Abstract:Eighteen linear triquinane sesquiterpenoids (LTSs), including seventeen previously undescribed ones (hirsutuminoids A-Q), were isolated from the fermentation of the fungus Stereum hirsutum (Willd.) Pers. The structures and absolute configurations of the isolates were characterized by extensive spectroscopic analysis (1D, 2D NMR, and HRMS data), together with comparing the experimental and calculated data of both electronic circular dichroism and NMR data, as well as X-ray crystallography. Based on the literature survey and efforts on constructing the absolute configurations of these LTSs in this study, one empirical rule about the orientations of substitutions at C-2/C-3/C-7/C-9 was summarized. Anti-inflammatory and cytotoxic bioassays showed that only hirsutuminoid B inhibited the nitric oxide (NO) production in RAW 264.7 macrophages with an IC50 value, 18.9 μM.

Abstract:Three new acetylenic aromatic compounds, sterehirsutynes A-C (1-3), along with three known congeners, frustulosinol (4), vibrayne (5), and sterehirsutinol (6), were isolated from the culture broth of Stereum hirsutum. Their structures were determined by detailed analyses of NMR and high-resolution mass. Chiral column analysis showed that compounds 2 and 3 were racemic mixtures. These new compounds were evaluated for porcine pancreatic lipase (PPL) inhibitory activities, and compounds 1 and (+/-) 3 showed moderate inhibitory activity against PPL, with IC50 values of 23.2 +/- 1.04 and 21.8 +/- 2.15 mu M, respectively.

Abstract:Five cadinane sesquiterpenoids, named stereumin A (1), B (2), C (3), D (4) and E (5) were isolated from the CHCl3 extract of the culture broth of the fungal strain CCTCC AF 207024. Based on the sequences at the internal transcribed spacer (ITS) region and partial 28S rDNA, this fungus was identified as a Stereum sp. The structures of the five compounds were elucidated using spectroscopic data from 1D, 2D NMR and HRESIMS experiments, and the structures of 1 and 2 were further confirmed by single-crystal X-ray diffraction analysis. Compounds 1–5 showed nematicidal activities against the nematode Panagrellus redivivus at 400 mg l−1. Among these five compounds, compounds 3 and 4 killed 84.4% and 94.9% of P. redivivus, respectively in 48 h.

Abstract:The mushroom Stereum hirsutum is parasitized by Tremella aurantia to form a heterogeneous basidiocarp Jin'er, which has been used as food and folk medicine in Chinese society. In present work, the S. hirsutum was fermented in YMG broth, and four novel mixed terpenes, stereumamides A-D (1-4), which are sesquiterpenes combined with α-amino acids to form quaternary ammonium hybrids, were isolated from the Stereum hirsutum FP-91666 and their structures were elucidated by spectroscopic data analysis. Stereumamides A and D showed antibacterial activity against Escherichia coli, Staphylococcus aureus, and Salmonella typhimurium, with the minimum inhibitory concentration (MIC) values of 12.5-25.0 μg/mL. The stereumamides A-D should be apparently the first example of naturally occurring a quaternary ammonium compound (QAC) conjugated by sesquiterpene with an α-amino acid. QAC is a common antibacterial agent in food industry, which is found in the mycelium of Stereum hirsutum would suggest that the complex basidiocarp is a functional food and veritable folk medicine.

Abstract:Three new isopimarane-type diterpenoids, botrysphins G-I (1–3), a new muurolane-type sesquiterpenoid, 11,12-dihydroxylentideusether (4), and two new triketides, 4-dechlorobotrysphone C (5) and 4,5-dihydroxy-3-methoxy-6-undecanoyloxy-2-cyclohexen-1-one (6), together with one known diterpenoid, sphaeropsidin A (7), one sesquiterpenoid, lentideusether (8), and one triketide sphaeropsidone (9), were isolated from culture of the fungus Botrysphaeria laricina associated with the moss Rhodobryum umgiganteum. The structures of the new compounds were established on the basis of extensive spectroscopic techniques including HRMS and 1D and 2D NMR data. Compounds 1 and 2 exhibited NO inhibitory activity with IC50 values of 13.9 μM and 41.9 μM, respectively. At the same time, these two compounds showed quinone reductase inducing activity with 2.7-fold of induction for 1 at 12.5 μM and 1.6-fold for 2 at 25.0 μM.

Abstract:From the ethyl acetate extract of the culture broth of Beltrania rhombica, two new eudesmane sesquiterpenes, named rhombidiol (1) and rhombitriol (2), were isolated and characterized along with five enantiomers of known sesquiterpenes: (-)-beta-eudesmol, (-)-pterocarpol, (-)-chrysanthemol, (-)-longilobol and (-)-5beta-hydroxy-beta-eudesmol. Their structures were determined by analysis of 1D and 2D NMR data and comparison of spectral data and physical data with those previously reported.

Abstract:A new ursane-type triterpenoid, 3 beta-hydroxy-urs-30-p-Z-hydroxycinnamoyloxy-12- en-28-oic-acid (1), together with three known triterpenoids, 3 beta-hydroxy-urs-30-p-E-hydroxycinnamoyloxy-12-en-28-oic-acid (2), 2 alpha, 3 beta, 19 alpha-trihydroxy-urs-12-en-28-oic-acid (3), and ursolic acid (4), four known lignans, pinoresinol (5), 9 alpha-hydroxypinoresinol (6), (+)-medioresinol (7), and (+)-kobusin (8), and two steroids, beta-sitosterol (9), and daucosterol (10), were isolated from the whole parts of Teucrium viscidum. Their structures were established by a combination of spectroscopic data analysis, besides comparison with literature data. Compounds 1-4 were evaluated for their inhibitory activities against 11 beta-hydroxysteroid dehydrogenase 1 (11 beta-HSD1).

