Abstract:Metal-catalyzed enantioselective allylation, which involves the substitution of allylic metal intermediates with a diverse range of different nucleophiles or S(N)2'-type allylic substitution, leads to the formation of C-H, -C, -O, -N, -S, and other bonds with very high levels of asymmetric induction. The reaction may tolerate a broad range of functional groups and has been applied successfully to the synthesis of many natural products and new chiral compounds.

Abstract:Recent years have witnessed the growing interest in the remote functionalization of alkenes for it offers a strategy to activate the challenging C-H bonds distant from the initiation point via alkene isomerization/functionalization. However, the catalytic enantioselective isomerization/functionalization with one single transition metal catalyst remains rare. Here we report a highly regio- and enantioselective cobalt-catalyzed remote C-H bond borylation of internal alkenes via sequential alkene isomerization/hydroboration. A chiral ligand featured twisted pincer, anionic, and non-rigid characters is designed and used for this transformation. This methodology, which is operationally simple using low catalyst loading without additional activator, shows excellent enantioselectivity and can be used to convert various internal alkenes with regio- and stereoisomers to valuable chiral secondary organoboronates with good functional group tolerance.

Abstract:The evolution of modern catalytic methodologies for asymmetric organic synthesis, including transition metal and organocatalysis, is briefly considered. The results of the authors’ research group comprise the development of convenient, efficient, and reusable metal-free amine-containing catalysts (aminocatalysts) for manufacturing enantiomerically enriched organic compounds. The efficiency of the obtained catalysts is demonstrated for asymmetric aldol reactions, Michael reactions, and domino reactions, including green chemistry processes. The applicability of the developed catalysts and processes for the synthesis of natural product analogs and the most active enantiomers of clinically used drugs is demonstrated.

Abstract:Carbon dioxide (CO2) is an important and appealing C1 building block in chemical synthesis due to its nontoxicity, abundance, availability and sustainability. Tremendous progress has been achieved in the chemical transformation of CO2 into high value-added organic chemicals. However, the asymmetric synthesis with CO2 to form enantioenriched molecules, especially the catalytic process, has lagged far behind. The enantioselective incorporation of CO2 into organic compounds is highly desirable, as the corresponding chiral products, such as carboxylic acids and amino acids, are common structural units in a vast array of natural products and biologically active compounds. Herein, we discuss recent progress toward the enantioselective incorporation of CO2 into organic molecules, which mainly rely on three strategies: 1) kinetic resolution or desymmetrization of epoxides with CO2 to form chiral cyclic carbonates and polycarbonates; 2) nucleophilic attack of O- or N-nucleophiles to CO2 in tandem with asymmetric C−O bond formation to prepare chiral cyclic carbonates and carbamates; 3) direct enantioselective nucleophilic attack of organometallic reagents to CO2 with asymmetric C−C bond formation. Finally, challenges and future outlook in this area are also presented.

Abstract:Saturated stereogenic centers containing C(sp 3 )–C(sp 3 ) bonds comprise a major portion of organic molecules. Over the past decades, transition-metal-catalyzed asymmetric C(sp 3 )–C(sp 3 ) cross-coupling has evolved into an efficient strategy for constructing such stereogenic centers. However, reaction modes to build asymmetric C(sp 3 )–C(sp 3 ) bonds remain limited. Herein, a nickel-catalyzed enantioselective cross-hydrodimerization between distinct alkenes to enable the enantioselective construction of alkyl–alkyl bonds has been developed. In this reaction mode, N -acyl enamines (enamides) and unactivated alkenes undergo oxidative enantioselective cross-hydrodimerization with excellent levels of chemo- and head-to-tail regioselectivity to give enantioenriched N -acyl α-branched amines by forging the C(sp 3 )–C(sp 3 ) bond with control of the enantioselectivity. The presence of both reducing and oxidizing reagents in the reaction allows the use of alkenes as sole precursors to forge enantioselective C(sp 3 )–C(sp 3 ) bonds, representing a new reaction mode for asymmetric alkyl–alkyl cross-coupling. The asymmetric cross-hydrodimerization between distinct alkenes provides a new strategy for constructing saturated stereogenic centers containing C(sp 3 )–C(sp 3 ) bonds. Abstract:Trifluoromethylated organic molecules are ubiquitous in pharmaceutics, materials science, and agrochemicals. In the past few decades, great efforts have been devoted to the development of convenient and efficient strategies for the synthesis of these compounds. β-Trifluoromethylated enones are readily available building blocks, which recognize as one of the attractive synthetic feedstock in organic transformations. Thus far, numerous new catalytic systems including novel chiral ligands and organocatalysts have been applied to asymmetric catalytic protocols using β-trifluoromethylated enones and various optically active compounds containing CF3 motif have been synthesized. Hence, this minreview describes the applications of β-trifluoromethylated enones in diverse asymmetric catalytic transformations since 2008, which incorporates the mechanistic aspects, involving the key mechanistic intermediates and the reaction pathways. Finally, the challenges of related fields and future perspectives are also pointed out.

