Surface Characteristics and Biological Responses of Si, Mg and Co3 Substituted Nano-Hydroxyapatite Coatings

Abstract:The aim of this study is to examine the correlation between the surface characteristics (surface charge, dissolution rate) and the biological functions (fibronectin adsorption, hydroxycarbonate apatite formation, and cell response) of the nanostructured Si, Mg, and CO3 substituted hydroxyapatite coatings (alone and co-substituted), synthesized by the liquid precursor plasma spraying process. The dissolution rates and hydroxycarbonate apatite formation of different substituted HA coatings have been examined under three different biological media: simulated body fluid, phosphate buffer saline and cell culture medium. In general, the dissolution rates for Ca, P and doped elements were low and the differences were very small among different substituted HA coatings. The hydroxycarbonate apatite formation was much more prominent in simulated body fluid than in phosphate buffer saline and cell culture medium, whereas only small differences were observed among different substituted hydroxyapatite coatings regardless of medium used. Both Si-HA and CO3-HA coatings have shown enhanced proliferation and differentiation of MC3T3-E1 osteoblasts, whereas a minor inhibitory effect on the cell differentiation was observed for the Mg-HA coating. Overall, our results suggested that the cellular responses of the substituted hydroxyapatite coatings did not correlate with the dissolution rates of Ca, P and the doped elements, which has been commonly suggested in literature. A stronger correlation between the cellular responses and the fibronectin adsorption behavior was observed, except for the CO3-HA coating, which indicated that complex mechanisms were involved including surface factors such as protein adsorption characteristics.
