Abstract:Mitochondrial viscosity serves as a critical indicator for assessing mitochondrial functionality and offers valuable insights into cellular homeostasis. Continuous, real-time monitoring of mitochondrial viscosity is indispensable for understanding and diagnosing diseases associated with these dynamic changes. In this study, we introduce a novel mitochondrial viscosity-responsive probe named "JL-JC" which is designed by using a molecular strategy, with a classic "D-π-A" molecular structure. Leveraging the distinctive twisted intramolecular charge transfer (TICT) properties of the probe, JL-JC exhibits exceptional sensitivity and a high signal-to-noise ratio, enabling precise detection of viscosity variations within its microenvironment while remaining unaffected by other factors. Upon rapid cellular uptake, JL-JC can efficiently evaluate the mitochondrial viscosity changes under diverse physiological and pathological conditions. Notably, this probe also enables viscosity imaging in zebrafish, offering insights into mitochondrial states in vivo. Our findings present JL-JC as a promising tool and potential diagnostic platform for mitochondria-related diseases.

Abstract:Cells of most eukaryotic species contain mitochondria, which play a role in physiological processes such as cellular senescence, metabolism, and autophagy. Viscosity is considered a key marker for many illnesses and is involved in several crucial physiological processes. Cyanide (CN-) can target cytochrome-c oxidase, disrupting the mitochondrial electron transport chain and causing cell death through asphyxiation. In this study, a fluorescent probe named HL-1, which targets mitochondria and measures viscosity and CN- levels, was designed and synthesized. HL-1 is viscosity-sensitive, with a linear correlation coefficient of up to 0.992. In addition, HL-1 was found to change color substantially during a nucleophilic addition reaction with CN-, which has a low detection limit of 47 nM. HL-1 not only detects viscosity and exogenous CN- in SKOV-3 cells and zebrafish but also monitors viscosity changes during mitochondrial autophagy in real time. Furthermore, HL-1 has been used successfully to monitor changes in mitochondrial membrane potential during apoptosis. Endogenous CN- in plant samples was quantified. HL-1 provides new ideas for studying viscosity and CN-.

Abstract:Hypochlorite (ClO−) and viscosity both affect the physiological state of mitochondria, and their abnormal levels are closely related to many common diseases. Therefore, it is vitally important to develop mitochondria-targeting fluorescent probes for the dual sensing of ClO− and viscosity. Herein, we have explored a new fluorescent probe, XTAP–Bn, which responds sensitively to ClO− and viscosity with off–on fluorescence changes at 558 and 765 nm, respectively. Because the emission wavelength gap is more than 200 nm, XTAP–Bn can effectively eliminate the signal crosstalk during the simultaneous detection of ClO− and viscosity. In addition, XTAP–Bn has several advantages, including high selectivity, rapid response, good water solubility, low cytotoxicity, and excellent mitochondrial-targeting ability. More importantly, probe XTAP–Bn is successfully employed to monitor the dynamic change in ClO− and viscosity levels in the mitochondria of living cells and zebrafish. This study not only provides a reliable tool for identifying mitochondrial dysfunction but also offers a potential approach for the early diagnosis of mitochondrial-related diseases.

Abstract:The level of sulfur dioxide (SO2) and viscosity in mitochondria play vital roles in various of physiological and pathological processes. Abnormalities in mitochondrial SO2 and viscosity are closely associated with numerous biological diseases. It is of great significance to develop novel fluorescence probe for simultaneous detection of SO2 and viscosity within mitochondria. Herein, we have developed a water-soluble, mitochondrial-targeted and near-infrared fluorescent probe, CMBT, for the simultaneous detection of SO2 and viscosity. The probe CMBT incorporates benzothiazolium salt as a mitochondrial targeting moiety and 7-diethylaminocoumarin as a rotor for viscosity detection, respectively. Based on the prompt reaction between nucleophilic HSO3−/SO32− and the backbone of benzothiazolium salt derivative, probe CMBT displayed high sensitivity and selectivity toward SO2 with a limit of detection as low as 0.17 μM. As viscosity increased, the twisted intramolecular charge transfer (TICT) process was restricted, resulting in fluorescence emission enhancement at 690 nm. Moreover, probe CMBT demonstrated exceptional mitochondrial targeting ability and was successfully employed to image variation of SO2 and viscosity in living cells and mice. The work highlights the great potential of the probe as a convenient tool for revealing the relationship of SO2 and viscosity in biological systems.

