Abstract:Density functional theory (DFT) was used to investigate the three-component coupling reactions of aldehydes, alkynes, and amines (A3 coupling) using N-heterocyclic carbene silver as the catalyst. This study reveals that the addition reaction between the catalyst N-heterocyclic carbene silver and phenylacetylene (PAE) forms Ag_PAE. Subsequently, one hydrogen atom of the Ag_PAE migrates to the nitrogen atom of the Amine. Thereafter, the amine aldehyde condensation reaction generates a molecule of water and an imine ion with (Path one) or without (Path two) another amine catalyst. Path one has a lower reaction barrier than Path two. Subsequently, the imine ion reacts with silver phenylacetylide to generate the A3 coupling reaction product propargylamine (PPA). Furthermore, the triple bond and −N3 group in PPA undergo a cycloaddition reaction and generate the final product (PR). The entire reaction is strongly exothermic, and, therefore, the reaction is easy to conduct. Moreover, conceptual density functional theory calculations confirm the reaction mechanism. Investigating the mechanism of these reactions will be helpful for understanding and developing new synthesis strategies for similar functional compounds.

Abstract:The N-heterocyclic carbene silver(I) complex 1,3-bis(4-methylbenzyl)imidazol-2-ylidene silver chloride was applied as the effective catalyst for the three-component carboxylative coupling of terminal alkynes, butyl iodide and carbon dioxide. The reaction proved to be highly efficient when using 2mol% of 1,3-bis(4-methylbenzyl)imidazol-2-ylidene silver(I) chloride as the catalyst in the presence of 1.5 equiv. of Cs2CO3 as the base in DMF. This reaction shows good selectivity and substituent-loading capability, in which various functionalized 2-alkynoates were obtained in good yields under very mild conditions.

Abstract:The novel efficient procedure has been developed for the conjugate addition of amines to electron deficient alkenes. A novel multi-SO3H functioned ionic liquid has been synthesized for the conjugate addition. The results showed that the catalyst was very efficient for the reactions with the excellent yields in several minutes. Operational simplicity, without need of any solvent, low cost of the catalyst used, high yields, reusability, excellent chemoselectivity, applicability to large-scale reactions are the key features of this methodology.

The novel ionic liquid [HO3S-bPy][FeCl4] with both Lewis and Brønsted acid sites has been synthesized successfully for the reactions. The results show that the catalyst was very efficient for the conjugate addition of amines to electron deficient alkenes with excellent yields in several minutes. This method has several key features including operational simplicity, no need of any solvent, low cost of the catalyst used, high yields, reusability, excellent chemoselectivity, and wide applicability.

Abstract:Silver (Ι) oxide nanoparticles immobilized on different solid supports were employed in catalyzing an aldehyde-amine-alkyne coupling reaction. It was found that the alumina-supported Ag2O was the most active catalyst. The scope of the corresponding reaction substrates was extended by virtue of these new catalysts. The feasibility of recycling of the alumina-supported Ag2O catalyst was also examined.

Abstract:A novel supramolecular complex [Ag(L)(NO3)] (1) was synthesized by reaction of the oxazoline-containing ligand 1,4-bis(4,5-dihydro-2-oxazolyl)benzene (L) with silver(I) nitrate and its structure was determined by single-crystal X-ray diffraction. Structurally speaking, the one-dimensional (1D) zigzag chains of 1, in which the Ag(I) is three-coordinated with distorted trigonal-planar coordination geometry, are linked together by the hydrogen bonding interactions to form a three-dimensional (3D) supramolecular structure. It was found that the silver supramolecule 1 catalyzes the three-component coupling reactions of aldehyde, alkyne, and amine in the air at room temperature to generate propargylic amines with high efficiency. No additional co-catalyst or activator is required. A variety of aldehydes and amines were used to demonstrate the versatility of the reaction.

Abstract:The preparation of allylic amines by transition metal catalysis has played an important role in synthetic chemistry; however, the main employment of prefunctionalized coupling partners through multistep synthesis from available starting materials limits the scope and applications of allylic amines. In this context, we develop a Ni-catalyzed three-component coupling of commercially available aldehydes, amines and alkenes. Avoiding the isolation of imine intermediate, this direct and modular approach delivers a number of versatile allylic amines in high to excellent yields. Diverse aliphatic aldehydes are well tolerated in this reaction, largely reducing imine-enamine tautomerization and self-condensation, hydroamination and hydroacylation of alkenes, and alkenylation of aldehydes, showing high regio-, chemo- and stereoselectivity. The practical applicability of this method is also demonstrated by the late-stage diversification of various natural products and drugs, as well as an array of product transformations. Preliminary asymmetric attempts provide enantio-enriched allylic amines in up to 48% ee by using chiral phosphoramidite ligand, offering the direction for further optimizations.

Abstract:Complex asymmetric synthesis can be realized by the chiral induction of amino acids in nature. It is of great significance to design a new biomimetic catalytic system for asymmetric synthesis. In this context, we report the preparation and characterization of the composite of polyacrylonitrile fiber (PANF) and metal-organic framework to catalyze the chiral synthesis of propargylamines. A confined microenvironment is established with N-heterocyclic carbene (NHC) silver complex-supported PANF and D-proline-encapsulated MIL-101(Cr). This novel supported catalyst demonstrated high activity in addition to excellent stereoselectivity in the three-component reaction between alkynes, aldehydes, and amines (A3). The regeneration can be realized by adsorption of D-proline again when the stereoselectivity decreases after recycle uses. By regulating the confined microenvironment on the composite, the activity and selectivity of the catalytic system are improved with turnover numbers of up to 2800 and 98% ee. The biomimetic catalytic system to A3 coupling reaction is systematically studied, and the synergistic catalytic mechanism between NHC-Ag and D-proline in the confined microenvironment is revealed.

Abstract:We report a synthesis of silver complexes bearing chelating bidentate N-heterocyclic carbene, with various substitutions at the terminal positions of the imidazole moiety of the NHC units. The long aliphatic substituents proved to be beneficial in terms of the synthetic efficiency of the complexes, compared to previously reported methyl substitution. The complexes demonstrated excellent suitability for the KA2 coupling reaction, providing quaternary carbon-containing propargylic amines in yields up to 95 %, under solvent-free conditions. The method showed high tolerance for a wide range of substrates, including naturally occurring ketones, underscoring its practicality. To our knowledge, this represents the first use of a well-defined silver species in KA2 coupling, marking an advancement in the field.

Abstract:The direct C-H and C-halogen activation for C-C bond formation is one of the most interesting reactions in organic chemistry. C-C bond formation using alkynes as a carbon nucleophilic source is a very useful method in synthesis. Herein, a copper(I) chloride catalyzed three-component coupling reaction of alkynes, dihalomethanes and amines through C-H and C-halogen activation to form propargylic amines under mild conditions has been established. The reaction can be conducted in water, in neat or in common organic solvents and it was applicable to both aromatic and aliphatic alkynes with good functional groups tolerance. It represents an excellent example of multi-component reactions (MCRs), provides an elegant method for the synthesis of propargylic amines which are frequent skeletal components and synthetically versatile key intermediates for the preparation of many nitrogen-containing biologically active compounds. From the mechanism point of view, this chemistry offers not only a new approach to propargylic amines with new C-C and C-N bonds formation through C-H and C-halogen activation, but also provides valuable mechanistic insight into the novel multi-component reactions.
