Clarke) is a widely distributed wild shrub. It is a well-known medicinal plant that has a relatively high content of solasodine (alkaloid) in its foliage, stems, fruit, and seeds. During field surveys from July to September 2018 in Mile and Jianshui counties, Yunnan Province, China, Dutch eggplants with leaf mosaic and mottling were observed around flue-cured tobacco (Nicotiana tabacum ‘Yunyan 87’) crops. Samples were collected from 11 symptomatic Dutch eggplants (six plants from Mile and five plants from Jianshui counties). These leaf samples were analyzed by transmission electron microscopy (TEM). Rod-shaped particles 17–18 nm in diameter and 300 nm in length were observed in all Dutch eggplant samples. The leaf samples were further analyzed using enzyme-linked immunosorbent assay (ELISA) kits (Agdia, U.S.A.) for the presence of tobacco mosaic virus (TMV), tomato mosaic virus, and potyviruses. Positive and negative controls (leaves of a healthy Dutch eggplant) were also used for the assays. Double-antibody sandwich (DAS)-ELISA detected only TMV in the symptomatic Dutch eggplants. Total RNA from leaves of all 11 samples was extracted using the TaKaRa MiniBEST Universal RNA Extraction kit (Takara Bio Inc., Japan) and RT-PCR was performed using TMV-specific primers (TMV-CP-R: 5′-TCAAGTTGCAGGACCAGA-3′ and TMV-CP-F 5′-ATGTCTTACAGTATCACTAC-3′) as described previously (Srivastava et al. 2015) and complete TMV coat protein (CP) gene (480 nt) was amplified. An amplicon from sample Mile-1 was gel-purified, cloned into pMD19-T vector (Takara Bio Inc., Japan), and sequenced. In BLASTn, the 480 bp nucleotide sequence of the amplicon was identical to TMV Bei Cang Zhu (KU198186), Xingren-2 (HE818450), Xingren-1 (HE818449), and Chengjiang (HE818410) isolates. The sequence of a TMV isolate Mile-1 was determined and annotated (GenBank accession no. MK584554). Mechanical transmission assays of sap from symptomatic Dutch eggplant resulted in systemic mosaic and mottled symptoms in N. tabacum ‘Yunyan 87’ and local necrotic lesions in N. glutinosa. These plants also tested positive for TMV by DAS-ELISA and RT-PCR methods (as described above). There are previous reports of mosaic and leaf distortion in Dutch eggplant caused either by TMV, as observed in India (Verma et al. 1972), or by potato virus Y, as observed in Israel (Marco. 1987). In 2007, Dutch eggplant was also reported as a host of tomato yellow leaf curl China virus in China (Liao et al. 2007).
