Abstract:A mild and efficient method for the synthesis of 2,5-disubstituted chalcogenophenes is reported. This method involves two steps of reactions via one-pot approach by employing commercially available and cost-effective alkynes and element chalcogen. Further, by using LiNH2 as the promoter and DMSO/EtOH as the solvent, Cu-catalyzed Glaser coupling-cycloaddition processes are dramatically enhanced that a diverse range of chalcogenophenes can be obtained in good to excellent yields under mild conditions. Controlled experiments show that trisulfur radical anion (S3 & sdot;-) is the key intermediate and DMSO/EtOH serves as the hydrogen source, while alcohol may facilitate the transition state. The mild and highly efficient constructions of 2,5-disubstitued thiophenes, selenophenes and tellurophenes are achieved by using a one-pot, two-step protocol that combine Cu-catalyzed Glaser coupling with LiNH2-mediated cycloaddition. And the DMSO/EtOH mixture solvent is found to short the reaction time and improve the reaction yield by promoting the production of S3 & sdot;- and stabilizing the transition state.image

Abstract:A concise and efficient synthetic approach was developed for the synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones via a one-pot, three-component reaction of 4-aminocoumarin, arylglyoxal monohydrates, and aromatic amines promoted by KHSO4 in toluene under reflux condition. The attractive features of this protocol are operational simplicity, short reaction time, easy purification of products and good to excellent yields.

Abstract:The Pd(II)-catalyzed intramolecular carbonylative cyclization reaction of aryl alkenes and aryl alkenols is reported for the synthesis of structurally diverse chromanes. PdCl2(CH3CN)(2) was used as the catalyst and CuCl2 as the oxidant under the balloon pressure of CO. The reaction is conducted under mild conditions, and chromane-type esters and lactones can be generated in a highly regio- and stereoselective manner.

Abstract:P-stereogenic tert- and sec-phosphines have wide applications in asymmetric catalysis, materials, and pharmaceutical chemistry, however, their practical synthesis still constitutes a significant challenge. Herein, a successful kinetic resolution of rac-secondary phosphine oxides via the enantioselective P-benzylation process catalyzed by the palladium/Xiao-Phos was designed. Both tert- and sec-phosphine oxides were delivered in good yield and excellent enantiopurity (selectivity factor up to 226.1). The appealing synthetic utilities are further demonstrated by the facile preparation of several valuable P-chiral compounds, precursors of bidentate ligands, as well as transition metal complexes.

Abstract:An efficient tandem process was developed to synthesize diversified benzo[a]xanthones from 2-methyl-3-(1-alkynyl)chromones with electron-deficient chromone-fused dienes. This unusual reaction, involving multiple steps and not requiring the use of transition metal catalysts or an inert atmosphere, results in the formation of three new C-C bonds and one C-O bond.

Abstract:Palladium serves as a multi‐functional catalyst which is controllable by tuning reaction conditions. This work demonstrated the utilization of a palladium catalyst for the synthesis of phenanthrenols by cascade palladium‐catalyzed Suzuki/Heck reaction between chalcone and 2‐bromophenylboronic acid, followed by Michael addition. The sequential reaction could be controlled by reactivity of the palladium catalyst in different solvents and concentrations of reagents. This protocol could be applied to a broad range of substrates to give products in low to good yields.

Abstract:Sonogashira-type cross-couplings are one of the most significant alkynylations in organic chemistry. One of the first palladium-catalyzed intramolecular Heck/Sonogashira reactions of alkenes with terminal alkynes is now reported. With this method, a variety of uniquely substituted chiral benzene-fused heterocycles bearing a propargyl-substituted all-carbon quaternary stereocenter were obtained in a straightforward, high-yielding, and highly stereoselective manner under mild conditions. Salient features of this process include the use of readily available substrates, high selectivities, a broad substrate scope as well as versatile product functionalizations.

Abstract:A tandem Prins/Friedel-Crafts cascade strategy has been developed for the synthesis of hexahydro-2H-benzo[g]chromene derivatives. Under action of equimolecular amount of BF3 etherate, delta,epsilon-unsaturated alcohols and arylacetaldehydes are coupled to form oxocarbenium ion intermediates, which are captured by the alkene moiety through a favored six-membered transition state. The Prins-cyclization resultant tertiary carbenium ions undergo consecutive intramolecular Friedel-Crafts cyclization to create the tricyclic products. This process enables a tandem formation of three new chemical bonds in one single operation with high yields and stereoselectivity in most cases. When cyclic substituents are involved, a variety of spiro derivatives could also be synthesized successfully, which further proved the robustness of this methodology.

