Abstract:2D MXene-Ti3C2Tx has demonstrated promising application prospect in various fields, however, failing to function properly in biosensor setup due to restacking and anodic oxidation problem. Herein, we reported a synthesis study of covalently grafting MXene by first-generation poly(amidoamine) (PAMAM) dendrimers with AuNPs self-adsorption (AuNPs/MXene@PAMAM) to consider it as a potential functional nanoplatform for electrochemical sensing. In this hybrid system, MXene provided a highly conductive substrate and specific 2D architecture; PAMAM not only acted as an efficient stabilizer simultaneously suppressing serious restacking and oxidation of MXene under anodic potential and consequently improving the electrochemical performance but also as a signal amplifier offering large specific surface areas and massive amino terminals to adsorb Au nanoparticles (AuNPs) for the final formation of 3D hierarchical nanoarchitecture. This functional platform containing abundant active sites was allowed to directly and covalently immobilize thiol-linked anti-cardiac troponin T (cTnT) antibody as the biorecognition element to fabricate the cTnT immunosensor. Preliminary results indicated that this immunosensor was validated with a fast and sensitive response toward cTnT in presence of [Fe(CN)(6)](3-/4-) redox marker and displayed a wide detection range from 0.1 to 1000 ng/mL and a limit of detection (LOD) of 0.069 ng/mL. This high-performance MXene-based nanobiosensing platform has the potential applicability in bioanalysis of cTnT or other biomarkers and opens a new pathway for expanding its application in the electrochemical biosensing field.

Abstract:CdIn2S4 and zinc tetrakis(4-carboxyphenyl)porphyrin (ZnTCPP) were synthesized by hydrothermal method, and an organic dye-sensitized inorganic semiconductor ZnTCPP/CdIn2S4 type II heterojunction was constructed on a fluorine-doped tin oxide (FTO) substrate electrode. A sandwich immunostructure for signal-attenuation photoelectrochemical (PEC) detection of cardiac troponin I (cTnI) was constructed using the ZnTCPP/CdIn2S4/FTO photoanode and a horseradish peroxidase (HRP)-ZnFe2O4-Ab2-bovine serum albumin (BSA) immunolabeling complex. The bioenzyme HRP and the HRP-like nanozyme ZnFe2O4 can co-catalyze the oxidation of 4-chloro-1-naphthol (4-CN) by H2O2 to produce an insoluble precipitate on the photoanode, thus notably reducing the anodic photocurrent for quantitative determination of cTnI. Under the optimal conditions, the photocurrent at 0 V vs. SCE in 0.1 M phosphate buffer solution (pH 7.40) containing 0.1 M ascorbic acid was linear with the logarithm of cTnI concentration from 500 fg mL-1 to 50.0 ng mL-1, and the limit of detection (LOD, S/N = 3) is 0.15 pg mL-1. Spiked recoveries were 95.1% ~ 104% for assay of cTnI in human serum samples.

Abstract:A self-powered photoelectrochemical (PEC) immunosensor triple enhanced photoelectric response was proposed for the sensitive cardiac troponin (cTnI) detection. The self-powered PEC platform was constructed by sulfur indium copper hollow nanocage as a photocathode and hollow nanometer sphere heterojunction (In 2 S 3 @CdIn 2 S 4 ) as a photoanode, where the coulomb force of distinct photoelectrodes as the driving force effectively improves the photoelectric response without redox-mediator or external potential. In addition, the reverse transfer of photo-generated charge carriers increases the photoelectric response capacity due to the internal electric field effect in type II heterojunction. What is more exciting is that carefully designed hollow materials playing energy conversion function possess compelling properties. Given the advantages of multiple reflection effects in the cavity, thin-shell configuration and large surface, the designed hollow materials possessed a higher photon capture rate, a shorter-range transfer for photon-generated charges, and more active site points for the associated redox reactions , which facilitated the entire photoelectric response process. Based on the above strategies, the proposed PEC immunosensor achieved triple enhanced photoelectric response to present high sensitivity for detecting cTnI. The present study has gone some way to design nanomaterials with superior photoelectric activity, which paves a way for detecting other disease markers in high sensitivity.

