Abstract:It is of keen importance of deep tissue imaging with high resolution in vivo. Here we present an opto-ultrasound imaging method which utilizes an ultrasound to confine the laser pulse in a very tiny spot as a guide star. The results show that the imaging depth is 2mm with a resolution of 10um. Meanwhile, the excitation power we used is less than 2mW, which indicates that our methods can be applied in vivo without optical toxicity and optical bleaching due to the excitation power.

Abstract:Photoacoustic tomography emerged as a promising tool for noninvasive biomedical imaging and diseases diagnosis. However, most of the current piezoelectric ultrasound transducers suffer optical opacity and tissue-mismatched acoustic impedance, hindering the miniaturization and integration of the system for multiscale and multimodal imaging. Here, a transparent polydimethylsiloxane (PDMS) encapsulated optical microfiber ultrasound sensor was demonstrated for photoacoustic imaging with scalable spatial resolution and penetration depth. The sensor comprised a microfiber loop sandwiched by a pair of in-line Bragg gratings, which formed an ultrasound-sensitive Fabry-Perot cavity allowing free delivery of ultrasound/light beams and unique needle-shaped ultrasound focusing along the penetration depth. The sensor with a detection limit of ∼ 700 Pa and a bandwidth of ∼ 10 MHz was applied for multiscale photoacoustic imaging of mouse ear and brain vasculatures. With advantages of flexibility, optical transparence and focusing capability, the sensor offers new opportunities for developing photoacoustic/ultrasound imaging devices for biomedical and clinic applications.

Video-rate functional photoacoustic microscopy at depths

Abstract:We report the development of functional photoacoustic microscopy capable of video-rate high-resolution in vivo imaging in deep tissue. A lightweight photoacoustic probe is made of a single-element broadband ultrasound transducer, a compact photoacoustic beam combiner, and a bright-field light delivery system. Focused broadband ultrasound detection provides a 44-μm lateral resolution and a 28-μm axial resolution based on the envelope (a 15-μm axial resolution based on the raw RF signal). Due to the efficient bright-field light delivery, the system can image as deep as 4.8 mm in vivo using low excitation pulse energy (28 μJ per pulse, 0.35 mJ/cm² on the skin surface). The photoacoustic probe is mounted on a fast-scanning voice-coil scanner to acquire 40 two-dimensional (2-D) B-scan images per second over a 9-mm range. High-resolution anatomical imaging is demonstrated in the mouse ear and brain. Via fast dual-wavelength switching, oxygen dynamics of mouse cardio-vasculature is imaged in realtime as well.
Functional photoacoustic microscopy for high-resolution and noninvasive in vivo imaging

Abstract:Although optical absorption is strongly associated with the physiological status of biological tissue, existing high-resolution optical imaging modalities, including confocal microscopy1,2, two-photon microscopy3,4 and optical coherence tomography5, do not sense optical absorption directly. Furthermore, optical scattering prevents these methods from imaging deeper than ∼1 mm below the tissue surface. Here we report functional photoacoustic microscopy (fPAM), which provides multiwavelength imaging of optical absorption and permits high spatial resolution beyond this depth limit with a ratio of maximum imaging depth to depth resolution greater than 100. Reflection mode, rather than orthogonal or transmission mode, is adopted because it is applicable to more anatomical sites than the others. fPAM is demonstrated with in vivo imaging of angiogenesis, melanoma, hemoglobin oxygen saturation (sO2) of single vessels in animals and total hemoglobin concentration in humans.

Single-shot hybrid photoacoustic-fluorescent microendoscopy through a multi-mode fiber with wavefront shaping

Abstract:We present a minimally-invasive endoscope based on a multimode fiber that combines photoacoustic and fluorescence sensing. From the measurement of a transmission matrix during a prior calibration step, a focused spot is produced and raster-scanned over a sample at the distal tip of the fiber by use of a fast spatial light modulator. An ultra-sensitive fiber-optic ultrasound sensor for photoacoustic detection placed next to the fiber is combined with a photodetector to obtain both fluorescence and photoacoustic images with a distal imaging tip no larger than 250um. The high signal-to-noise ratio provided by wavefront shaping based focusing and the ultra-sensitive ultrasound sensor enables imaging with a single laser shot per pixel, demonstrating fast two-dimensional hybrid imaging of red blood cells and fluorescent beads.

