Abstract:A highly regio- and stereoselctive palladium-catalyzed domino reaction of functionalized aryl allyl ethers has been developed. Various aryl allyl ethers derived from the phosphine-catalyzed addition of electron-deficient allenes with phenol are found to be efficient substrates for the synthesis of 2-substituted 2,3-dihydrobenzofurans and indolines. It is the first example of aryl allyl ether used as an ideal and practical precursor of hard to get functionalized 1,3-butadiene for the heterocyclic compound synthesis.

Abstract:Reactions between 1,4-dibromo-1,3-butadienes and 2,5-disubstituted pyrroles afforded cyclopenta[c]pyridine derivatives in high yield, catalyzed by palladium and a cyclopentadiene-phosphine ligand (L1). Insertion of one terminal carbon of the butadienyl skeleton into one C═C double bond in the pyrrole ring resulted in ring expansion, along with a 1,2-shift of an alkyl or an aryl substituent on the butadienes.

Abstract:An efficient synthesis of diversified indolizine derivatives was developed via CuBr-catalyzed reaction of pyridines, methyl ketones and alkenoic acids under solvent free conditions in oxygen atmosphere. This synthesis involves cascade processes of copper-catalyzed bromination of the methyl ketone, 1,3-dipolar cycloaddition of the pyridinium ylide with the alkenoic acid, followed by oxidative decarboxylation and dehydrogenative aromatization of the primary cycloadduct. By this protocol, a wide range of indoliznes with different substitution patterns were selectively prepared in one pot from simple substrates in good to excellent yields.

Abstract:An efficient Pd-catalyzed cleavage of C(sp(3))-N bonds in secondary amines and a consequent C(sp(2))-N and C(sp(3))-N coupling process was developed. Various secondary amines could be used to react with alkenyl or aryl dibromides, affording pyrroles and indoles in high yields. Cyclopentadiene-phosphine ligands, a new type of P-olefin ligand, were found to be able to promote the efficiency of this Pd-catalyzed process remarkably. A reactive Pd complex coordinated with a cyclopentadiene-phosphine ligand was successfully isolated and structurally characterized.

Abstract:A direct and convenient strategy for the assembly of Indolo[1,2-f]phenanthridine via Pd-catalyzed tandem cyclization reaction is presented. The current strategy delivers a range of Indolo[1,2-f]phenanthridine by utilizing readily available 1-(2-iodophenyl)-1H-indole and commercially available o-bromobenzoic acids as the starting materials, in which two C-C bonds were formed through Pd-catalyzed C-H bond activation and decarboxylation.

Abstract:An unprecedented Pd-catalyzed highly stereoselective synthetic strategy for the construction of a broad spectrum of polycyclic fused indoline skeletons employing N-halobenzoyl o-haloanilines and N-(prop-2-yn-1-yl)anilines as the substrates in the presence of Pd(PPh3)4 (10 mol%), Cu2O (20 mol %), BINOL-based phosphoramidite L (12 mol %), and K2CO3 (2.0 equiv) in 1,2-dichloroethane at 110 °C for 19 h under an argon atmosphere has been described. This Palladium-catalyzed protocol, which proceeds through a sequential intramolecular dearomative Heck annulation/phenyl electrophilic addition/dehydrogenation sequences, provides a straightforward, powerful, and atom-economical approach for accessing elaborated indoline quinoline derivatives in moderate-to-good yields.

Abstract:A novel cyclization reaction of pyridinium 1,4-zwitterionic thiolates and propiolic acid derivatives mediated by triethylamine is described, which allows the facile synthesis of indolizines under mild reaction conditions. The net transformation involves an acetylide-driven formal [5 + 1] annulation reaction followed by a spontaneous ring-contraction/sulfur extrusion process of transient pyridothiazine intermediates.

Abstract:The synthesis of pyridothiazines was achieved using pyridinium 1,4-zwitterionic thiolates and α-functionalized bromoalkanes via a formal [5+1] pathway. Further studies demonstrated that the synthesis of indolizines could be accomplished via a stepwise [(5+1)-1] pathway with unstable pyridothiazines as the transient intermediates from the same starting materials. The net transformation involves a sequence of annulation, oxidation and ring-contraction/desulfuration.

Abstract:A palladium-catalyzed dialkylation of 1,3-dien-5-ynes was developed using alkenyl double bonds as the initiator and terminator for the synthesis of functionalized indene derivatives. The reactions were performed under mild reaction conditions, affording the corresponding multi-substituted indene derivatives in high efficiency via unprecedented 5- endo cyclization and alkylation processes. It was found that the substituent location at the alkenyl double bond was essential for the chemoselective synthesis of the indene and naphthalene derivatives, respectively.

Abstract:Here we report B(C6F5)3/CPA‐catalyzed enantioselective aza‐Diels–Alder reaction of 3,3‐difluoro‐2‐Aryl‐3H‐indoles with unactivated dienes to access chiral 10,10‐difluoro‐tetrahydropyrido[1,2‐a]indoles. This protocol allows the formation of pyrazole‐based C2‐quaternary indolin‐3‐ones with high enantioselectivities and regioselectivities. Moreover, gram‐scale synthesis of the 10,10‐difluoro‐tetrahydropyrido[1,2‐a]indole skeleton was successfully achieved without any reduction in both yield and enantioselectivity.

Abstract:A novel palladium-catalyzed cascade reaction of o-iodostyrenes, internal alkynes and formates to access various indene-1-acetates was developed. The method enables the construction of three C-C and one C-O bonds through sequential double carbopalladation and aryloxycarbonylation under a palladium/air system. This protocol exhibited high regioselectivity and broad functional group tolerance. In addition, the synthetic utility of this protocol has been successfully demonstrated by the gram-scale synthesis and the late-stage modification of a series of complex bioactive molecules.

Abstract:3‐Idolenone is a key intermediate in the synthesis of many drugs and plays an important role in synthetic chemistry and biochemistry. A new method for the synthesis of trifluoromethylated 3‐indolenones by carbonylation catalyzed by Pd(0) is described. In the absence of an additive, 1‐chloro‐3,3,3‐trifluoro‐propylene (an inexpensive ozone‐friendly CF3 structural unit) reacts with indole and carbon monoxide to synthesize trifluoromethylatedindolenone in good yield, regioselectivity and chemoselectivity and with strong resistance to basal functional groups such as alkynes, aldehydes and esters. Notably, in addition to indole compounds being well transformed into the corresponding products, pyrrole, heteroindole can also is also suitable for the corresponding chemical transformation. This study provides a scientific method for the late construction of trifluoromethylated indole/pyrrole 3‐ketene.

Abstract:A direct synthesis of multi-substituted pyrrolo[1,2-a]pyrazines via palladium(II)-catalyzed C(sp)–C(sp2) cascade coupling and intramolecular cyclization in the presence of ligand was developed. This reaction originates from phenylboronic acids and readily synthesized 2-carbonyl- or 2-formylpyrroloacetonitriles, and affords products in good to excellent yields for a diversity of substrates. Additionally, a possible mechanism for the transformation is proposed.