Abstract:A novel photoelectrochemical (PEC) immunosensor for the determination of cardiac troponin I (cTnI) was constructed. The flower-like stannic oxide (SnO 2 ) with large specific surface area was prepared by hydrothermal synthesis. Nitrogen-doped carbon quantum dots (NCQDs) with excellent surface property were used as a sensitizer for SnO 2 . Bismuth oxyiodide (BiOI) is a narrow band gap (1.83 eV) nanomaterial, which was firstly modified on NCQDs-sensitized SnO 2 through in situ growth method. After NCQDs with small size and BiOI nanoparticles are successively combined with SnO 2 , the SnO 2 /NCQDs/BiOI microflower was obtained, which possessed good photochemical properties. Using visible light as excitation source and ascorbic acid (AA) as electron donor, the ultrasensitive and quantitative determination of cTnI was realized by detecting the changes of photocurrent under different concentrations of cTnI. The PEC immunosensor showed a large-scaled response (0.001–100 ng mL −1 ) and a low detection limit (0.3 pg mL −1 ) under optimised experimental conditions. The sensor has potential clinical value in the prediction and diagnosis of cardiovascular diseases in elderly patients with diabetes. Graphical abstract

Abstract:An interference-free photoelectrochemical (PEC) immunoassay was developed for cardiac troponin I (cTnI) detection. Covalent linkage of cTnI antibody to carboxymethylated (CM-) dextran pre-immobilized onto a gold nanoparticles (AuNPs)-modified TiO2 nanotube array (NTA) affords five consecutive analyte captures with surface regenerations in between. Changes in the photocurrents at this photoanode before and after cTnI captures can be well fitted with the Langmuir isotherm from 0.220 pM to 2.20 nM cTnI. Owing to the inherently high sensitivity of the PEC detection, the detection limit (2.20 pg/mL) is lower than the range attainable with the enzyme-linked immunosorbent assay (ELISA) (6.00–40.0 pg/mL). Furthermore, CM-dextran prevents species in complex biological matrices from nonspecifically adsorbing onto the sensor surface, a feature not attainable with uncoated semiconductor electrodes or those coated with non-hydrogel-based chemical modifiers. The excellent anti-fouling property of dextran hydrogel allowed us to validate the accuracy of our regenerable sensors through a comparison of PEC immunoassays of patient sera to those of ELISA.

Abstract:Photoelectrochemical (PEC) immunoassay has received increasing attention owing to its good analytical performance and attractive potential for future protein assay. This Letter represents a novel and general strategy for elegant PEC immunoassay of the important cardiac marker troponin T (cTnT) at neutral conditions. Specifically, we first developed an efficient CdS quantum dots (QDs)/TiO2 nanoparticles (NPs) photoelectrode, on the basis of which an exquisite β-galactosidase (β-Gal) catalytic system was integrated with sandwich immunobinding for probing cTnT. In pH 7.4, β-Gal could catalyze the conversion of p-aminophenyl galactopyranoside (PAPG) to p-aminophenol (PAP), which could be easily photo-oxidized to p-quinone imine (PQI). Because the resulting photocurrent was directly related with the target concentration, an innovative PEC immunoassay could be realized for cTnT detection. The neutral operating condition of this protocol would greatly contribute to its wide applicability for protein assay. This work provides the first PEC immunoassay toward cardiac marker and, more significantly, opens a different perspective for future PEC immunoassay development through a general sensing protocol.