Abstract:Optical-resolution photoacoustic microscopy offers high-resolution, label-free hemodynamic and functional imaging to many biomedical applications. However, long-standing technical barriers, such as limited field of view, bulky scanning probes, and slow imaging speed, have limited the application of optical-resolution photoacoustic microscopy. Here, we present freehand scanning photoacoustic microscopy (FS-PAM) that can flexibly image various anatomical sites. We develop a compact handheld photoacoustic probe to acquire 3D images with high speed, and great flexibility. The high scanning speed not only enables video camera mode imaging but also allows for the first implementation of simultaneous localization and mapping (SLAM) in photoacoustic microscopy. We demonstrate fast in vivo imaging of some mouse organs, and human oral mucosa. The high imaging speed greatly reduces motion artifacts and distortions from tissue moving, breathing, and unintended handshaking. We demonstrate small-lesion localization in a large region of the brain. FS-PAM offers a flexible high-speed imaging tool with an extendable field of view, enabling more biomedical imaging applications.

Whispering-gallery Optical Microprobe for Photoacoustic Imaging

Abstract:Optical ultrasonic probes, exemplified by Fabry–Perot cavities on optical fibers, have small sizes, high sensitivity, and pure optical characteristics, making them highly attractive in high-resolution ultrasonic/photoacoustic imaging, especially in near-field or endoscopic scenarios. Taking a different approach, we demonstrate an ultrasensitive and broadband ultrasound microprobe formed by an optical whispering-gallery-mode polymer microcavity coupled to a U-shaped microfiber. With the high-quality (Q) factors (>106), the noise equivalent pressure of the ultrasound microprobe reaches 1.07 mPa/√Hz with a record broadband response of 150 MHz and a large detection angle of 180°. Our results show that this optical microprobe can overcome the strong decay resulting from ultrasound diverging and medium absorption through short working distances. We further demonstrate high-quality in vivo whole-body photoacoustic imaging of a zebrafish larva. Our implementation provides a new strategy for developing miniature ultrasound detectors and holds great potential for broad applications.

Abstract:Imaging of the cells and microvasculature simultaneously is beneficial to the study of tumor angiogenesis and microenvironments. We designed and built a fiber-optic based photoacoustic microscopy (PAM) and confocal fluorescence microscopy (CFM) dual-modality imaging system. To explore the feasibility of this all-optical device for future endoscopic applications, a microelectromechanical systems (MEMS) scanner, a miniature objective lens, and a small size optical microring resonator as an acoustic detector were employed trying to meet the requirements of miniaturization. Both the lateral resolutions of PAM and CFM were quantified to be 8.8μm. Axial resolutions of PAM and CFM were experimentally measured to be 19μm and 53μm, respectively. The experiments on ex vivo animal bladder tissues demonstrate the good performance of this system in imaging not only microvasculature but also cellular structure, suggesting that this novel imaging technique holds potential for improved diagnosis and guided treatment of bladder cancer.
Dual-foci Fast-Scanning Photoacoustic Microscopy with 3.2-Mhz A-line Rate

Abstract:We report fiber-based dual-foci fast-scanning OR-PAM that can double the scanning rate without compromising the imaging resolution, the field of view, and the detection sensitivity. To achieve fast scanning speed, the OR-PAM system uses a single-axis water-immersible resonant scanning mirror that can confocally scan the optical and acoustic beams at 1018 Hz with a 3-mm range. Pulse energies of 45∼100-nJ are sufficient for acquiring vascular and oxygen-saturation images. The dual-foci method can double the B-scan rate to 2036 Hz. Using two lasers and stimulated Raman scattering, we achieve dual-wavelength excitation on both foci, and the total A-line rate is 3.2-MHz. In in vivo experiments, we inject epinephrine and monitor the hemodynamic and oxygen saturation response in the peripheral vessels at 1.7 Hz over 2.5 × 6.7 mm2. Dual-foci OR-PAM offers a new imaging tool for the study of fast physiological and pathological changes.

Abstract:SignificancePhotoacoustic (PA) imaging is an emerging biomedical imaging modality that can map optical absorption contrast in biological tissues by detecting ultrasound signal. Piezoelectric transducers are commonly used in PA imaging to detect the ultrasound signals. However, piezoelectric transducers suffer from low sensitivity when the dimensions are reduced and are easily influenced by electromagnetic interference. To avoid these limitations, various optical ultrasound sensors have been developed and shown their great potential in PA imaging.AimOur study aims to summarize recent progress in optical ultrasound sensor technologies and their applications in PA imaging.ApproachThe commonly used optical ultrasound sensing techniques and their applications in PA systems are reviewed. The technical advances of different optical ultrasound sensors are summarized.ResultsOptical ultrasound sensors can provide wide bandwidth and improved sensitivity with miniatured size, which enables their applications in PA imaging.ConclusionsThe optical ultrasound sensors are promising transducers in PA imaging to provide higher-resolution images and can be used in new applications with their unique advantages.

