Abstract:Apoptosis, also called programmed cell death, has attracted great attention in recent years. After its discovery by Carl Vogt in 1842, apoptosis research was dormant for more than a century. Its rediscovery in the second half of this century, and the coining of the term apoptosis in 1972 by Kerr, Wyllie, and Currie, ignited an unparalleled interest in this field of science. The number of publications related to apoptosis has been growing exponentially every year ever since. This is mainly due to three major advances, two of which have been made recently and one that is currently seen. First, studies with the small nematode Caenorhabditis elegans have identified a number of apoptosis regulating genes--the first evidence that cell death is an active process under genetic control. Many of these genes have mammalian homologs that, like their worm counterparts, seem to regulate mammalian apoptosis. Second, elucidation of the signal transduction pathways of apoptosis has lead especially to the identification of specific death signaling molecules such as a new family of cysteine proteases, the caspases. Third, it has now become clear that many diseases are characterized by dysregulation of apoptotic programs. Many of these programs involve a family of receptors and their ligands, the death receptor/ligand family. The hope now is to interfere with apoptosis regulation in these systems and to develop new therapeutic concepts.

Abstract:Cold and apoptosis are tightly correlated and the research on cold-induced apoptosis has become a hotspot within the transplantation medicine area. The signal transduction involved in the cold-induced apoptosis is anfractuous, either mediated or modulated by reactive oxygen species (ROS), Ca2+, Fe2+ and cold-relevant genes in vivo. Based on the worldwide research progress, we try to generally summarize the signal transduction and modu- lation mechanism involved in the course of cold-induced apoptosis.

Abstract:Apoptosis is a programmed,physiological mode of cell death that plays an important role in maintaining the homeostasis of internal environment.The accurate detection of apoptosis is the first step to analysis the relative mechanisms of apoptosis and to assess the impact of apoptosis on the respective tissue and the disease.Morphologic is the most intuitionistic method to detect the cell apoptosis,and is still considered to be golden standard.DNA ladder,TUNEL,and ELISA are used to detect the DNA fragmentation.The decrease of mitochondria membrane potential is the early event of cell apoptosis.The detection of apoptosis-related genes include the proapoptosis genes and anti-apoptosis genes.Flow cytometry is a easy and fast method developed in recent years to detect the cell apoptosis,and it can be used for qualitative and quantitative analysis of apoptosis.As a important signal molecular,the concentration of Ca2 closely related to cell apoptosis.

Abstract:Apoptosis, or programmed cell death, is a complex, genetically-determined process involved in the development and maintenance of homeostasis in multicellular organisms. Dysregulation of apoptosis has been implicated in a number of diseases, including cancer and autoimmune disease. Thus, the investigation of apoptotic regulation has evoked considerable interest. Many apoptotic proteins have been shown to be post-translationally modulated, such as by protein cleavage, translocation, protein-protein interaction, and various post-translational modifications, which fall precisely within the range of proteomic analysis. Recently, contemporary proteomic technologies have achieved significant advances and have accelerated research in functional and chemical proteomics, which have been applied to the field of apoptosis research and have the potential to be a driving force for the field. This review highlights some of the major achievements in the application of proteomics in apoptosis research and discusses new directions and challenges for the near future.

Abstract:Apoptosis is an active process of cellular self-destruction that is involved in normal development. Two kinds of anti-apoptosis genes p35 and iap have been identified in baculovirus genome. Both of them function at different point in the death pathway. Extensive studies have been made about the structures and function mechanisms of the genes and their products. This review highlights the recent advances in the researches of the genes and their function mechanisms and explores the possible utilizations in the future.

Abstract:Apoptosis is a process of programmed cell death that is controlled by genes. Normally, there are three regulation pathways involved in the process of apoptosis, including the signaling of intracel-lular mitochondria, endoplasmic reticula and extracellular death receptors. Recent studies showed that NF-κB is a key regulator in the process of apoptosis. NF-κB plays a promotional and a inhibitory role as well in the regulation of apoptosis, closely related to the the inhibitor of apoptosis proteins family, the B cell lymphoma/ lewkmia-2 family, tumor necrosis factor receptor associated factors, c-Jun N-terminal kinase, tumor necrosis factor related apoptosis inducing ligand and Fas-associated death domain protein-like interleukin-1β. Thus, investigation of the mechanism regarding NF-κB in apoptosis regulation is of great importance for apoptosis-related drug development. The paper reviews the recent research progress in the function of NF-κB in apoptosis pathway regulation.

Abstract:Apoptosis is closely associated with many diseases. Detection of apoptosis can be achieved by morphology, biochemistry, molecular biology, immunology, and other techniques. However, as technologies are increasingly used for the detection of apoptosis, many researchers are confused about how to choose a suitable method to detect apoptosis. Selection of a suitable detection method for apoptosis will help clinical diagnosis and prevention of diseases. This article reviews the selection of optimal apoptosis-detection methods based on research purposes and technique principles.

