Abstract:The separation of micro and nanoscale colloids is a necessary step in most biological microassay techniques, and is a common practice in microchemical processing. Chemical waves are frequently encountered in biochemical systems driven far from equilibrium. Here, we put forward a strategy for separating small suspending colloids by means of their surfing on substrate chemical wavefronts. The colloids with catalytic activities sensitive to the substrates are activated to show self-propulsion and consequently exhibit a chemotactic response to the traveling wavefronts, which results in their spontaneous separation from the multicomponent complex mixture via self-diffusiophoresis. The dynamics of the process is analyzed through a particle-based simulation. In addition, it is found that separation can be carried out according to particle size. The mechanisms underpinning the chemical and physical separation processes are discussed, and the dependencies on the reaction rate constant and particle size are presented. The results may prove relevant for further experimental and theoretical studies of separation in complex active environments.

Abstract:Nanoseparation and concomitant purification of nanoparticles by ultracentrifugation in a nonhydroxylic organic density gradient has been demonstrated by separating several typical colloidal nanoparticles, including Au, Ag, and CdSe. Successful separation of Au nanowires from their spherical counterparts showed that colloidal particles can be separated not only by size but also morphology. In addition to extending the range of colloidal systems which can be separated and providing monodisperse samples that cannot be obtained by synthesis optimization alone, this method simplifies the postsynthesis treatment process and facilitates subsequent bulk assembly of the monodisperse colloids. Dissolution of organic polymers in the gradient medium both enhances the separation efficiency and also allows the direct fabrication of functional composite films with discrete monodisperse nanoparticles embedded inside.

Abstract:The experimental conditions necessary for the synthesis of well-defined nanoparticles are often difficult to control. There is thus a compelling need for post-synthesis separation of nanoparticles polydispersed in size and shape. We demonstrate here both theoretically and experimentally that gold nanorods with diverse aspect ratios can be separated using density gradient centrifugation. By analysing the force balance of a Brownian rod falling in a Stokes flow, we derive a rigorous and predictive model that reveals the quantitative dependency of the nanorod sedimentation rates on their mass and shape. The calculations show that while mass dependency is still the dominating factor during centrifugation, the shape factor is not insignificant. Relatively heavier but long and thin rods could sediment slower than certain size of lighter spheres, and some rods and spheres with different masses and shapes may never be separated. This mass and shape dependency is exploited to separate as-prepared gold nanorod colloids by sucrose gradient centrifugation. Two layers of nanorods with narrow aspect-ratio distributions are obtained.

Abstract:The authors describe a multiphase density gradient centrifugation method for the separation and purification of hybrid nanoparticles (NPs). The method was applied to the purification of NPs consisting of magnetite gold NPs of type Fe3O4@Au and coated with 1-dodecylthiol. A density gradient has been established by forming cushions of a multicomponent solution composed of (a) a nonlinear gradient of sucrose in the of 5–50 % concentration range, (b) a constant concentration of ficoll (a neutral, highly branched, high-mass, hydrophilic polysaccharide; 30 % w/w), and (c) a constant concentration of percoll (colloidal silica particles coated with polyvinylpyrrolidone; 15 % w/w). The final separation profiles are adjusted via dynamic differential centrifugation at 4480×g for 30 min. The mixture of NPs to be separated is added and centrifuged at 2520×g for 40 min. This will separate the NPs into five fractions together with other secondary components. The fractions were characterized by FTIR, XPS, and SEM-EDX techniques. Abstract:The separation and detection of microparticles within complex samples pose substantial challenges due to the intricate variations in size and concentration. A strategy employing gravity-assisted gradient size exclusion principle based on controllable gap sizes on the surface of silicon nanowire arrays (SiNWAs) has been established to achieve the separation of microparticles with diverse sizes. The formation of gradient gap sizes was accomplished by meticulously investigating the impact of oxidation-reduction reactions through metal-assisted chemical etching. Particles of different sizes were initially aggregated at the accumulation base, followed by a sequential size exclusion process within the finely regulated 0.9-12.5 μm gradient-gap-sized separation region facilitated with gravity-assisted, leading to a comprehensive separation of microparticles based on their respective size differences, progressing from small to large. The effective separation of four model-sized microparticles demonstrated a separation degree of ≥2.7, purity of ≥96.1 %, RSDs of ≤4.6 %, and a separation capacity of up to 107 particles. The separation efficacy of this gradient-sized chip was verified by evaluating the more complex atmospheric particulates with varying sizes, which exhibited separation degree ranging between 2 and 10. This method offers a precise separation range, easily adjustable separation sizes, and simple operation, rendering it a versatile tool for separating complex samples.

