Abstract:Fungi are important resource for the discovery of novel bioactive natural products. This study investigated the metabolites produced by Mariana-Trench-associated fungus Aspergillus sp. SY2601 in EY liquid and rice solid media, resulting in the isolation and structure determination of 28 metabolites, including five new compounds, asperindopiperazines A–C (1–3), 5-methoxy-8,9-dihydroxy-8,9-deoxyaspyrone (21), and 12S-aspertetranone D (26). Structures of the new compounds were elucidated based on extensive NMR spectral analyses, HRESIMS data, optical rotation, ECD, and 13C NMR calculations. The new compound 12S-aspertetranone D (26) exhibited antibacterial activity against both methicillin-resistant Staphylococcus aureus and Escherichia coli with MIC values of 3.75 and 5 μg/mL, respectively.

Abstract:Termitomyces albuminosus is the symbiotic edible mushroom of termites and cannot be artificially cultivated at present. In the project of exploring its pharmaceutical metabolites by microbial fermentation, four new selinane type sesquiterpenoids-teucdiol C (1), D (2), E (3), and F (4), together with two known sesquiterpenoids teucdiol B (5) and epi-guaidiol A (6)-were obtained from its fermented broth of T. albuminosus. Their structures were elucidated by the analysis of NMR data, HR Q-TOF MS spectral data, CD, IR, UV, and single crystal X-ray diffraction. Epi-guaidiol A showed obvious anti-acetylcholinesterase activity in a dose-dependent manner. The experimental results displayed that T. albuminosus possess the pharmaceutical potential for Alzheimer's disease, and it was an effective way to dig new pharmaceutical agent of T. albuminosus with the microbial fermentation technique.

Abstract:Mariana Trench sediments are enriched in microorganisms, however, the structures and bioactivities of their secondary metabolites are not very known. In this study, a fungus Penicillium sp. SY2107 was isolated from a sample of Mariana Trench sediment collected at a depth of 11000 m and an extract prepared from the culture of this fungus in rice medium showed antimicrobial activities. Chemical investigation on this active extract led to the isolation of 16 compounds, including one novel meroterpenoid, named andrastone C. Structure of the new compound was elucidated based on high-resolution electrospray ionization mass spectroscopy (HRESIMS) data, extensive nuclear magnetic resonance (NMR) spectroscopic analyses and a single crystal X-ray diffraction. The crystal structure of a known meroterpenoid andrastone B was also reported in this study. Both andrastones B and C exhibited antimicrobial activities against methicillin-resistant Staphylococcus aureus (MRSA), Escherichia coli, and Candida albicans with minimum inhibitory concentration (MIC) values in a range from 6 to 13 mu g/mL.

Abstract:Five cadinane sesquiterpenoids, named stereumin A (1), B (2), C (3), D (4) and E (5) were isolated from the CHCl3 extract of the culture broth of the fungal strain CCTCC AF 207024. Based on the sequences at the internal transcribed spacer (ITS) region and partial 28S rDNA, this fungus was identified as a Stereum sp. The structures of the five compounds were elucidated using spectroscopic data from 1D, 2D NMR and HRESIMS experiments, and the structures of 1 and 2 were further confirmed by single-crystal X-ray diffraction analysis. Compounds 1–5 showed nematicidal activities against the nematode Panagrellus redivivus at 400 mg l−1. Among these five compounds, compounds 3 and 4 killed 84.4% and 94.9% of P. redivivus, respectively in 48 h.

Abstract:Four new alkyl aromatics, penixylarins A–D (1–4), along with the known biogenetically related 1,3-dihydroxy-5-(12-hydroxyheptadecyl)benzene (5) and 1,3-dihydroxy-5-(12-sulfoxyheptadecyl)benzene (6), were isolated from a mixed culture of the Antarctic deep-sea-derived fungus Penicillium crustosum PRB-2 and the mangrove-derived fungus Xylaria sp. HDN13-249. UPLC-MS data and an analysis of structural features showed that compounds 1 and 2 were produced by collaboration of the two fungi, while compounds 3–6 could be produced by Xylaria sp. HDN13-249 alone, but in noticeably increased quantities by cocultivation. Compounds 2, 3, 5, and 6 showed antibacterial activity against a panel of strains, and compound 3 possessed potential antituberculosis effects (MIC = 6.25 μM against Mycobacterium phlei).

Abstract:Streptomide (1), a new amide analogue, streptomynone (2), a new quinolinone, and ten known compounds including three aliphatic acids (3‐5), two amides (6‐7), four cyclic dipeptides (8‐11), and an adenosine (12) were also obtained from the fermentation broth of a Streptomyces isolate collected from a sediment sample of soil sample collected in Bendong Village, Huadong Town, Chuxiong, China. Their structures were determined by analysis of the 1D/2D‐NMR and HR‐ESI‐MS spectra. Compound 12 presented weak antimicrobial activities against Candida albicans and Aligenes faecalis (MIC = 64 μg/mL). Compounds 7 and 12 showed weak cytotoxic activity against MHCC97H.

