Abstract:A self-powered photoelectrochemical (PEC) immunosensor triple enhanced photoelectric response was proposed for the sensitive cardiac troponin (cTnI) detection. The self-powered PEC platform was constructed by sulfur indium copper hollow nanocage as a photocathode and hollow nanometer sphere heterojunction (In 2 S 3 @CdIn 2 S 4 ) as a photoanode, where the coulomb force of distinct photoelectrodes as the driving force effectively improves the photoelectric response without redox-mediator or external potential. In addition, the reverse transfer of photo-generated charge carriers increases the photoelectric response capacity due to the internal electric field effect in type II heterojunction. What is more exciting is that carefully designed hollow materials playing energy conversion function possess compelling properties. Given the advantages of multiple reflection effects in the cavity, thin-shell configuration and large surface, the designed hollow materials possessed a higher photon capture rate, a shorter-range transfer for photon-generated charges, and more active site points for the associated redox reactions , which facilitated the entire photoelectric response process. Based on the above strategies, the proposed PEC immunosensor achieved triple enhanced photoelectric response to present high sensitivity for detecting cTnI. The present study has gone some way to design nanomaterials with superior photoelectric activity, which paves a way for detecting other disease markers in high sensitivity.

Abstract:Photoelectrochemical (PEC) immunoassay is a burgeoning and promising bioanalytical method. However, the practical application of PEC still exist some challenges such as the inevitable damage of biomolecules caused by the PEC system and the unsatisfactory sensitivity for biomarkers with low abundance in real sample. To solve the problems, we integrated the cosensitized structure of Ag2S/ZnO nanocomposities as photoelectrode with photogenerated hole-induced chemical redox cycling amplification (CRCA) strategy to develop a split-type PEC immunosensor for cardiac troponin I (cTnI) with high sensitivity. Initially, the immunoreaction was carried out on the 96-well plates in which alkaline phosphatase (ALP) could catalyze ascorbic acid 2-phosphate (AAP) to generate the signal-reporting species ascorbic acid (AA). Subsequently, the AA participated and the tris (2-carboxyethyl) phosphine (TCEP) mediated chemical redox cycling reaction took place on the photoelectrode, thus leading to signal amplification. Under the optimized conditions, the immunosensor demonstrated a detection limit (LOD) of 3.0 × 10−15 g mL−1 with a detection range of 1.0 × 10−14 g mL−1 to 1.0 × 10−9 g mL−1 for cTnI. Impressively, the proposed method could determine the cTnI in human serum samples with high sensitivity and satisfactory accuracy. Considering the virtues of the photoelectrode and the chemical redox cycling strategy, the method would hold great potential for highly sensitive biosensing and bioanalysis.

Abstract:CdIn2S4 and zinc tetrakis(4-carboxyphenyl)porphyrin (ZnTCPP) were synthesized by hydrothermal method, and an organic dye-sensitized inorganic semiconductor ZnTCPP/CdIn2S4 type II heterojunction was constructed on a fluorine-doped tin oxide (FTO) substrate electrode. A sandwich immunostructure for signal-attenuation photoelectrochemical (PEC) detection of cardiac troponin I (cTnI) was constructed using the ZnTCPP/CdIn2S4/FTO photoanode and a horseradish peroxidase (HRP)-ZnFe2O4-Ab2-bovine serum albumin (BSA) immunolabeling complex. The bioenzyme HRP and the HRP-like nanozyme ZnFe2O4 can co-catalyze the oxidation of 4-chloro-1-naphthol (4-CN) by H2O2 to produce an insoluble precipitate on the photoanode, thus notably reducing the anodic photocurrent for quantitative determination of cTnI. Under the optimal conditions, the photocurrent at 0 V vs. SCE in 0.1 M phosphate buffer solution (pH 7.40) containing 0.1 M ascorbic acid was linear with the logarithm of cTnI concentration from 500 fg mL-1 to 50.0 ng mL-1, and the limit of detection (LOD, S/N = 3) is 0.15 pg mL-1. Spiked recoveries were 95.1% ~ 104% for assay of cTnI in human serum samples.

