Abstract:The first and enantioselective total synthesis of (+)-plumisclerinA, a novel unique complex cytotoxic marine diterpenoid, has been accomplished. Around the central cyclopentane anchorage, a sequential ring-formation protocol was adopted to generate the characteristic tricycle[4.3.1.0(1,5)]decane and trans-fused dihyrdopyran moiety. Scalable enantioselective La-III-catalyzed Michael reaction, palladium(0)-catalyzed carbonylation and SmI2-mediated radical conjugate addition were successfully applied in the synthesis, affording multiple grams of the complex and rigid B/C/D-ring system having six continuous stereogenic centers and two all-carbon quaternary centers. The trans-fused dihyrdopyran moiety with an exo side-chain was furnished in final stage through sequential redox transformations from a lactone precursor, which overcome the largish steric strain of the dense multiring system. The reported total synthesis also confirms the absolute chemistries of natural (+)-plumisclerinA.

Abstract:A concise synthesis of the tricyclo[4.3.1.03,7]decane caged core of palhinine alkaloids was developed with SmI2-mediated cyclization and light-initiated radical addition–fragmentation as key steps. Compared with the reported racemic routes which are all based on Diels–Alder-type key reactions, our strategy would be more readily accessible to the asymmetric total syntheses of the palhinine alkaloids.

Abstract:Efficient formal syntheses of (±)-Asterisca-3(15),6-diene, a natural product with a bicyclo[6.3.0]undecane skeleton, and (±)-Pentalenene, a natural product with a tricyclo[6.3.0.04,8]undecane skeleton, have been achieved by using Rh(I)-catalyzed [(5+2)+1] cycloaddition. The [(5+2)+1] reaction provides an expeditious approach to reach the bicyclic cyclooctenone 4, which was quickly transformed (via hydroboration then oxidation) to diketone 14, a key advanced intermediate for the total synthesis of (±)-Asterisca-3(15),6-diene. Through further transformations, 14 was converted to diene 18, an advanced intermediate for the total synthesis of (±)-Pentalenene.

Abstract:Inherently chiral tetraoxacalix[2]arene[2]pyridines containing C(2) symmetry were synthesized efficiently from a macrocyclic condensation reaction of resorcinol derivatives with 2,6-dichloro-3-nitropyridine in a one-pot reaction manner, while tetraoxacalix[2]arene[2]pyridine with an ABCD-substitution pattern was prepared in a good yield by means of a stepwise fragment coupling approach. Postmacrocyclization chemical manipulations led to functionalized tetraoxacalix[2]arene[2]pyridines. A racemic sample was resolved into enantiopure (+)- and (-)-inherently chiral compounds.

Abstract:π-Conjugated chiral shape-persistent molecular nanocarbons hold great potential as chiroptical materials, though their synthesis remains a considerable challenge. Here, we present a simple approach using Suzuki coupling of a [5]helicene building block with various aromatic units, enabling the one-pot synthesis of a series of chiral macrocycles with persistent figure-eight and Möbius shapes. Single-crystal structures of 7 compounds were solved, and 22 enantiomers were separated by preparative chiral HPLC. A notable pyrene-bridged figure-eight macrocycle, with its rigid, fully π-conjugated and overcrowded structure, exhibited pure excimer emission and outstanding circularly polarized luminescence (CPL) properties, including a large dissymmetric factor (|glum| = 3.8 × 10−2) and significant CPL brightness (BCPL = 710.5 M-1cm-1). This method provides a versatile synthetic platform for producing various chiral D2-symmetric figure-eight macrocycles and singly or triply twisted Möbius macrocycles with C2 and D3 symmetry, offering tunable chiroptical properties for CPL applications.

Abstract:A sterically congested all-carbon quaternary center was installed for the first time via a Pd-catalyzed cascade diarylation with aryl bromides and acyclic 1,3-diketones. This method was used as a key step in the total synthesis of (±)-spiroaxillarone A. Computational experimental results indicated that the selective diarylation is accelerated by the higher free-energy barriers of the endothermic transmetalation and reductive elimination in the first arylation step.

Abstract:Indole-containing polyaromatic scaffolds are widely found in natural products, pharmaceutical agents, and π-conjugated functional materials. Often, the synthesis of these highly valuable molecules requires a multistep sequence. Therefore, a simple, one-step protocol to access libraries of polyaromatic indole scaffolds is highly desirable. Herein we describe the direct synthesis of polysubstituted indolo[2,1-a]isoquinoline analogues via a double C-H annulation cascade using triazene as an internally cleavable directing group. Evidence from HRMS and theoretical calculations suggests that an unprecedented 1,2-alkyl migration might be responsible for the in situ cleavage of the directing group. Both kinetic isotope effects and DFT calculations suggested that the alkyne insertion step is rate-limiting for the second C,N annulation reaction.

Abstract:The core skeleton syntheses 3'H-spiro[cyclohexane-1,1'-isobenzofuran]-2,5- dien-4-one and their derivatives (3a-3w) of spirobisnaphthalene urnucratins, plecmillins and lignan kadsulignan, were explored using the Suzuki–Miyaura coupling product biphenols as the precursors and the intramolecular oxidative dearomatization with highvalance iodine reagent PIFA as the key step in 46-91% yields, a wide range of functional groups were tolerated. Then using this strategy in combination with the Diels-Alder reaction, two skeleton molecules (5ah and 6d) of the natural product urnucratins and plecmillins were successfully constructed in 78% and 39% overall yields, which provided a potential methodology for the total synthesis of Type D spirobisnaphthalene urnucratins and other natural products with the similar structures.