Abstract:Marine fungi are a promising source of novel bioactive natural products with diverse structure. In our search for new bioactive natural products from marine fungi, three new phenone derivatives, asperphenone A-C (1-3), have been isolated from the ethyl acetate extract of the fermentation broth of the mangrove-derived fungus, Aspergillus sp. YHZ-1. The chemical structures of these natural products were elucidated on the basis of mass spectrometry, one- and two-dimensional NMR spectroscopic analysis and asperphenone A and B were confirmed by single-crystal X-ray crystallography. Compounds 1 and 2 exhibited weak antibacterial activity against four Gram-positive bacteria, Staphylococcus aureus CMCC(B) 26003, Streptococcus pyogenes ATCC19615, Bacillus subtilis CICC 10283 and Micrococcus luteus, with the MIC values higher than 32.0 mu M.

Abstract:Three new mycophenolic acid derivatives, penicacids L–N (1–3), together with four known analogues, were isolated from a fungus Penicillium sp. HN‐66 derived from a South China Sea marine sediment. The structures of compounds 1–3 were determined on the basis of HR‐ESI‐MS, NMR (1H, 13C, HSQC and HMBC) data analyses, and comparison of optical rotations. Antimicrobial activities of 1–7 were tested. The results showed that compounds 1–3 and 5–7 had weak inhibitory effects against E. coli ATCC 25922 with the MIC values of 50 μg/mL.

Abstract:Abstract A total of 14 compounds including a novel tetrasubstituted benzene derivative peniprenylphenol A were isolated from a scaled-up culture of the Indonesian mangrove sediment-derived fungus Penicillium chrysogenum ZZ1151 in rice medium. Structures of the isolated compounds were determined based on their NMR spectroscopic analyses, HRESIMS data, optical rotation calculations and comparison with the reported data. New peniprenylphenol A (1) was found to have antimicrobial activities against human pathogenic methicillin-resistant Staphylococcus aureus (MRSA), Escherichia coli and Candida albicans with MIC values of 6, 13, and 13 µg/mL, respectively. The known compounds of penicimumide (2), preparaherquamide (5), uridine (6), thymine (7), 1,2-seco-trypacidin (8) communol G (9), clavatol (10), 4-hydroxybenzeneacetic acid methyl ester (11), 2,5-dihydroxyphenylacetic acid methyl ester (12), 2-hydroxyphenylacetic acid methyl ester (13) and 4-hydroxyphenylethanone (14) showed antimicrobial activity against at least one of the three tested pathogens with MIC values of 3–25 µg/mL. Abstract:To study fermentation from strain Aspergillus sp. YQ-13, dwelling in sediment Kueishantao hydrothermal vents off Taiwan. Compounds were separated and purified by silica gel column, preparative HPLC techniques. Their structures were identified by the physicochemical properties and spectral analysis. The isolates were identified as 3-hydroxy-2-(2-hydroxy-6-methoxy-4-methylbenzoyl)-5-methoxy-benzoic acid methyl ester (1), myristic acid (2), orcinol (3), 1,2-seco-trypacidin (4), leporin A (5), chaetominine (6), 5-hydroxy-2-hydroxymethyl-4H-pyran-4one (7) and N-methyl-2-pyrolidinone (8). Compound 1 was identified as a new natural product belonging to diphenyl ketone family. Known compounds 5 and 6 exhibited antibiotic activity with MIC value of around 1 to 25 μg mL−1 against Bacillus subtilis, Klebsiella pneumoniae, methicillin resistant Staphylococcus aureus (MRSA), Pseudomonas aeruginosa, Staphylococcus aureus, Escherichia coli and Acinetobacter Bauman. Compounds 1, 4, 5 and 6 were tested by the methods of DPPH and FRAP assay, showing moderate antioxidant activities.

Abstract:An intense relativistic electron beam (IREB) was injected into an un-magnetized axially decaying plasma. Observed were excitation of rapidly oscillating electric fields such that W = ε0<;E2>;/2nTe >; 1 which lasted as long as about 30 times longer than the beam duration, broadening of the distribution of beam electrons about the initial energy, acceleration of plasma electrons, current enhancement and the total microwave power radiated in the frequency range 18-40 GHz. The experimental data are discussed in connection with the strong Langmuir turbulence.

Abstract:In our ongoing study of fungal bioactive natural products, 12 previously undescribed triquinane sesquiterpene glycosides, namely, antrodizonatins A-L (1-12), and four known compounds (13-16) have been obtained from the fermentation of the basidiomycete Antrodiella zonata. The structures were established unambiguously via extensive spectroscopic analysis and theoretical calculations of electronic circular dichroism spectra. This is the first report of triquinane sesquiterpene glycosides. Compounds 1, 5, and 12 displayed antibacterial activity against Staphylococcus aureus with MIC50 values of 35, 34, and 69 μM, respectively.

