Abstract:Performance of nanoprobes can often determine the detection level of Lateral immunochromatography. Traditional probes were limited by the quantity and orientation of antibodies, immune activity of the Fab region or binding strength between protein and substrate. This study developed a new efficient and robust technology to construct fluorescent nanoprobes with oriented modified antibodies, based on specific binding of the Fc region of antibody with streptococcal protein G (SPG) on the surface of polystyrene microspheres (MS) and subsequent covalent cross-linking at binding sites to firm them. Lateral flow immunoassay using these probes was applied for the detection of cardiac troponin I (cTnI). The significantly improved detection sensitivity demonstrated that antibody orientation on MS surfaces effectively enhanced immunological activities of probes compared with random immobilizing methods. Furthermore, performance evaluation results of lateral flow test strips met clinical requirements perfectly, including limit of detection (0.032 ng/mL), linearity ( R > 0.99), repeatability (CV < 10%), correlation ( R > 0.99), and heat aging stability. This research also employed heterophilic blocking reagent (HBR) to actively block redundant binding sites of SPG for the first time in order to eliminate false positive interferences, improving the sensitivity and precision of test results further.

Abstract:Acute myocardial infarction (AMI) is a major health problem leading to high rates of mortality and morbidity.Biomarker cardiac troponin I (cTnI) has shown high sensitivity and specificity towards AMI detection,and has been regarded as "gold standard".An ultrasensitive method to detect cTnI with low concentration in human fluid is essential.In this paper,we developed an aptamer-based assay coupled with rolling circle amplification (RCA) and molecular beacon probe for sensitive detection of cTnI.In this strategy,aptamer acts as a bridge to communicate between oligonucleotides and cTnI.RCA reaction produces a single-stranded tandem repeated copy of the circular template,which are recognized by fluorescence molecular beacon probe.With this strategy,highly sensitive and specific detection of cTnI was realized with the lowest detectable concentration of 7.24 pg/mL.The developed aptamer-RCA assay can be a promising tool in clinical samples analysis.The assay can also analyze other disease-related biomarkers by replacing the aptamer.

Abstract:Magnetic fluorescent nanoparticles have demonstrated notable advantages as multifunctional immunoassays in lateral flow biosensor (LFB), contributing to the enhancement of both the flexibility and practicability of LFB. Herein, we synthesized flower-shaped magnetic fluorescent Nanoparticles (FMNP@QDs) with heightened fluorescence emission. The distinctive core/shell architecture of FMNP@QDs comprised the high saturation magnetization core encapsulated by oleic acid-modified Fe3O4, the flower-shaped shell made up of polymer colloids, and quantum dots (QDs) uniformly distributed on the outside. After surface modified with PEG-COOH and conjugate with cardiac troponin I (cTnI) antibodies, the FMNP@QDs were successfully applied in LFB for highly sensitive cTnI detection in samples. The limit of detection (LOD) of the proposed LFB for cTnI was 0.0097ng/mL under fluorescent mode and 0.026ng/mL under magnetic mode. A high level of concordance was observed between the FMNP@QDs based fluorescent LFB and the commercial CLIA method. The results indicated that the FMNP@QDs based LFB has a substantial potential for point-of-care test (POCT) technology, owing to its simple operating procedures, affordability, and enhanced sensitivity.

