Effect of Human Atrial Fibrillation on Gene Expression of Calcium Regulatory Proteins in Atrial Sarcoplasmic Reticular

Abstract:Objective: To investigate the effect of human atrial fibrillation (AF) on gene expression of calcium regulatory proteins in atrial sarcoplasmic reticular (SR). Methods: A total of 36 patients with rheumatic heart disease were included. Atrial tissue was obtained from the right atrial appendage during open heart surgery. The mRNA amount of calcium regulatory proteins genes was measured by reverse transcription-polymerase chain reaction and normalized to the mRNA levels of GAPDH. Results: The mRNA of SR Ca2+-ATPase and of ryanodine receptor in patients with persistent AF for more than 6 months were significantly decreased compared with patients with sinus rhythm and persistent AF for less than 6 months. (P all < 0.01), whereas no alterations of the mRNA levels for SR phospholamban calsequestrin were observed (P all > 0.05). Conclusions: The mRNA expression of SR Ca2+-ATPase and ryanodine receptor are down-regulated only in the patients with long-term persistent AF. The change may be a contributory factor for atrial electrical and mechanical dysfunction caused by AF.
