Transformation of Three Nematode- Trapping Fungi by Restriction Enzyme- Mediated Integration

Abstract:Three nematode-trapping fungi,Monacrosporium candidum,M.sphaeroides and Arthrobotrys vermicola,were transformed successfully by plasmids carrying hygromycin B phosphotransferase (hygB) gene,using the method of restriction enzyme-mediated integration(REMI).For the three fungi,thetransformation frequency was 10~18,25~35 and 0.2~3 transformants μg~(-1) DNA;and the drug resistance stability of their transformants was 12.5%,82.5% and 87.5%,respectively.The mycelial growth of the three wild types was restrained on PDA medium containing 150μg/ml hygromycin B,but most of their transfomants could grow on the medium containing 700μg/ml hygromycin B.There was no obvious difference between the transformants and their wild types on the mycelial growth rate,morphology of mycelia and trapping devices,and trapping frequency,by testing 40 randomly selected transformants and the wild type from each fungus.
