Abstract:Hemorrhagic snake venom specially induces apoptosis of VEC (vascular endothelial cells). Five apoptosis-inducing proteins had been purified and characterized from crude snake venom. Two of these are L-amino acid oxidase (LAO), the others belong to metalloprotease/disintegrin family. LAO catalyzes H2O2 production by oxidizing some plasma membrane proteins of VEC, disintegrins interfere with binding of integrins with their ligands. The expression of p53 and bcl-2 increases during VEC apoptosis induced by snake venom, moreover, the mRNA of bcl-2 is spliced into two fragments. It has been proved that one of adhesion-dependent signal molecules, alphavbeta3, and one of phospholipid signal molecules, PC-PLC (phosphatidylcholine-specific phospholipase C), are involved in above apoptosis-inducing signal transudation pathway. These results throw light on finding out specific component from protein is snake venom. This component is able to induce tumor vascular endothelial cells apoptosis. This review summarized progress of research on hemorrhagic snake venoms.

Abstract:Apoptosis is a programmed,physiological mode of cell death that plays an important role in maintaining the homeostasis of internal environment.The accurate detection of apoptosis is the first step to analysis the relative mechanisms of apoptosis and to assess the impact of apoptosis on the respective tissue and the disease.Morphologic is the most intuitionistic method to detect the cell apoptosis,and is still considered to be golden standard.DNA ladder,TUNEL,and ELISA are used to detect the DNA fragmentation.The decrease of mitochondria membrane potential is the early event of cell apoptosis.The detection of apoptosis-related genes include the proapoptosis genes and anti-apoptosis genes.Flow cytometry is a easy and fast method developed in recent years to detect the cell apoptosis,and it can be used for qualitative and quantitative analysis of apoptosis.As a important signal molecular,the concentration of Ca2 closely related to cell apoptosis.

Abstract:Objective To discuss the antitumor effect of snake venom toxins and its application in medical field .Methods To review the research progress of composition ,the antitumor effect and mechanism of snake venom toxins .Results It was demonstrated that the snake venom toxins can stop various tumor cells from growing ,and have the functions of killing the tumor cells ,inducing cell death ,and inhibition of angiogenesis .Conclusion It is an important direction in antitumor drug re-search and development to deeply research the composition and the mechanisms of action of the snake venom toxins .
Involvement of Gene Expressions in Apoptosis of Vascular Endothelial Cells Induced by Rattlesnake Venom

Abstract:Formation of apoptotic bodies is a typical character of apoptotic cell death, but how the processes are controlled is not known. In this study, we compared two apoptosis inducing systems in vascular endothelial cells (VEC). We found that the formation of apoptotic bodies during apoptosis induced by rattlesnake venom, which is an unique and specific apoptosis inducer to vascular endothelial cells, was much faster than that induced by deprivation of survival factors (aFGF and serum). When we blocked the synthesis of mRNAs in cells treated with rattlesnake venom by DRB (5, 6-dichloro-1-beta-D-ribofuranosylbenzimidazole), an inhibitor of transcription, the formation of apoptotic bodies was dramatically inhibited. We examined the expression of p53 gene and found that its expression was much higher in apoptosis induced by rattlesnake venom than that in apoptosis induced by deprivation of aFGF and serum. Our results suggest that gene expression is important and p53 gene may play a major role in inducing the formation of apoptotic bodies in VEC.

Abstract:Hemorrhagic snake venom induces apoptosis in vascular endothelial cells (VEC). Vascular apoptosis-inducing protein 1 (VAP1), which is identified as an apoptosis toxin against vascular endothelial cells, induces apoptosis accompanied by severe cell fragmentation compared with that of apoptosis due to other inducers. The mechanism of this morphologic feature is not known. In this report, we examine the roles of the caspases in the apoptosis induced by VAP1. Measurement of the caspase activities shows that activation of caspases occurred in this type of cell death. In the presence of certain caspase inhibitors, the severe cell fragmentation was strongly inhibited. The other hand, cell death induced by VAP1 was not affected by caspase inhibitors. These data suggest that the severe cell fragmentation induced by the snake toxin is a special characteristic of this apoptosis. Apoptosis with severe cell fragmentation may be regarded as a new category of endothelial cell apoptosis.

