Abstract:2D MXene-Ti3C2Tx has demonstrated promising application prospect in various fields, however, failing to function properly in biosensor setup due to restacking and anodic oxidation problem. Herein, we reported a synthesis study of covalently grafting MXene by first-generation poly(amidoamine) (PAMAM) dendrimers with AuNPs self-adsorption (AuNPs/MXene@PAMAM) to consider it as a potential functional nanoplatform for electrochemical sensing. In this hybrid system, MXene provided a highly conductive substrate and specific 2D architecture; PAMAM not only acted as an efficient stabilizer simultaneously suppressing serious restacking and oxidation of MXene under anodic potential and consequently improving the electrochemical performance but also as a signal amplifier offering large specific surface areas and massive amino terminals to adsorb Au nanoparticles (AuNPs) for the final formation of 3D hierarchical nanoarchitecture. This functional platform containing abundant active sites was allowed to directly and covalently immobilize thiol-linked anti-cardiac troponin T (cTnT) antibody as the biorecognition element to fabricate the cTnT immunosensor. Preliminary results indicated that this immunosensor was validated with a fast and sensitive response toward cTnT in presence of [Fe(CN)(6)](3-/4-) redox marker and displayed a wide detection range from 0.1 to 1000 ng/mL and a limit of detection (LOD) of 0.069 ng/mL. This high-performance MXene-based nanobiosensing platform has the potential applicability in bioanalysis of cTnT or other biomarkers and opens a new pathway for expanding its application in the electrochemical biosensing field.

Abstract:Photoelectrochemical (PEC) immunoassay is a burgeoning and promising bioanalytical method. However, the practical application of PEC still exist some challenges such as the inevitable damage of biomolecules caused by the PEC system and the unsatisfactory sensitivity for biomarkers with low abundance in real sample. To solve the problems, we integrated the cosensitized structure of Ag2S/ZnO nanocomposities as photoelectrode with photogenerated hole-induced chemical redox cycling amplification (CRCA) strategy to develop a split-type PEC immunosensor for cardiac troponin I (cTnI) with high sensitivity. Initially, the immunoreaction was carried out on the 96-well plates in which alkaline phosphatase (ALP) could catalyze ascorbic acid 2-phosphate (AAP) to generate the signal-reporting species ascorbic acid (AA). Subsequently, the AA participated and the tris (2-carboxyethyl) phosphine (TCEP) mediated chemical redox cycling reaction took place on the photoelectrode, thus leading to signal amplification. Under the optimized conditions, the immunosensor demonstrated a detection limit (LOD) of 3.0 × 10−15 g mL−1 with a detection range of 1.0 × 10−14 g mL−1 to 1.0 × 10−9 g mL−1 for cTnI. Impressively, the proposed method could determine the cTnI in human serum samples with high sensitivity and satisfactory accuracy. Considering the virtues of the photoelectrode and the chemical redox cycling strategy, the method would hold great potential for highly sensitive biosensing and bioanalysis.

Abstract:Here, a sandwich-type electrochemical immunosensor was constructed to detect cardiac troponin I (cTnI) via asymmetric bowl-shaped PdAgFe mesoporous nanospheres (PdAgFe ABMS) as a signal amplification label and core-shell cubic Au/Co-LDH@ZIF-67 as a substrate material. PdAgFe ABMS prepared by dual-template directional anisotropic island growth method has abundant mesoporous channels to accelerate molecular mass transfer in solution. In particular, the asymmetric bowl-like structure allows more active sites to be exposed, which improves the utilization of atoms to stabilize and high current response signals. The synergistic effect between PdAgFe increased the activation energy of the catalytic reaction and further amplified the current signal. In addition, the biosensing interface based on Au/Co-LDH@ZIF-67 not only exhibits a high electron transfer rate, but also can capture more bioactive molecules. Under optimal conditions, the constructed immunosensor was detected to exhibit a low limit of detection (LOD, 4.47 fg/mL) and a wide detection range (10 fg/mL - 100 ng/mL). This work provides an accurate and convenient scheme for the clinical detection of cTnI.

