Abstract:Abstract While a great number of ortho C–H functionalization reactions have been developed and several breakthroughs have been achieved in meta C–H activation, para C–H functionalization is still in its infancy stage. In this article, a versatile strategy for the synthesis of para-substituted arenes has been developed via a tandem process consisting of palladium-catalyzed C–H functionalization and subsequent copper-catalyzed protodecarboxylation of benzoic acids. Both electron-withdrawing and electron-donating functionalities can be introduced into the para positions of arenes bearing a variety of substituents.

Abstract:An efficient Pd(II)-catalyzed synthesis of benzyl benzoates via direct functionalization of benzyl C(sp(3))-H bonds was developed. The method features a broad substrate scope. This method features high tolerance of functional groups, mild reaction condition, and high chemoselective when there are multiple active C(sp(3))-H bonds. A plausible oxidative coupling mechanism was proposed on the basis of mechanistic studies.

Abstract:A novel and efficient palladium‐catalyzed cross‐coupling benzylation reaction of pentafluorobenzene through selective C−H functionalization has been developed. A series of carbonates and polyfluorobenzenes have been examined and found to give moderate to good yields. Abstract:Site selectivity control is of vital importance in the direct functionalization of inert C-H bonds. Reported here is a novel example of remote regiodivergent ortho- and meta-C-H bond functionalizations of phenylethylamine derivatives by using a novel 2-cyanobenzoyl group as the original directing functionality, where the regioselectivity was adjusted by a methylation. The potential of the method was exemplified by sequential functionalizations of both ortho- and meta-C-H bonds of a phenylethylamine derivative in a streamlined manner.

Abstract:Direct para-selective C-H functionalization of arenes remains a daunting challenge and is still significantly restricted to a few scaffolds. Herein, we report an unprecedented pyridine-based para-directing template (DT) assisted, Pd-catalyzed para-C-H alkenylation of three classes of arenes, i.e. phenylpropanoic acids, 2-phenyl benzoic acids and benzyl alcohols, with a series of alkenes including perfluoroalkenes. Notably, the pyridine-based para-DT could be easily synthesized and readily recycled under mild conditions. These results may find application in rapid construction of para-substituted arenes and stimulate the exploration of novel methods for para-C-H functionalization of arenes.

Abstract:An unprecedented method for the palladium (II)-catalyzed direct halogenation of benzothiadiazole derivatives is reported here, which would allow the rational tuning of their electronic properties by further transformations. The key strategy is the combination of the double functions of benzothiadiazole as both an important moiety of functional materials and modifiable directing group. Various mono/poly-chlorinated, brominated or iodinated benzothiadiazole products could be prepared efficiently which can be converted into a wide range of valuable polyaromatic compounds with potential appli-cations in material chemistry.

Abstract:The direct ortho arylation of 4-methyl-N-phenylpyridin-2-amines via palladium(II)-catalyzed C-H activation is described. Treatment of 4-methyl-N-phenylpyridin-2-amine with potassium aryltrifluoroborate using 10 mol % of palladium(II) acetate as the catalyst, 2 equiv of copper(II) acetate as the oxidant, and 1 equiv of p-benzoquinone in tert-butyl alcohol gave ortho-arylated products in modest to excellent yields. This reaction shows good functional group compatibility. A series of H-1 NMR titration experiments and controlled experiments elucidating the reaction mechanism were carried out. The key intermediate, 4-methyl-N-phenylpyridin-2-amine palladacycle, was isolated and characterized by X-ray crystallography. The advanced transformations of ortho-phenylated 4-methyl-N-phenylpyridin-2-amine to N-(4-methylpyridin-2-yl)-9H-carbazole, biphenyl-2-amine, and 3-methyl-6-phenylpyrido[1,2-a]benzimidazole were successfully demonstrated as potential synthetic applications.

Abstract:Dual activation of both C-I and vicinal C-H bonds of diaryliodonium salts allowing for diarylation is a subject of rapid construction of π-extended frameworks. Here, we report palladium-catalyzed cascade of C8-arylation/intramolecular Friedel-Crafts acylation of α-naphthoic acids in the synthesis of benzanthrone derivatives. The step-economical protocol tolerates various substrates, which resulted in a potential molecular library for developing functional polycyclic scaffolds. The approach relies on the synergistic action of strong acid with palladium catalysts to form two bonds in a one-pot procedure.