Abstract:Three new holostan-type triterpene glycosides, arguside F (1), impatienside B (2), and pervicoside D (3), together with a known saponin, holothurin B ( 4) were isolated from the sea cucumber Holothuria (Microthele) axiloga H. L. Clark. On the basis of spectroscopic data and chemical reactions, the structures of the new compounds were elucidated. Compound 2 showed significant antifungal activities against six strains (1 < or = MIC(80) < or = 4 microg/mL). The stereochemistry of holothurin B (4) isolated from the title sea cucumber was also solved through X-ray diffraction analysis.

Abstract:In the course of our screening program for novel metabolites from tunicate-derived fungi, novel sesquiterpenoids, named JBIR-27 (1) and -28 (2), together with known sporogen-AO1 and phomenone, were isolated from the culture broth of Penicillium sp. SS080624SCf1. The structures of 1 and 2 were determined to be eremophilane analogs on the basis of extensive NMR and MS analyses. Sporogen-AO1, phomenone and 2 showed cytotoxicity against human cervical carcinoma cell line HeLa at IC(50) values of 8.3, 19 and 92 microM, respectively, whereas 1 was inactive at a concentration of 80 microM.

Abstract:In our ongoing study of fungal bioactive natural products, 12 previously undescribed triquinane sesquiterpene glycosides, namely, antrodizonatins A-L (1-12), and four known compounds (13-16) have been obtained from the fermentation of the basidiomycete Antrodiella zonata. The structures were established unambiguously via extensive spectroscopic analysis and theoretical calculations of electronic circular dichroism spectra. This is the first report of triquinane sesquiterpene glycosides. Compounds 1, 5, and 12 displayed antibacterial activity against Staphylococcus aureus with MIC50 values of 35, 34, and 69 μM, respectively.

Abstract:The fungal strains Pseudogymnoascus are a kind of psychrophilic pathogenic fungi that are ubiquitously distributed in Antarctica, while the studies of their secondary metabolites are infrequent. Systematic research of the metabolites of the fungus Pseudogymnoascus sp. HSX2#-11 led to the isolation of six new tremulane sesquiterpenoids pseudotremulanes A–F ( 1 – 6 ), combined with one known analog 11,12-epoxy-12β-hydroxy-1-tremulen-5-one ( 7 ), and five known steroids ( 8 – 12 ). The absolute configurations of the new compounds ( 1 – 6 ) were elucidated by their ECD spectra and ECD calculations. Compounds 1 – 7 were proved to be isomeride structures with the same chemical formula. Compounds 1 / 2 , 3 / 4 , 1 / 4 , and 2 / 3 were identified as four pairs of epimerides at the locations of C-3, C-3, C-9, and C-9, respectively. Compounds 8 and 9 exhibited cytotoxic activities against human breast cancer (MDA-MB-231), colorectal cancer (HCT116), and hepatoma (HepG2) cell lines. Compounds 9 and 10 also showed antibacterial activities against marine fouling bacteria Aeromonas salmonicida . This is the first time to find terpenoids and steroids in the fungal genus Pseudogymnoascus .

Abstract:Two undescribed protostane triterpenoids, 11‐deoxy‐13(17),15‐dehydro‐alisol B 23‐acetate (2) and alisol S (3), together with 21 known ones (1, 4‐23), were isolated from the dried rhizome of Alisma plantago‐aquatica. Of these compounds, 13(17),15‐Dehydro‐alisol B 23‐acetate (1) and 11‐deoxy‐alisol Q 23‐acetate (2) are two protostane trierpenoids containing conjugated double bonds in the five‐membered ring D that are rarely found from nature resource, while alisol S (3) is a protostane triterpenoid with undescribed tetrahydrofuran moiety linked via C20‐O‐C24 at the side chain. Additionally, compound 18 is a new natural product, and cycloartenol triterpenoid 23 is a non protostane triterpenoid firstly isolated from genus Alisma. Their structures were elucidated by extensive spectral analysis of the UV, IR, MS, 1D and 2D NMR, and comparison of the experimental and calculated CD curves.

Abstract:Three novel andrastin-type meroterpenoids, penicimeroterpenoids A–C (1–3), possessing two unprecedented skeletons consisting of fused 6/5/6/6/7 and 6/5/6/6/4 polycyclic systems, were obtained from the marine-derived fungus Penicillium sp. Abstract:Studies of the fermentation broth of fungus Antrodiella gypsea led to the isolation of a new bisabolane-type sesquiterpenoid that was named gypseatriol (1), together with the known compound 2,10-dodecadiene-1,6,7-triol (2). The structure of this new metabolite was assigned by analysis of 2D NMR and HR-EI-MS. Absolute configuration was assigned by single crystal X-ray diffraction analysis. Compound 1 was evaluated for its antifungal activity on Candida albicans.

Abstract:Two new trichothecenes, named 8α-hydroxyroridin H and myrothecin A, along with six known compounds, 8α-acetoxy roridin H, isororidin K, verrucarin A, verrucarin J, verrucarin L and 8α-acetoxy verrucarin L, were isolated from the fermentation broth of a halotolerant fungus Myrothecium sp. GS-17, which was separated from the soil sample of a salina. Structure elucidation and NMR signal assignments were achieved on the basis of spectroscopy. In addition, compounds 1 and 2 were active against plant pathogenic fungi Rhizoctonia solani and Fusarium oxysporum.

Their structures were determined by spectroscopic methods, the in situ dimolybdenum CD method, and modified Mosher's method. The bioassays results showed that 2 exhibited weak cytotoxicity against K-562 cells.