Abstract:A novel photoelectrochemical (PEC) immunosensor for the determination of cardiac troponin I (cTnI) was constructed. The flower-like stannic oxide (SnO 2 ) with large specific surface area was prepared by hydrothermal synthesis. Nitrogen-doped carbon quantum dots (NCQDs) with excellent surface property were used as a sensitizer for SnO 2 . Bismuth oxyiodide (BiOI) is a narrow band gap (1.83 eV) nanomaterial, which was firstly modified on NCQDs-sensitized SnO 2 through in situ growth method. After NCQDs with small size and BiOI nanoparticles are successively combined with SnO 2 , the SnO 2 /NCQDs/BiOI microflower was obtained, which possessed good photochemical properties. Using visible light as excitation source and ascorbic acid (AA) as electron donor, the ultrasensitive and quantitative determination of cTnI was realized by detecting the changes of photocurrent under different concentrations of cTnI. The PEC immunosensor showed a large-scaled response (0.001–100 ng mL −1 ) and a low detection limit (0.3 pg mL −1 ) under optimised experimental conditions. The sensor has potential clinical value in the prediction and diagnosis of cardiovascular diseases in elderly patients with diabetes. Graphical abstract

Abstract:An interference-free photoelectrochemical (PEC) immunoassay was developed for cardiac troponin I (cTnI) detection. Covalent linkage of cTnI antibody to carboxymethylated (CM-) dextran pre-immobilized onto a gold nanoparticles (AuNPs)-modified TiO2 nanotube array (NTA) affords five consecutive analyte captures with surface regenerations in between. Changes in the photocurrents at this photoanode before and after cTnI captures can be well fitted with the Langmuir isotherm from 0.220 pM to 2.20 nM cTnI. Owing to the inherently high sensitivity of the PEC detection, the detection limit (2.20 pg/mL) is lower than the range attainable with the enzyme-linked immunosorbent assay (ELISA) (6.00–40.0 pg/mL). Furthermore, CM-dextran prevents species in complex biological matrices from nonspecifically adsorbing onto the sensor surface, a feature not attainable with uncoated semiconductor electrodes or those coated with non-hydrogel-based chemical modifiers. The excellent anti-fouling property of dextran hydrogel allowed us to validate the accuracy of our regenerable sensors through a comparison of PEC immunoassays of patient sera to those of ELISA.

Abstract:An electrochemical immunosensor based on the novel high efficiency catalytic cycle amplification strategy for the sensitive detection of cardiac troponin I (cTnI). With its variable valence metal elements and spiny yolk structure, the Cu2O/CuO@CeO2 nanohybrid exhibits high speed charge mobility and exceptional electrochemical performance. Notably, fluorite-like cubic crystal CeO2 shell would undergo redox reaction with Cu2O core, which successfully ensures the continuous recycling occurrence of "fresh" Cu (II)/Cu (I) and Ce (Ⅳ)/Ce (Ⅲ) pairs at the electrode interface. The "fresh" active sites continue to emerge constantly, resulting in a significant increase in the current signal. In light of the electrochemical characterization, the electron transfer pathway and catalytic cycle mechanism among CeO2, Cu2O and CuO were further discussed. The developed electrochemical immunosensor detected cTnI from 100 fg/mL to 100 ng/mL with a LOD of 15.85 fg/mL under optimal conditions. The analysis results indicate that the immunosensor would hold promise for broad application prospects in the biological detection for other biomarkers.

Abstract:Based on the necessity and urgency of cardiac troponin I (cTnI) detection for the diagnosis of myocardial infarction, a novel unlabeled photoelectrochemical (PEC) immunosensor has been developed to detect cTnI rapidly and sensitively. Silicon nanowire arrays (SiNWs) were prepared via metal-assisted chemical etching. To improve the stability and sensitivity towards cTnI sensing, the surface of silicon nanowire arrays were coated with polydopamine by an in situ growth method. PDA was uniformly modified on the nanowire surface to provide a reliable active site for antibody binding. The linear dynamic range of the cTnI detection method was 0.005-10 ng mL(-1), and the detection limit was 1.47 pg mL(-1). The designed PEC immunosensor exhibited good sensitivity, selectivity, stability and reproducibility. The electrode enabled label-free detection and provided a new route to realize point-of-care testing of cTnI.