Abstract:A self-powered photoelectrochemical (PEC) immunosensor triple enhanced photoelectric response was proposed for the sensitive cardiac troponin (cTnI) detection. The self-powered PEC platform was constructed by sulfur indium copper hollow nanocage as a photocathode and hollow nanometer sphere heterojunction (In 2 S 3 @CdIn 2 S 4 ) as a photoanode, where the coulomb force of distinct photoelectrodes as the driving force effectively improves the photoelectric response without redox-mediator or external potential. In addition, the reverse transfer of photo-generated charge carriers increases the photoelectric response capacity due to the internal electric field effect in type II heterojunction. What is more exciting is that carefully designed hollow materials playing energy conversion function possess compelling properties. Given the advantages of multiple reflection effects in the cavity, thin-shell configuration and large surface, the designed hollow materials possessed a higher photon capture rate, a shorter-range transfer for photon-generated charges, and more active site points for the associated redox reactions , which facilitated the entire photoelectric response process. Based on the above strategies, the proposed PEC immunosensor achieved triple enhanced photoelectric response to present high sensitivity for detecting cTnI. The present study has gone some way to design nanomaterials with superior photoelectric activity, which paves a way for detecting other disease markers in high sensitivity.

Abstract:Cardiac troponin I (cTnI) is a gold-standard marker for the detection of acute myocardial infarction (AMI). Accurate and timely detection of cTnI is particularly important for AMI diagnosis and therapy. Herein, based on the dual-capacity (electro-catalysis and catalytic degradation) of @COF to toluidine blue (TB), we developed novel electrochemical and enzyme-linked immunosorbent assay (ELISA) methods for ultrasensitive detection of cTnI. In the electrochemical sensor, polyaniline-coated cerium oxide loaded with gold nanoparticles (@CeO 2 ) were used as a substrate platform for immobilizing antibodies (Ab1), and @COF loaded with the affinity peptide (Pep) of cTnI and TB (Pep/TB/@COF) was the probe material for signal amplification of the sensor. Using the 'peptide-cTnI-Ab1′ sandwich biosensor, we successfully detected cTnI in a concentration range of 0.00001–50 ng·mL −1 with an ultralow detection limit (LOD) of 3.3 fg·mL −1 . In addition, a sandwich ELISA was constructed by 96-well plate using Pep/@COF as the probe molecule for cTnI detection, showing a detection range of 0.00001–5 ng·mL −1 and a LOD of 3.3 fg·mL −1 . Both detection methods presented excellent accuracy, selectivity, reproducibility, and stability, providing a potential application in clinical diagnosis of AMI.

Abstract:A novel photoelectrochemical (PEC) immunosensor for the determination of cardiac troponin I (cTnI) was constructed. The flower-like stannic oxide (SnO 2 ) with large specific surface area was prepared by hydrothermal synthesis. Nitrogen-doped carbon quantum dots (NCQDs) with excellent surface property were used as a sensitizer for SnO 2 . Bismuth oxyiodide (BiOI) is a narrow band gap (1.83 eV) nanomaterial, which was firstly modified on NCQDs-sensitized SnO 2 through in situ growth method. After NCQDs with small size and BiOI nanoparticles are successively combined with SnO 2 , the SnO 2 /NCQDs/BiOI microflower was obtained, which possessed good photochemical properties. Using visible light as excitation source and ascorbic acid (AA) as electron donor, the ultrasensitive and quantitative determination of cTnI was realized by detecting the changes of photocurrent under different concentrations of cTnI. The PEC immunosensor showed a large-scaled response (0.001–100 ng mL −1 ) and a low detection limit (0.3 pg mL −1 ) under optimised experimental conditions. The sensor has potential clinical value in the prediction and diagnosis of cardiovascular diseases in elderly patients with diabetes. Graphical abstract

Abstract:Enhancement in the signals of electrochemical immunosensors via advanced nanomaterials is beneficial for developing immunoassay of cardiac troponin I (cTnI) with high sensitivity and selectivity. By virtue of K3[Fe(CN)6] as a signal probe, an advanced label-free electrochemical immunosensor was fabricated for ultrasensitive detection of cTnI with trimetallic alloyed AuPtPd porous fluffy-like nanodendrites (AuPtPd FNDs), followed by assembly of the antibody (Ab). By virtue of the structural advantages, electronic effects and synergetic catalysis of the trimetals, such nanostructures showed large specific surface area to increase the loading of the cTnI Ab and amplify the electrochemical signals of the probe. The sensor displayed a wide linear range (0.01 ~ 100.0 ng mL−1) and a low detection limit (LOD = 3 pg mL−1) under the optimal conditions, which showed great potential for detecting cTnI in serum samples. Furthermore, this biosensor provides an effective detection platform for analysis of other cardiac markers in practical samples.

