Abstract:The preparation of allylic amines by transition metal catalysis has played an important role in synthetic chemistry; however, the main employment of prefunctionalized coupling partners through multistep synthesis from available starting materials limits the scope and applications of allylic amines. In this context, we develop a Ni-catalyzed three-component coupling of commercially available aldehydes, amines and alkenes. Avoiding the isolation of imine intermediate, this direct and modular approach delivers a number of versatile allylic amines in high to excellent yields. Diverse aliphatic aldehydes are well tolerated in this reaction, largely reducing imine-enamine tautomerization and self-condensation, hydroamination and hydroacylation of alkenes, and alkenylation of aldehydes, showing high regio-, chemo- and stereoselectivity. The practical applicability of this method is also demonstrated by the late-stage diversification of various natural products and drugs, as well as an array of product transformations. Preliminary asymmetric attempts provide enantio-enriched allylic amines in up to 48% ee by using chiral phosphoramidite ligand, offering the direction for further optimizations.

Abstract:Despite the prominence of C−N bond forming cross-coupling reactions as a strategy to assemble molecular fragments, aminative coupling approaches, in which two fragments are assembled directly at the heteroatom, represents a rarely exploited retrosynthetic strategy. Herein, we report the design, synthesis, and implementation of an anomeric amide reagent capable of promoting highly regioselective aminative alkene-arene and alkene-alkene coupling reactions. This transformation follows a sequence of catalyst-free chloroamination, N-deprotection, and formal nitrene functionalization, all in one-pot. Due to the simplicity of both the protocol and the building blocks required, high-throughput experimentation (HTE) was employed to rapidly and efficiently explore a wide range of chemical space and determine the limits of reactivity. In addition, alternative reactivity modes from the functionalized intermediates delivered by this protocol demonstrate the divergent nature of this aminative coupling strategy.

Abstract:Density functional theory (DFT) was used to investigate the three-component coupling reactions of aldehydes, alkynes, and amines (A3 coupling) using N-heterocyclic carbene silver as the catalyst. This study reveals that the addition reaction between the catalyst N-heterocyclic carbene silver and phenylacetylene (PAE) forms Ag_PAE. Subsequently, one hydrogen atom of the Ag_PAE migrates to the nitrogen atom of the Amine. Thereafter, the amine aldehyde condensation reaction generates a molecule of water and an imine ion with (Path one) or without (Path two) another amine catalyst. Path one has a lower reaction barrier than Path two. Subsequently, the imine ion reacts with silver phenylacetylide to generate the A3 coupling reaction product propargylamine (PPA). Furthermore, the triple bond and −N3 group in PPA undergo a cycloaddition reaction and generate the final product (PR). The entire reaction is strongly exothermic, and, therefore, the reaction is easy to conduct. Moreover, conceptual density functional theory calculations confirm the reaction mechanism. Investigating the mechanism of these reactions will be helpful for understanding and developing new synthesis strategies for similar functional compounds.

Abstract:A nickel-catalyzed Kumada coupling of aniline derivatives was developed by selective cleavage of aryl C-N bonds under mild reaction conditions. Without preinstallation of an ortho directing group on anilines, the cross-coupling reactions of Boc-protected aromatic amines with aryl Grignard reagents afforded unsymmetric biaryls. Mechanistic studies by DFT calculations revealed that the nickel-mediated C-N bond cleavage is the rate-limiting step.