Abstract:Photoelectrochemical (PEC) immunoassay has received increasing attention owing to its good analytical performance and attractive potential for future protein assay. This Letter represents a novel and general strategy for elegant PEC immunoassay of the important cardiac marker troponin T (cTnT) at neutral conditions. Specifically, we first developed an efficient CdS quantum dots (QDs)/TiO2 nanoparticles (NPs) photoelectrode, on the basis of which an exquisite β-galactosidase (β-Gal) catalytic system was integrated with sandwich immunobinding for probing cTnT. In pH 7.4, β-Gal could catalyze the conversion of p-aminophenyl galactopyranoside (PAPG) to p-aminophenol (PAP), which could be easily photo-oxidized to p-quinone imine (PQI). Because the resulting photocurrent was directly related with the target concentration, an innovative PEC immunoassay could be realized for cTnT detection. The neutral operating condition of this protocol would greatly contribute to its wide applicability for protein assay. This work provides the first PEC immunoassay toward cardiac marker and, more significantly, opens a different perspective for future PEC immunoassay development through a general sensing protocol.

Abstract:Considering the fact that acute myocardial infarction has shown a trend towards younger age and has become a major health problem, it is necessary to develop rapid screening devices to meet the needs of community health care. Herein, we developed an artificial neural network-assisted solar-powered photoelectrochemical (SP-PEC) sensing platform for rapid screening of cardiac troponin I (cTnI) protein in the prognosis of patients with acute myocardial infarction (AMI) by integrating a self-powered photoelectric signal output system with low-cost screen-printed paper electrodes functionalized with ultrathin Bi2O2S (BOS) nanosheets. An integrated solar-powered PEC immunoassay with micro-electro-mechanical system (MEMS) was constructed without an excitation light source. The quantification of cTnI protein was obtained by the electrical signal changes caused by the electro-oxidation process of H2O2, generated by the classical split immune reaction, on the electrode surface. The test electrodes were developed as dual working electrodes, one for target cTnI testing and the other for evaluating light intensity, to reduce the temporal inconsistency of sunlight. The photoelectrodes were discovered to exhibit satisfactory negative response to target concentrations in the dynamic range of 2.0 pg mL-1-10 ng mL-1 since being regressed in an improved artificial neural network (ANN) model using the pooled dataset of target signals affected by the light source. The difference of hot electron and hole transfer behavior in different thickness of nano-materials was determined by finite element analysis (FEA), which provided a theoretical basis for the development of efficient PEC sensors. This work presents a unique perspective for the design of a revolutionary low-cost bioassay platform by inventively illuminating the PEC biosensor's component process without the use of light.
Variability in potassium removal by hemodialysis.

Abstract:The extent to which plasma potassium determines potassium removal in hemodialysis was examined in 8 end-stage renal disease patients during 51 treatments. Dialyzers, treatment time, blood and dialysate flow were held constant. Dialysate composition was also uniform except that 24 treatments utilized glucose-free dialysate and the remaining 27 a 200 mg/dl glucose bath. At either level of dialysate glucose, approximately 40% of the potassium removal during dialysis could not be accounted for by plasma-dialysate potassium gradient, body weight or serum carbon dioxide content, although glucose-free dialysate tended to increase potassium removal by a mean of 28%. The large, unexplained variability in potassium removal suggests that therapeutic manipulation of potassium flux across cell membranes may improve the management of potassium balance in hemodialyzed patients.

Abstract:Here, a sandwich-type electrochemical immunosensor was constructed to detect cardiac troponin I (cTnI) via asymmetric bowl-shaped PdAgFe mesoporous nanospheres (PdAgFe ABMS) as a signal amplification label and core-shell cubic Au/Co-LDH@ZIF-67 as a substrate material. PdAgFe ABMS prepared by dual-template directional anisotropic island growth method has abundant mesoporous channels to accelerate molecular mass transfer in solution. In particular, the asymmetric bowl-like structure allows more active sites to be exposed, which improves the utilization of atoms to stabilize and high current response signals. The synergistic effect between PdAgFe increased the activation energy of the catalytic reaction and further amplified the current signal. In addition, the biosensing interface based on Au/Co-LDH@ZIF-67 not only exhibits a high electron transfer rate, but also can capture more bioactive molecules. Under optimal conditions, the constructed immunosensor was detected to exhibit a low limit of detection (LOD, 4.47 fg/mL) and a wide detection range (10 fg/mL - 100 ng/mL). This work provides an accurate and convenient scheme for the clinical detection of cTnI.