Abstract:An electrochemical immunosensor based on the novel high efficiency catalytic cycle amplification strategy for the sensitive detection of cardiac troponin I (cTnI). With its variable valence metal elements and spiny yolk structure, the Cu2O/CuO@CeO2 nanohybrid exhibits high speed charge mobility and exceptional electrochemical performance. Notably, fluorite-like cubic crystal CeO2 shell would undergo redox reaction with Cu2O core, which successfully ensures the continuous recycling occurrence of "fresh" Cu (II)/Cu (I) and Ce (Ⅳ)/Ce (Ⅲ) pairs at the electrode interface. The "fresh" active sites continue to emerge constantly, resulting in a significant increase in the current signal. In light of the electrochemical characterization, the electron transfer pathway and catalytic cycle mechanism among CeO2, Cu2O and CuO were further discussed. The developed electrochemical immunosensor detected cTnI from 100 fg/mL to 100 ng/mL with a LOD of 15.85 fg/mL under optimal conditions. The analysis results indicate that the immunosensor would hold promise for broad application prospects in the biological detection for other biomarkers.

Abstract:CdIn2S4 and zinc tetrakis(4-carboxyphenyl)porphyrin (ZnTCPP) were synthesized by hydrothermal method, and an organic dye-sensitized inorganic semiconductor ZnTCPP/CdIn2S4 type II heterojunction was constructed on a fluorine-doped tin oxide (FTO) substrate electrode. A sandwich immunostructure for signal-attenuation photoelectrochemical (PEC) detection of cardiac troponin I (cTnI) was constructed using the ZnTCPP/CdIn2S4/FTO photoanode and a horseradish peroxidase (HRP)-ZnFe2O4-Ab2-bovine serum albumin (BSA) immunolabeling complex. The bioenzyme HRP and the HRP-like nanozyme ZnFe2O4 can co-catalyze the oxidation of 4-chloro-1-naphthol (4-CN) by H2O2 to produce an insoluble precipitate on the photoanode, thus notably reducing the anodic photocurrent for quantitative determination of cTnI. Under the optimal conditions, the photocurrent at 0 V vs. SCE in 0.1 M phosphate buffer solution (pH 7.40) containing 0.1 M ascorbic acid was linear with the logarithm of cTnI concentration from 500 fg mL-1 to 50.0 ng mL-1, and the limit of detection (LOD, S/N = 3) is 0.15 pg mL-1. Spiked recoveries were 95.1% ~ 104% for assay of cTnI in human serum samples.

Abstract:A novel multiple amplification strategy for electrochemical immunoassay was developed elaborately. The realization of this strategy is based on the high efficiency catalysis of boron nitrogen double-doped carbon loaded trimetallic PtPdCu mulberry-like nanospheres (PtPdCu/BNC) and the satisfactory conductivity of gold nanoparticles modified with sulfur nitrogen co-doped hollow porous carbon (). Single crystal anisotropic Pt octahedral seeds were synthesized with sodium citrate as shape-directing agent, and then three metals were grown in situ to prepare the trimetallic PtPdCu mulberry nanospheres, which had excellent utilization of atoms and a significant number of catalytic active centers. Boron nitrogen double-doped carbon (BNC) with abundant free electrons and high electron density micro-mesoporous can be used as the supported dispersion material of PtPdCu MBNSs, further catalytically enrich H 2 O 2 . Moreover, the etching vulcanization of small ZIF template and the co-doping of sulfur and nitrogen endow sulfur nitrogen co-doped hollow porous carbon (SNHC) with shorter the diffusion distance of charge carrier and lower mass transfer resistance. Under optimal conditions, the developed immunosensor exhibits good sensing performance with a detection limit of 4.27 fg⋅mL −1 , and the wide linear dynamic range (10 fg⋅mL −1 –100 ng⋅mL −1 ). This study provides a possible strategy for early diagnosis and postoperative monitoring of AMI.