Abstract:Apoptosis is one of the main types of programmed cell deaths(PCD)and involves a series of biochemical events that lead to a variety of morphological changes and death.The initial and progress of apoptosis is precisely regulated.This review will summarize current knowledge of the signal transduction pathways of apoptosis.It is now well-established that the apoptotic signals generally involve the extrinsic or intrinsic pathways of apoptosis.The extrinsic pathway originates at the membrane and engages cell surface receptors whereas the intrinsic pathway predominantly involves mitochondria.In the intrinsic pathway,the cell signal induced changes of mitochondrial membrane permeability and the loss of membrane potential.Many proteins factors released,and then cytoplasmic cytochrome C and caspase-9 form of apoptosis.The activated caspase-9 cut caspase-3,then cell dead at last.In the case of extrinsic pathway,several receptors exist including Fas,TNFR-1,DR3,DR4,DR5 and DR6.These receptors contain an intracellular region of approximately 80 amino acids that is designated as death domain.The domain is an important structure that plays a key role in the transduction of apoptotic signals.The interaction between Fas and its ligand(FasL)triggers the formation of a death-inducing signaling complex(DISC),which subsequently recruits and activates caspase-8;this in turn activates other procaspases and culminates in the cleavage of cellular substrates and apoptosis.During the process of tumor cell lines apoptosis Inducted by chemotherapy.It is easy to see the increasing of the Fas receptors and inducing of FasL expression,it can inhibite apoptosis when the blocking Fas /FasL.Tumor necrosis factor(TNF)-related apoptosis-inducing ligand(TRAIL)is a type II transmembrane protein belonging to the TNF family of ligands.TRAIL has been suggested as a safe and tumorselective anticancer agent with low toxicity to normal tissues,and is thought to play a role in tumor immune surveillance by NK,T cells and

Abstract:At present,there are many methods for detecting apoptosis in vitro,but these methods just are traumatic in material extraction and tissue biopsy by sacrificing of animals in vitro,which limits their clinical application and transformation.In vivo detection methods have become the focus of current research because of non-invasive and real-time monitoring of apoptosis in vivo.Radionuclide apoptotic cell imaging technology among has good research prospects in non-invasive,early stage,dynamic,sensitive,quantitative,and detection intravital.It is the most widely studied and mature technology for detecting apoptosis intravital.Radionuclide apoptosis imaging has been widely used in the detection of apoptosis in cardiovascular diseases,central nervous system diseases,organ transplant rejection,and the evaluation of efficacy and prognosis of malignant tumor after radiotherapy and chemotherapy.This article reviews the progress of radionuclide imaging probes in apoptosis.

Abstract:Cell death is of great significance in maintaining tissue homeostasis and bodily functions. With considerable research coming to the fore, it has been found that programmed cell death presents in multiple modalities in the body, which is not only limited to apoptosis, but also can be divided into autophagy, pyroptosis, ferroptosis, mitotic catastrophe, entosis, netosis, and other ways. Different forms of programmed cell death have disparate or analogous characteristics with each other, and their occurrence is accompanied by multiple signal transduction and the role of a myriad of regulatory factors. In recent years, scholars across the world have carried out considerable in-depth research on programmed cell death, and new forms of cell death are being discovered continually. Concomitantly, the mechanisms of intricate signaling pathways and regulators have been discovered. More critically, cancer cells tend to choose distinct ways to evade cell death, and different tumors adapt to different manners of death. Therefore, targeting the cell death network has been regarded as an effective tumor treatment strategy for a long time. The objective of our paper is to review the signaling pathways and gene regulation in several typical types of programmed cell death and their correlation with cancer.

Abstract:Apoptosis, or programmed cell death, is involved in numerous human conditions including neurodegenerative diseases, ischemic damage, autoimmune disorders and many types of cancer, and is often confused with other types of cell death. Therefore strategies that enable visualized detection of apoptosis would be of enormous benefit in the clinic for diagnosis, patient management, and development of new therapies. In recent years, improved understanding of the apoptotic machinery and progress in imaging modalities have provided opportunities for researchers to formulate microscopic and macroscopic imaging strategies based on well-defined molecular markers and/or physiological features. Correspondingly, a large collection of apoptosis imaging probes and approaches have been documented in preclinical and clinical studies. In this review, we mainly discuss microscopic imaging assays and macroscopic imaging probes, ranging in complexity from simple attachments of reporter moieties to proteins that interact with apoptotic biomarkers, to rationally designed probes that target biochemical changes. Their clinical translation will also be our focus.

Abstract:The discovery of apoptosis has prompted scientists in biology and medicine to explore the mechanisms and clinical significance of this fascinating phenomenon. In the last decade, accumulating evidence has revealed that apoptosis plays a pivotal role in almost every aspect of growth, differentiation and development. Although it is known that apoptotic cells are rapidly scavenged as a means to prevent inflammation, it remains to be determined whether the clearance of apoptotic cells is related to immune regulation. A review of the recent literature combined with our own work shows that apoptosis is not only a programmed and conserved process of cell death, but also exerts unique effects on the immune system, i.e. apoptotic cell-mediated immune regulation (AMIR). It seems that phagocytes, along with their cytokines released upon binding, phagocytosing and processing apoptotic cells, lie at the heart of AMIR. Investigation of AMIR will not only bring about a better understanding of many important physiological and pathological mechanisms, but also provide new hope for patients of transplantation and autoimmune diseases.