Abstract:Over the last decade, considerable progress has been made in the study of sphingolipids with the development of biological techniques. Sphingolipids play important roles in diverse physiological process, including cytoskeleton migration, angiogenesis, embryonic development and signal transduction. Except for this, the lastest evidence has suggested that sphingolipids and their metabolite (ceramide, sphingosine, sphingosine 1-phosphate) can induce apoptosis in a wide variety of tumor cell lines such as LoVo HT29, Bel7402, A549, CNE2 cells. This paper is attempted to review the recent advances of investigation into the relationship between sphingolipids and apoptosis.
Emerging anticancer potential and mechanisms of snake venom toxins: A review

Abstract:Animal-derived venom, like snake venom, has been proven to be valuable natural resources for the drug development. Previously, snake venom was mainly investigated in its pharmacological activities in regulating coagulation, vasodilation, and cardiovascular function, and several marketed cardiovascular drugs were successfully developed from snake venom. In recent years, snake venom fractions have been demonstrated with anticancer properties of inducing apoptotic and autophagic cell death, restraining proliferation, suppressing angiogenesis, inhibiting cell adhesion and migration, improving immunity, and so on. A number of active anticancer enzymes and peptides have been identified from snake venom toxins, such as L-amino acid oxidases (LAAOs), phospholipase A2 (PLA2), metalloproteinases (MPs), three-finger toxins (3FTxs), serine proteinases (SPs), disintegrins, C-type lectin-like proteins (CTLPs), cell-penetrating peptides, cysteine-rich secretory proteins (CRISPs). In this review, we focus on summarizing these snake venom-derived anticancer components on their anticancer activities and underlying mechanisms. We will also discuss their potential to be developed as anticancer drugs in the future.

Abstract:Apoptosis, or programmed cell death, is a complex, genetically-determined process involved in the development and maintenance of homeostasis in multicellular organisms. Dysregulation of apoptosis has been implicated in a number of diseases, including cancer and autoimmune disease. Thus, the investigation of apoptotic regulation has evoked considerable interest. Many apoptotic proteins have been shown to be post-translationally modulated, such as by protein cleavage, translocation, protein-protein interaction, and various post-translational modifications, which fall precisely within the range of proteomic analysis. Recently, contemporary proteomic technologies have achieved significant advances and have accelerated research in functional and chemical proteomics, which have been applied to the field of apoptosis research and have the potential to be a driving force for the field. This review highlights some of the major achievements in the application of proteomics in apoptosis research and discusses new directions and challenges for the near future.

Abstract:Apoptosis, also called programmed cell death, has attracted great attention in recent years. After its discovery by Carl Vogt in 1842, apoptosis research was dormant for more than a century. Its rediscovery in the second half of this century, and the coining of the term apoptosis in 1972 by Kerr, Wyllie, and Currie, ignited an unparalleled interest in this field of science. The number of publications related to apoptosis has been growing exponentially every year ever since. This is mainly due to three major advances, two of which have been made recently and one that is currently seen. First, studies with the small nematode Caenorhabditis elegans have identified a number of apoptosis regulating genes--the first evidence that cell death is an active process under genetic control. Many of these genes have mammalian homologs that, like their worm counterparts, seem to regulate mammalian apoptosis. Second, elucidation of the signal transduction pathways of apoptosis has lead especially to the identification of specific death signaling molecules such as a new family of cysteine proteases, the caspases. Third, it has now become clear that many diseases are characterized by dysregulation of apoptotic programs. Many of these programs involve a family of receptors and their ligands, the death receptor/ligand family. The hope now is to interfere with apoptosis regulation in these systems and to develop new therapeutic concepts.
Effect of Bitis gabonica and Dendroaspis angusticeps snake venoms on apoptosis-related genes in human thymic epithelial cells