Abstract:In this work, Ag nanoparticles (Ag NPs) decorated CoS nanoflowers (CoS NFs) were firstly used as bis-coreaction accelerator for signal amplification to establish an highly sensitive electrochemiluminescence (ECL) immunosensor for cardiac troponin T (cTnT) detection. Due to the synergistic catalysis between CoS NFs and Ag NPs, the decomposition rate of the co-reaction reagent H2O2 was largely improved, generating more ROS for signal amplification. Using the N-(4-aminobutyl)-N-ethylisoluminol (ABEI) as ECL luminophore, a new signal tag was acheived according to the assembly of ABEI functionalized Ag NPs (ABEI-Ag) on the CoS NFs via Ag-S bond, which integrated with the ECL luminophore and bis-co-reaction accelerator. In addition, to fabricate sensing interface, primary antibodies (Ab1) was immobilized on the glassy carbon electrode which decorated with Au nanoparticles (Au NPs), almost providing a zero background signal. As a result, this developed immunosensor for cTnT possessed a linear range from 0.1 fg mL(-1) to 100 pg mL(-1) and the limit of detection down to 0.03 fg mL(-1) for ultrasensitive detection of cTnT, which was expected to be applicable to cTnT detection in clinic acute myocardial infarction. Abstract:Cardiac troponin I (cTnI), a principal biomarker of acute myocardial infarction, possesses superior cardiac specificity and can provide a long detection window for cardiac injury. Although the potential of cTnI as a cardiac biomarker is promising, sensitive and rapid detection of cTnI remains challenging. In this work, a novel colorimetric biosensor was developed for cTnI detection based on multifunctional hybrid nanoprobes coupled with triple-aptamer-modified tetrahedral DNA nanostructures (TAPT-TDNs). First, we evaluated the affinity of four TAPT-TDNs with three vertices modified by two kinds of aptamers in different proportions. The four TAPT-TDNs exhibited much better affinity than the single-stranded aptamer for cTnI, which was attributed to the fact that TAPT-TDNs could improve the capture efficiency of cTnI with excellent interface density and constant support. Second, the selected TAPT-TDNs with highest affinity were immobilized on Fe3O4-Au magnetic nano-particles as capture probes (named Fe3O4-Au-TAPT-TDNs). TAPT-TDNs and HRP were both labeled on NH2-MIL-101(Fe) to form signaling probes (named MIL-101(Fe)-HRP-TAPT-TDNs). Then, with the help of capture probes and signaling probes, the target protein was recognized and captured to form capture probe-cTnI-signaling probe sandwich-like structures, causing a decrease in signaling probes in the supernatant with magnetic separation and eventually leading to a visual color change. This colorimetric aptasensor displays excellent analytical perfor-mance toward cTnI with a detection limit of 27 pg & BULL;mL-1. The anisotropic aptamer-mediated colorimetric sensor may serve as an alternative to conventional immunoassays for quick and accurate detection of cTnI in clinical samples.

Abstract:Cardiac Troponin I (cTnI) is a preferred biomarker to diagnose acute myocardial infarction which is one of the leading risks to health globally due to its short term. However, clinical analyzers are difficult to achieve its on-site quantitative detection. A novel label-free aptasensor was constructed to realize ultrasensitive and rapid recognition of cTnI. A nanocubic AuNPs/In2O3 composite was designed to provide synergistic effects of abundant active sites and signal magnification for aptamers grafting. Relying on a conductance-dependence strategy, this aptasensor can achieve the quantitative detection within 10 min, which is much faster than state-of-the-art analyzers, as well as exhibiting an ultrawide linear range of 0.1-1000 ng/mL and a low detection limit of 0.06 ng/mL with an excellent selectivity in the analysis of human serum.

Abstract:Electrochemiluminescence (ECL) has emerged as one of the most important methods for in vitro diagnosis and detection, but it is still limited in sensitivity for ultrasensitive biodetections. Fast and ultrasensitive detection of biomolecules is critical, especially for the clinical detection of cardiac troponin I (cTnI) for cardiac infarction diagnosis. In this study, an effective tactic was developed to enhance ECL efficiency of the luminol system, by combined use of Co2+-based metal organic frameworks (MOF), zeolitic imidazolate frameworks (ZIF-67), and luminol-capped Ag nanoparticles (luminol-AgNPs). The integration leads to a pronounced ∼115-fold enhancement in luminol ECL. On the basis of this fascinating sensing platform, a robust label-free ECL immunosensor was constructed for ultrasensitive detection of cTnI, the main marker of myocardial infarction, with good stability and a detection limit as low as 0.58 fg mL-1 (S/N = 3).

Abstract:Despite its simplicity and specificity, enzyme-linked immunosorbent assay (ELISA) requires conjugation of the enzyme to an antibody and preservation of enzymatic activity during storage and assay. In light of the high porosity and enormous surface area possessed by metal-organic frameworks (MOFs), we constructed an immunosensor by integrating antibody and fluorescent probe (coumarin or COU) with zeolitic imidazolate framework-8 (ZIF-8), in which about 34 COU molecules can be encapsulated per ZIF-8. After coating of ZIF-8 with the secondary antibody specific to cardiac troponin I (cTnI), the final MOF@COU/Ab(2) composite renders an enzyme-free assay that amplifies the detection signal via release of alkaline-hydrolyzed COU from the MOF interior. Owing to the isolation of the fluorescent probe before the readout step, our immunosensors are highly stable and reproducible, with a relative standard deviation of less than 1.5% for repetitive assays in a 30-day period. Our MOF-based immunoassay is also highly sensitive and possesses a wide dynamic range (11.1 fM to 35.6 pM). The detection limit was estimated to be 4.4 fM (0.099 pg.mL(-1)), which is similar to 180-fold lower than that of ELISA. We quantified cTnI in clinical serum samples, yielding results highly comparable to those measured by ELISA. Particularly remarkable is that the low detection limit enabled us to quantify cTnI in sera of healthy persons, including levels that are close to the high-risk threshold but not detectable by ELISA. The generality of this cost-effective and stable fluorescent immunosensor renders versatilities for sensitive detections of many other protein biomarkers.