Abstract:Phosphorous-containing heterocycles have attracted particular interest because they exhibit markedly different properties than common oxygen-, nitrogen-, and/or sulfur-based heterocyclic compounds. However, the study of such molecules remains difficult because of a lack of synthetic methods with which to synthesize them, especially with respect to the construction of P-stereogenic centers. Here, we present a simple, versatile method for the synthesis of PN-heterocycles bearing P(V) centers from phosphanamines through palladium-catalyzed intramolecular C–P coupling. Under the developed catalytic systems, chiral PN-heterocycles can be produced from P(III)-stereogenic precursors without loss of stereochemical integrity. Computational studies revealed the detailed reaction pathway and the origin of stereospecificity. The methods showed excellent functional-group tolerance, allowing access to diverse, optoelectronically relevant products that are stable under air and moisture.

Abstract:Palladium-catalyzed enantioselective domino Heck/intramolecular C–H functionalization reaction, as a valuable strategy for creating molecular diversity, has remained a prominent challenge. Here, we describe a Pd/XuPhos catalyst for asymmetric domino Heck/intermolecular C–H alkylation of unactivated alkenes with diverse polyfluoro- and heteroarenes in a highly chemo- and enantioselective manner. This process enables efficient synthesis of various dihydrobenzofurans, indolines and indanes, which are of interest in pharmaceutical research and other areas. Late-stage modifications of the core structures of natural products are also well showcased. Moreover, synthetic transformations create a valuable platform for preparing a series of functionalized molecules. Several control experiments for mechanistic study are conducted to pursue a further understanding of the reaction.

Abstract:Comprehensive SummaryA rhodium‐catalyzed denitrogenative carbonylation of 1,2,3‐thiadiazole with alkynes to construct (benzo)thiochromenones is developed. Both terminal and internal alkynes were compatible in this [3+2+1] cycloaddition, affording a broad range of poly‐substituted thiochromenones. Unsymmetrical alkyne substrates exhibited good regioselectivity. It is noteworthy that the present strategy could be used to construct both monocyclic and benzo‐thiochromenones, and it is a novel example of denitrogenative transannulation with CO insertion.

Abstract:We developed a novel protocol for the preparation of functionalized chromeno[2,3-c]pyrrol-9(1H)-one derivatives (CMPOs) 3 & 3' from 3-formylchromones with proline derivatives. This strategy involved an unprecedented cascade reaction entailing simple heating of a mixture of substrates 1 with 2 and toluene, followed by oxidation with oxygen and catalysis by CuBr. As a result, a series of CMPOs 3 & 3' were produced via cleavage of two bonds (2 C−C) and formation of three bonds (1 C−N and 2 C−C bonds) in a single step. This protocol can be used to synthesize functionalized CMPOs for combinatorial and parallel syntheses of CMPOs in a one-pot reaction.

Abstract:Tetrasubstituted alkenes are one of the most important classes of aggregation-induced emission luminogens with wide applications in analytical chemistry, bioimaging science, luminescent materials and cancer treatment. However, general methods for the assembly of tetrasubstituted alkenes, especially for unsymmetrical species, remain elusive. The established methods typically require prefunctionalized substrates, sensitive organometallic reagents and multiple-step operation. Herein, we report a dual phosphine/rhodium catalysis for the direct synthesis of unsymmetrical tetrasubstituted alkenes from readily available cyclopropenones, aryl halides and water. This reaction was achieved in a highly efficient and chemoselective manner by a synergistic merger of phosphine-mediated hydration with rhodium-catalyzed arylation, furnishing a diverse set of carboxyl acid–based tetrasubstituted alkenes (>80 examples). The resulting carboxylate groups can be employed as universal tags for the downstream synthesis of unsymmetrical tetraarylethenes. A range of the obtained compounds were demonstrated to be solid-state luminogens with obvious aggregation-induced emission properties. The success of late-stage functionalization of bioactive compounds further illustrate the synthetical utility of this protocol in material development and drug discovery.
