Abstract:A Pd-catalyzed annulative π-extension reaction of 1-arylpyrroles using 1,2-dihalobenzenes as the coupling partners was investigated. The higher reactivity of pyrrole C2−H bond compared to C−H bonds of the aryl unit of 1-arylpyrroles allows selective synthesis of pyrrolo[1,2-f]phenanthridines via successive palladium-catalyzed direct intermolecular followed by intramolecular direct arylation steps. From 1-bromo-2-iodobenzenes bearing substituents at C4- or C5-positions and ortho-, meta- or para-substituted 1-arylpyrroles, the access to pyrrolo[1,2-f]phenanthridines containing substituents at C5, C6, C7, C8, C10 and/or C11 positions is possible.

Abstract:A novel palladium-catalyzed decarboxylative coupling reaction of vinyl benzoxazinanones with arynes which may feature an intramolecular nucleophilic attack of an amino group at the central carbon of π-allylpalladium intermediate has been developed. The cis-5,5a,6,10b-tetrahydroindeno[2,1-b]indoles were generated in moderate to good yields. One key to the success of the present reaction was to achieve comparable rates for the palladium-catalyzed decarboxylation and aryne formation steps.

Abstract:A direct and convenient strategy for the assembly of Indolo[1,2-f]phenanthridine via Pd-catalyzed tandem cyclization reaction is presented. The current strategy delivers a range of Indolo[1,2-f]phenanthridine by utilizing readily available 1-(2-iodophenyl)-1H-indole and commercially available o-bromobenzoic acids as the starting materials, in which two C-C bonds were formed through Pd-catalyzed C-H bond activation and decarboxylation.

Abstract:A novel palladium-catalyzed cascade reaction of o-iodostyrenes, internal alkynes and formates to access various indene-1-acetates was developed. The method enables the construction of three C-C and one C-O bonds through sequential double carbopalladation and aryloxycarbonylation under a palladium/air system. This protocol exhibited high regioselectivity and broad functional group tolerance. In addition, the synthetic utility of this protocol has been successfully demonstrated by the gram-scale synthesis and the late-stage modification of a series of complex bioactive molecules.

Abstract:Ethyl and phenyl oxanilates were readily prepared from N-(2-bromobenzyl)anilines and oxalyl chloride monoethyl and monophenyl esters, respectively. It was found that the ethyl oxanilate survived in the presence of K2CO3 in DMA at 120 degrees C and underwent an intramolecular direct arylation using Pd(OAc)(2)-dppf, furnishing the 5,6-dihydrophenanthridine derivative. In contrast, the corresponding phenyl oxanilates decomposed upon exposure to K2CO3 in DMA at 120 degrees C and were transformed into N-arylisoindolin-1-ones via Pd(OAc)(2)-dppf-catalyzed intramolecular decarbonylative coupling. Except for the 4-methoxy-substituted oxanilic acid phenyl ester, other phenyl oxanilates possessing electron-withdrawing (NO2, Cl) and weak electron-donating (Me) substituents provided the N-arylisoindolin-1-ones in 43-80% yields.

Abstract:A facile strategy for the synthesis of valuable indolines has been developed, involving the palladium(II)/Brønsted acid co-catalyzed annulation of readily available (2-aminophenyl) methanols and sulfoxonium ylides. The protocol allows for the direct utilization of the OH group as a leaving group, tolerates alkyl and aryl groups on the N atom of the aniline moiety, operates under mild reaction conditions, and exhibits good efficiency.

Abstract:An efficient [bis-(trifluoroacetoxy)iodo]benzene (PIFA)-mediated oxidative C-N bond formation is developed for the synthesis of 1H-indazoles from readily available arylhydrazones. The reaction tolerates a wide range of functional groups and has broad scope of substrates. Moreover, this method is a relative green and reliable method for rapid preparation of substituted 1H-indazoles under mild conditions.

Abstract:An efficient one-pot three-component palladium-catalyzed domino reaction of aryl iodide, 2-bromophenylboronic acid, and norbornadiene to produce phenanthrenes has been developed. Norbornadiene serves both as the activator of ortho-C-H bond and the source of ethylene via a retro-Diels-Alder reaction. The method features inexpensive and readily available substrates, a broad range of functional groups, and good yields.

Abstract:A palladium-catalyzed synthesis of polysubstituted indoles and isoquinolines through the coupling of aryl bromides with 2-alkynyl arylazides or 2-alkynyl benzylazides has been developed. This method provides straightforward access to indoles and isoquinolines with high efficiency and excellent functional group compatibility. In this transformation, the iminophosphorane in situ generated from azides is served as the nucleophile that attacks the alkyne moiety in the cyclization process.

Abstract:A Pd-catalyzed dearomative arylvinylation reaction of indoles with N-arylsulfonylhydrazones as coupling partners is developed, which proceeds via a domino sequence involving dearomative carbopalladation, Pd-carbene migratory insertion, and regioselective β-hydride elimination. A range of 3-vinylindolines bearing vicinal quaternary and tertiary carbon stereocenters is achieved in moderate yields. The enantioselective reaction was established by using a chiral BINOL-based phosphoramidite ligand, which led to the arylvinylation products in good enantiomeric ratios and excellent diastereoselectivities.

Abstract:A practical process for the synthesis of 2-unsubstituted indoles and azaindoles has been developed by the palladium-catalyzed direct annulation of electron-poor o-chloro/bromoanilines and o-chloroaminopyridines with aldehydes. Coupled with the previous results of Jia and Zhu, this allows rapid access to a variety of 2-unsubstituted indoles and azaindoles starting from simple and easily accessible precursors.

Abstract:An efficient and novel route for the synthesis of 1H-indol-2-yl-(4-aryl)-quinolin-2(1H)-one 1 via palladium-catalyzed site-selective cross-coupling reaction and cyclization process was described. Reaction of 3-bromo-4-trifloxy-quinolin-2(1H)-one 3 with arylboronic acid catalyzed by PdCl2(PPh3)2 afforded 3-bromo-4-aryl-quinolin-2(1H)-one 4, which then reacted with 2-ethynylaniline 5 via Pd-catalyzed Sonogashira coupling and CuI-mediated cyclization leading to the desired 1H-indol-2-yl-(4-aryl)-quinolin-2(1H)-one 1 in good yields.