Abstract:We present a microfluidic assay device integrated with Au nanorods for rapid and sensitive detection of Cardiac troponin I (cTnI), the current gold standard biomarker for diagnosing acute myocardial infarction. The Au nanorods, fabricated using oblique angle deposition (OAD), significantly enhance fluorescent signals, thereby eliminating the need for high-resolution lithography technologies. Leveraging this device, rapid and sensitive detection of cTnI is realized with a limit of detection (LOD) of 6.9 pg mL−1 within 6 minutes, referring to a sensitivity enhancement of 4000-fold compared to glass substrates. Given its simple fabrication method, ease of operation, and highly sensitive immunoassay performance, this device holds great potential for clinical and point-of-care testing (POCT) applications.
A Biomimetic Chip with Dendrimer-Encapsulated Platinum Nanoparticles for Enhanced Electrochemiluminescence Detection of Cardiac Troponin I

Abstract:The measurement of cardiac troponin I (cTnI) is of vital importance for the early diagnosis of acute myocardial infarction. In this study, an enhanced electrochemiluminescent immunoassay for the highly sensitive and precise determination of cTnI was reported. A biomimetic chip with nepenthes peristome surface microstructures to achieve single-layer microbead arrays and integrated microelectrode arrays (MEAs) for ECL detection was microfabricated. Ru@SiO2 nanoparticles were prepared as signal amplificators labeling immunomagnetic beads. Dendrimer-encapsulated platinum nanoparticles (Pt DENs) were electrochemically modified on ITO MEAs. The resulting Pt DEN-modified ITO MEAs preserved good optical transparency and exhibited an approximately 20-fold ECL signal amplification compared to that obtained from bare ITO. The method made full use of the biomimetic chip with Pt DENs to develop single-layer immunomagnetic bead arrays with increasingly catalyzed electrochemical oxidation of the [Ru(bpy)3]2+–TPA system. Consequently, a limit of detection calculated as 0.38 pg/mL (S/N = 3) was obtained with excellent selectivity, demonstrating significant potential for the detection of cTnI in clinical diagnostics.

Abstract:Cardiac troponin (cTn) is a specific and sensitive biomarker for diagnosis of myocardial injury. Hence, numerous kinds of biosensors for cTn have been reported. Electrochemical methods possess inherent advantages over other kinds of sensors because they are specific, sensitive, and simple. By combining the advantages of electrochemical biosensors with those of nanomaterials, some interesting electrochemical biosensor for cTn can be obtained where the nanomaterials trigger substantial signal amplification. This review (with 101 refs.) summarizes the state of the art in electrochemical biosensing of cTn based on the use of nanomaterials. Following an introduction into the field, the use of nanomaterials in electrochemical sensing is briefly discussed. A next section covers strategies for signal amplification by using nanomaterials, with subsections on the use of nanowires, nanotubes, graphenes, and various other nanoparticles. The article concludes with a discussion of the prospects of nanomaterial-based signal amplification and on future research directions.

Abstract:The accurate and rapid detection of cardiac troponin I (cTnI) at an early stage can prevent people from getting acute myocardial infarction to a great extent. Here, an ultrasensitive sandwich-type electrochemical immunosensor for the quantitative detection of cTnI is designed based on a novel signal amplification strategy. Gold nanoparticles (Au NPs) doped covalent organic frameworks (COFs) and electron mediator toluidine blue (TB-Au-COFs) behave as labels for achieving signal amplification. Note that polypyrrole modified titanium dioxide nanoparticles (TiO2-PPy) with a high electronic conductivity and a large specific surface area can promote the performance of the immunosensor. For the immobilization of primary antibodies, Au NPs decorated TiO2-PPy (TiO2-PPy-Au) is used as the substrate material to improve electron transfer. The proposed method exhibits a linear range from 0.5 pg mL−1 to 10.0 ng mL−1 and a low detection limit of 0.17 pg mL−1 (S/N = 3). The immunosensor exhibits good stability, acceptable reproducibility and accuracy, indicating potential applications in clinical diagnostics.

