Abstract:Gap junctions provide a communication pathway between adjacent cells. They are formed by paired connexons that reside in the plasma membrane of their respective cell and their activity can be modulated by the bilayer composition. In this work, we study the dynamic behavior of a Cx26 connexon embedded in a POPC lipid bilayer, studying: the membrane protein interactions and the ion flux though the connexon pore. We analyzed extensive atomistic molecular dynamics simulations for different conditions, with and without calcium ions. We found that lipid-protein interactions were mainly mediated by hydrogen bonds. Specific amino acids were identified forming hydrogen bonds with the POPC lipids (ARG98, ARG127, ARG165, ARG216, LYS22, LYS221, LYS223, LYS224, SER19, SER131, SER162, SER219, SER222, THR18 and TYR97, TYR155, TYR212, and TYR217). In the presence of calcium ions, we found subtle differences on the HB lifetimes. Finally, these MD simulations are able to identify and explain differential chlorine flux through the pore depending on the presence or absence of the calcium ions and its distribution within the pore.

Abstract:Calcium ions play a key role in cell signaling processes, yet the molecular nature of their interaction with heterogenous membranes is difficult to study with traditional techniques. Parameters were developed for sodium, calcium and chloride ions for use with the CgProt force field for molecular dynamics simulations of membrane‐protein systems. Similar to atomistic simulations, highly attractive behavior was observed for calcium with zwitterionic POPC/POPE membrane interfaces. Both sodium and calcium preferentially bound to the phosphate region of anionic POPG bilayers. The addition of CaCl2 to implicit solvent simulations had the effect of stabilizing the lipid bilayer. Sub‐molar concentrations of calcium adsorb onto POPE and POPG membranes and enable low or zero tension simulations. Close agreement in the area per lipid was obtained compared to results from atomistic simulations, indicating the utility of the model for large scale membrane simulations. Abstract:Diffusion of multivalent metallic ions into aqueous solution of rigid, negatively charged macromolecules of high concentration is an effective approach to prepare macroscopically anisotropic hydrogels. However, the mechanism for superstructure formation is still not clear. By observing the mixing process of a small drop of CaCl2 solution with solution of a rigid polyanion, poly(2,2′-disulfonyl-4,4′-benzidine terephthalamide) (PBDT), under the polarizing optical microscope, the diffusion profile of Ca2+ and detailed anisotropic gelation process of PBDT are revealed. Diffusion of Ca2+ into the surrounding PBDT solution immediately induces the formation of physical liquid crystalline (LC) gel with concentric alignment of PBDT. The thickness d of this region increases with diffusion time t, obeying the diffusion law d ∼ t1/2. Abstract:Calcium cation (Ca2+) is a key element to the cell membrane functions. Its effects on liquid crystal vesicle deposition have already been learnt. In this study, it is found that Ca2+ can also influence the gel vesicle deposition by controlling the vesicle rupture and fusion on SiO2 at temperatures lower than the main transition temperature. Particular analyses were given to the vesicle–SiO2 and inter-vesicle attractions that are originated from the Ca2+ bridging effect. It is concluded that the aggregate condition of vesicles should be taken into consideration when dealing with vesicle deposition on a solid substrate.
Asymmetrical Calcium Ions Induced Stress and Remodeling in Lipid Bilayer Membranes

Abstract:Ca2+ ions play crucial roles in regulating many chemical and biological processes, but their impact on lipid bilayer membranes remains elusive, especially when the impacts on the two leaflets are asymmetrical. Using a recently developed multisite Ca2+ model, we performed molecular dynamics simulations to study the impact of Ca2+ on the properties of membranes composed of POPC and POPS and observed that both the structure and fluidity of the membranes were significantly affected. In particular, we examined the influence of asymmetrically distributed Ca2+ on asymmetric lipid bilayers and found that imbalanced stress in the two leaflets was generated, with the negatively charged leaflet on the Ca2+-rich side becoming more condensed, which in turn induced membrane curvature that bent the membrane away from the Ca2+-rich side. We employed continuum mechanics to study the large-scale deformations of a spherical vesicle and found that the vesicle can go through vesiculation to form a multi-spherical shape in which a number of spheres are connected with infinitesimal necks, depending on the specific Ca2+ distributions. These results provide new insights into the underlying mechanisms of many biological phenomena involving Ca2+-membrane interactions and may lead to new methods for manipulating the membrane curvature of vesicles in chemical, biological, and nanosystems.

