Abstract:A new eremophilane sesquiterpene, capnodiumone (1), along with five known eremophilane sesquiterpenes (2-6) and eight other compounds (7-14), have been isolated from a mangrove-associated fungus Capnodium sp. SZ-F22. The chemical structures were elucidated on the basis of extensive spectroscopic analysis. The broth extract of the fungus exhibited a good inhibitory effect on the mycelium growth against Fusarium graminearum at 100g/mL, however, all the 14 compounds showed no expected antifungal activity. Abstract:Objective: To explore the secondary metabolites of the higher fungus Stereum hirsutum. Methods: The structures of these compounds were elucidated by extensive spectroscopic analyses and NMR calculations. Results: Three new bisabolane sesquiterpenoids, sterene acids A–C (1–3), and one new phenolic derivative, sterephenol A (4), were isolated from the higher fungus S. hirsutum. Conclusion: Compound 1 is the first example of bisabolane sesquiterpenoid bearing an allene group. It is also the first time to discover bisabolane sesquiterpenoids from the fungus S. hirsutum.

Abstract:Five cadinane sesquiterpenoids, named stereumin A (1), B (2), C (3), D (4) and E (5) were isolated from the CHCl3 extract of the culture broth of the fungal strain CCTCC AF 207024. Based on the sequences at the internal transcribed spacer (ITS) region and partial 28S rDNA, this fungus was identified as a Stereum sp. The structures of the five compounds were elucidated using spectroscopic data from 1D, 2D NMR and HRESIMS experiments, and the structures of 1 and 2 were further confirmed by single-crystal X-ray diffraction analysis. Compounds 1–5 showed nematicidal activities against the nematode Panagrellus redivivus at 400 mg l−1. Among these five compounds, compounds 3 and 4 killed 84.4% and 94.9% of P. redivivus, respectively in 48 h.

Abstract:A new eudesmane-type sesquiterpenoid, artemihedinic acid N (1), together with four known compounds, aspterric acid (2), two naphtho-γ-pyrones, peninaphones A (3) and B (4), and a diphenyl ether (5), were isolated from the rice medium fermentations of the mangrove rhizosphere soil-derived fungus Penicillium sp. HK1-22. The structure of 1 was determined by combined spectroscopic methods including 1D and 2D NMR spectral data. The antibacterial activities of all isolated metabolites were evaluated.

Abstract:The marine fungus Chondrostereum sp. was collected from a soft coral Sarcophyton tortuosum from the South China Sea. This fungus was cultured in glucose-peptone-yeast (GPY) medium and the culture broth was extracted with EtOAc. By the method of 1H NMR pre-screening and tracing the diagnostic proton signals of the methyl groups, two new hirsutane-type sesquiterpenoids, chondrosterins N and O (1 and 2) were isolated from the metabolite extracts. Their structures were elucidated on the basis of MS, 1D and 2D NMR data.

Abstract:Two new sesterterpenes together with related known terpestacin were isolated from the cultural broth of a marine fungal strain (5XNZ5-4) that lives in the gut of a marine crab. The new compounds were identified on the basis of analyses of NMR and mass spectral data. This is the first report on the oxidative derivatives of terpestacin. The new compounds showed no activity in antibacterial and cytotoxicity assays.

Abstract:Two isocoumarin derivatives, stoloniferol A (1) and B (2), a known 5a, 8a-epidioxy-23-methyl-(22E, 24R)-ergosta-6, 22-dien-3 β-ol (3), and a known dihydrocitrinone (4) were isolated from the ethyl acetate extract of the sea squirt-derived fungus, Penicillium stoloniferum QY2-10, and a halophilic fungus, Penicillium notatum B-52, respectively. Their structures were elucidated by spectroscopic methods and optical rotation. The stereochemistry of 2 was determined on the basis of different NOE experiments and chemical transformation. Compound 3 showed cytotoxicity against P388 cells, with an IC 50 value of 4.07 μ.M.

Abstract:Four new compounds including three andrastin-type meroterpenoids hemiacetalmeroterpenoids A-C (1-3), and a drimane sesquiterpenoid astellolide Q (15), together with eleven known compounds (4-14) were isolated from the cultures of the marine-derived fungus Penicillium sp. N-5, while compound 14 was first isolated from a natural source. The structures of the new compounds were determined by analysis of detailed spectroscopic data, and the absolute configurations were further decided by a comparison of the experimental and calculated ECD spectra. Hemiacetalmeroterpenoid A (1) possesses a unique and highly congested 6,6,6,6,5,5-hexa-cyclic skeleton. Moreover, the absolute configuration of compound 14 was also reported for the first time. Compounds 1, 5 and 10 exhibited significant antimicrobial activities against Penicillium italicum and Colletrichum gloeosporioides with MIC values ranging from 1.56 to 6.25 mu g/mL.

Abstract:Eighteen linear triquinane sesquiterpenoids (LTSs), including seventeen previously undescribed ones (hirsutuminoids A-Q), were isolated from the fermentation of the fungus Stereum hirsutum (Willd.) Pers. The structures and absolute configurations of the isolates were characterized by extensive spectroscopic analysis (1D, 2D NMR, and HRMS data), together with comparing the experimental and calculated data of both electronic circular dichroism and NMR data, as well as X-ray crystallography. Based on the literature survey and efforts on constructing the absolute configurations of these LTSs in this study, one empirical rule about the orientations of substitutions at C-2/C-3/C-7/C-9 was summarized. Anti-inflammatory and cytotoxic bioassays showed that only hirsutuminoid B inhibited the nitric oxide (NO) production in RAW 264.7 macrophages with an IC50 value, 18.9 μM.