Abstract:This paper demonstrates the use of aqueous multiphase systems (MuPSs) as media for rate-zonal centrifugation to separate nanoparticles of different shapes and sizes. The properties of MuPSs do not change with time or during centrifugation; this stability facilitates sample collection after separation. A three-phase system demonstrates the separation of the reaction products (nanorods, nanospheres, and large particles) of a synthesis of gold nanorods, and enriches the nanorods from 48 to 99% in less than ten minutes using a benchtop centrifuge.

Abstract:Mesoporous titanium dioxide sub-microspheres were prepared using aerosol techniques with a size distribution from 80 nm to 3 mu m. Both theoretical and experimental results showed that non-equilibrium sucrose density gradient centrifugation is an effective way to size-partition these titanium dioxide nanoparticles from a continuous and broad particle size range. The sucrose serves as a multifunctional solution and plays three significant roles during the metal oxide fractionation. First, the high viscosity and density make the sedimentation rate of nanomaterials sensitive to particle size, which leads to particle fractionation in solution. Second, sucrose greatly decreases aggregation among nanoparticles during the separation by acting as a non-ionic capping agent. No other capping agent or surfactant is required. Finally, the density gradient stratifies the nanoparticles with a similar size into well-defined layers, so that the size-selected particles are relatively easy to collect. In addition, the unique biocompatibility of sucrose makes this fractionation method an ideal candidate for biological applications of nanoparticles. Post-aerosol synthesis separation of mesoporous metal oxide nanoparticles using a non-equilibrium density gradient has proven to be an effective, scalable way to access a large fraction of TiO2 sub-microspheres within a narrow size range and a low polydispersity index.

Abstract:Gold nanoparticles (AuNPs) have found widespread applications in life sciences. While synthesis of monodispersed AuNPs has been fairly convenient by using chemical reduction of chloroauric acid by sodium citrate, we found that AuNPs of high quality and high concentrations were not readily obtained via this method. As an example, we showed that monodispersed 13-nm AuNPs were readily synthesized at relatively low concentrations ( e. g. 3.5 nM); in contrast, 13-nm AuNPs of 17 nM obtained by the direct reduction method were irregularly shaped and not well dispersed. In this work, we demonstrated that AuNPs of high concentration could be prepared by a two-step approach, i.e. chemical reduction at low concentrations and subsequent centrifugation. Compared to the direct reduction method, this new two-step method led to AuNPs with high salt resistance and high stability, which are essential for the preparation of DNA-AuNPs conjugates for DNA biodetection.

Abstract:Density-gradient centrifugation is a label-free approach that has been extensively used for cell separations. Though elegant, this process is time-consuming (>30 min), subjects cells to high levels of stress (>350 g) and relies on user skill to enable fractionation of cells that layer as a narrow band between the density-gradient medium and platelet-rich plasma. We hypothesized that microfluidic adaptation of this technique could transform this process into a rapid fractionation approach where samples are separated in a continuous fashion while being exposed to lower levels of stress (<100 g) for shorter durations of time (<3 min). To demonstrate proof-of-concept, we designed a microfluidic density-gradient centrifugation device and constructed a setup to introduce samples and medium like Ficoll in a continuous, pump-less fashion where cells and particles can be exposed to centrifugal force and separated via different outlets. Proof-of-concept studies using binary mixtures of low-density polystyrene beads (1.02 g/cm³) and high-density silicon dioxide beads (2.2 g/cm³) with Ficoll-Paque (1.06 g/cm³) show that separation is indeed feasible with >99% separation efficiency suggesting that this approach can be further adapted for separation of cells.