Synthesis of Size-Controlled Acid-Resistant Hybrid Calcium Carbonate Microparticles As Templates for Fabricating “Micelles-Enhanced” Polyelectrolyte Capsules by the LBL Technique

Abstract:Size-controlled, low-dispersed calcium carbonate microparticles were synthesized in the presence of the amphiphilic block copolymer polystyrene-b-poly(acrylic acid) (PS-b-PAA) by modulating the concentration of block copolymer in the reactive system. This type of hybrid microparticles have acid-resistant properties. By investigating the aggregation behaviors of PS-b-PAA micelles by transmission electron microscopy (TEM), the mechanism of hybrid calcium carbonate formation illustrated that the block copolymer served not only as "pseudonuclei" for the growth of calcium carbonate nanocrystals, but also forms the supramicelle congeries, a spherical framework, as templates for calcium carbonate nanocrystal growth into hybrid CaCO(3) particles. Moreover, this pilot study shows that the hybrid microparticle is a novel candidate as a template for fabricating multilayer polyelectrolyte capsules, in which the block copolymer is retained within the capsule interior after core removal under soft conditions. This not only facilitates the encapsulation of special materials, but also provides "micelles-enhanced" polyelectrolyte capsules.
Size-Selective Adhesion of Calcium Oxalate Monohydrate Crystals to Lipid Membranes

Abstract:The retention of calcium oxalate monohydrate (COM) crystals on cell membranes is pivotal in kidney stone formation. However, the mechanisms underlying COM attachment to neutral lipid membranes remain unclear. In this study, we demonstrate that COM exhibits size-selective adhesion to fluid lipid membranes composed of lipids with distinct sizes. Specifically, the (100) facet of COM induces the formation of new domains and establishes strong adhesion in the 18:1 (Δ9-Cis) PC (DOPC) membrane, while the (010) facet induces domains with strong adhesion in the 16:0-14:0 PC membrane. This selectivity is linked to the compatibility of the area per lipid in DOPC with the unit cell area of the (100) facet and the area per lipid in 16:0-14:0 PC with the (010) facet. Our Raman spectroscopic analyses reveal that the lipid acyl chains within these induced domains exhibit a higher degree of ordering compared to the typical fluid state of the membrane. This ordered structural alignment, combined with the lateral size-matching effect, suggests the potential formation of molecular arrays within the lipid bilayer that are in harmony with the lattice dimension of COM. To elucidate the strong adhesion between calcium oxalate and the phospholipid head group in the absence of a direct molecular structural correspondence, we propose that crystal water associated with COM can form hydrogen bonds with the phospholipid head group. Using structure visualization software, we demonstrate the feasibility of such hydrogen bonding networks. The formation of this network could serve to stabilize and enhance the attachment of COM to the lipid membrane. This mediation by water molecules offers a plausible explanation for the pronounced affinity at the interface.

“Annular Ring” Microtubes Formed by SDS@2β-CD Complexes in Aqueous Solution

Abstract:Traditional aqueous self-assembly of tubular structures (as well as other aggregates) usually relies on the hydrophobic effect, a relatively weak and nondirectional interaction. The resultant aggregates are inherently soft, fluid, and less-ordered. Alternatively, we report a novel kind of nonamphiphilic self-assembly of microtubes in aqueous solutions of cyclodextrin/ionic surfactant (CD/IS) complexes. This self-assembly is driven exclusively by H-bonds, relatively strong, directional interactions. The CD/IS microtubes feature an unbundling nature, ultralong persistence lengths, highly monodispersed diameters, and remarkable rigidity. Every single CD/IS microtube is constituted by a set of coaxial, equally spaced, hollow cylinders, resembling the annular rings of trees (thus termed as “annular ring” microtubes). Furthermore, bearing in mind the fundamental difference between the amphiphilic counterpart in driving forces, this H-bond-driven hydrophilic self-assembly is envisioned to complement its counterpart and expand the field of molecular self-assembly.

Abstract:Cyclodextrin (CD)/surfactant complexes were usually believed to be quite soluble in water and unable to form aggregates because of the hydrophilic outer surface. However, in this work, SDS@2β-CD complex is found to be able to self-assemble into well-defined lamellar structures in concentrated aqueous solution. The lamellae possess unprecedented in-plane solid-crystalline order in addition to classical lamellar liquid-crystalline order. Such a combination in orderliness makes the lamellar phase an intermediate phase between a solid and a liquid crystal. Upon dilution, the lamellae transform to microtubes and then to vesicles. The three classes of SDS@2β-CD aggregates share a consistent building block, the channel-type crystalline bilayer membrane. Moreover, since the outer surface of SDS@2β-CD is hydrophilic, its self-assembly behavior, unlike traditional amphiphilic assembly, does not rely on the hydrophobic effect. Therefore, the present nonamphiphilic self-assembly of SDS@2β-CD is envisioned to open new possibilities for self-assembly chemistry.