Abstract:Cardiac troponin I (cTnI), as a cardiac biomarker, holds significant importance in the diagnosis of acute myocardial infarction. However, the current detection methods mostly require specialized personnel and large analytical instruments, making it difficult to achieve convenient and on-site testing. This situation leads to delayed disease diagnosis and treatment, that increases patient suffering, and reduces the cure rate. This strategy presents the development of a portable visual DNA hydrogel colorimetric sensing platform based on a smartphone. Utilizing dual-mode detection with ultraviolet signals and solution colorimetry, the platform achieves ultra-sensitive and real-time detection of cTnI. Furthermore, optimization of detection conditions, such as the amount of polyacrylamide, reaction time, aptamer concentration, encapsulation of the nanozyme, incubation time, and reaction temperature, were performed based on this platform. The proposed ultraviolet and visual detection platforms exhibited good linear relationships with the signal within the ranges of 0.003 ng mL −1 to 10.00 ng mL −1 and 0.01 ng mL −1 to 7.00 ng mL −1 , with detection limits of 2.57 pg mL −1 and 0.013 pg mL −1 , respectively. Additionally, utilizing 3D printing technology, a portable detection device was designed and employed for the detection of cTnI concentrations in human serum samples. Whatever in the initial and spiked samples, the results showed high sensitivities. The sensitivity and convenience of the sensor in detecting cTnI make it promising for home testing of patients, with broad market prospects.

Abstract:The sensitive and accurate detection of cardiac troponin I (cTnI) is critical for myocardial infarction diagnosis. In this work, a dual-aptamer-based electrochemical (EC) biosensor was designed for cTnI detection based on the DNA nanotetrahedron (NTH) capture probes and multifunctional hybrid nanoprobes. First, the NTH-based Tro4 aptamer probes were anchored on a screen printed gold electrode (SPGE) surface through the Au-S bond, providing an enhanced spatial dimension and accessibility for capturing cTnI. Then, the hybrid nanoprobes were fabricated by using magnetic Fe3O4 nanoparticles as nanocarriers to load a large amount of cTnI-specific Tro6 aptamer, natural horseradish peroxidase (HRP), HRP-mimicking Au@Pt nanozymes and G-quadruplex/hemin DNAzyme. This signaling nanoprobes are capable of specifically recognizing the target cTnI based on the Tro6 aptamer and amplifying the signals to improve the detection sensitivity via enzymatic processes. We found the remarkable enhanced effect of EC signal to be attributed to the co-catalysis effect of hybrid nanozymes, HRP and DNAzyme. The target cTnI was sandwiched between the two types of aptamers (Tro4 and Tro6) on the electrode interface. Finally, this EC aptasensing platform exhibited great analytical performance with a wide dynamic range of 0.01-100 ng mL-1 and a low detection limit of 7.5 pg mL-1 for cTnI. The high selectivity, sensitivity and reliability of EC aptasensor can provide great potential in the clinic disease diagnostics.

Abstract:Troponin I, as a cardiac-specific biomarker, is a very valuable tool in the identification of acute coronary syndromes. In this work, an economical technique for easy and sensitive diagnosis of cardiac troponin I (cTnI) was developed nanoprobe on the basis of graphene oxide quenching of the fluorescence of bovine serum albumin–capped fluorescent nickel (BSA@NiNCs) nanoclusters. The developed nanoprobe nickel nanocluster exhibited superparamagnetism. The nanoprobe showed a fluorescence turn-on behavior toward incremental addition of in cTnI, with a detection limit of 0.012 ng/mL. We successfully used the proposed sensing technique for cTnI sensing in serum samples.

Abstract:Real-time label-free electrical detection of proteins, including cardiac troponin (cTn), is demonstrated using functionalized SnO₂ nanobelt field-effect transistors (FETs) with integrated microfluidics. Selective biomolecular functionalization of the active SnO₂ nanobelt channel and effective passivation of the substrate surface were realized and verified through fluorescence microscopy. The validation/verification of the sensing scheme was initially demonstrated via detection of biotin-streptavidin binding: devices with single biotinylated SnO₂ nanobelts showed pronounced conductance changes in response to streptavidin binding. Importantly, the pH-dependence of the conductance changes was fully consistent with the charged states of streptavidin at different pH. Moreover, the specificity of the sensors' electrical responses was confirmed by co-labeling with quantum dots. Finally, the sensing platform was successfully applied for detection of the cardiac troponin I (cTnI) subunit within cTn, a clinically important protein marker for myocardial infarction.

