Abstract:We have constructed a novel fluorescent probe PPF‐1 for detecting viscosity in vitro and in vivo. Lipid droplet, an intracellular lipid reservoir, is vital for energy metabolism and signal transmission in cells. The viscosity directly affects the metabolism of lipid droplets, and the abnormal viscosity is associated with the occurrence and development of various diseases. Therefore, it is indispensable to develop techniques that can detect viscosity changes in intracellular lipid droplets. Based on twisted intramolecular charge transfer (TICT) mechanism, a novel small‐molecule lipid droplet‐targeted viscosity fluorescence probe PPF‐1 was designed. The probe was easy to synthesize, it had a large Stokes shift, stable optical properties, and low bio‐toxicity. Compared to being in methanol solution, the fluorescence intensity of PPF‐1 in glycerol solution was increased 26.7‐fold, and PPF‐1 showed excellent ability to target lipid droplets. Thus, the probe PPF‐1 could provide an effective means of detecting viscosity changes of lipid droplets and was of great value for physiological diagnosis of related diseases, pathological analysis, and medical research.

Abstract:Lipid droplets (LDs) represent highly dynamic organelles found in almost all organisms and play a pivotal role in regulating the balance of intracellular lipid homeostasis. Viscosity directly affects the metabolism of lipid droplets, and the abnormal change of viscosity will lead to a series of related diseases. Therefore, developing a tool that can monitor changes in intracellular lipid droplet viscosity in real-time is of great significance for disease prevention, diagnosis, and treatment. In this study, a viscosity-sensitive fluorescent probe CA-LD based on an intramolecular twisted charge transfer (TICT) mechanism was developed. The CA-LD probe exhibited strong fluorescence emission at approximately 640 nm and showed a 161-fold increase in fluorescence intensity in 99% glycerol compared to water. Additionally, the probe CA-LD demonstrated the advantages of good anti-interference, excellent biocompatibility and good photostability. Significantly, because of the probe's sensitivity to viscosity, the probe CA-LD successfully distinguished normal cells from cancer cells and has been able to visualize changes in lipid droplets' viscosity induced by oleic acid. In addition, the probe CA-LD was effectively employed to monitor changes in the viscosity of cells and zebrafish induced by different drugs. Therefore, the probe CA-LD can provide an effective means for the detection of lipid droplet viscosity changes and is of great value for physiological diagnosis, pathological analysis and medical research on related diseases.

Abstract:Lipid droplets (LDs) are simple intracellular storage sites for neutral lipids and exhibit important impact on many physiological processes. For example, the changes in the polar microenvironment inside LDs could affect physiological processes, such as lipid metabolism and storage, protein degradation, signal transduction, and enzyme catalysis. Herein, a new fluorescent chemo-sensor (Couoxo-LD) was formulated by our molecular design strategy. The probe could be applied to effectively label intracellular lipid droplets. Intriguingly, Couoxo-LD demonstrated positive sensitivity to both polarity and viscosity, which might be attributed to its D-π-A structure and the twisted rotational behavior of the carbon–carbon double bond (TICT). Additionally, Couoxo-LD was successfully implemented in cellular imaging due to its excellent selectivity, pH stability, and low biotoxicity. In HeLa cells, the co-localization curve between Couoxo-LD and commercial lipid droplet dyes overlapped at 0.93. The results indicated that the probe could selectively sense LDs in HeLa cells. Meanwhile, Couoxo-LD can be applied for in vivo imaging of zebrafish.

Abstract:A near-infrared fluorescent probe (IC-V) for detecting viscosity is constructed. The probe has a large Stokes shift (170 nm) and an about 180-fold increase in fluorescence intensity at 700 nm. In addition, IC-V can not only distinguish cancer cells from normal cells, but also monitor viscosity in normal mice and tumor-bearing mice.

Abstract:A new two-photon fluorescent probe (MCN) for viscosity imaging was developed based on a 6-substituted quinoline framework. MCN showed an excellent "off-on" fluorescence response (ca. 90-fold enhancement) with viscosity increasing in the glycerol-water viscosity system. MCN showed great sensitivity to viscosity (R-2 = 0.98, x = 0.65), which gave rise to cell imaging for micro-viscosity or real-time cell imaging during apoptosis with low cytotoxicity under two-photon excitation (lambda(ex) = 800 nm). Fluorescence lifetime imaging (FLIM) of living HeLa cells stained with MCN revealed that the intracellular average viscosity value was 73.45 +/- 21.55 cP in cytosol. Imaging in living tissue slices indicated that MCN can work in deep tissue (similar to 130 mu M) under two-photon excitation. Moreover, MCN also showed the capacity for in vivo imaging viscosity in zebrafish with obvious fluorescence emission.