Miniature All-Optical Probe for Photoacoustic and Ultrasound Dual-Modality Imaging

Abstract:Photoacoustic (PA) imaging forms an image based on optical absorption contrasts with ultrasound (US) resolution. In contrast, US imaging is based on acoustic backscattering to provide structural information. In this study, we develop a miniature all-optical probe for high-resolution PA-US dual-modality imaging over a large imaging depth range. The probe employs three individual optical fibers (F1-F3) to achieve optical generation and detection of acoustic waves for both PA and US modalities. To offer wide-angle laser illumination, fiber F1 with a large numerical aperture (NA) is used for PA excitation. On the other hand, wide-angle US waves are generated by laser illumination on an optically absorbing composite film which is coated on the end face of fiber F2. Both the excited PA and backscattered US waves are detected by a Fabry-Pérot cavity on the tip of fiber F3 for wide-angle acoustic detection. The wide angular features of the three optical fibers make large-NA synthetic aperture focusing technique possible and thus high-resolution PA and US imaging. The probe diameter is less than 2 mm. Over a depth range of 4 mm, lateral resolutions of PA and US imaging are 104−154 μm and 64−112 μm, respectively, and axial resolutions of PA and US imaging are 72−117 μm and 31−67 μm, respectively. To show the imaging capability of the probe, phantom imaging with both PA and US contrasts is demonstrated. The results show that the probe has potential for endoscopic and intravascular imaging applications that require PA and US contrast with high resolution.

Abstract:High-speed optical-resolution photoacoustic microscopy (OR-PAM), integrating the merits of high spatial resolution and fast imaging acquisition, can observe dynamic processes of the optical absorption-based molecular specificities. However, it remains challenging for the evaluation to morphological and physiological parameters that are closely associated with photoacoustic spectrum due to the inadequate ultrasonic frequency response of the routinely-employed piezoelectric transducer. By utilizing the galvanometer for fast optical scanning and our previously-developed surface plasmon resonance sensor as an unfocused broadband ultrasonic detector, high-speed spectroscopic photoacoustic imaging was accessed in the OR-PAM system, achieving an acoustic bandwidth of ∼125 MHz and B-scan rate at ∼200 Hz over a scanning range of ∼0.5 mm. Our system demonstrated the dynamic imaging of the moving phantoms' structures and the simultaneous characterization of their photoacoustic spectra over time. Further, fast volumetric imaging and spectroscopic analysis of microanatomic features of a zebrafish eye ex vivo was obtained label-freely.
Photoacoustic Mesoscopy with Lower Cost: Using Photothermal Tunable Fabry-Perot Interferometric Ultrasound Sensor

Abstract:Photoacoustic mesoscopy is an emerging noninvasive imaging modality, which offers high resolution 3D images with optical absorption contrast at depths beyond the light diffusion limit. Ultrasound sensor based on a Fabry-Perot (FP) polymer cavity has the following advantages: broadband frequency response, wide angular coverage and small footprint. We present a photoacoustic mesoscope based on a tunable Fabry-Perot interferometer, which offers the potential for reducing system cost and making array of such sensor. A cw diode laser working at 650nm was used to heat the sensor, offering an active tune range of 5nm by elongating the cavity. Ex-vivo and in-vivo imaging experiments demonstrated the imaging capability of this PA mesoscope, showing great potential in biological and medical applications.
Reflection-mode Subwavelength-resolution Photoacoustic Microscopy for Label-free Microvascular Imaging in Vivo

Abstract:We developed a reflection-mode subwavelength-resolution photoacoustic microscopy system capable of imaging optical absorption contrast in vivo. The simultaneous high-resolution and reflection-mode imaging capacity of the system was enabled by delicately configuring a miniature high-frequency ultrasonic transducer tightly under a water-immersion objective with numerical aperture of 1.0. At 532-nm laser illumination, the lateral resolution of the system was measured to be ~320 nm. With this system, subcellular structures of red blood cells and B16 melanoma cells were resolved ex vivo; microvessels, including individual capillaries, in a mouse ear were clearly imaged label-freely in vivo, using the intrinsic optical absorption from hemoglobin. The current study suggests that, the optical-absorption contrast, subwavelength resolution, and reflection-mode ability of the developed photoacoustic microscopy may empower a wide range of biomedical studies for visualizing cellular and/or subcellular structures.

Abstract:Abstract The combination of photoacoustic (PA) imaging and ultrasound localization microscopy (ULM) with microbubbles has great potential in various fields such as oncology, neuroscience, nephrology, and immunology. Here we developed an interleaved PA/fast ULM imaging technique that enables super-resolution vascular and physiological imaging in less than 2 seconds per frame in vivo. By using sparsity-constrained (SC) optimization, we accelerated the frame rate of ULM up to 37 times with synthetic data and 28 times with in vivo data. This allows for the development of a 3D dual imaging sequence with a commonly used linear array imaging system, without the need for complicated motion correction. Using the dual imaging scheme, we demonstrated two in vivo scenarios challenging to image with either technique alone: the visualization of a dye-labeled mouse lymph node showing nearby microvasculature, and a mouse kidney microangiography with tissue oxygenation. This technique offers a powerful tool for mapping tissue physiological conditions and tracking the contrast agent biodistribution non-invasively.