Abstract:Based on the necessity and urgency of cardiac troponin I (cTnI) detection for the diagnosis of myocardial infarction, a novel unlabeled photoelectrochemical (PEC) immunosensor has been developed to detect cTnI rapidly and sensitively. Silicon nanowire arrays (SiNWs) were prepared via metal-assisted chemical etching. To improve the stability and sensitivity towards cTnI sensing, the surface of silicon nanowire arrays were coated with polydopamine by an in situ growth method. PDA was uniformly modified on the nanowire surface to provide a reliable active site for antibody binding. The linear dynamic range of the cTnI detection method was 0.005-10 ng mL(-1), and the detection limit was 1.47 pg mL(-1). The designed PEC immunosensor exhibited good sensitivity, selectivity, stability and reproducibility. The electrode enabled label-free detection and provided a new route to realize point-of-care testing of cTnI.

Abstract:The content of cardiac troponin I (CTnI) in human blood is the key factor in judging acute myocardial infarction (AMI). In order to detect the content of CTnI, we constructed a sandwich-type electrochemical immunosensor based on hydrogen peroxide (H2O2) as a signal source. Dendritic platinum-copper alloy nanoparticles (DPCN) loaded on molybdenum disulfide (MoS2) nanosheets (DPCN/MoS2) as secondary antibodies (Ab2) label provided signal amplification. The hollow three-dimensional (3D) pyramid-shaped structure of DPCN exposed abundant active sites and exhibited excellent catalytic properties. MoS2 nanosheets with flower-like structure and a larger specific surface area can effectively load more DPCN. The combination of MoS2 and DPCN enhanced the catalytic performance of DPCN/MoS2 towards H2O2 reduction and realized signal amplification. For the substrate material, the two-dimensional (2D) metal-organic framework (Co-BDC, 1,4-benzenedicarboxylate is abbreviated as BDC) was hybridized with MoS2 nanosheets to load gold nanoparticles (Au NPs). The obtained Au/Co-BDC/MoS2 had low catalytic activity and excellent electrical conductivity, which was used to load primary antibodies (Ab1) to effectively enhance the sensitivity. Under the best conditions, we constructed the immunosensor with the detection range of 10 fg/mL to 100 ng/mL and the limit of detection (LOD) of 3.02 fg/mL. At the same time, the content of CTnI in human serum was tested with satisfactory results. Therefore, the constructed immunosensor has important significance in the sensitive and accurate detection of CTnI and early diagnosis of AMI.

Abstract:An interference-free photoelectrochemical (PEC) immunoassay was developed for cardiac troponin I (cTnI) detection. Covalent linkage of cTnI antibody to carboxymethylated (CM-) dextran pre-immobilized onto a gold nanoparticles (AuNPs)-modified TiO2 nanotube array (NTA) affords five consecutive analyte captures with surface regenerations in between. Changes in the photocurrents at this photoanode before and after cTnI captures can be well fitted with the Langmuir isotherm from 0.220 pM to 2.20 nM cTnI. Owing to the inherently high sensitivity of the PEC detection, the detection limit (2.20 pg/mL) is lower than the range attainable with the enzyme-linked immunosorbent assay (ELISA) (6.00–40.0 pg/mL). Furthermore, CM-dextran prevents species in complex biological matrices from nonspecifically adsorbing onto the sensor surface, a feature not attainable with uncoated semiconductor electrodes or those coated with non-hydrogel-based chemical modifiers. The excellent anti-fouling property of dextran hydrogel allowed us to validate the accuracy of our regenerable sensors through a comparison of PEC immunoassays of patient sera to those of ELISA.