A duple nanozyme stimulating tandem catalysis assisted multiple signal inhibition strategy for photoelectrochemical bioanalysis

Abstract:A sandwich-type photoelectrochemical (PEC) immunosensor was developed for cytokeratin 19 fragment 21-1 (CYFRA21-1) ultrasensitive detection relying on a multiple signal inhibition strategy. In this work, Bi10O6S9 nanosheet arrays (BiOS) sensitized strontium titanate nanocubes (SrTiO3 NCs) was used as photoactive matrix. Bi10O6S9 with the large specific surface area and narrow band gap could improve the utilization of light to provide strong initial PEC signal. Besides, bovine serum albumin stabilized gold nanoparticles (BSA@Au NPs) anchored on Cu7S4 polyhedron composite (BSA-Au@Cu7S4 polyhedron) was utilized as an effective PEC signal regulator. As a duple nanozyme with the glucose oxidase (GOx)-like activity and the peroxidase-like activity, BSA@Au NPs induced intriguing tandem biocatalytic precipitation (BCP). On the one hand, the BCP could produce precipitations which obstructed the electron transfer between the photoelectrode and electron donor, leading a decrease of PEC signal; On the other hand, the competitive light absorption between BSA-Au@Cu7S4 polyhedron and photoactive matrix caused a further reduction of PEC signal. Generally, the PEC signal was quenched sharply taking advantage of multiple signal inhibition strategy including tandem BCP strategy, competitive absorption of light and steric hindrance, which improved the detection sensitivity of PEC immunosensor. The suggested PEC immunosensor displayed a good linearity in the range of 0.1 pg/mL -50 ng/mL for CYFRA21-1 detection with a low detection limit of 1.12 fg/mL (S/N = 3). The skillful strategy has infinite potential in biological samples analysis.

Abstract:As an inflammatory marker, procalcitonin (PCT) is more representative than other traditional inflammatory markers. In this work, a highly efficient photoelectrochemical (PEC) immunosensor was constructed based on the photoactive material Bi2S3/Ag2S to realize the sensitive detection of PCT. Bi2S3 was prepared by a hydrothermal method, and Ag2S quantum dots were deposited on the ITO/Bi2S3 surface via in situ reduction. Bi2S3 is a kind of admirable photoelectric semiconductor nanomaterial on account of its moderate bandgap width and low binding rate of photogenerated electron holes, which can effectively convert light energy into electrical energy. Therefore, based on the energy level matching principle of Bi2S3 and Ag2S, a labeled Bi2S3/Ag2S PEC immunosensor was constructed, and the sensitive detection of PCT was successfully established. The linear detection range of the PEC immunosensor was 0.50 pg∙mL−1 to 50 ng∙mL−1, and the minimum detection limit was 0.18 pg∙mL−1. Compared with the traditional PEC strategy, the proposed PEC immunosensor is simple, convenient, and has good anti-interference, sensitivity, and specificity, which could provide a meaningful theoretical basis and reference value for the clinical detection of PCT.

Abstract:The development of photoelectrochemical (PEC) sensors with novel principles is of significance in realizing sensitive and low-cost detection. This work uses CuO NPs labeled antibody to construct a simple and sensitive sandwich-type immunobiosensor for the detection of protein. The detection signal is produced by dissolving the CuO NPs to release copper ions, which are then added on a quantum dots (QDs) modified F-doped tin oxide to quench the photocurrent of QDs via copper ion-induced formation of exciton trapping. The formed exciton trapping blocks the escape of photoelectron and thus leads to a "signal off" PEC method for sensitive immunoassay. The proposed method shows a detectable range from 0.05 to 500 ng/mL for α-fetoprotein (AFP) with a detection limit (LOD) of 0.038 ng/mL. This work further extends the application of exciton trapping-based PEC biosensing strategy in bioanalysis. The sensitive analytical performance of the designed route implies a promising potential of the PEC sensing in clinical diagnosis.
Electrochemical/colorimetric dual-mode sensing strategy for cardiac troponin I detection based on zirconium nitride functionalized covalent organic frameworks

