Abstract:A new Ti-III-mediated reductive epoxide-opening/ Beckwith-Dowd rearrangement process efficiently assembles the bicyclo[3.2.1]octane framework of highly oxidized grayanane diterpenoids. By incorporation of a Cu(tbs)(2)-catalyzed (tbs=N-tert-butylsalicylaldiminato) intramolecular cyclopropanation, a diastereoselective oxidative dearomatization-induced Diels-Alder cycloaddition and a MeReO3-catalyzed Rubottom oxidation, this approach has enabled the first total syntheses of rhodomolleins XX and XXII in 23 and 22 steps, respectively.

Abstract:A regio-, diastereo- and enantioselective 1,3-dipolar cycloaddition reaction between nitrones and electron-deficient alkenes has been developed employing C 1 -symmetrical Ti(OTos) 2 - and TiCl 2 -TADDOLate as catalysts. In the presence of powdered 4 Å molecular sieves and 100 mol% of Ti(OTos) 2 -TADDOLate as the catalyst, the nitrones smoothly react with alkenes at room temperature to give isoxazolidines in high conversion, giving the trans-diastereomer of the isoxazolidines with de's up to >90% and ee up to 58%. Furthermore, C 1 -symmetrical TiCl 2 -TADDOLate has a better chiral induction, giving the trans-diastereomer of the isoxazolidines with high enantioselectivity up to 70% ee. Abstract:The development of an efficient diastereoselective synthesis of the oxabicyclo[3.2.1]octane ring system bearing two oxygenated quaternary chiral centres represents a significant challenge. This motif can be found in a wide range of natural products with significant biological activities. Here we report the synthesis of such kind of scaffold using a cyclohexane- trans -1,4-diol with an alkyne side chain in the presence of Au(I) catalyst. This is a domino process in which two C–H, two C–O and one C–C bond is assembled through a sequence of cyclization/semi-pinacol rearrangements. This strategy has been successfully applied to the asymmetric formal total synthesis of (+)-cortistatins.

Abstract:The asymmetric denitrogenative cycloaddition has emerged as a powerful tool to build chiral aza-heterocyles. However, only one example of asymmetric denitrogenative cycloaddition of benzotriazole with unsaturated hydrocarbons has been explored so far, because the ring-opening of benzotriazole to generate α-imino metal carbenoid species is a thermodynamically unfavorable process. We herein report an efficient asymmetric denitrogenative cycloaddition of benzotriazoles with cyclic and acyclic 1,3-dienes enabled by Pd and chiral sulfonamide phosphine ligand. A variety of substituted hexahydrocarbazoles and indolines were delivered in good yields with high ee values. Interestingly, a pair of enantiomers could be obtained with the use of Xu1 and PC2 with the same absolute configuration. The synthetic utilities of the optically active hexahydrocarbazoles were also showcased.

Abstract:Multiple contiguous quaternary carbon stereocenters (CQS) are highly challenging, yet important structural motifs in organic synthesis. Here, we describe a visible light induced catalytic [2 + 2] cycloaddition approach that constructed up to four CQS in a pentacyclic fused ring system diastereoselectively, from the readily accessible dienamides with pendent heteroaryls. Variously substituted dienamides have been cyclized with heteroaryls to provide a range of novel CQS-containing scaffolds (26 examples, up to 96% yield and >20:1 dr ratio). Mechanistic studies revealed that it may proceed through an uncommon β -C radical initiated 7- endo cyclization from the biradical intermediate.

Abstract:Enantioselective construction of contiguous quater-nary centers is a daunting challenge in the synthetic community. Substituted 3-(methoxycarbonyl)cyclobutenone, possessing strained C=C double bond, exhibits both good stability and reactivity. Here, we report a chiral oxazaborolidinium ion (COBI)-catalyzed enantioselective Diels-Alder reaction of 2,3-disubsti-tuted cyclobutenones with a variety of dienes to access bicyclo[4.2.0]octane derivatives, bearing two contiguous angular quaternary centers. Further transformations, including alkylation, cross-coupling reaction and oxyallyl cation reaction afforded the molecules with intriguing structural patterns. The formal total synthesis of estrone was also completed in three steps from enantio-enriched cycloadducts.

