Abstract:meta/ortho-Selective C-Ar-H (di) alkylation reactions of azoarenes have been achieved via [Ru(p-cymene)Cl-2](2) catalyzed ortho-metalation using various types of alkyl bromides. Particularly, dual meta-alkylation of azoarene and reduction offer an attractive strategy for the synthesis of meta-alkylanilines, which are difficult to access via traditional aniline functionalization methods.

Abstract:The first example of transition-metal-catalyzed ortho/meta-selective dual C-H functionalizations of arenes in one reaction is described. In this transformation, ortho-C-H chlorination and meta-C-H sulfonation of 2-phenoxypyri(mi)dines were achieved simultaneously under catalysis by [Ru(p-cymene)Cl2]2. The other reactant, namely, an arylsulfonyl chloride, played the role of both a sulfonation and chlorination reagent. More importantly, the arylsulfonyl chloride was also an oxidant in the process. Mechanistic studies indicated that six-membered ruthenacycles were the key intermediate in the reaction.

Abstract:The meta-C-Ar-H bond formylation of arenes has been achieved using CHBr3 as a formyl source in the presence of [Ru(p-cym)(OAc)(2)] as a catalyst. This method provides efficient access to the preparation of various meta-substituted aromatic compounds, such as alcohols, ethers, amines, nitriles, alkenes, halogens, carboxylic acids, and their derivatives, through transformation of the versatile formyl group. Furthermore, mechanistic studies show that the key active species is a pentagonal ruthenacycle complex.

Abstract:A ruthenium‐catalyzed C(sp2)–H acyloxylation of 2‐aroyl pyridine derivatives with simple sodium carboxylate utilizing transformable directing groups is described. This protocol features broad functional group tolerance and chemo‐ and regio‐selectivity, providing the acyloxylation products in 45%‐84%yield. Furthermore, the synthetic utility of this protocol was demonstrated by the late‐stage functionalization of pharmaceutical compounds. Notably, the acyloxylation products could be further transformed into a variety of useful heterocycles under mild conditions.

Abstract:An efficient approach for the remote C-H alkylation of arenes, employing a variety of N-directing groups is described. This method facilitates the straightforward synthesis of valuable phenylethylamine derivatives by exclusively cleaving the benzylic C-N bond in aziridines. Furthermore, these products can easily remove the protecting groups, resulting in a variety of meta-substituted compounds, such as amines and ketones, which hold significance in synthetic chemistry.

Abstract:AbstractThe prevalence ofC‐aryl glycosides in biologically active natural products and approved drugs has long motivated the development of efficient strategies for their selective synthesis. Cross‐couplings have been frequently used, but largely relied on palladium catalyst with prefunctionalized substrates, while ruthenium‐catalyzedC‐aryl glycoside preparation has thus far proven elusive. Herein, we disclose a versatile ruthenium(II)‐catalyzedmeta‐C−H glycosylation to accessmeta‐C‐aryl glycosides from readily available glycosyl halide donors. The robustness of the ruthenium catalysis was reflected by mild reaction conditions, outstanding levels of anomeric selectivity and exclusivemeta‐site‐selectivity.

Abstract:Free radical cyclization has emerged as one of the most important reaction types, which is widely used in natural product synthesis, pharmaceutical chemistry, and materials science. This report described the combination of radical cyclization and ruthenium-catalyzed meta-selective C–H functionalization for the synthesis of arylpyrrolidone derivatives. This method exhibited the highly meta-site selectivity of the primary, secondary, and tertiary alkyl radicals formed by intramolecular addition. A wide spectrum of directing groups bearing diversified N-heterocycles performed well, including biologically active molecules. Abstract:A ruthenium-catalyzed sulfonamide-directed ortho aryl C-H alkenylation/annulation to afford five-membered sultam by using of molecular oxygen as an oxidant is reported in this article. Compared to the previous transition-metal-catalyzed C-H alkenylation of aryl sulfonamides, no excess metal salt oxidant was required in this method. A wide sulfonamide substrates scope and good regioselectivity and site-selectivity make this Ru-catalyzed method more attractive. Importantly, this method can not only be used to effectively prepare the biologically active five-membered sultam molecules but also be efficiently applied to the late-stage modification of sulfonamide drugs.

Abstract:We report ruthenium( II)-catalyzed ortho-C-H arylation of N,N-dialkylthiobenzamides with boronic acids. The method employs [RuCl2(p-cym)](2) in the presence of Cu(OTf)(2) and Ag2O oxidant. The reaction represents the first example of Ru-catalyzed C-H arylation directed by sulfur-containing groups and a rare example of C-H arylation directed by the versatile thiobenzamide moiety. As a further advantage, the method is performed in sustainable and eco-friendly 2-MeTHF as a solvent.

Abstract:A chiral ruthenium catalyst was developed for the enantioselective sulfoxidation of the title compounds. The catalyst combines two elements of chirality, a chiral pybox ligand and a chiral bicylic lactam unit, to which the ligand is attached. The latter unit was shown to improve significantly the performance of the catalyst by exposing one of the two enantiotopic sulfur atoms to the active site via hydrogen-bond mediated coordination. Ten differently substituted substrates were converted into the respective sulfoxides in yields of 52-71 % and with ≥90 % ee.

Abstract:A removable oxime-assisted meta-C-H nitration of arenes is reported. Mechanistic investigations and DFT calculations reveal a new monomeric octahedral ruthenium(II) complex is responsible for the meta-selective nitration. Dioxygen as a cooxidant is crucial for achieving high conversion and good yields. Moreover, the utility of the present reaction protocol is further showcased by the late-stage modification of the clinical CNS drugs Diazepam and Fluvoxamine.

Abstract:The first example of transition metal-catalyzed meta-selective CAr-H nitration of arenes is described. With the use of Ru3(CO)12 as the catalyst and Cu(NO3)2·3H2O as the nitro source, a wide spectrum of arenes bearing diversified N-heterocycles or oximido as the directing groups were nitrated with meta-selectivity exclusively. Mechanism studies have demonstrated the formation of a new 18e-octahedral ruthenium species as a key ortho-CAr-H metalated intermediate, which may be responsible for the subsequent meta-selective electrophilic aromatic substitution (SEAr). Moreover, this approach provides a fast-track strategy for atom/step economical synthesis of many useful pharmaceutical molecules.