Abstract:Viscosity, at the subcellular level, plays a crucial role as a physicochemical factor affecting microenvironment homeostasis. Abnormal changes in mitochondrial viscosity often lead to various diseases in the organism. Based on the twisted intramolecular charge transfer mechanism, four hemicyanine dye fluorescent probes (HT-SA, HT-SA-S, HT-Bzh, and HT-NA) were designed and synthesized for viscosity response. The single bond between the nitrogen-containing heterocycle and the carbon-carbon double in the structure of the probe bond served as the viscosity response site. Finally, the probe HT-Bzh was screened as the optimal mitochondrial viscosity probe according to its responsiveness, targeting, and interference resistance. The fluorescence intensity of the probe HT-Bzh increased 22-fold when the viscosity was increased from 13.75 to 811.2 cP. In summary, all four viscosity probes we have developed can be used in different applications depending on the external environment, providing a valuable reference for the design of potential tools to address viscosity monitoring in biological systems.

Abstract:Viscosity is one of the key factors of the mitochondrial microenvironment, and any abnormal change in the viscosity is not conducive to the normal functioning of the mitochondria. Therefore, it is extremely crucial to monitor the mitochondrial viscosity for disease diagnoses. In this study, a novel fluorescent probe YPE based on twisted intramolecular charge transfer (TICT) was synthesized via simple synthetic steps. The probe has the characteristics of excellent targeting of mitochondria, good adaptation to a wide pH range, outstanding photostability, and good biocompatibility. Furthermore, we used the probe to investigate the changes in the mitochondrial viscosity under various drug stimulations and distinguish between normal and cancer cells via fluorescence imaging. It provides a powerful fluorescence tool for understanding the abnormal changes in the mitochondria.

Abstract:Mitochondria are important subcellular organelles involved in many cellular activities. Therefore, it is very important to monitor the concentration of various substances in mitochondria. In this work, we constructed a dual-response mitochondria-targeted fluorescent probe HBTN for the detection of viscosity and HOCl in vitro and in vivo. HBTN not only has a long emission wavelength with an emission peak of 680 nm, but also has a large Stokes shift of 278 nm. The fluorescence intensity of probe HBTN at 680 nm has a good linear relationship with solution viscosity, which can be used for quantitative detection of viscosity. In addition, the probe HBTN enables ratiometric detection of HOCl with a low detection limit (DL = 24.5 nM) and rapid response (<20 min). More importantly, the probe HBTN has been successfully applied to the detection of viscosity and exogenous/endogenous HOCl in the mitochondria of MCF-7 cells. Furthermore, the probe HBTN has been implemented in imaging viscosity and HOCl in zebrafish. HBTN is expected to be a practical tool for monitoring viscosity and HOCl in vivo.

Abstract:At the organelle level, pathogenesis due to abnormal concentrations of cysteine (Cys) is of great significance for the early diagnosis and treatment of related diseases. Generally speaking, organelle localization requires the participation of specific target groups, which increases the difficulty of synthesis. Herein, through simple synthesis, a novel biflavone derivative (BFD) that exhibits excited-state intramolecular proton transfer (ESIPT) was obtained and successfully located in mitochondria without target groups. The probe BFD can distinguish Cys from Hcy and GSH with a rapid response (< 5 s) and showed visual detection for Cys with a large Stokes shift (about 260 nm). Because of its nanomorphology in solution and surface functional groups, the probe BFD can enter the cell smoothly and achieve mitochondrial localization. Owing to its excellent optical performance, the probe BFD was successfully applied to the imaging of endogenous Cys in HeLa cells and zebrafish.

Abstract:Mitochondria, as an important organelle in cells, play an important role in the production of adenosine triphosphate (ATP) and cell signal transduction, as well as control of the cell cycle and growth. Abnormal viscosity is a pivotal factor for indicating mitochondrial dysfunction. In addition, abnormal changes in mitochondrial viscosity have been confirmed to be closely related to many diseases and malfunctions, such as cell malignancy, atherosclerosis, diabetes, and Alzheimer's disease. Therefore, the development of mitochondrial viscosity response probe and its application in mitochondrial viscosity measurement are considered as a new tool to understand diseases. So far, most of the fluorescent probes used to detect mitochondrial viscosity have emission wavelengths in the visible region, making them less penetrable to tissue. In this work, we report a novel near-infrared fluorescent probe (MN-V) for the detection of mitochondrial viscosity in live cells, which exhibited near infrared emission (795 nm) and high sensitivity towards viscosity changes. The fluorescence intensity of the probe at 795 nm has a 37 times increase from pure PBS solution to glycerol. There was a linear relationship between the fluorescence intensity and the media viscosity. The probe MN-V was applicable to detection of the viscosity changes in living cells and zebrafish. Most importantly, with the probe we found that during cancer cells ferroptosis process was accompanied by an increase in mitochondrial viscosity.

