Abstract:The synthesis and purification of a calcium formyl-5, 6, 7, 8-tetrahydrofolate is described. The yield is about 40%~42%. The compounds were indentified on the basis of their analytical and spectroscopic data. The factors affecting the reaction and purifaication were discussed.

Abstract:OBJECTIVE:To establish a new approach to synthesis and purification of calcium 5-formytetrahydrofolate.METHODS:Target compound was synthesized by the use of folic acid as starting material via reduction, formylation, hydrolysis and salt formationRESULTS:The structure of calcium 5-formytetrahydrofolate was confirmed by UV, 1H-NMR and elemental analysis and the overall yield was 54%-59%, 14%-17% higher than the reported yield.CONCLUSION:A convenient synthetic route was developed and it would be suitable for industrial production.

Abstract:Hundred-gram scale of highly selective catalytic hydrogenation of folic acid has been developed, which is adopted continuous-flow technology with Raney Ni as a catalyst. Through optimization of the reaction condition, a high conversion rate of folic acid (> 99%) and a high selectivity (99%) of tetrahydrofolate have been achieved. Additionally, a high-purity calcium-6 S -5-methyltetrahydrofolate (6 S -5-MTHF.Ca) has been synthesized from tetrahydrofolate obtained by continuous hydrogenation through chiral resolution, methylation, salting and recrystallization (purity: 99.5%, de : 97.6%). Compared to known methods, this method provides a feasible procedure using simple, inexpensive, and readily available reagents, making it a step-economical and cost-effective alternative strategy for production of tetrahydrofolate and its active derivatives.

Abstract:The backbone of folate analogue was constructed via Wittig reaction between 2,4-diamino-6-hydroxymethylpyrido[3,2-d]pyrimidine and diethyl-N[4-(formyl)benzoyl]-L-glutamate. Four new 4-amino-N-8,N-10-deazatetrahydrofolate analogues were obtained by hydrogenating of pyridine ring and linking different substituent on N-5 position. The compounds were characterized by H-1 NMR, C-13 NMR and MS techniques. The inhibitory activities against human thymidylate synthetase were determined. The results showed that the bioactivities of the compounds were related with different substituents on N-5 position. The inhibitory rates of two compounds against leukemia cell line HL-60 were above 60% at 0.1 mu mol.L-1.

Abstract:Carbon-14 labelled Dufulin, diethyl ((2-fluorophenyl)((4-methylbenzo[2-14C]thiazol-2-yl) amino)methyl)phosphonate was synthesized from barium [14C]carbonate via Grignard reaction, Curtius arrangement, nucleophilic addition, thionation reaction, cyclization reaction, etc. The labelled product (38.3 mCi) was obtained after RP-HPLC purification in the overall radiochemical/chemical yield of 10%. The product was characterized by ESI-MS,1H NMR and HPLC-FSA. Its technical data (radiochemical purity > 98%, chemical purity > 98%, specific activity 58.0 mCi/mmol) was determined by TLC-IIA, HPLC-LSC, HPLC-FSA/PDA/MS and LSC. This carbon-14 labelled Dufulin can be used as radiotracer in the study on its metabolism and environmental behavior by employing radioisotope tracing techniques.

Abstract:Two versions of carbon-14 labelled dufulin, diethyl ((2-fluoropheny1((4-methyl[phenyl-U-C-14(6)]benzo[d]thiazol-2-yl)amino)methyl)phosphonate (2) and diethyl ((2-fluorophenyl)((4-methylbenzo[d]thiazol-2-yl)amino)[C-14]methyl)phosphonate (3), were separately synthesized from barium [C-14]carbonate via ethyne cyclotrimerization, carboxylation and methylation of benzene ring, amination, cyclization, nucleophilic substitution, Grignard reaction, oxidation, reduction reaction, etc. The labelled products were obtained independently after reverse phase high-performance liquid chromatography (RP-HPLC) purification in the overall radiochemical/chemical yield of 31% and 67% and identified by using H-1 NMR and flow scintillation analyzer/photo-diode array/mass spectrometry (HPLC-FSA/PDA/MS). Their technical data was determined by using TLC-IIA (isotope imaging analysis), HPLC-LSC (liquid scintillation counter), HPLC-FSA/PDA/MS and LSC, respectively. The result demonstrates that radiochemical/chemical purity of 2 and 3 is more than 98% and the specific activities of 2 and 3 are 25.5 and 55.5 mCi/mmol, respectively. The carbon-14 labelled products can be used as radiotracer in the study on its metabolism, residue and environmental behavior by employing radioisotope tracing techniques.