Abstract:Self-assembly behavior of an ABC triblock copolymer, poly(ethylene glycol)-b-polystyrene-b-poly(ε-caprolactone) (PEG-b-PS-b-PCL), in aqueous media is presented. The formed micelle structures were analyzed by using transmission electron microscopy, scanning electron microscopy, and laser light scattering. Various fascinating multicompartmental aggregates, including multilamellar vesicles, cylinder-containing vesicles, entrapped vesicles, and porous large compound micelles, were prepared from four copolymers with various block lengths of PEG and PS. The phase separation of hydrophobic PS and PCL blocks, as well as the hydrophilic/hydrophobic balance, plays a crucial role on the self-assembly behaviors. The mechanism regarding the formation of these fascinating aggregates is also suggested.

Abstract:While lipids form soft, fluidic membranes (soft assembly), proteins can readily assemble into rigid, crystalline structures such as viral capsids and bacterial compartments (lattice assembly). The key difference has to do with the driving forces, where the former is driven by the weak, directionless hydrophobic effect and the latter, by a combination of relatively strong, directional intermolecular interactions. In synthetic systems, the lipid assembly has been massively replicated but the protein assembly has been rarely rivaled. Herein, we briefly review these two kinds of assemblies with special emphasis on a recently reported lattice self-assembly system of cyclodextrin complexes. The complexes arrange themselves into an in-plane, rhombic lattice that develops into lamellar, tubular, and polygonal structures depending on concentration. We will then cover the formation mechanisms, driving forces, and an application of the tubes in particle encapsulation. We hope that this short review would draw people's attention to this emerging field of lattice self-assembly.
Molecular‐Curvature‐Induced Spontaneous Formation of Curved and Concentric Lamellae Through Nucleation

Abstract:Spontaneous formation of concentric lamellae was observed in self- assembling giant surfactants consisting of a fluorinated polyhedral oligomeric silsesquioxane (FPOSS) head and flexible polymer tail(s). Owing to the asymmetrical sizes of the head and tail blocks and the rectangular molecular interface, the giant surfactants assumed a truncated- wedge- like molecular shape, which induced morphological curvature during self- assembly, thus resulting in the formation of curved and concentric lamellae. These curved/concentric lamellae were observed in FPOSS- based giant surfactants with different architectures and compositions. The spontaneous curvature formation not only promotes our fundamental understanding of assembly principles, but also provides a promising and efficient approach to the fabrication of a wide range of high- performance devices.

Abstract:The morphologies of biomimetic mineralized octacalcium phosphate (OCP) are extremely versatile. Numerous studies have addressed the gel phases, surfactants, polyelectrolytes and thin layers as convenient media to produce OCP nanostructures with various morphologies. However, hardly any studies have to date discussed the concept of controlling the structural evolution of these nanostructures by organic molecule mediation. This paper aims at opening the pathway to such OCP assemblies with levels of morphological expression and evolution from an aqueous solution consisting of highly diluted acidic macromolecule and amphiphilic small molecule as co-mediators. This approach differs in that the usual organic molecules are used to shape the single OCP morphology, and specifically establish an evolutionary relationship between the lamellar clusters and spherical aggregates. We present a possible model that gives qualitative description for the morphology of OCP grown in a variety of molar ratios between acidic macromolecule and small molecule surfactant. We believe that extension of the present approach to other biominerals and inorganic materials will prepare a large variety of highly flexible biomimetic nanostructures with elaborate superstructures across nanoscale to macroscopic dimensions.

Single Lipid Bilayers Constructed on Polymer Cushion Studied by Sum Frequency Generation Vibrational Spectroscopy

Abstract:Planar solid supported single lipid bilayers on mica, glass, or other inorganic surfaces have been widely used as models for cell membranes. To more closely mimic the cell membrane environment, soft hydrophilic polymer cushions were introduced between the hard inorganic substrate and the lipid bilayer to completely avoid the possible substrate-lipid interactions. In this article, sum frequency generation (SFG) vibrational spectroscopy was used to examine and compare single lipid bilayers assembled on the CaF(2) prism surface and on poly (L-lactic acid) (PLLA) cushion. By using asymmetric lipid bilayers composed of a hydrogenated 1,2-dipalmitoyl-sn-glycerol-3-phosphoglycerol (DPPG) leaflet and a deuterated 1,2-dipalmitoyl-(d62)-sn-glycerol-3-phosphoglycerol (d-DPPG) leaflet, it was shown that the DPPG lipid bilayers deposited on the CaF(2) and PLLA surfaces have similar structures. SFG has also been applied to investigate molecular interactions between an antimicrobial peptide Cecropin P(1) (CP1) and the lipid bilayers on the above two different surfaces. Similar results were again obtained. This research demonstrated that the hydrophilic PLLA cushion can serve as an excellent substrate to support single lipid bilayers. We believe that it can be an important cell membrane model for future studies on transmembrane proteins, for which the possible inorganic substrate-bilayer interactions may affect the protein structure or function.
Hierarchical self-assembly of a PS-b-P4VP/PS-b-PNIPAM mixture into multicompartment micelles and their response to two-dimensional confinement