Abstract:As the development of genetic and bioinformatic, the strategy of "bottom-up", combined with genome and transcriptome techniques, was considered as an efficient and practical method to break through the limitation of traditional discovery of natural products. Generally, comparative transcriptome analysis could be useful to guide the optimization of fungal cultivation conditions in which the transcriptional level of interesting compounds is higher. The transcriptome analysis of the algal endophytic fungus Penicillium chermesinum EN-480 indicated that fermentation of this fungus in modified rice solid medium could produce some metabolites different from those cultivated in other media. Four new meroterpenoids (compounds 1-4, namely, chermesins E-H) were characterized and their structures were determined by HRESIMS and NMR spectra. The absolute configurations were confirmed by NOESY experiments, X-ray diffraction analysis, and comparison of ECD cotton effects. Antimicrobial activities against human- and aqua-bacteria as well as against plant-pathogenic fungi were assayed. The plausible biosynthesis pathway of these compounds was discussed.

Abstract:Chemical investigation of the culture extract of an endophyte Xylaria curta YSJ-5 from Alpinia zerumbet (Pers.) Burtt. et Smith resulted in the isolation of eight previously undescribed compounds including five eremophilane sesquiterpenes xylarcurenes A-E, one norsesquiterpene xylarcurene F, and two α-pyrone derivatives xylarpyrones A-B together with eight known related derivatives. Their chemical structures were extensively established based on the 1D- and 2D-NMR spectroscopic analysis, modified Mosher's method, electronic circular dichroism calculations, single-crystal X-ray diffraction experiments, and the comparison with previous literature data. All these compounds were tested for in vitro cytotoxic, anti-inflammatory, α-glucosidase inhibitory, and antibacterial activities. As a result, 6-pentyl-4-methoxy-pyran-2-one was disclosed to display significant antibacterial activity against Staphylococcus aureus and methicillin-resistant S. aureus with minimal inhibitory concentration value of 6.3 μg/mL.

Abstract:Objective To investigate the antibacterial chemical constituents of the marine-derived fungus Aspergillus violaceus WZXY-m64-17.Methods After large-scale solid fermentation, the secondary metabolites were isolated from the fermentation extract based on column chromatography over silica gel, Sephadex-LH 20, C18 ODS and semi-prepared HPLC.Their structures were elucidated through spectroscopic analysis of NMR, MS, UV and IR, as well as comparison with the data of literature.The antibacterial activities were assayed using broth dilutionmethod.Results Six compounds were isolated from the EtOAc extract and their structures were identified as 2-hydroxyemodin (1), emodin (2), nidurufin (3), versicolorin B (4), sterigmatocystin (5), terrequinone A (6).Compounds 4 and6 were found to inhibit the growth of Staphylococcus aureus and MRSA M6.Conclusion The fermentation extract of the fungus Aspergillus violaceus WZXY-m64-17 could produce secondary metabolites, and some of which showed antibacterial activity.

Abstract:Two new sulfur-containing phenolic compounds, 7-hydroxy-5-hydroxymethyl-2H-benzo[1,4]thiazin-3-one (1) and 2,5-dihydroxy-3-methanesulfinylbenzyl alcohol (2), along with two known compounds, 3-chloro-2,5-dihydroxybenzyl alcohol (3) and 2-hydroxy-6-methylbenzoic acid (4), were isolated from the mycelial solid culture of a soil-derived Ampelomyces fungus by antibacterial assay-guided fractionation. Their structures were elucidated on the basis of spectroscopic analysis. Compounds 1-3 showed structure and microbial dependent antibacterial activities.

Abstract:Four new isocoumarins, alternariethers A‐C (1‐3) and alternariester (4) were separated from the fermentation of the fungus Alternaria malorum FL39, purified from Myoporum bontioides. Their structures were ascertained using NMR and HR‐ESI‐MS spectroscopy. For compound 4, the absolute configuration was solved with the help of ECD calculation and the DP4+ method. Compared with the positive control triadimefon, compound 1 showed more potent antifungal effects on Colletotrichum musae . The antifungal effects of compounds 1, 2, and 3 on Fusarium oxysporum and Fusarium graminearum, of compound 4 on F. oxysporum, were equal to those of triadimefon. Except for compound 4 which was inactive against Escherichia coli with O78 serotype, all compounds showed moderate or weak antibacterial activity against Staphylococcus aureus and E. coli with O6 or O78 serotype.

Abstract:Five new bisabolane sesquiterpenes, a new polyketide, along with seven known compounds, were isolated from endophyte Schizophyllum commune associated with a famous medicinal and edible plant, Gastrodia elata. Most compounds 1-12, and extract indicated antifeedant activities against silkworm with feeding deterrence index (FDI) of 21-85 %, at concentrations of 20 μg/cm2 , 40 μg/cm2 , respectively. Compound 6 indicated obvious insecticidal activity with fatality rate of 60 %, at the concentration of 20 μg/cm2 . Five bisabolane sesquiterpenes, two ergosterols, and a glyceride showed insecticidal synergism by combining with abamectin. Interesting, ergosterol peroxide (13) distributed widely in mushrooms and fungi, was found to have feeding attractant activities on insects and antifungal activity against entomopathogen Beauveria bassiana. The reciprocal relationship should be occurred between S. commune and pests for the fungus produced ergosterol peroxide to attract the pests propagating spore, and its anti-entomopathogen activity was also benefit for the health of insects.