Abstract:The disruption of lipid droplet function is associated with the pathogenesis of various diseases. Clarifying the response behavior of lipid droplets to the microenvironment at the cellular level is of great significance. Plant lipids not only exist in phospholipids in cell membranes, but also in aromatic essential oils. Monitoring the level of lipid droplets in plant cells using fluorescent probes provides a simple method for screening lipid-rich varieties. We synthesized a polarity-viscosity responsive coumarin fluorescent probe, Cou-CN, which achieved sensitive detection of polarity and viscosity in dilute solution environments by constructing this simple probe with ICT and TICT properties and verifying it using Gaussian computational simulation. Cou-CN exhibited good lipid droplet illumination effects in HepG2 cells with a correlation coefficient of 0.92 compared to the commercial lipid droplet dye BODIPY. Additionally, co-staining the probe with the lipophilic commercial dye Nile Red in tobacco root stem seedling cells resulted in a high correlation coefficient of 0.9.

Abstract:Lipid droplets (LDs) are involved in a variety of complex life activities in cells, which are closely related to human diseases including obesity, diabetes and cancer. LDs are generally considered as a kind of organelle with extremely low water content and high viscosity. Herein, two novel probes (E)-2-(3-(2-(7-(diethylamino)-2-oxo-2H-chromen-3-yl)vinyl)-5,5-dimethylcyclohex-2-en-1-ylidene)malononitrile (LDP-1) and (E)-2-(3-(4-(diethylamino)styryl)-5,5-dimethylcyclohex-2-en-1-ylidene)malononitrile (LDP-2) were synthesized, which exhibited large Stokes shifts of 4758 cm−1 and 3986 cm−1 respectively. The two probes emitted strongly in hydrophobic and high viscosity environments. Moreover, bio-probes LDP-1 and LDP-2 exhibited low biological toxicity, and good selectivity. The two probes were also fit for selectively monitoring LDs distribution in living HeLa cells.

Abstract:The viscosity of lysosome is reported to be a key indicator of lysosomal functionality. However, the existing mechanical methods of viscosity measurement can hardly be applied at the cellular or subcellular level. Herein, a BODIPY-based two-photon fluorescent probe was presented for monitoring lysosomal viscosity with high spatial and temporal resolution. By installing two morpholine moieties to the fluorophore as target and rotational groups, the TICT effect between the two morpholine rings and the main fluorophore scaffold endowed the probe with excellent viscosity sensitivity. Moreover, Lyso-B succeeded in showing the impact of dexamethasone on lysosomal viscosity in real time.

Abstract:The irregular cellular viscosity is associated with disease and malfunction at the cellular level, developing simple but effective fluorescence probe for detection of intracellular viscosity is of great significance. Herein, we report a benzothiazole-based fluorescent probe HBTD-V for sensitive response to viscosity change in biological systems. HBTD-V displays weak emission in low-viscosity media but strong fluorescence in high-viscosity media. Linear fitting of the viscosity titration results provided a good linear relationship between the fluorescence intensity (logI583) and viscosity (logη) within the viscosity range of 13.8–221.5 cp, indicating the capability of the probe for quantitative viscosity detection. Probe HBTD-V also holds some advantages such as excellent photostability, good anti-interference ability, and good mitochondrial targeting ability. Moreover, probe HBTD-V has been successfully employed to image the viscosity variations in living MCF-7 cells, mice lung organ and tissues, as well as in living mice body. The results demonstrate that HBTD-V can serve as a sensitive fluorescence probe for investigating the vital role of viscosity changes in complex biological systems.