Abstract:Cardiac troponin I (cTnI) monitoring is of great value in the clinical diagnosis of acute myocardial infarction (AMI). In this paper, a highly sensitive electrochemical aptamer sensor using polystyrene (PS) microspheres as the electrode substrate material in combination with Prussian blue (PB) and gold nanoparticles (AuNPs) was demonstrated for the sensitive and label-free determination of cTnI. PS microspheres were synthesized by emulsion polymerization and then dropped onto the glassy carbon electrode (GCE); PB and AuNPs were electrodeposited on the electrode in corresponding electrolyte solutions step by step. The PS microsphere substrate provided a large surface area for the loading mass of the biological affinity aptamers, while the PB layer improved the electrical conductivity of the modified electrode, and the electroactive AuNPs exhibited excellent catalytic performance for the subsequent electrochemical measurements. In view of the above mentioned AuNPs/PB/PS/GCE sensing platform, the fabricated label-free electrochemical aptamer sensor exhibited a wide detection range of 10 fg/mL~1.0 μg/mL and a low detection limit of 2.03 fg/mL under the optimal conditions. Furthermore, this biosensor provided an effective detection platform for the analysis of cTnI in serum samples. The introduction of this sensitive electrochemical aptamer sensor provides a reference for clinically sensitive detection of cTnI.

Stable PDMS-AuNPs composite film was simply prepared with dropping HAuCl4 solution in PDMS microchip, gold nanoparticles were both used as nuclei for silver enhancement and as basis for adsorption of antibody, immunoassay was implemented after BSA blocking. Because of the inhibition ability difference for silver enhancement between BSA and antibody-antigen complex, darkness of silver deposition was relative to the amount of antigen. The proposed colorimetric method was applied for clinical sample cTnI detection, results were consistent with that from enzyme-linked immunosorbent assay (ELISA). The composite film could be coupled with digital transmission of images for remote monitoring system in diagnosis, food control, and environmental analysis.
Dual-mode of electrochemical-colorimetric imprinted sensing strategy based on self-sacrifice beacon for diversified determination of cardiac troponin I in serum

Abstract:Diversified determination of cardiac troponin I (cTnI) in serum is of great significance for the clinical diagnose and daily monitoring of acute myocardial infarction. Herein, an electrochemical-colorimetric dual-mode imprinted sensing strategy was proposed. Firstly, the aptamer-functionalized Fe3+-polydopamine (3+-PDA) was constructed as the self-sacrifice beacon. Furthermore, an epitope magnetic molecularly imprinted polymer was prepared to recognize cTnI. Once the cTnI was captured, the beacon was then bound to it through the aptamer, which formed a sandwich-like complex. Under acidic condition, the 3+-PDA was disintegrated to release large amount of Fe3+, and further converted to Prussian blue (PB). On one hand, the electrochemical response of the PB was sensitive to the cTnI amount. On the other hand, the addition of the yellow K3[Fe(CN)6] into different amounts of the produced PB was able to result in different colors, which enabled the visual detection of cTnI. Under the optimal condition, the sensing strategy exhibited a linear range from 1.0 × 10−2 to 1.0 × 103 ng mL−1, and the detection limits were 3.2 and 7.4 pg mL−1 for the electrochemical and colorimetric modes, respectively. Besides, It is demonstrated that the dual-mode strategy shows favorable selectivity and stability in the assays, indicating a diversified application prospect in both clinic analysis and daily care.

Abstract:Due to the short course and high mortality of acute myocardial infarction (AMI) which will cause irreversible myocardial necrosis to induce cardiac arrest, its early diagnosis within minutes is always a challenge in clinical emergency. In this work, an ultrafast and ultrasensitive aptasensor is designed to achieve quantitatively detection of cardiac troponin I (cTnI) which is proved as a highly specific and sensitive biomarker to AMI, and within only 4 min, the cTnI concentration can be accurately reported. This aptasensor is constructed via an in situ growth of the oriented Pt‐polyaniline (PANI) needle‐like nanoarrays providing both high electrocatalysis and abundant active sites as the signal magnifier and DNA carrier. A target‐induced strand release strategy is proposed to remarkably accelerate the recognition process of cTnI through the specific design of DNA stand for binding. The as‐prepared aptasensor can exhibit an ultrawide linear detection range of cTnI from 1 pg mL−1 to 100 ng mL−1 satisfying the clinical demand for AMI diagnosis, together with excellent selectivity among various biomarkers. Based on its fast recognition ability, this aptasensor presents a high accuracy in the practical detection of human serum to prove its promising ability in the early and on‐site diagnosis of AMI.