Abstract:Photoelectrochemical (PEC) immunoassay is a burgeoning and promising bioanalytical method. However, the practical application of PEC still exist some challenges such as the inevitable damage of biomolecules caused by the PEC system and the unsatisfactory sensitivity for biomarkers with low abundance in real sample. To solve the problems, we integrated the cosensitized structure of Ag2S/ZnO nanocomposities as photoelectrode with photogenerated hole-induced chemical redox cycling amplification (CRCA) strategy to develop a split-type PEC immunosensor for cardiac troponin I (cTnI) with high sensitivity. Initially, the immunoreaction was carried out on the 96-well plates in which alkaline phosphatase (ALP) could catalyze ascorbic acid 2-phosphate (AAP) to generate the signal-reporting species ascorbic acid (AA). Subsequently, the AA participated and the tris (2-carboxyethyl) phosphine (TCEP) mediated chemical redox cycling reaction took place on the photoelectrode, thus leading to signal amplification. Under the optimized conditions, the immunosensor demonstrated a detection limit (LOD) of 3.0 × 10−15 g mL−1 with a detection range of 1.0 × 10−14 g mL−1 to 1.0 × 10−9 g mL−1 for cTnI. Impressively, the proposed method could determine the cTnI in human serum samples with high sensitivity and satisfactory accuracy. Considering the virtues of the photoelectrode and the chemical redox cycling strategy, the method would hold great potential for highly sensitive biosensing and bioanalysis.

Abstract:Here, a sandwich-type electrochemical immunosensor was constructed to detect cardiac troponin I (cTnI) via asymmetric bowl-shaped PdAgFe mesoporous nanospheres (PdAgFe ABMS) as a signal amplification label and core-shell cubic Au/Co-LDH@ZIF-67 as a substrate material. PdAgFe ABMS prepared by dual-template directional anisotropic island growth method has abundant mesoporous channels to accelerate molecular mass transfer in solution. In particular, the asymmetric bowl-like structure allows more active sites to be exposed, which improves the utilization of atoms to stabilize and high current response signals. The synergistic effect between PdAgFe increased the activation energy of the catalytic reaction and further amplified the current signal. In addition, the biosensing interface based on Au/Co-LDH@ZIF-67 not only exhibits a high electron transfer rate, but also can capture more bioactive molecules. Under optimal conditions, the constructed immunosensor was detected to exhibit a low limit of detection (LOD, 4.47 fg/mL) and a wide detection range (10 fg/mL - 100 ng/mL). This work provides an accurate and convenient scheme for the clinical detection of cTnI.

Abstract:Cardiac troponin I (cTnI) is a gold-standard marker for the detection of acute myocardial infarction (AMI). Accurate and timely detection of cTnI is particularly important for AMI diagnosis and therapy. Herein, based on the dual-capacity (electro-catalysis and catalytic degradation) of @COF to toluidine blue (TB), we developed novel electrochemical and enzyme-linked immunosorbent assay (ELISA) methods for ultrasensitive detection of cTnI. In the electrochemical sensor, polyaniline-coated cerium oxide loaded with gold nanoparticles (@CeO 2 ) were used as a substrate platform for immobilizing antibodies (Ab1), and @COF loaded with the affinity peptide (Pep) of cTnI and TB (Pep/TB/@COF) was the probe material for signal amplification of the sensor. Using the 'peptide-cTnI-Ab1′ sandwich biosensor, we successfully detected cTnI in a concentration range of 0.00001–50 ng·mL −1 with an ultralow detection limit (LOD) of 3.3 fg·mL −1 . In addition, a sandwich ELISA was constructed by 96-well plate using Pep/@COF as the probe molecule for cTnI detection, showing a detection range of 0.00001–5 ng·mL −1 and a LOD of 3.3 fg·mL −1 . Both detection methods presented excellent accuracy, selectivity, reproducibility, and stability, providing a potential application in clinical diagnosis of AMI.

Abstract:An electrochemical immunosensor based on the novel high efficiency catalytic cycle amplification strategy for the sensitive detection of cardiac troponin I (cTnI). With its variable valence metal elements and spiny yolk structure, the Cu2O/CuO@CeO2 nanohybrid exhibits high speed charge mobility and exceptional electrochemical performance. Notably, fluorite-like cubic crystal CeO2 shell would undergo redox reaction with Cu2O core, which successfully ensures the continuous recycling occurrence of "fresh" Cu (II)/Cu (I) and Ce (Ⅳ)/Ce (Ⅲ) pairs at the electrode interface. The "fresh" active sites continue to emerge constantly, resulting in a significant increase in the current signal. In light of the electrochemical characterization, the electron transfer pathway and catalytic cycle mechanism among CeO2, Cu2O and CuO were further discussed. The developed electrochemical immunosensor detected cTnI from 100 fg/mL to 100 ng/mL with a LOD of 15.85 fg/mL under optimal conditions. The analysis results indicate that the immunosensor would hold promise for broad application prospects in the biological detection for other biomarkers.