Abstract:Skeletal diversity in diversity-oriented synthesis has proven to be especially challenging. A rhodium-catalyzed intramolecular tandem reaction of vinyloxirane-alkynes leading to four structurally distinct classes of mono-, bi-, and tricyclic carbocycles and heterocycles was developed. [Rh(NBD)(2)]+BF4- is identified to be an efficient catalyst for these transformations. Using this highly efficient catalyst, hetero-[5 + 2] cycloadditions, tandem hetero-[5 + 2] cycloaddition/Claisen rearrangements, and subsequent cyclopropane ring-opening reactions of vinyloxiranes with monoalkynes afford 2,S-dihydrooxepins, tetrasubstituted vinylcyclopropanes, and multifunctional five-membered rings, respectively, under mild conditions with high stereoselectivity and yield. Moreover, hetero-[5 + 2] cycloaddition/Claisen rearrangement/[5 + 2] cycloaddition reactions of vinyloxiranes with diynes for step-economical construction of linearly fused 5-7-5 tricyclic skeletons has also been developed. The complete transfer of chirality from readily available vinyloxiranes to the corresponding products provides a highly efficient and practical access to these chiral cyclic compounds.

Abstract:The Cu-catalyzed asymmetric carboboration of cyclohexadienone-tethered allenes has been achieved through regioselective β-borylation of the allenes and subsequent conjugate addition to cyclohexadienones, affording cis-bicyclic frameworks with acceptable yields and high to excellent enantioselectivities. Further conjugate borylation of the carboboration products proved to be a favorable kinetic resolution process, which improved the overall enantioselectivity. Abstract:A tandem Prins/Friedel-Crafts cascade strategy has been developed for the synthesis of hexahydro-2H-benzo[g]chromene derivatives. Under action of equimolecular amount of BF3 etherate, delta,epsilon-unsaturated alcohols and arylacetaldehydes are coupled to form oxocarbenium ion intermediates, which are captured by the alkene moiety through a favored six-membered transition state. The Prins-cyclization resultant tertiary carbenium ions undergo consecutive intramolecular Friedel-Crafts cyclization to create the tricyclic products. This process enables a tandem formation of three new chemical bonds in one single operation with high yields and stereoselectivity in most cases. When cyclic substituents are involved, a variety of spiro derivatives could also be synthesized successfully, which further proved the robustness of this methodology.

Abstract:A tandem reaction involving a Rh(I)-catalyzed two-component [(5+2)+1] cycloaddition and an aldol condensation has been developed to construct the tricyclo[6.3.0.0(2,6)]undecane skeleton and its heteroatom-imbedded analogues. Meanwhile, this method has been successfully applied to natural product synthesis for the first time. The present strategy enables a straightforward approach to the natural linear triquinane skeleton, as demonstrated by concise and step economical syntheses of hirsutene and 1-desoxyhypnophilin, whereby the linear triquinane core is diastereoselectively established in one manipulation with correct placement of all stereocenters, including two quarternary centers. This first application of the Rh(I)-catalyzed [(5+2)+1] cycloaddition in natural product synthesis highlights the efficiency of this methodology for constructing complex fused ring systems.

Abstract:The development of protocols for constructing chiral medium-sized heterocycles with high efficiency and excellent stereocontrol is of great interest owing to their ubiquitous occurrence in natural products and biologically active pharmaceuticals. Nonetheless, current synthetic approaches are limited due to unfavorable enthalpy and entropy factors, as well as transannular interactions. The present work addresses this issue by designing an asymmetric allylation/ring expansion reaction of 2-(1-hydroxyallyl)phenols and cyclobutanone carboxamides enabled by sequential iridium/zinc/bifunctional squaramide catalysis, affording a series of 8-membered benzo[b]oxocines in high yields with high diastereo- and enantioselectivities. Mechanistic investigation reveals that the enantioselectivity is controlled by the chiral iridium catalyst, while density functional theory calculations demonstrate that the diastereoselectivity is controlled by the chiral bifunctional squaramide catalyst. Abstract:An attractive and novel methodology involving Pd/Cu-catalyzed tandem head-to-tail dimerization/cycloisomerization of terminal ynamides for the synthesis of 3,5-disubstituted oxazolones was developed. Under Pd(PPh3)2Cl2/CuI cooperative catalyzed reaction conditions, it provided efficient access to 5-vinyloxazolones with exceptional functional group tolerance and good chemoselectivity. The control experiments demonstrated that Pd(PPh3)2Cl2 serves a key role in the dimerization of terminal ynamides and shows low catalytic activity in the intramolecular cyclization. Moreover, the hetero-Diels-Alder reaction of product 5-vinyloxazolones was also described, which provided polycyclic oxazolones in good yield.