Abstract:Interesting double-channel microfluidic chip integration with a sandwich-type cathodic photo-electrochemical (PEC) biosensor is utilized for ultrasensitive and efficient detection of cardiac troponin I (cTnI) based on a signal amplification strategy. The Pd nanoparticles loading on the I-doped bismuth oxybromide with oxygen vacancies (Pd/I:BiOBr-OVs) as a sensing platform can effectively enhance cathodic photocurrent response by improving the visible light absorption ability with I doping, facilitating the efficiency separation of photogenerated electron-hole pairs with OVs, and increasing the electron-transfer rate with Pd loading, where the photogenerated electron could be captured by dissolved O2 to boost generation of a superoxide anion radical (•O2-). To further enhance the PEC response, a novel superoxide dismutase loaded on gold@polyaniline (SOD-Au@PANI) as a signal amplification label is developed for incubating the detection antibody (dAb). It is particularly noteworthy that SOD can effectively catalyze dismutation of the •O2- to produce H2O2 and O2, and Au@PANI with a good reduction and catalytic property can catalyze the produced H2O2 into H2O and O2. Then, the produced O2 that has been dissolved or adsorbed can capture more photogenerated electrons, resulting in more electron-hole pairs to separate, so as to the cathodic photocurrent signal of this system which can be amplified more significantly. Therefore, a signal amplification cathodic PEC biosensor is prepared for sensitively detecting cTnI, in which a good linearity ranging from 0.1 pg/mL to 100 ng/mL with a low detection limit of 0.042 pg/mL is obtained. Furthermore, the proposed biosensor exhibits excellent sensitivity and high selectivity, which could be extended to detect other disease markers in biological analysis and early disease diagnosis.

Abstract:Enhancement in the signals of electrochemical immunosensors via advanced nanomaterials is beneficial for developing immunoassay of cardiac troponin I (cTnI) with high sensitivity and selectivity. By virtue of K3[Fe(CN)6] as a signal probe, an advanced label-free electrochemical immunosensor was fabricated for ultrasensitive detection of cTnI with trimetallic alloyed AuPtPd porous fluffy-like nanodendrites (AuPtPd FNDs), followed by assembly of the antibody (Ab). By virtue of the structural advantages, electronic effects and synergetic catalysis of the trimetals, such nanostructures showed large specific surface area to increase the loading of the cTnI Ab and amplify the electrochemical signals of the probe. The sensor displayed a wide linear range (0.01 ~ 100.0 ng mL−1) and a low detection limit (LOD = 3 pg mL−1) under the optimal conditions, which showed great potential for detecting cTnI in serum samples. Furthermore, this biosensor provides an effective detection platform for analysis of other cardiac markers in practical samples.

Abstract:The accurate detection of biomarkers for acute myocardial infarction (AMI) plays an important role in clinical diagnosis and management process. In this work, we developed an electrochemical biosensor by using magnetic metal organic framework (MMOF) nanocatalysts and DNA nanotetrahedron (NTH) based dual-aptamer probes for nonenzymatic detection of cardiac troponin I (cTnI), a gold standard biomarker for the early diagnosis of AMI. Firstly, the NTH-assisted dual-aptamer (Tro4 and Tro6) capture probes were immobilized on the screen-printed gold electrode (SPGE) for the highly enhanced capture the target cTnI with steady support and optimized interface density. Then, the MMOF Fe3O4@UiO-66 nanozymes were decorated by bimetallic Cu@Au nanoparticles and two kinds of aptamer. This fabricated nonenzymatic nanoprobe1 (NP1) can be applied for recognizing the cTnI specifically and amplifying the current signal by catalyzing the oxidation of hydroquinone (HQ) to benzoquinone (BQ) with H2O2. The target proteins were captured to fabricate a supersandwich-like structure on a SPGE interface. Furthermore, the nanoprobe2 (NP2) of Cu@Au nanozymes labeled with dual-complementary DNA (cDNA) to the dual-aptamer, were anchored on the NP1 through DNA hybridization, leading to the formation of cluster-based nanoprobes for further enhancing detection sensitivity. Finally, this enzyme-free electrochemical aptasensor exhibited great analytical performance with a dynamic range of 0.05-100 ng/mL, a low detection limit of 16 pg/mL, high selectivity and good repeatability. The fabricated aptasensor has great potential development in the field of clinic disease diagnostics for AMI.
