Effects of Atrial Fibrillation on Expressions of Connexin40 and Connexin43 in Atrial Myocytes in Rheumatic Heart Disease Patients

Abstract:Objective To investigate the influence of atrial fibrillation (AF) on connexin40 (Cx40) and connexin43 (Cx43) in the left and right atrial myocytes when the patients with rhematic heart disease suffered from AF or not. Methods A total of 31 patients undergoing cardiac surgery for correction of valvular defect were divided into three groups: Group Ⅰ, AF lasting ≤6 months (n=11), Group Ⅱ, AF lasting 6 months (n=12), control group, without AF (n=8). Atrial muscle samples of the left and right atrial appendage were cut off during heart valve replacement. Expressions of Cx40 and Cx43 were detected by immunoblotting and immunohistologic analyses. Results ①No obvious change of Cx43 and Cx40 in the left and right atrium of each patient in each group was measured. ②Compared with that in the control group, the level of Cx40 was decreased signficantly in GroupⅠ (P0.05) and more signficantly in Group Ⅱ (P0.01). ③No significant difference in the level of Cx43 was found in the three groups. ④There was an increase in the immunostaining of Cx40 at the lateral membrane and low density area of immunostaining of the atrial cells in the expriment groups. ⑤No change of expression of the Cx43 signal was found in the three groups. Conclusion ① No obvious difference in the level and distribution of Cx43 and Cx40 is found between the left and right atria. ② Down-regulation of level and unevern distribution of Cx40 may be associated with the occurrence and maintenance of AF.