Abstract:A suitable electron donor, which guarantees the stability of the whole system, is considered as the driving force of the PEC sensor. Nowadays, searching appropriate electron donor is still one of the orientations to explorate in the field of sensor. Na48[H496Mo368O1464S48]·ca.1000H2O (abbr. {Mo368}), as a type of polyoxometalate, has perfect morphology, definite size and unique electronic property. Due to the prominent water solubility, {Mo368} usually releases small cations and exists as large anions in the ultrapure water. The interesting property endows {Mo368} with excellent reducibility, which provides great feasibility to become an outstanding electron donor. In addition, FeOOH prepared through a simple operation owns high adsorption capacity, which ensures the fastness of other materials. Subsequently, the narrow band-gap of Bi2S3 and the unique noble metal properties of Au nanoparticles are utilized to co-sensitize FeOOH to improve the light-harvesting capability and photoelectric conversion efficiency. Combined with the specificity recognition of antigen and antibody, a novel photoelectrochemical sensor is constructed with a wide detection range of 1.00 pg mL−1 - 100 ng mL−1 and low detection limit (0.76 pg mL−1), which achieves the sensitive detection of cardiac troponin I in early diagnosis of cardiovascular disease.

Abstract:Based on the necessity and urgency of cardiac troponin I (cTnI) detection for the diagnosis of myocardial infarction, a novel unlabeled photoelectrochemical (PEC) immunosensor has been developed to detect cTnI rapidly and sensitively. Silicon nanowire arrays (SiNWs) were prepared via metal-assisted chemical etching. To improve the stability and sensitivity towards cTnI sensing, the surface of silicon nanowire arrays were coated with polydopamine by an in situ growth method. PDA was uniformly modified on the nanowire surface to provide a reliable active site for antibody binding. The linear dynamic range of the cTnI detection method was 0.005-10 ng mL(-1), and the detection limit was 1.47 pg mL(-1). The designed PEC immunosensor exhibited good sensitivity, selectivity, stability and reproducibility. The electrode enabled label-free detection and provided a new route to realize point-of-care testing of cTnI.

Abstract:The sensitivity and specificity of a semiconductor photoelectrochemical (PEC) aptamer biosensor are determined by the separation and transport of the photoinduced carriers as well as aptamer probe immobilization. In this study, an in situ thermal transformation organic polymer strategy was employed to produce an ∼2.5 nm carbon quantum layer on the surface of the BiVO 4 (BVO) photoanode. Experimental tests and theoretical calculations have revealed that this carbon quantum layer-coated BVO(C@BVO) heterostructure could generate surface charge depletion regions through an interface nephelauxetic effect. These charge depletion regions facilitated the efficient immobilization of DNA aptamer probes of the acute myocardial infarction biomarker cardiac troponin I (cTnI), while showing almost no immobilization capability on a pure-phase C quantum layer or BVO crystals. Simultaneously, the formation of the C@BVO heterostructure also enhanced the directional transport of photo-generated holes from BVO to the C quantum layer. Due to the above reasons, the C@BVO PEC aptamer biosensor achieved a linear detection range for cTnI from 10 −14 g L −1 to 10 −10 g L −1 , with a record detection limit (LOD) of ∼0.33 × 10 −14 g L −1 ( N > 3). Meanwhile, the biosensor also demonstrated well the detection reproducibility and specificity for cTnI detection. Therefore, the strategy of using a carbon quantum layer-coated PEC electrode shows good potential to develop PEC biosensors with high sensitivity, specificity, and robustness.