Abstract:Mitochondria are the powerhouses in cells, providing the energy needed for cellular activities. However, the abnormalities in the mitochondrial microenvironment (e.g., the increased viscosity) can lead to mitochondrial dysfunctions and diseases. Herein, we develop a series of near-infrared (NIR) fluorescence probes for the detection of viscosity. After screening, probe CQ-4 is selected since it shows a great fluorescence enhancement (89-fold) in the NIR window. Its specific response to viscosity is not influenced by pH, polarity and biological species. Under stimulation with monensin or nystatin, CQ-4 can measure the cellular viscosity changes with good biocompatibility. In addition, we can observe an increase of viscosity during starvation. CQ-4 is applied to distinguish cancer cells from normal cells based on the viscosity differences. Furthermore, the probe has been successfully applied to image viscosity in inflamed and tumor-bearing mice in vivo. Therefore, CQ-4 may contribute to the future study about viscosity in the physiological and pathological processes.

Abstract:Mitochondria are crucial for physiological activities, and alterations in mitochondrial function will lead to diverse human diseases. However, the tracking and long-term visualizing mitochondria are still deficient, which limits the research related to mitochondria. Inspired by the exceptional interfacial architecture of mitochondria, we proposed the interface-targeting model for designing fluorescent probes that could track and long-term visualize mitochondria with high selectivity in living cells, tissues, and zebrafish. And (E)-4-(2-(7-(diethylamino)-2-oxo-2H-chromen-3-yl)vinyl)-1-dodecylpyridin-1-ium iodide (CVP) with a cationic pyridinium unit and a C12-chain for targeting mitochondria was synthesized in this work. Thanks to the C12-chain, CVP has excellent permeability in tissues and zebrafish. In comparison to traditional mitochondrial probes, CVP stained mitochondria in short time and long-term track mitochondria without being affected by mitochondrial membrane potential because of the distal long alkyl chain, which enhanced the binding affinity of CVP to mitochondria. The phospholipid-biomimetic structure of CVP endowed it with high selectivity to mitochondria, which decreased the background noise. So CVP could stain mitochondria in tissues and zebrafish with high fidelity through no washing procedures. Particularly, four kinds of mitochondria were visualized by CVP in tissues. In addition, CVP can be applied to track the mitophagy behavior in situ and real-time. All of them demonstrated that the interface-targeting model is an effective strategy for designing mitochondrial probes with high selectivity.

Abstract:Carbon monoxide (CO) is an important gas transporter and is closely associated with a series of physiological and pathological processes. So far, it is clear that plenty of fluorescent probes have been prepared, but most of them still have some disadvantages and is unsuitable for imaging in vivo. Herein, a novel fluorecent probe named HPQ-BI-CO is synthesized and used to detect CO. In this probe, the benindole group is introduced to extend the wavelength. This probe possess a variety of advantages such as long emission wavelength, high sensitivity and selectivity, and rapid response capability. Moreover, the probe possesses a low cytotoxicity, which make it apply in the imaging of exogenous and endogenous CO in living 4T1 cells. Furthermore, this probe can be utilized as a promising tool for fluorescent bioimaging of CO in vivo like zebrafish.

Abstract:Carbon monoxide (CO) not only causes damage to life and health as an environmental pollutant, but also undertakes many physiological functions in organisms. In particular, developing means that can be used for the determination of CO in organelles will provide insight into the vital role it plays. Studies have shown that mitochondrial respiration is closely related to CO concentrations, so it is critical to develop tools for CO detection in mitochondria. Here, we use a rhodamine derivative that can target mitochondria as fluorophores to construct a mitochondrial-labeled CO fluorescence probe (Rh-CO) with high sensitivity (detection limit: 9.4 nM), excellent water-solubility, and long emission (λem = 630 nm). Prominently, the probe has outstanding mitochondria-targeting capabilities. Moreover, we used transient glucose deprivation (TGD) and heme to stimulate endogenous CO production in living cells and zebrafish, respectively, and the probe exhibited excellent imaging capabilities. All in all, we expect this probe to contribute to a deeper understanding of the role played by CO in mitochondria.

Abstract:Mitochondria, the main source of energy of cells, play a significant role in aerobic respiration process. Some stimulants can result in changes of mitochondrial microenvironments such as viscosity, pH and polarity. Abnormal changes of mitochondrial viscosity have been shown to relate to pathological activities and diseases. Therefore, it is critical to focus our attention on mitochondrial viscosity under different conditions. A novel organic water-soluble molecule called JLQL that could monitor viscosity was conveniently synthesized in two steps. The near-infrared sensor with maximum emission wavelength of 734.6 nm and the Stokes shift of 134.6 nm consisted of a fluorophore and a mitochondrial-targeting moiety as an acceptor group; the two were connected by a double bond. The fluorescence intensity of the sensor increased 175 times with the enhancement of viscosity of a PBS-glycerol system. The interference of other microenvironments such as pH and polarity and other interference analytes could be reduced. JLQL could sensitively and selectively differentiate different levels of mitochondrial viscosity induced by monensin or nystatin. Furthermore, the probe may provide an attractive way to monitor real-time changes of viscosity during mitophagy. Possessing the above properties, JLQL can potentially be employed as a powerful tool for the observation of mitochondrial viscosity.

