Abstract:We transformed the nematode-trapping fungus Arthrobotrys vermicola with hygromycin B phosphotransferase gene (hygB) using restriction enzyme-mediated integration (REMI).The transformation frequency was 1-3 transformants μg~(-1) DNA and the transformants stability of hygromycin B resistance was 87.5 %. A mitotic stable transformant Tav36, without obvious difference in vegetative growth and pathogenicity to nematodes compared with that of the wild type, was selected for the detection and monitoring fungal growth and pathogenicity activity in natural soil. Results proved that Tav36 could grow normally in natural soil and trap nematodes, however, the vegetative growth and the pathogenicity to nematodes of Tav36 in natural soil were restrained by fungistasis of soil.