Abstract:Aryl halides are key building blocks in organic synthesis for the construction of valuable natural products, medicinal and agricultural chemicals via transition metal-catalyzed coupling or substitution reactions. Halogenation is one of the most fundamental and important reactions in organic synthesis. Electrochemical transition-metal-catalyzed C-H functionalization has emerged as a powerful tool for molecular synthesis with the prospect of avoiding the use of costly and toxic oxidants or reductants, thereby reducing the footprint of undesirable, toxic byproducts. The palladium-catalyzed electrochemical C-H chlorination of benzamide derivatives directed by PIP amine directing group under divided cells has been demonstrated, in which readily available inorganic halides salts serve as halogen sources. The reaction features a broad substrate scope, high functional group tolerance, and compatibility of thiophene substrates. This reaction could be conducted on a gram scale, which is important for future application. Additionally, the sequential bromination and chlorination of C(sp(2))-H bond constructs highly functionalized aromatic carboxylic acid derivatives. The typical procedure is as follows: The electrolysis was carried out in an H-type divided cell (anion-exchange membrane), with a RVC anode (10 mm x 10 mm x 12 mm) and a platinum cathode (10 mm x 10 mm x 0.2 mm). The anodic chamber was charged with Pd(OAc)(2) (5.6 mg, 0.025 mmol, 10 mol%) and benzamide derivative (0.25 mmol, 1.0 equiv.) and dissolved in DMF (10 mL). LiCl (847.8 mg, 20.0 mmol) was added in the cathodic chamber and dissolved in water (10 mL). Then the reaction mixture was electrolyzed under a constant current of 5 mA at 90 degrees C until the complete consumption of the starting material as monitored by TLC or H-1 NMR. After the reaction, EtOAc (50 mL) was added to dilute the mixture and then washed with water (20 mL x 3) and then with brine (20 mL). The organic fraction was dried over Na2SO4 and concentrated. The resulting residue was purified by silica gel flash chromatography to give the chlorination product.

Abstract:Palladium-catalyzed regioselective alpha-amino C-H functionalization via the isomerization of alpha,beta-unsaturated carbonyls including esters, ketones, and amides has been established, providing an easy access to a wide array of tricyclic 1,2,3,4-tetrahydro-b-carbolines, azepinoindoles, 2,3,4,5-tetrahydro- 1H-pyrido [4,3-b] indoles, 4,5,6,7- tetrahydro- 1H-pyrrolo [2,3-c] - pyridines, 1,2,3,4-tetrahydropyrazino-[1,2-a]indoles, pyran-fused indoles, and tetrahydroisoquinolines in good to excellent yields. This transformation showed high regioselectivity, excellent functional group tolerance, and scalability. Moreover, this methodology was also employed as the key step for the total synthesis of desbromoarborescidines A, B, and C. Preliminary mechanistic studies revealed that the palladium catalyst not only formed [Pd-H] to promote the isomerization of a unsaturated carbonyls but also played a role as a Lewis acid for the final protonation/cyclization.

Abstract:Pd(II)-catalyzed remote C-H olefination of aromatic tertiary amines was achieved with high meta selectivity. With the assistance of an elaborated template, C-H functionalization of unreactive aryl tertiary amines, hindered by the p-π conjugation between the lone-pair electrons of the nitrogen atom and the phenyl ring, was realized with high meta regioselectivity via a quaternary ammonium salt assembly. The results demonstrate that apart from the distance and geometry of the template, the conformation of the arene substrate also plays a crucial role in the templated-assisted remote C-H functionalization.

Abstract:A palladium-catalyzed Suzuki cross-coupling of benzyl ammonium triflates via activation of a Csp 3 –N bond to construct a Csp 3 –Csp 2 bond is described. Various boronic acids can be coupled with a range of benzylamine-derived quaternary ammonium salts in 1:1 molar ratio by the bench-stable N -heterocyclic carbene palladium complex, SIPr-PdCl 2 -Py to afford diarylmethane derivatives. This reaction exhibits a wide substrate scope and functional group tolerance. Direct arylation of benzylamine in a one-pot process and the gram-scale reaction can be performed successfully.
