondrial viscosity.

Abstract:Bacteriophages have been induced from strains in the Mycobacterium avium, Mycobacterium intracellulare, Mycobacterium scrofulaceum serocomplex by exposure of cultures to UV light or treatment with mitomycin C. One-sixth of the strains examined, representing all but one of the 31 authenticated serotypes, were found to possess phages lytic for a Mycobacterium smegmatis indicator strain. Four single-plaque isolated phages, TM4, TM9, TM10 and TM20, were purified and shown to have a similar morphology on electron micrographs. They had an isometric head of diameter 50-58 nm and a flexible non-contractile tail about 170 nm in length with a terminal bulb. All had an identical buoyant density in CsCl of 1.521 g cm-3 and extreme sensitivity to chloroform. The induced phages differed in host range and possessed the unique ability to lyse other members of the serocomplex. Interest in these phages centres on a possible role in mediating genetic interrelationships between members of the serocomplex.

Abstract:Viscosity is a crucial indicator of the flow state of proteins, lipids, and polysaccharides in the cell microenvironment and plays a vital role in maintaining normal cellular activities. Abnormal viscosity in any part of the cell constituents can lead to various diseases in the organism. For instance, abnormal mitochondrial viscosity can lead to diseases such as sugar in diabetes and Parkinson's disease. Therefore, real-time monitoring of changes in mitochondrial viscosity in both pathological and physiological environments is relevant. This study describes a water-soluble xylan-based near-infrared fluorescence probe that can detect changes in cellular viscosity. The designed mitochondria-targeting near-infrared fluorophores were introduced into modified xylan to form a viscosity sensing fluorescent probe (NI-XylV). The fluorescence intensity of NI-XylV at 590, 670 nm gradually increases with the increase in viscosity caused by environmental changes, enabling the sensitive detection of viscosity changes in mitochondria within living cells. NI-XylV exhibits good photostability, biocompatibility, excellent mitochondrial targeting, and broad application prospects as a biobased fluorescence probe.

Abstract:Based on OR logic gate, we proposed a smart near-infrared (NIR) fluorescent probe, named VPCPP, for simultaneously monitoring local microviscosity, micropolarity, and carboxylesterases (CEs) in living cells through blue and red channels. This proposed probe was capable of distinguishing cancer cells from normal cells and had good potential for identifying living liver cell lines. Furthermore, the fluctuations of the three analytes of interest in different cell status was successfully explored. Particularly, facilitated with high-content analysis (HCA) and VPCPP, a simple and efficient high-throughput screening (HTS) platform was first constructed for screening antitumor drugs and studying their effect on the analytes. For the first time, we found that sorafenib-induced ferroptosis led to an increase in the microviscosity and up-regulation of CEs at the same time. Additionally, the procedure that aristolochic acid (AA) induced the overexpression of CEs was verified. Besides, VPCPP was utilized for imaging the variations of the two microenvironment parameters and CEs in the inflammation model. Near-Infrared Fluorescent Probe Reveals Elevated Mitochondrial Viscosity during Acute Alcoholic Liver Injury

Abstract:Acute alcoholic liver injury (AALI) has become an important cause of liver disease worldwide, and there is an urgent need to develop noninvasive and sensitive methods to detect and evaluate AALI. We report herein three novel but readily available mitochondrial targeting fluorescence probes (ICR, ICJ, and ICQ) for AALI detection. These probes contain different electron-donating groups, among which ICQ exhibits NIR fluorescence (740 nm), a large Stokes shift (110 nm), and a sensitive response to viscosity (73-fold enhancement in fluorescence from water to glycerol), making it suitable for in vivo imaging. ICQ also exhibits an excellent ability to image mitochondrial viscosity changes in cells. More importantly, ICQ can target the liver selectively and image the viscosity changes in the liver noninvasively. Through establishing an AALI mouse model, ICQ was successfully applied to the in situ imaging changes in liver viscosity during the AALI process. The results showed a significant increase in liver viscosity in AALI mice, indicating that viscosity can serve as a marker for AALI, and ICQ is a promising noninvasive and sensitive tool for detecting and evaluating AALI.
Bivariate Tracking of NIR Phototherapeutic Probe That Illuminates the Deterioration Process of NAFLD-HCC

Abstract:Non-alcoholic fatty liver disease (NAFLD) has evolved to become a major cause of hepatocellular carcinoma (HCC). Visualization of NAFLD-HCC deterioration process imaging is essential to understand the underlying pathophysiological processes. However, currently relevant probes with short emission wavelengths, univariate and the inability to achieve theranostics functionality have encountered obstacles in further evaluating the NAFLD-HCC process. Here, we present a carboxylesterase (CE)-activated NIR fluorescent probe (BODJ) which has lipid droplets (LDs)-targeting ability and emits at a wavelength of 858 nm with a fluorescence quantum yield of 19.06%. CE-activated BODJ was used as a visual tool to successfully visualize both NAFLD deterioration processes and HCC in situ based on changes in the average number of LDs and the associated fluorescence intensity fluctuations. Imaging results showed that changes associated with CE and LDs in the modelled cells varied during the transition from nonalcoholic fatty liver to nonalcoholic steatohepatitis and later progression to HCC, highlighting the close association between bivariate and disease. We also demonstrate that BODJ has photodynamic (PDT) and photothermal therapy (PTT) capabilities, allowing image-guided dual phototherapy to damage HCC in situ. This NIR probe, which takes advantage of bivariate to track the deterioration process that illuminates NAFLD-HCC and has dual phototherapy capabilities, provides new ideas for the design of probes related to the diagnosis and treatment of hepatic metabolic diseases.

Abstract:Elucidating the intrinsic relationship between disease and mitochondrial viscosity is great significance for understanding the diagnosis and treatment of fatty liver, inflammation and cancer. However, the development of a single mitochondrial viscosity fluorescent probe capable of visualizing multiple disease models and achieving photodynamic cancer therapy function is severely deficient and remains a great challenge. Herein we intelligently designed a mitochondria-targeted and viscosity-sensitive near-infrared (NIR) fluorescent probe POTA-OH, containing a p -hydroxydiphenylamino substituted oxanthracene moiety as the strong electron donor (D), a pyridinium cationic unit as the electron acceptor (A) and the mitochondria-targeted group. With increasing the viscosity level, POTA-OH displayed a significant "turn-on" fluorescence behavior at 720 nm, and an excellent linear relationship with viscosity from 0.89 cP to 945 cP. Meanwhile, POTA-OH could achieve effective reactive oxygen species production upon a 635 nm red laser irradiation. Using POTA-OH, we have demonstrated the visualization of fatty liver tissues, inflammation living mice, tumor living mice and even clinic cancer patient tissues samples via mitochondrial viscosity imaging. Furthermore, we revealed that POTA-OH has the potential of simultaneous PDT treatment in vitro and immediate evaluation of its therapeutic effect. Importantly, high-efficient PDT of POTA-OH has also successfully achieved for in vivo PDT antitumor, making it a promising integrated reagent for cancer diagnosis, treatment and even evaluation of PDT efficacy.