Abstract:The development of photoelectrochemical (PEC) biosensors plays a critical role in enabling timely intervention and personalized treatment for cardiac injury. Herein, a novel approach is presented for the fabrication of highly sensitive PEC biosensor employing Bi2O3/MgIn2S4 heterojunction for the ultrasensitive detection of heart fatty acid binding protein (H-FABP). The Bi2O3/MgIn2S4 heterojunction, synthesized through in-situ growth of MgIn2S4 on Bi2O3 nanoplates, offers superior attributes including a larger specific surface area and more homogeneous distribution, leading to enhanced sensing sensitivity. The well-matched valence and conduction bands of Bi2O3 and MgIn2S4 effectively suppress the recombination of photogenerated carriers and facilitate electron transfer, resulting in a significantly improved photocurrent signal response. And the presence of the secondary antibody marker (ZnSnO3) introduces steric hindrance that hinders electron transfer between ascorbic acid and the photoelectrode, leading to a reduction in photocurrent signal. Additionally, the competition between the ZnSnO3 marker and the Bi2O3/MgIn2S4 heterojunction material for the excitation light source further diminishes the photocurrent signal response. After rigorous repeatability and selectivity tests, the PEC biosensor exhibited excellent performance, and the linear detection range of the biosensor was determined to be 0.05 pg/mL to 100 ng/mL with a remarkable detection limit of 0.029 pg/mL (S/N = 3).
Copper(II)-Tannic Acid@Cu with In Situ Grown Gold Nanoparticles as a Bifunctional Matrix for Facile Construction of Label-Free and Ultrasensitive Electrochemical cTnI Immunosensor

Abstract:Sensitive detection of cardiac troponin I (cTnI) is of great significance in the diagnosis of a fatal acute myocardial infarction. A redox-active nanocomposite of copper(II)-tannic acid@Cu (CuTA@Cu) was herein prepared on the surface of a glassy carbon electrode by electrochemical deposition of metallic copper combined with a metal stripping strategy. Then, HAuCl4 was in situ reduced to gold nanoparticles (AuNPs) by strong reductive catechol groups in the TA ligand. The AuNPs/CuTA@Cu composite was further utilized as a bifunctional matrix for the immobilization of the cTnI antibody (anti-cTnI), producing an electrochemical immunosensor. Electrochemical tests show that the immunoreaction between anti-cTnI and target cTnI can cause a significant reduction of the electrochemical signal of CuTA@Cu. It can be attributed to the insulating characteristic of the immunocomplex and its barrier effect to the electrolyte ion diffusion. From the signal changes of CuTA@Cu, cTnI can be analyzed in a wide range from 10 fg mL-1 to 10 ng mL-1, with an ultralow detection limit of 0.65 fg mL-1. The spiked recovery assays show that the immunosensor is reliable for cTnI determination in human serum samples, demonstrating its promising application in the early clinical diagnosis of myocardial infarction.
Ultrasensitive Photoelectrochemical Biosensing Platform for Detecting N-Terminal Pro-brain Natriuretic Peptide Based on SnO2/SnS2/mpg-C3N4 Amplified by PbS/SiO2.

Abstract:A sandwich-type photoelectrochemical (PEC) immunosensor for detecting N-terminal pro-brain natriuretic peptide (NT-proBNP) was constructed on the basis of SnO2/SnS2/mpg-C3N4 nanocomposites and PbS/SiO2, with the former as a photoactive matrix and the latter as an efficient quencher. SnO2/SnS2/mpg-C3N4 was synthesized via in situ growth of SnO2 and SnS2 on mesoporous graphene like C3N4 nanocomposites (mpg-C3N4). Specifically, SnO2/SnS2/mpg-C3N4 exhibited intense PEC signal responses, which are tens of times stronger than its each single component. Because of its superior performance, SnO2/SnS2/mpg-C3N4 was applied as a photoactive matrix and signal indicator for fabricating PEC immunosensor. Interestingly, the excellent PEC signals from SnO2/SnS2/mpg-C3N4 could be reduced severely with the addition of PbS/SiO2. Hence, the secondary antibody bioconjugates (PbS/SiO2-Ab2) were prepared as an efficient quencher. The mechanism of the quench reaction was further discussed in detail. On the basis of the interaction between the matrix and the quencher, the NT-proBNP immunosensor was fabricated and a wide linear range of 0.1 pg·mL-1 to 50 ng·mL-1 was obtained with a low detection limit of 0.05 pg·mL-1. Additionally, the PEC immunosensor manifested good stability, reproducibility, and selectivity, which could underlie robust platforms for detecting multitudinous biomarkers or other targets of interest.