Abstract:Saturated stereogenic centers containing C(sp 3 )–C(sp 3 ) bonds comprise a major portion of organic molecules. Over the past decades, transition-metal-catalyzed asymmetric C(sp 3 )–C(sp 3 ) cross-coupling has evolved into an efficient strategy for constructing such stereogenic centers. However, reaction modes to build asymmetric C(sp 3 )–C(sp 3 ) bonds remain limited. Herein, a nickel-catalyzed enantioselective cross-hydrodimerization between distinct alkenes to enable the enantioselective construction of alkyl–alkyl bonds has been developed. In this reaction mode, N -acyl enamines (enamides) and unactivated alkenes undergo oxidative enantioselective cross-hydrodimerization with excellent levels of chemo- and head-to-tail regioselectivity to give enantioenriched N -acyl α-branched amines by forging the C(sp 3 )–C(sp 3 ) bond with control of the enantioselectivity. The presence of both reducing and oxidizing reagents in the reaction allows the use of alkenes as sole precursors to forge enantioselective C(sp 3 )–C(sp 3 ) bonds, representing a new reaction mode for asymmetric alkyl–alkyl cross-coupling. The asymmetric cross-hydrodimerization between distinct alkenes provides a new strategy for constructing saturated stereogenic centers containing C(sp 3 )–C(sp 3 ) bonds. Abstract:The oxime or hydrazone formation is a classic condensation reaction between aldehydes/ketones and hydroxyl amine or hydrazine.It is a simple,yet fundamental coupling reaction that has been widely applied in the ligations and conjugations of biomolecules and material synthesis.However,the reactions are usually sluggish and normally require acidic conditions with large excess of substrates to facilitate conversion,which limit their wide applications.Recent studies have shown that aniline as a nucleophilic catalyst can significantly accelerate the oxime/hydrazone formation reaction,preliminarily solving the reaction rate issue under mild and bio-compatible conditions.Therefore,it has been the most common coupling method in the modification of biological macromolecules.In this review,the design and development of aromatic amine catalyst in recent years as well as the catalytic mechanism and structure activity relationship are summarized.The application of highly active aniline catalysis in bioconjugations and material synthesis was also included together with a prospect for future development.

Abstract:The transition-metal catalysed three-component coupling of an alkyne, an aldehyde and an amine has been became a widely used method for preparing propargylic amines. Here, we report an unexpected copper(I)-catalysed E -stereoselective reduction of propargylic amines in situ formed from readily available terminal alkynes, aldehydes and 3-pyrroline or isoindoline via [1,5]-hydride transfer, affording E -allylic amines. Abstract:A Cu-catalyzed diastereo- and enantioselective borylative coupling reaction of 1,3-dienes with imines was realized. Branched homoallylic amines are readily prepared in a syn-selective manner with high regio-, diastereo- and enantioselectivity. Moreover, these three-component coupling reactions feature good functional-group compatibility and easy access to the substrates and catalyst.

Abstract:A catalytic asymmetric dehydrogenative cross-coupling reaction between enones and tertiary amines enabled by synergistic photoredox and chiral primary amine catalysis is reported. The reaction was proposed to proceed via the interception of iminium ion intermediate, in situ generated from photoredox oxidation, by dienamine at alpha-position, followed by isomerization, leading to aza-Morita-Baylis-Hillman-type products with good diastereo- and enantioselectivity.

Abstract:Aromatic enamines: para-Vinylanilines were identified as unique nucleophiles, closely resembling enamines, to undergo CC coupling with aldehydes catalyzed by a simple primary–tertiary diamine/Brønsted acid. Abstract:Molecules with an allylic amine motif provide access to important building blocks and versatile applications of biologically relevant chemical space. The need for diverse allylic amines requires the development of increasingly general and modular multicomponent reactions for allylic amine synthesis. Herein, we report an efficient catalytic multicomponent coupling reaction of simple alkenes, aldehydes, and amides by combining nickel catalysis and Lewis acid catalysis, thus providing a practical, environmentally friendly, and modular protocol to build architecturally complex and functionally diverse allylic amines in a single step. The method is remarkably simple, shows broad functional-group tolerance, and facilitates the synthesis of drug-like allylic amines that are not readily accessible by other methods. The utilization of accessible starting materials and inexpensive Ni(II) salt as the alternative precatalyst offers a significant practical advantage. In addition, the practicality of the process was also demonstrated in an efficient, gram-scale preparation of the prostaglandin agonist.