Abstract:Individual single–walled carbon nanotubes (SWNTs) with distinct electronic types are crucial for the fabrication of SWNTs–based electronic and magnetic devices. Herein, the water–soluble polymethyl(1–butyric acidyl)silane (BA–PMS) was synthesized via the hydrosilylation reaction between 3–butenoic acid and polymethylsilane catalyzed by 2,2′–azodibutyronitrile. As a new dispersant, BA–PMS displayed a quite good dispersing capacity to arc–discharged SWNTs and moderate selectivity for metallic species. The application of sucrose–DGU, the density gradient ultracentrifugation with sucrose as the gradient medium, to the co–surfactants (BA–PMS and sodium dodecyl sulfonate) individually dispersed SWNTs yielded metallic SWNTs of 85.6% purity and semiconducting SWNTs of 99% purity, respectively. This work paves a path to the DGU separation of the SWNTs dispersed by polymer–based dispersants with hydrophobic alkyl chains.

Abstract:The separation and purification of a sample of interest is essential for subsequent detection and analysis procedures, but there is a lack of effective separation methods with which to purify nano-sized particles from the sample media. In this paper, a microfluidic system based on negative magnetophoresis is presented for the high-resolution separation of nanoparticles. The system includes on-chip magnetic pole arrays and permalloys that symmetrically distribute on both sides of the separation channel and four permanent magnets that provide strong magnetic fields. The microfluidic system can separate 200 nm particles with a high purity from the mixture (1000 nm and 200 nm particles) due to a magnetic field gradient as high as 10,000 T/m being generated inside the separation channel, which can provide a negative magnetophoretic force of up to 10 pN to the 1000 nm particle. The overall recovery rate of the particles reaches 99%, the recovery rate of 200 nm particles is 84.2%, and the purity reaches 98.2%. Compared with the existing negative magnetophoretic separation methods, our system not only exhibits high resolution on particle sizes (800 nm), but also improves the sample processing throughput, which reaches 2.5 μL/min. The microfluidic system is expected to provide a new solution for the high-purity separation of nanoparticles, as well as nanobiological samples.

Abstract:Over the past decades, nanoparticles have increased in implementation to a variety of applications ranging from high-efficiency electronics to targeted drug delivery. Recently, microfluidic techniques have become an important tool to isolate and enrich populations of nanoparticles with uniform properties (<i><i>e.g.</i></i>, size, shape, charge) due to their precision, versatility, and scalability. However, due to the large number of microfluidic techniques available, it can be challenging to identify the most suitable approach for isolating or enriching a nanoparticle of interest. In this review article, we survey microfluidic methods for nanoparticle isolation and enrichment based on their underlying mechanisms, including acoustofluidics, dielectrophoresis, filtration, deterministic lateral displacement, inertial microfluidics, optofluidics, electrophoresis, and affinity-based methods. We discuss the principles, applications, advantages, and limitations of each method. We also provide comparisons with bulk methods, perspectives for future developments and commercialization, and next-generation applications in chemistry, biology, and medicine.This article has not yet been cited by other publications.

Abstract:The microfluidic passive control of microparticles largely relies on the hydrodynamic effects of the carrier media such as Newtonian fluids and viscoelastic fluids. Yet the viscoelastic/Newtonian interfacial effect has been scarcely investigated, especially for high-resolution particle separation. Here we report a microfluidic co-flow of Newtonian (water or PBS) and viscoelastic fluids (PEO) for the size-dependent separation of microparticles. The co-flow condition generates a stable viscoelastic/Newtonian interface, giving rise to the wall-directed elastic lift forces that compete with the center-directed lift forces, and efficiently hinders the migration of microparticles from the Newtonian to the viscoelastic fluid in a size-dependent manner. An almost complete separation of a binary mixture of 1 μm and 2 μm polystyrene particles is achieved by the co-flow of water and a very dilute PEO solution (100 ppm), whereas the sole use of water or PEO could not lead to an efficient separation. This co-flow microfluidic system is also applied for the separation of Staphylococcus aureus (1 μm) from platelets (2-3 μm) with >90% efficiencies and purities.