Abstract:sulfomethyltetrazole 5 thiol disodium salt, a side chain of cefonicid,was synthesized from sodium bisulfite,ammonium hydroxide and formaldehyde through five steps of reactions. The yield of product was 10.8% in the optimum condition. The process was more economic in which the iodomethane is substituted to the bromoethane in the second reaction.

Abstract:p-Trifluoromethylthiophenol was synthesized from p-hydroxylphenylthiol in DMF initiated by ·CF_3 which was formed from CF_3Br using Na_2S_2O_4 as a stimulant. The purity of the product was 98% and the yield was 81.5%.

Abstract:To conquer disadvantages in existing synthetic routes of diethyl 4-amino-8-deazatetrafolate,just as high polarity and poor solubility of some intermediates as well as unsatisfying overall yield,a novel synthesis method with protecting groups introduced for 2,4-diamino groups was reported.Starting from 6-acetoxymethyl-2,4-dichloropyridopyrimidine,diethyl 4-amino-8-deazatetrafolate as a key intermediate of anti-folate inhibitors was obtained by a series of successive reactions including protection,condensation,reduction and debenzylation.Relatively shorter reaction time,higher yield and convenient work-up were achieved using this method.The regioselectivity of benzylation and reduction method of pyridine ring were also investigated.This novel synthesis route is of significance for synthesis of tetrahedronfolate analogues.

Abstract:OBJECTIVE To study the synthesis of finasteride. METHODS The ring A of 3-oxo-4-androstene-17β-carboxylic acid was opened with KMnO 4-NaIO 4. Then the product compound reacted with NH 3 and hydrogenated with Pd/C to get 3-oxo-4-aza-5α-androsta-17β-carboxylic acid, which was esterified with anhydrous CH 3OH, dehydrogenated with DDQ/BSTFA, and reacted with t-butylamino magnesium bromide to get finasteride. RESULTS The structures of all the intermediates and finasteride were verified by IR, 1HNMR and MS. CONCLUSION This method was successful without using those expensive reagents such as PtO 2, (PhSeO) 2O and 2,2′-dipyridyl disulphide. The column chromatography was not necessary for all steps. The yield of finasteride is much higher than reported yield.

Abstract:Folic acid (FA) has shown great potential in the fields of targeted drug delivery and disease diagnosis due to its highly tumor-targeting nature, biocompatibility, and low cost. However, FA is generally introduced in targeted drug delivery systems through macromolecular linkage via complex synthetic processes, resulting in lower yields and high costs. In this work, we report a general protocol for synthesizing thiolated folate derivatives. The small molecule thiolated folate (TFa) was first synthesized with a purity higher than 98.20%. First, S-S-containing diol was synthesized with a purity higher than 99.44 through a newly developed green oxidation protocol, which was carried out in water with no catalyst. Then, folic acid was modified using the diol through esterification, and TFa was finally synthesized by breaking the disulfide bond. Further, the synthesized TFa was utilized to modify silver nanoparticles. The results showed that TFa could be easily bonded to metal particles. The protocol could be extended to the synthesis of a series of thiolated derivatives of folate, such as mercaptohexyl folate, mercaptoundecyl folate, etc., which would greatly benefit the biological applications of FA.