Abstract:The irregular cellular viscosity is associated with disease and malfunction at the cellular level, developing simple but effective fluorescence probe for detection of intracellular viscosity is of great significance. Herein, we report a benzothiazole-based fluorescent probe HBTD-V for sensitive response to viscosity change in biological systems. HBTD-V displays weak emission in low-viscosity media but strong fluorescence in high-viscosity media. Linear fitting of the viscosity titration results provided a good linear relationship between the fluorescence intensity (logI583) and viscosity (logη) within the viscosity range of 13.8–221.5 cp, indicating the capability of the probe for quantitative viscosity detection. Probe HBTD-V also holds some advantages such as excellent photostability, good anti-interference ability, and good mitochondrial targeting ability. Moreover, probe HBTD-V has been successfully employed to image the viscosity variations in living MCF-7 cells, mice lung organ and tissues, as well as in living mice body. The results demonstrate that HBTD-V can serve as a sensitive fluorescence probe for investigating the vital role of viscosity changes in complex biological systems.

Construction of a mitochondria-targeted probe to monitor cysteine levels in cancer cells and zebrafish

Abstract:Cysteine (Cys) plays an indispensable role as an antioxidant in the maintenance of bioredox homeostasis. We have constructed an efficient fluorescent probe Mito-Cys based on the binding of indole and naphthol. The acrylic ester group serves as a recognition switch for specific detection of Cys, which undergoes Michael addition and intramolecular cyclization reactions, thereby ensuring the chemical kinetics priority of Cys compared to other biothiols. The probe has good water solubility, large Stokes shift (137 nm), with a detection limit of 21.81 nM. In addition, cell imaging experiments have shown that the probe has excellent mitochondrial targeting ability (R = 0.902). The probe can distinguish between Cys, homocysteine (Hcy) and glutathione (GSH), and can detect Cys specifically and quickly (100 s) to ensure accurate quantitative analysis of Cys changes in cells. More importantly, the probe confirms that ferroptosis inducing factors trigger thiol starvation in mitochondria, which helps to gain a deeper understanding of the physiological and pathological functions related to Cys and ferroptosis.

Abstract:Viscosity is a crucial indicator of the flow state of proteins, lipids, and polysaccharides in the cell microenvironment and plays a vital role in maintaining normal cellular activities. Abnormal viscosity in any part of the cell constituents can lead to various diseases in the organism. For instance, abnormal mitochondrial viscosity can lead to diseases such as sugar in diabetes and Parkinson's disease. Therefore, real-time monitoring of changes in mitochondrial viscosity in both pathological and physiological environments is relevant. This study describes a water-soluble xylan-based near-infrared fluorescence probe that can detect changes in cellular viscosity. The designed mitochondria-targeting near-infrared fluorophores were introduced into modified xylan to form a viscosity sensing fluorescent probe (NI-XylV). The fluorescence intensity of NI-XylV at 590, 670 nm gradually increases with the increase in viscosity caused by environmental changes, enabling the sensitive detection of viscosity changes in mitochondria within living cells. NI-XylV exhibits good photostability, biocompatibility, excellent mitochondrial targeting, and broad application prospects as a biobased fluorescence probe.

Abstract:The visual detection of mitochondrial viscosity is of considerable significance in that it is associated with various cellular functions. Herein, a novel red-emitting viscosity probe YD-Mito constructed by linking phenylamine derivatives and benzo[e]indolium through a C–C double bond has been designed and synthesized. It was found that YD-Mito exhibited a special response to viscosity accompanying with a sharp fluorescence enhancement (20-fold) at 620 nm. Importantly, YD-Mito can precisely localized in mitochondria independent from membrane potential and visualized the mitochondrial viscosity. Therefore, probe YD-Mito could provide a powerful tool for the determination of mitochondrial viscosity.

Abstract:As an important cellular signaling molecule, carbon monoxide (CO) is of great significance in maintaining the physiological activities of biological systems. Exploring techniques for detecting of CO at the subcellular fraction is crucial for understanding its various cellular functions. In this work, a simple and effective fluorescence probe (MNP-CO) with mitochondria-targeting was reported for detecting CO in mitochondria. The nitro group was chosen as the recognizing moiety, triphenylphosphine as the mitochondrial target unit and naphthalimide as a fluorescent group. This probe displays high selectivity, high sensitivity and excellent photo-stability. Moreover, biological imaging experiments have shown that MNP-CO has excellent fluorescence imaging ability for CO in cells and zebrafish, and has mitochondrial targeting ability.
