Abstract:Autophagy and apoptosis are two pivotal mechanisms in mediating cell survival and death. Cross-talk of autophagy and apoptosis has been documented in the tumorigenesis and progression of cancer, while the interplay between the two pathways in colorectal cancer (CRC) has not yet been comprehensively summarized. In this study, we outlined the basis of apoptosis and autophagy machinery firstly, and then reviewed the recent evidence in cellular settings or animal studies regarding the interplay between them in CRC. In addition, several key factors that modulate the cross-talk between autophagy and apoptosis as well as its significance in clinical practice were discussed. Understanding of the interplay between the cell death mechanisms may benefit the translation of CRC treatment from basic research to clinical use.

Abstract:Cell apoptosis exists in the whole life of multicellular organisms ,which is capable to maintain homeostasis of tissues and organs through removing unwanted or damaged cells. Eukaryotic cells mediate apoptosis through four main pathways:the death receptor-mediated extrinsic pathway,the intrinsic mitochondrial pathway,granzyme B-mediated pathway and endoplasmic reticulum stress-mediated pathway. There are four important protein molecules involved in apoptotic pathways:caspases (the proteases which execute cell death),adapter proteins (caspase activators),Bcl-2 family proteins and Inhibitor of apoptosis proteins (IAPs). Cell apoptosis plays an important role in establishment of ovarian reserve and abnormal apoptosis inducing some diseases,such as endometriosis,ovarian cancer,et al. In this review we will discuss the apoptotic pathways and their mechanisms mentioned above.

Abstract:The aetiology and pathogenesis of endometriosis remain obscure.Endometriosis is a commom gynecological disease,but it has some malignant behavors,such as hyperplasia,invasion,metastasis and recurrence.It is still a difficult problem for the preventation and treatment of endometriosis.Apoptosis is a distinctive form of programmed cell death,it plays a critical role in maintaining tissue homeostasis and represents a normal function to eliminate excess or dysfunctional cells.Accumulated evidence suggests that abnormal apoptosis can promote the occurrence and development of endometriosis.This study is a current review of the literature focused on the apoptosis pathways,expression of apoptosis in normal endometrium and the alterations in regulation of apoptosis in eutopic and ectopic endometrium from women with endometriosis.The role of apoptosis in the treatment of endometriosis is reviewed to link the basic research findings with clinical applications.

Abstract:Cold and apoptosis are tightly correlated and the research on cold-induced apoptosis has become a hotspot within the transplantation medicine area. The signal transduction involved in the cold-induced apoptosis is anfractuous, either mediated or modulated by reactive oxygen species (ROS), Ca2+, Fe2+ and cold-relevant genes in vivo. Based on the worldwide research progress, we try to generally summarize the signal transduction and modu- lation mechanism involved in the course of cold-induced apoptosis.

Abstract:Apoptosis is a programmed,physiological mode of cell death that plays an important role in maintaining the homeostasis of internal environment.The accurate detection of apoptosis is the first step to analysis the relative mechanisms of apoptosis and to assess the impact of apoptosis on the respective tissue and the disease.Morphologic is the most intuitionistic method to detect the cell apoptosis,and is still considered to be golden standard.DNA ladder,TUNEL,and ELISA are used to detect the DNA fragmentation.The decrease of mitochondria membrane potential is the early event of cell apoptosis.The detection of apoptosis-related genes include the proapoptosis genes and anti-apoptosis genes.Flow cytometry is a easy and fast method developed in recent years to detect the cell apoptosis,and it can be used for qualitative and quantitative analysis of apoptosis.As a important signal molecular,the concentration of Ca2 closely related to cell apoptosis.