Abstract:Relying on high-sensitivity refractive index sensing and a highly constrained evanescent field of surface plasmon resonance (SPR), broadband photoacoustic (PA) pressure transients were measured using an SPR sensor instead of routinely used piezoelectric ultrasonic transducers. An acoustic cavity made from stainless steel and having a designed ellipsoidal inner surface redirected laser-induced PA waves from the PA excitation spot to the SPR sensor. By incorporating the SPR sensor with the acoustic cavity, we developed optical-resolution photoacoustic microscopy (OR-PAM) with multiple advantages, including reflection-mode signal capture, improved PA detection sensitivity, increased PA spectral bandwidth as broad as ∼98 MHz, and micrometer-scale lateral resolution. This allowed label-free volumetric PA imaging of vasculature in not only the thin ear but also the thick forelimb of living mice. With these combined advantages, our OR-PAM system potentially offers more opportunities for biomedical investigation, for example, when studying microcirculations in the eye and cortex.

Abstract:A miniature all-optical probe for high-resolution photoacoustic (PA)-ultrasound (US) imaging using a large synthetic aperture is developed. The probe consists of three optical fibers for PA excitation, US generation, and detection of acoustic waves, respectively. The fiber for PA excitation has a large numerical aperture (NA) for wide-angle laser illumination. On the other hand, the fiber with a carbon black-polydimethylsiloxane composite coated on the end face of the optical fiber is used for wide-angle US transmission through laser-US conversion. Both the excited PA and backscattered US signals are detected by a fiber-tip Fabry-Perot cavity for wide-angle acoustic detection. The probe outer diameter is only similar to 2 mm. The synergy of the three optical fibers makes a large-NA synthetic aperture focusing technique for high-resolution PA and US imaging possible. High PA lateral resolutions of 104-154 mu m and high US lateral resolutions of 64-112 mu m over a depth range of > 4 mm are obtained. Compared with other existing miniature PA-US probes, to our knowledge, our probe achieves by far the best performance in terms of lateral resolutions and imaging depth range. The constructed probe has potential for endoscopic and intravascular imaging applications that require PA and US contrasts with high resolutions over a large depth range. Abstract:Among the numerous endogenous biological molecules, information on lipids is highly coveted for understanding both aspects of developmental biology and research in fatal chronic diseases. Due to the pronounced absorption features of lipids in the extended near-infrared region (1650-1850 nm), visualisation and identification of lipids become possible using multi-spectral photoacoustic (optoacoustic) microscopy. However, the spectroscopic studies in this spectral region require lasers that can produce high pulse energies over a broad spectral bandwidth to efficiently excite strong photoacoustic signals. The most well-known laser sources capable of satisfying the multi-spectral photoacoustic microscopy requirements (tunability and pulse energy) are tunable nanosecond optical parametric oscillators. However, these lasers have an inherently large footprint, thus preventing their use in compact microscopy systems. Besides, they exhibit low-repetition rates. Here, we demonstrate a compact all-fibre, high pulse energy supercontinuum laser that covers a spectral range from 1440 to 1870 nm with a 7 ns pulse duration and total energy of 18.3 µJ at a repetition rate of 100 kHz. Using the developed high-pulse energy source, we perform multi-spectral photoacoustic microscopy imaging of lipids, both ex vivo on adipose tissue and in vivo to study the development of Xenopus laevis tadpoles, using six different excitation bands over the first overtone transition of C-H vibration bonds (1650-1850 nm).

Abstract:Optical whispering-gallery microresonators have attracted considerable interest for ultrasensitive ultrasound detection and photoacoustic imaging because of the combination of high quality factors and small cavity sizes. In the last decade, ultrasonic sensors with on-chip microcavities have been extensively developed; however, they are unsuitable for the near-field photoacoustic microscopy of micro/nanoscale objects in complex biological environments and endoscopic imaging. In this work, we developed ultrasonic sensors using two types of encapsulated microsphere resonators with different cavity materials. A noise equivalent pressure of as low as 160 Pa at 20 MHz was achieved with the acoustic response up to 70 MHz at-6 d B. Furthermore,the microsensor was used for photoacoustic microscopy in which we successfully performed 3 D imaging of hairs and leaf veins.The microsphere ultrasonic sensor has considerable potential as a probe-type ultrasonic detector for near-field photoacoustic microscopy of micro/nanoscale objects such as subcellular structures and high-resolution endoscopic photoacoustic imaging with its high sensitivity and wide bandwidth.