Abstract:Hierarchical self-assembly offers an elegant and energy-efficient bottom-up strategy for the fabrication of complex materials with precisely controllable morphologies and internal structures. Herein, pupa-like multicompartment micelles (MCMs) were readily fabricated via the hierarchical self-assembly of a polystyrene-block-poly(4-vinylpyridine) (PS-b-P4VP) and polystyrene-block-poly(N-isopropylacrylamide) (PS-b-PNIPAM) block copolymer mixture in a THF/water mixture solvent, which were unable to be obtained by any of the individual block copolymers (BCPs). This means that the hierarchical self-assembly is a result of the synergistic cooperation between the two BCPs. Moreover, a kinetic study revealed that the MCMs were formed by hierarchical self-assembly of small spherical micelles (SSMs), which were co-assembled from the PS-b-P4VP/PS-b-PNIPAM mixture. Subsequently, we investigated the self-assembly of a PS-b-P4VP/PS-b-PNIPAM mixed solution confined in the nanopores of an anodic aluminum oxide (AAO) template. In such two-dimensional confinement, long multicompartment micelles (LMCMs) with a period multilayer structure were obtained. Notably, the confinement effect of nanopores on the hierarchical self-assembly could be distinguished into two different situations according to the activity of secondary assembly of the preformed SSMs from different BCP compositions, i.e., dynamic and static confinement. The dynamic confinement can affect the Brownian movement of SSMs and thus promotes their fusion to form spherical micelles with larger size compared with the SSMs formed under an unconfined condition. For the situation where AAO nanopores were partially filled with the preformed SSMs, the static confinement could decrease the stretching of BCP chains along the short axis of LMCMs and thus induce the formation of long range ordered multilayer nanostructures. These results illustrated that the synergistic effect played an important role in the hierarchical assembly of BCPs; meanwhile, such hierarchical self-assembly could be further manipulated by cylindrical confinement to rationally tune the nanostructures and dimensions of the BCP assemblies.

Abstract:Polymer-supported phospholipid bilayers (PLBs) are popular model systems for the study of transmembrane proteins under conditions close to cellular membrane environments. In this work, by combining the techniques of dissipative quartz crystal microbalance and atomic force microscopy, we investigate the deposition of vesicles on a hydrated cationic poly(diallyldimethylammonium chloride) (PDDA) layer as a function of phospholipid composition and sodium chloride concentration. The vesicles used consist of phospholipid mixtures with varying amounts of net negative charge. Uniform PLBs are formed by either increasing the negative charge density of the vesicles or decreasing sodium chloride concentration, suggesting that the electrostatic attraction between the vesicle and PDDA layer is the driving force for the formation of the PLBs. Our results indicate that the PLB formation is a fast adsorption-rupture process of the vesicles, without passing through a critical vesicle density. We further contend that the fluctuating PDDA support plays a central role for this process. This work provides a framework for understanding the key factors that influence the formation of PLBs.

Abstract:Supported lipid bilayers (SLBs) are useful structures for mimicking cellular membranes, and they can be integrated with a variety of sensors. While there are a variety of methods for forming SLBs, many of these methods come with limitations in terms of the lipid compositions that can be employed and the substrates upon which the SLBs can be deposited. Here we demonstrate the use of an all-aqueous chaotropic agent exchange process that can be used to form SLBs on two different substrate materials: SiO2, which is compatible with traditional SLB formation by vesicle fusion and Al2O3, which is not compatible with vesicle fusion. When examined with quartz crystal microbalance with dissipation monitoring, the SLBs generated by chaotropic agent exchange (CASLBs) have similar frequency and dissipation shifts to SLBs formed by the vesicle fusion technique. The CASLBs block nonspecific protein adsorption on the substrate and can be used to sense protein-lipid interactions. Fluorescence microscopy was used to examine the CASLBs, and we observed long-range lateral diffusion of fluorescent probes, which confirmed the CASLBs were composed of a continuous, planar lipid bilayer. Our CASLB method provides another option for forming planar lipid bilayers on a variety of surfaces, including those that are not amenable to the widely used vesicle fusion method.