Abstract:Background: Certain environmental toxins permanently damage the thymic epithelium, accelerate immune senescence and trigger secondary immune pathologies. However, the exact underlying cellular mechanisms and pathways of permanent immune intoxication remain unknown. The aim of the present study was to demonstrate gene expressional changes of apoptosis-related cellular pathways in human thymic epithelial cells following exposure to snake venom from Bitis gabonica and Dendroaspis angusticeps. Methods: Snake venoms were characterized by analytical methods including reversed phase high-performance liquid chromatography and sodium dodecyl sulphate-polyacrylamide gel electrophoresis, then applied on human thymic epithelial cells (1889c) for 24 h at 10 μg/mL (as used in previous TaqMan Array study). Gene expressional changes restricted to apoptosis were assayed by TaqMan Array (Human Apoptosis Plate). Results: The most prominent gene expressional changes were shown by CASP5 (≈ 2.5 million-fold, confirmed by dedicated quantitative polymerase chain reaction) and CARD9 (0.016-fold) for B. gabonica, and BIRC7 (6.46-fold) and CASP1 (0.30-fold) for D. angusticeps. Conclusion: The observed apoptotic environment suggests that pyroptosis may be the dominant pathway through which B. gabonica and D. angusticeps snake venoms trigger thymic epithelial apoptosis following envenomation.

Abstract:The main approaches,the recent achievements and the present problems in snake venomics research were summarized in this paper. In recent years,the elucidation of the panorama of snake venom composition by utilizing high-throughput proteomic approaches has become a hotspot at the field of proteomics research.The protocols for snake venomics studies involve fractionation of the crude venom,followed by proteomic characterization and bioinformatics analysis.Methods for crude venom separation mainly include Two-dimensional gel electrophoresis(2-DE),Gel filtration and RP-HPLC;Proteomic characterization consists of Matrix-assisted laser desorption/ionization mass spectrometry(MALDI-TOF MS),Liquid chromatography and tandem mass spectrometry(LC-MS/MS),Electron spray ionization-tandem mass spectrometry(ESI-MS/MS) and Electron spray ionization-Fourier transform ion cyclotron resonance-mass spectrometry(ESI-FTICR MS);Bioinformatics identification mostly involves BLAST query of peptides interpreted by utilizing Mascot or MS-Fit search engine on protein specific internet sites.Up to now,based on proteomics approaches,venomes from more than one hundred species(subspecies) of fifty-eight Genus among three families(Elapidae,Colubridae and Viperidae) have been studied and more than ten protein families have been identified.These findings will undoubtedly contribute to a deeper understanding of snake venom evolution and diversification,the mechanism of the envenomation and the design of new drugs in search of leading compounds.In the future for snake venomics research,related high-throughput approaches for absolute quantification of snake venome and characterization of identified functional proteins remain to be developed.

Abstract:An increasing important area in immunology is the process cell death mechanism, enabling the immune system triggered thru extrinsic or intrinsic signals to effectively remove unwanted or virus infected cells called apoptosis. A recently isolated infectious Snakehead fish vesiculovirus (SHVV), comprising negative strand RNA and encoded viral matrix (M) proteins, is responsible for causing cytopathic effects in infected fish cells. However, the mechanism by which viral M protein mediates apoptosis has not been elucidated. Therefore, in the present experiments, it was investigated the regulatory potential of apoptosis signals during SHVV infection. By employing the model of SHVV infection in SSN-1 cells, the accelerated apoptosis pathway involves an intrinsic pathway requiring the activation of caspase-9 but not caspase-3 or-8. In the groups of infection (SHVV) or treatment (hydrogen peroxide) were induced apoptotic morphological changes and indicated the activation of the main caspases, i.e.; executioner caspase-3, initiators caspase-8 and caspase-9 using colorimetric assays. Turning to the role of viral M protein when it was overexpressed in SSN-1 cells, it was indicated that the viral M gene alone has the ability to induce apoptosis. To elucidate the mechanism of apoptosis in SSN-1 cells, the activation inhibitors of main caspases were used showing that inhibiting of caspase-3 or caspase-8 activation did not seize induction of apoptosis in virus-infected SSN-1 cells. However, the inhibiting of caspase-9 activation reduced significantly the apoptosis initiation process and sharply the expression of viral M gene, suggesting that SHVV plays a major role in the early induction of apoptosis by caspase-9. Interestingly, there were also differences in the mitochondrial membrane potential after the apoptotic induction of caspases, which confirm that caspase-9 is primarily responsible for the cleavage of caspases during apoptosis. Taken together, these findings can therefore be assumed that viral M protein induces apoptosis via the intrinsic apoptotic pathway in SHVV infecting SSN-1 cells.