Abstract:Cardiac troponin I (cTnI) is a gold-standard marker for the detection of acute myocardial infarction (AMI). Accurate and timely detection of cTnI is particularly important for AMI diagnosis and therapy. Herein, based on the dual-capacity (electro-catalysis and catalytic degradation) of @COF to toluidine blue (TB), we developed novel electrochemical and enzyme-linked immunosorbent assay (ELISA) methods for ultrasensitive detection of cTnI. In the electrochemical sensor, polyaniline-coated cerium oxide loaded with gold nanoparticles (@CeO 2 ) were used as a substrate platform for immobilizing antibodies (Ab1), and @COF loaded with the affinity peptide (Pep) of cTnI and TB (Pep/TB/@COF) was the probe material for signal amplification of the sensor. Using the 'peptide-cTnI-Ab1′ sandwich biosensor, we successfully detected cTnI in a concentration range of 0.00001–50 ng·mL −1 with an ultralow detection limit (LOD) of 3.3 fg·mL −1 . In addition, a sandwich ELISA was constructed by 96-well plate using Pep/@COF as the probe molecule for cTnI detection, showing a detection range of 0.00001–5 ng·mL −1 and a LOD of 3.3 fg·mL −1 . Both detection methods presented excellent accuracy, selectivity, reproducibility, and stability, providing a potential application in clinical diagnosis of AMI.

Abstract:Cardiac troponin I (cTnI) is an extremely sensitive biomarker for early indication of acute myocardial infarction (AMI). Abstract:A suitable electron donor, which guarantees the stability of the whole system, is considered as the driving force of the PEC sensor. Nowadays, searching appropriate electron donor is still one of the orientations to explorate in the field of sensor. Na48[H496Mo368O1464S48]·ca.1000H2O (abbr. {Mo368}), as a type of polyoxometalate, has perfect morphology, definite size and unique electronic property. Due to the prominent water solubility, {Mo368} usually releases small cations and exists as large anions in the ultrapure water. The interesting property endows {Mo368} with excellent reducibility, which provides great feasibility to become an outstanding electron donor. In addition, FeOOH prepared through a simple operation owns high adsorption capacity, which ensures the fastness of other materials. Subsequently, the narrow band-gap of Bi2S3 and the unique noble metal properties of Au nanoparticles are utilized to co-sensitize FeOOH to improve the light-harvesting capability and photoelectric conversion efficiency. Combined with the specificity recognition of antigen and antibody, a novel photoelectrochemical sensor is constructed with a wide detection range of 1.00 pg mL−1 - 100 ng mL−1 and low detection limit (0.76 pg mL−1), which achieves the sensitive detection of cardiac troponin I in early diagnosis of cardiovascular disease.

Abstract:Dysregulation of human tumor necrosis factor α (TNF-α) production has been implicated in a variety of human diseases. Serum level of TNF-α is frequently detected in clinical diagnosis of cancer patients. Herein, we present an ultrasensitive photoelectrochemical (PEC) sandwich immunosensor for TNF-α detection by utilizing anti-TNF-α nanobody (Nb) orientedly immobilized to the CdS and Mn cosensitized TiO2 nanorod arrays electrode. Using highly oriented Nb as receptor molecules and a secondary anti-TNF-α nanobody conjugated with ZnS nanoparticles as the signal amplification element, the PEC immunosensor for TNF-α was developed by monitoring the changes in the photocurrent intensity of the electrode resulting from immunoreaction. The proposed method has the linear range from 2.0pg/mL to 200ng/mL and the detection limit was 1.0pg/mL. The method also reveals high specificity to detect TNF-α, with acceptable intra-assay precision and excellent stability. The results for the detection of ten serum samples from lung cancer patients and normal human have good agreement with the clinical data.