Abstract:Abnormal viscosity is closely related to the occurrence of many diseases, such as cancer. Therefore, real-time detection of changes in viscosity in living cells is of great importance. Fluorescent molecular rotors play a critical role in detecting changes in cellular viscosity. Developing red emission viscosity probes with large Stokes shifts and high sensitivity and specificity remains an urgent and important topic. Herein, a novel viscosity-sensitive fluorescent probe (TCF-VIS1) with a large stokes shift and red emission was prepared based on the 2-dicyanomethylene-3-cyano-4,5,5-trimethyl-2,5-dihydrofuran (TCF) skeleton. Due to intramolecular rotation, the probe itself does not fluorescence at low viscosity. With the increase in viscosity, the rotation of TCF-VIS1 is limited, and its fluorescence is obviously enhanced. The probe has the advantages of simple preparation, large Stokes shift, good sensitivity and selectivity, and low cytotoxicity, which make it successfully used for viscosity detection in living cells. Moreover, TCF-VIS1 showed its potential for cancer diagnosis at the cell level and in tumor-bearing mice by detecting viscosity. Therefore, the probe is expected to enrich strategies for the detection of viscosity in biological systems and offer a potential tool for cancer diagnosis.

Abstract:As a pivotal parameter of microenvironment, viscosity is closely related to many diseases and dysfunction in vivo. Lipid droplets, as organelles that store and metabolize lipids, are involved in various physiological processes related to metabolism in cells. Diabetes is one of the metabolic diseases that may lead to dysfunction of organs, which is associated with abnormal viscosity changes. It is of great significance for the study of related diseases to clarify the relationship between viscosity variations in vivo and monitor the changes of lipid droplets. Here, a near-infrared fluorescent probe DPX ((E)− 6-(dimethylamino)− 4-(4-(diphenylamino)benzylidene)− 9-methyl-1,2,3,4-tetrahydroxanthylium) with AIE characteristics was designed, which shows an "off-on" fluorescence response to viscosity based on twisting intramolecular charge transfer (TICT) mechanism. The probe DPX features excellent mitochondria and lipid droplets targeting, and sensitively monitor accumulation of LDs induced by oleic acid and mitochondrial viscosity variations in HeLa cells. More importantly, DPX has been successfully applied to visualize viscosity changes of diabetic liver injury in vivo, which may be benefit for development of diabetes related clinical research.

Abstract:Viscosity is an important microenvironmental indicator that plays an important role in the process of information transmission in various regions. Herein, two coumarin-based viscosity-sensitive fluorescent probes (CHB, CHN) were synthesized and the photophysical properties of the two probes were studied. The fluorescence quantum yields of CHB and CHN in glycerol can be as high as 25.2% and 18.3% respectively. The two probes can linearly detect viscosity in the viscosity logarithm range of 0.83-2.07, which is not interfered with pH, metal ions, anions and biomolecules. Fluorescent confocal cell experiments show CHB and CHN have good targeting ability to mitochondrion, lysosome, Endoplasmic reticulum and Golgi apparatus, and can be used to detect viscosity in mitochondrion/lysosome.

Abstract:Viscosity and hypoxia, as microenvironment parameters, play important roles in maintaining normal biological processes and homeostasis. Therefore, simultaneous and sensitive detection of these elements with simple and effective methods could offer precise information in biology. Here, we report a two-site lysosome-targeting fluorescent probe, NVP, for monitoring viscosity and nitroreductase with dual emission channels (emission shift is 86 nm). The NVP probe has displayed highly sensitive and selective responses towards viscosity and nitroreductase, respectively. Significantly, the fluctuations of viscosity and NTR have been detected in vitro and in vivo. We expect that the dual-responsive fluorescent NVP probe will become a potential molecular tool for the exploration of deeper functions of viscosity and hypoxia.

Abstract:Pathophysiological micro-environments such as viscosity, polarity and hypoxia are tightly correlated with regulating diffusion, transportation, and intermolecular interactions in living cells. However, the interplay and crosstalk among these key factors have not been clearly explored yet due to the lack of practical tools. In this work, based on logic-gate, we developed a versatile fluorescent probe (VPHPP) for simultaneously detecting viscosity, polarity and Nitroreductases (NTRs) in living cells. This probe was able to distinguish cancer cells from normal cells with signals in blue channel (lambda em = 540 nm) for NTRs and red channel (lambda em > 747 nm) for viscosity/polarity, which seemed more accurate (or more information at least) to define the cell types than testing only one factor. Meanwhile, the dynamic changes of viscosity, polarity and NTRs were simultaneously observed under hypoxic conditions in living cells for the first time. Furthermore, the detection system could image the simultaneous down-regulation of NTRs and the elevation of viscosity level (decreasing polarity degree) induced by dicoumarin in living cells under a confocal microscopy. Last but not least, VPHPP could achieve the imaging in tumor-bearing mouse model. In brief, VPHPP could be used as a promising visualizing agent in image-guided surgery.