Abstract:Apoptosis is one of the main types of programmed cell deaths(PCD)and involves a series of biochemical events that lead to a variety of morphological changes and death.The initial and progress of apoptosis is precisely regulated.This review will summarize current knowledge of the signal transduction pathways of apoptosis.It is now well-established that the apoptotic signals generally involve the extrinsic or intrinsic pathways of apoptosis.The extrinsic pathway originates at the membrane and engages cell surface receptors whereas the intrinsic pathway predominantly involves mitochondria.In the intrinsic pathway,the cell signal induced changes of mitochondrial membrane permeability and the loss of membrane potential.Many proteins factors released,and then cytoplasmic cytochrome C and caspase-9 form of apoptosis.The activated caspase-9 cut caspase-3,then cell dead at last.In the case of extrinsic pathway,several receptors exist including Fas,TNFR-1,DR3,DR4,DR5 and DR6.These receptors contain an intracellular region of approximately 80 amino acids that is designated as death domain.The domain is an important structure that plays a key role in the transduction of apoptotic signals.The interaction between Fas and its ligand(FasL)triggers the formation of a death-inducing signaling complex(DISC),which subsequently recruits and activates caspase-8;this in turn activates other procaspases and culminates in the cleavage of cellular substrates and apoptosis.During the process of tumor cell lines apoptosis Inducted by chemotherapy.It is easy to see the increasing of the Fas receptors and inducing of FasL expression,it can inhibite apoptosis when the blocking Fas /FasL.Tumor necrosis factor(TNF)-related apoptosis-inducing ligand(TRAIL)is a type II transmembrane protein belonging to the TNF family of ligands.TRAIL has been suggested as a safe and tumorselective anticancer agent with low toxicity to normal tissues,and is thought to play a role in tumor immune surveillance by NK,T cells and

Abstract:The human body generates 10–100 billion cells every day, and the same number of cells die to maintain homeostasis. The genetically controlled, autonomously ordered cell death mainly proceeds by apoptosis. Apoptosis is an important way of programmed cell death in multicellular organisms, timely and effective elimination of apoptotic cells plays a key role in the growth and development of organisms and the maintenance of homeostasis. During the clearance of apoptotic cells, transcription factors bind to specific target promoters and act as activators or repressors to regulate multiple genes expression, how transcription factors regulate apoptosis is an important and poorly understood aspect of normal development. This paper summarizes the regulatory mechanisms of transcription factors in the clearance of apoptotic cells to date.

Abstract:The process of programmed cell death, or apoptosis, is generally characterized by distinct morphological characteristics and energy-dependent biochemical mechanisms. Apoptosis is considered a vital component of various processes including normal cell turnover, proper development and functioning of the immune system, hormone-dependent atrophy, embryonic development and chemical-induced cell death. Inappropriate apoptosis (either too little or too much) is a factor in many human conditions including neurodegenerative diseases, ischemic damage, autoimmune disorders and many types of cancer. The ability to modulate the life or death of a cell is recognized for its immense therapeutic potential. Therefore, research continues to focus on the elucidation and analysis of the cell cycle machinery and signaling pathways that control cell cycle arrest and apoptosis. To that end, the field of apoptosis research has been moving forward at an alarmingly rapid rate. Although many of the key apoptotic proteins have been identified, the molecular mechanisms of action or inaction of these proteins remain to be elucidated. The goal of this review is to provide a general overview of current knowledge on the process of apoptosis including morphology, biochemistry, the role of apoptosis in health and disease, detection methods, as well as a discussion of potential alternative forms of apoptosis.

Abstract:Apoptosis,or programmed cell death,is a default and well-controlled process during the cell development.It reflects the net balance of interactions among various pro-apoptotic and anti-apoptotic factors.Regulations of apoptosis can occur at the levels of epigenetical regulation,transcription,translation,post translational modifications or transport.Also it can take place within different cellular compartments,such as nucleus,cytosol,plasma member and mitochondria.This short review intends to exemplify some new apoptosis mechanisms using newly-defined proor anti-apoptotic factors recently characterized in our laboratory.For example,we characterized a novel activating mechanism for caspase-8;we described a specific phosphorylation on E3 ligase Pirh2,which led to the change in its target p53 stability;we also demonstrated how a E3 ligase Pirh2 can act as holder for intermediate filament,thus to stabilize mitochondria,avoiding triggering apoptosis.In addition,the signal transduction pathway that apoptotic signals flow from nucleus to cytosol and finally to mitochondria is also described.All these new findings,together with what had been reported,demonstrate that it truly exists a complicated network for apoptotic signaling within the cell,once it breaks,tumorgenesis begins.

Abstract:Apoptosis is a sequential order of cell death occurring regularly to ensure a homeostatic balance between the rate of cell formation and cell death. However, a misplaced of this balancing function can contribute to an abnormal cell growth / proliferation or autoimmune disorders etc. Apoptosis is therefore said to be crucial from the point of development of an embryo throughout the growth of an organism contributing to the renewal of tissues and also the getting rid of inflammatory cells. This review seeks to elaborate on the recent overview of the mechanism involved in apoptosis, some element and signal contributing to its function and inhibition together with how their malfunction contribute to a number of cancer related cases.