Abstract:Since its advent two decades ago, asymmetric organo/transition-metal combined catalysis (AOMC), including cooperative catalysis and relay catalysis, has leveraged redistribution of chemical bonds to build up molecular complexity and enantio-differentiation to form individual enantiomers with activations from versatile organocatalysts and transition-metal complexes. The goal of this perspective is to provide readers with the fundamental attributes of AOMC-orthogonality, kinetics, mechanism, and selectivity-to understand how an organocatalyst and a transition-metal complex would collaborate to enable fruitful new reaction development and what are the intrinsic pathways of unproductive events, such as catalyst self-quenching. In closing, future opportunities of AOMC have been directed toward the prediction of effective catalyst combination, introducing enzyme catalysis, and a focus on transient radical intermediate, to animate this area in the years to come.

Abstract:Asymmetric catalysis has been recognized as the most enabling strategy for accessing chiral molecules in enantioenriched forms. Catalytic asymmetric dearomatization is an emerging and dynamic research subject in asymmetric catalysis, which has received considerable attention in recent years. The direct transformations from readily available aromatic feedstocks to structurally diverse three-dimensional polycyclic molecules make catalytic asymmetric dearomatization reactions of broad interest for both organic synthesis and medicinal chemistry. However, the inherent difficulty for the disruption of aromaticity demands a large energy input during the dearomatization process, which might be incompatible with the conditions generally required by asymmetric catalysis. In this Outlook, we will discuss representative strategies and examples of catalytic asymmetric dearomatization reactions of various aromatic compounds and try to convince readers that by overcoming the above obstacles, catalytic asymmetric dearomatization reactions could advance chemical sciences in many respects.Recent advances in catalytic asymmetric dearomatization provide diverse polycyclic molecular scaffolds, novel mechanistic insights, and unprecedented disconnection strategies for total synthesis.This article has not yet been cited by other publications.

Abstract:The transition metal catalyzed asymmetric hydrogenation of carbon-carbon double bonds is recognized as one of the most straightforward methods for the preparation of stereopure compounds. Chiral cyclic motifs have widespread applications in organic synthesis and can also be prepared via this strategy. This review summarizes the recent advances (2016-2023) in the stereoselective metal catalyzed hydrogenation of cyclic α,β-unsaturated ketones, lactams and lactones since considerable developments in this regard were made. The applications of these methodologies in synthesis are also outlined where relevant.

Abstract:Asymmetric organocatalysis has developed spectacularly over the last decades, and its general importance was recently highlighted by the 2021 Nobel Prize in Chemistry. This Perspective aims at providing a brief historical introduction to this field, as well as discussing selected advanced recent examples underscoring the diversity and potential of this still growing and vibrant field. Asymmetric organocatalysis has experienced a long and spectacular way since the early reports over a century ago by von Liebig, Knoevenagel and Bredig, showing that small (chiral) organic molecules can catalyze (asymmetric) reactions. This was followed by impressive first highly enantioselective reports in the second half of the last century, until the hype initiated in 2000 by the milestone publications of MacMillan and List, which finally culminated in the 2021 Nobel Prize in Chemistry. This short Perspective aims at providing a brief introduction to the field by first looking on the historical development and the more classical methods and concepts, followed by discussing selected advanced recent examples that opened new directions and diversity within this still growing field.

Abstract:Chiral organoboronates have played a critical role in organic chemistry and in the development of materials science and pharmaceuticals. Much effort has been devoted to exploring synthetic methodologies for the preparation of these compounds during the past few decades. Among the known methods, asymmetric catalysis has emerged as a practical and highly efficient strategy for their straightforward preparation, and recent years have witnessed remarkable advances in this respect. Approaches such as asymmetric borylative addition, asymmetric allylic borylation and stereospecific cross-coupling borylation, have been extensively explored and well established employing transition-metal catalysis with a chiral ligand. This review provides a comprehensive overview of transition metal-catalysed asymmetric borylation processes to construct carbon-boron, carbon-carbon, and other carbon-heteroatom bonds. It summarises a range of recent achievements in this area of research, with considerable attention devoted to the reaction modes and the mechanisms involved.

Abstract:The merger of photoredox and transition-metal catalysis has evolved as a robust platform in organic synthesis over the past decade. The stereoselective 1,4-functionalization of 1,3-enynes, a prevalent synthon in synthetic chemistry, could afford valuable chiral allene derivatives. However, tremendous efforts have been focused on the ionic reaction pathway. The radical-involved asymmetric 1,4-functionalization of 1,3-enynes remains a prominent challenge. Herein, we describe the asymmetric three-component 1,4-dialkylation of 1,3-enynes via dual photoredox and chromium catalysis to provide chiral allenols. This method features readily available starting materials, broad substrate scope, good functional group compatibility, high regioselectivity, and simultaneous control of axial and central chiralities. Mechanistic studies suggest that this reaction proceeds through a radical-involved redox-neutral pathway.