Abstract:Viscosity and pH are two crucial microenvironment parameters, and their abnormal fluctuations at the cellular level can act as key indicators of various diseases diagnosis. Considering that the traditional viscosity/pH meters cannot work in the cellular environment, developing some viscosity/pH-sensitive fluorescent probes is urgently demanded. Herein, a novel dual-functional camphor-based fluorescent probe PN-SP for viscosity and pH detection was constructed based on spiropyran open/closed-ring switching strategy. PN-SP exhibited an intense green emission in response to alkaline pH while a bright red fluorescence was observed in high-viscosity media, which could avoid some problems caused by the use of multiple probes such as cross-talk spectrum and different localizations. The sensing performances of PN-SP were also demonstrated for monitoring viscosity and pH fluctuations in living HeLa cells by means of different emisson channels. Therefore, we believed that this work will provide guidance for designing some dual-functional fluorescent probes.

Abstract:Viscosity is a crucial factor influencing diffusion-controlled processes in biological systems. An abnormal increase or decrease in the viscosity level is closely related to various human diseases, such as malignant tumors, coronary heart disease, and cerebral thrombosis. We report a "turn-on" chitosan-based viscosity fluorescent probe (CS-V) for cell-level viscosity measurement. Under viscous conditions, the fluorescence intensity of the probe at a wavelength of 530 nm increased by approximately 56 times and was sensitive to changes in the viscosity level. The probe CS-V was constructed using chitosan (CS) with low toxicity and high biological activity, combined with mitochondrial targeting groups and luminescent groups. As the first chitosan-based probe for viscosity detection, it not only exhibited excellent selectivity and rapid response but also had lower toxicity, better biocompatibility and excellent photostability than small-molecule probes, and can be used for long time fluorescence imaging.

Abstract:Viscous microenvironments are ubiquitous in living organisms. An oxidative stress attack could cause the fluctuation of lymph or blood viscosity, which is closely related to many common diseases. However, the dual-function detection of reactive oxygen species and viscosity is still ineffective due to design defects. Herein, by rational linking a propeller-shaped triphenylamine rotator and a rigid planar anthracene, we report the first dual-ratiometric-response probe for ClO- and viscosity detection. The probe itself shows weak fluorescence background in low-viscous solution or at non-aggregate state, which can largely switch on its blue (414 nm) and yellow fluorescence (600 nm) respectively in response to ClO- and viscosity. Their sensing mechanisms were elucidated through the interruption of FRET and TICT processes for ClO- and viscosity detection, respectively. In addition to its mitochondria targeting ability, TPP-AN was employed for mitochondrial viscosity and ClO- imaging in HeLa cells, providing a potentially useful probe for clinical assays.

Abstract:Viscosity in biological systems is a critical factor for various physiological process, including signal transduction and metabolisms of substance and energy. Abnormal viscosity has been proven as a key feature of many diseases, thereby real-time monitoring of viscosities in cells and in vivo is of great significance for the diagnosis and therapy of related diseases. Up to date, it is still challenging to monitor viscosity cross-platform from organelles to cells to animals with a single probe. Here, we report a benzothiazolium-xanthene probe with rotatable bonds that switch on the optical signals in high viscosity environment. The enhancements of absorption, fluorescence intensity and lifetime signals allow to dynamically monitoring the viscosity change in mitochondria and cells, while near infrared absorption and emission facilitate imaging the viscosity with both fluorescence and photoacoustic imaging in animals. The cross-platform strategy is capable of monitoring the microenvironment with multifunctional imaging across various levels.

Abstract:Mitochondria are the powerhouses in cells, providing the energy needed for cellular activities. However, the abnormalities in the mitochondrial microenvironment (e.g., the increased viscosity) can lead to mitochondrial dysfunctions and diseases. Herein, we develop a series of near-infrared (NIR) fluorescence probes for the detection of viscosity. After screening, probe CQ-4 is selected since it shows a great fluorescence enhancement (89-fold) in the NIR window. Its specific response to viscosity is not influenced by pH, polarity and biological species. Under stimulation with monensin or nystatin, CQ-4 can measure the cellular viscosity changes with good biocompatibility. In addition, we can observe an increase of viscosity during starvation. CQ-4 is applied to distinguish cancer cells from normal cells based on the viscosity differences. Furthermore, the probe has been successfully applied to image viscosity in inflamed and tumor-bearing mice in vivo. Therefore, CQ-4 may contribute to the future study about viscosity in the physiological and pathological processes.
