Changes of Protein Expression of Connexin 40, 43 in Human Atrial Fibrillation

Abstract:Objective:To determine whether atrial expression of connexin 40 and connexin 43 is altered in patients with atrial fibrillation(AF) . Methods: Atrial tissue samples of 39 patients undergoing cardiac surgery were examined. 24 patients had AF, 15 patients had no history of AF. Connexin 40 and connexin 43 expression were analyzed at the protein level by Western blot and immunohistochemistry. Results: Western blot analysis for connexin 43,40 demonstrated that AF did not induce significant change in connexin 43 expression level in the right atrial appendage(RAA) . In contrast, connexin 43 content did increase in the left atrium of patients with AF in comparison with those who had sinus rhythm (SR). Connexin 40 protein content in the RAA and left atrium of patients with AF were significantly higher than that of patients with SR. Immunohis-tology revealed that connexin 43, connexin 40 of AF patients acculmated in the intracellular site,both connexins were redistributed. Conclusions:The up-regulation of connexin 43, 40 and spatial remodeling of connexins may have correlation with the initiation or maintenance of atrial fibrillation.