Abstract:A number of 8-deazatetrahydrofolates bearing electrophilic groups on N(5) were designed and synthesized based on the action mechanism of methionine synthase, and their biological activities were investigated as well. Compounds (11b, 12b and 16) showed the most active against methionine synthase (IC(50): 8.11 μM, 1.73 μM, 1.43 μM). In addition, the cytotoxicity to human tumor cell lines and dihydrofolate reductase (DHFR) inhibition by target compounds were evaluated.
Extraction, Purification and Detection by Liquid Chromatography−Electrospray-Ionization Mass Spectrometry of Tetrahydrofolate Metabolites in Arabidopsis thaliana

Abstract:The interconversions of C1-substituted folates form the core of plant one carbon metabolism. An analytical method for extraction, purification and liquid chromatography−electrospray-ionization mass spectrometry detection of low level, labile tetrahydrofolate monoglutamates from Arabidopsis thaliana was developed in this study. Under gold fluorescent light, folates were extracted by homogenizing the leaves in extraction buffer and heating the homogenate. A portion of leaf homogenate was incubated with crude preparation of rat plasma conjugase to remove the polyglutamate tails in order to simplify the mixture for analysis. The resulting extract containing the monoglutamyl folates was then applied to an agarose affinity column containing folate binding protein which was purified from milk whey. After elution from the affinity column, the folates were separated and identified by liquid chromatography−electrospray- ionization mass spectrometry on a 150×0.32 mm Symmetry 300 C18 column using isocratic elution with mobile phase (0.1% formic acid/95% acetonitrile, containing 0.1% formic acid, 88:12) at a flow rate of 0.3 µl/min. In positive ion mode, ions representing 5-methyl-tetrahydrofolic acid, 5-formyl-tetrahydrofolic acid and 5,10-methenyl-tetrahydrofolic acid were detected and identified. It is concluded that this folate extraction procedure, combined with folate binding protein affinity column purification is applicable to liquid chromatography−electrospray-ionization mass spectrometry and this method is appropriate for analysis of tetrahydrofolate metabolites in plant leaf tissues. Abstract:The cofactor tetrahydrofolate (THF) is used to reduce, oxidize, and transfer one-carbon (1C) units required for the synthesis of nucleotides, glycine, and methionine. Measurement of intracellular THF species is complicated by their chemical instability, signal dilution caused by variable polyglutamation, and the potential for interconversion among these species. Here, we describe a method using negative mode liquid chromatography-mass spectrometry (LC-MS) to measure intracellular folate species from mammalian cells. Application of this method with isotope-labeled substrates revealed abiotic interconversion of THF and methylene-THF, which renders their separate quantitation particularly challenging. Chemical reduction of methylene-THF using deuterated sodium cyanoborohydride traps methylene-THF, which is unstable, as deuterated 5-methyl-THF, which is stable. Together with proper sample handling and LC-MS, this enables effective measurements of five active folate pools (THF, 5-methyl-THF, methylene-THF, methenyl-THF/10-formyl-THF, and 5-formyl-THF) representing the biologically important 1C oxidation states of THF in mammalian cells.
Highly efficient heterogeneous procedure for the synthesis of fructone fragrancy

Abstract:A novel heterogeneous strong acid catalyst was synthesized through the copolymerization of p -toluenesulfonic acid and paraformaldehyde and utilized for the synthesis of fructone. The results showed that the catalyst was very efficient for the reaction with the yield over 95%. The advantages of extremely high density of acidity, high thermal and chemical stability, low cost for the simple synthetic procedure, and reusability made the catalyst one of the best choices for the reaction.

Abstract:The research present an improved synthetic method of 7-chloro-6-fluoro-1-(2,4-difluorophenyl) -1,4-dihydro-4-oxo-quinoline-3-carboxylic acid, the important intermediate of temafloxacin. The reaction was started with 2,4-dichloro-5-fluorobenzoyl acetoformic ethyl ester, via amination, cyclization, hydrolysis and decarbonylation. The reaction temperature was moderate and the operation was easily controlled. Moreover, three new intermediates 4,5 and 6 were synthesized by the suggested method.

Abstract:Based on our previous work, seven N-5-substituted 8,10-dideazatetrahydrofolate analogues and one 8-deazatetrahydrofolate analogue were designed and synthesized as human dihydrofolate reductase (hDHFR) inhibitors. All compounds were assayed versus DHFR and five different cancer cell lines. The biological assay indicated that replacing N-10 with carbon would significantly increase inhibitory activities against DHFR and cytotoxicities against cancer cell lines. Compound 19a with 4-amino and N-5-formyl showed great antitumour activities against HL-60, Bel-7402 and BGC823 which were much better than MTX.