Molecular and Functional Analyses of Characterized Sesquiterpene Synthases in Mushroom-Forming Fungi

Abstract:Sesquiterpenes are a type of abundant natural product with widespread applications in several industries. They are biosynthesized by sesquiterpene synthases (STSs). As valuable and abundant biological resources, mushroom-forming fungi are rich in new sesquiterpenes and STSs, which remain largely unexploited. In the present study, we collected information on 172 STSs from mushroom-forming fungi with experimentally characterized products from the literature and sorted them to develop a dataset. Furthermore, we analyzed and discussed the phylogenetic tree, catalytic products, and conserved motifs of STSs. Phylogenetic analysis revealed that the STSs were clustered into four clades. Furthermore, their cyclization reaction mechanism was divided into four corresponding categories. This database was used to predict 12 putative STS genes from the edible fungi Flammulina velutipes. Finally, three FvSTSs were selected to experimentally characterize their functions. FvSTS03 predominantly produced Δ-cadinol and FvSTS08 synthesized β-barbatene as the main product; these findings were consistent with those of the functional prediction analysis. A product titer of 78.8 mg/L β-barbatene was achieved in Saccharomyces cerevisiae via metabolic engineering. Our study findings will help screen or design STSs from fungi with specific product profiles as functional elements for applications in synthetic biology.

Abstract:The mushroom Stereum hirsutum is parasitized by Tremella aurantia to form a heterogeneous basidiocarp Jin'er, which has been used as food and folk medicine in Chinese society. In present work, the S. hirsutum was fermented in YMG broth, and four novel mixed terpenes, stereumamides A-D (1-4), which are sesquiterpenes combined with α-amino acids to form quaternary ammonium hybrids, were isolated from the Stereum hirsutum FP-91666 and their structures were elucidated by spectroscopic data analysis. Stereumamides A and D showed antibacterial activity against Escherichia coli, Staphylococcus aureus, and Salmonella typhimurium, with the minimum inhibitory concentration (MIC) values of 12.5-25.0 μg/mL. The stereumamides A-D should be apparently the first example of naturally occurring a quaternary ammonium compound (QAC) conjugated by sesquiterpene with an α-amino acid. QAC is a common antibacterial agent in food industry, which is found in the mycelium of Stereum hirsutum would suggest that the complex basidiocarp is a functional food and veritable folk medicine.

Abstract:Four (1-4) hitherto unknown and seven (5-11) known ergostane-type sterols were isolated from the EtOH extract of the dried fruiting bodies of the polypore macrofungus Fomes fomentarius. On the basis of spectroscopic analysis, the structures of polyhydroxylated sterols 1-4 were elucidated to be (22E,24R)-3β,5α,6β,14α-tetrahydroxyergosta-7,9(11),22-triene (fomentarol A, 1), (22E,24R)-3β,5β,6α,7α-tetrahydroxy-8α,9α-dihydroergosta-14,22-diene (fomentarol B, 2), (22E,24R)-3β,5α-dihydroxy-6β-ethoxyergosta-7,22-diene (fomentarol C, 3), and (22E,24S)-3β,25-dihydroxy-15α-O-β-D-glucopyranosylergosta-7,22-dien-6-one (fomentarol D, 4), respectively. Rings A/B and B/C are in turn cis-fused in compound 2, which is uncommon in natural ergostane-type sterols. The potential biogenetic relationship of 2 and other ergostane-type sterols isolated from F. fomentarius was briefly discussed. Moderate cytotoxic effects of the isolated sterols against a small panel of human cancer cell lines were also established.

Abstract:A new chloro-trinoreremophilane sesquiterpene 1, three new chlorinated eremophilane sesquiterpenes 2-4, together with a known compound, eremofortine C (5), were isolated from an Antarctic deep-sea derived fungus, Penicillium sp. PR19N-1. Structures were established using IR, HRMS, 1D and 2D NMR techniques. In addition, the plausible metabolic network of these isolated products is proposed. Compound 1 showed moderate cytotoxic activity against HL-60 and A549 cancer cell lines.

Abstract:To study the constituents and structures of secondary metabolite of Penicillium verruculosum YL-52.The compounds was isolated by extraction,silica gel and Sephadex LH-20 column chromatography.Their structures were elucidated by IR,ESI-MS,1H NMR,13C NMR and DEPT.Five compounds were obtained from the ethyl acetate,of which 2 and 3 were identified as Ursolic acid and Evernic acid.The two identified compounds ursolic acid(2) and evernic acid(3) were both firstly found in the fermentation liquid of Penicillium verruculosum.

Abstract:Two new ring A‐cleaved lanostane‐type triterpenoids, glometenoid A (1) and glometenoid B (2), together with four known steroids, (20S,22E,24R)‐ergosta‐7,22‐dien‐3β,5α,6β‐triol (3), (3β,5α,8α,22E)‐ergosta‐6,22‐diene‐3,5,8‐triol (4), 5α,8α‐epidioxy‐22E‐ergosta‐6,22‐dien‐3β‐ol (5), and ergosterol (6), were isolated from the endophytic fungus Glomerella sp. F00244. Their structures were elucidated by comprehensive spectroscopic analyses of NMR and MS data. Their antimicrobial activities were evaluated against pathogenic bacteria Bacillus subtilis ATCC 9372, Staphylococcus aureus ATCC 25923, Bacillus pumilus CMCC (B) 63202, Micrococcus luteus CMCC28001, and pathogenic fungi Candida albicans AS2.538 and Aspergillus niger ACCC30005, but no inhibition was observed at a concentration of 20 μg/ml. Further cytotoxicity assessment revealed that compound 1 exhibited weak antiproliferative activity against ovarian cancer HeLa cell.
