Abstract:Direct C-H functionalization of aryl ethers remains challenging owing to their low reactivity and selectivity. Herein, a novel strategy for nondirected C-H alkenylation of aryl ethers promoted by a dual ligand catalyst was demonstrated. This catalytic system readily achieved the highly efficient alkenylation of alkyl aryl ethers (anisole, phenetole, n-propyl phenyl ether, n-butyl phenyl ether and benzyl phenyl ether), cyclic aryl ethers (1,4-benzodioxan, 2,3-dihydrobenzofuran, dibenzofuran), and diphenyl oxides. Moreover, the proposed methodology was successfully employed for the late-stage modification of complex drugs containing the aryl ether motif. Interestingly, the compounds developed herein displayed fluorescent properties, which would facilitate their biological applications.

Abstract:An efficient Pd(II)-catalyzed synthesis of benzyl benzoates via direct functionalization of benzyl C(sp(3))-H bonds was developed. The method features a broad substrate scope. This method features high tolerance of functional groups, mild reaction condition, and high chemoselective when there are multiple active C(sp(3))-H bonds. A plausible oxidative coupling mechanism was proposed on the basis of mechanistic studies.

Abstract:The Suzuki-Miyaura coupling is among the most important C-C bond-forming reactions available due to its reliability, chemoselectivity, and diversity. Aryl halides and pseudohalides such as iodides, bromides, and triflates are traditionally used as the electrophilic coupling partner. The expansion of the reaction scope to nontraditional electrophiles is an ongoing challenge to enable an even greater number of useful products to be made from simple starting materials. Herein, we present how an NHC-based Pd catalyst can enable Suzuki-Miyaura coupling where the C(acyl)-O bond of aryl esters takes on the role of electrophile, allowing the synthesis of various ketone-containing products. This contrasts known reactions of similar esters that provide biaryls via nickel catalysis. The underlying cause of this mechanistic divergence is investigated by DFT calculations, and the robustness of esters compared to more electrophilic acylative coupling partners is analyzed.

Abstract:Palladium-catalyzed C-H functionalization of acyldiazomethanes with aryl iodides has been developed. This reaction is featured by the retention of the diazo functionality in the transformation, thus constituting a novel method for the introduction of diazo functionality to organic molecules. Consistent with the experimental results, the density functional theory (DFT) calculation indicates that the formation of Pd-carbene species in the catalytic cycle through dinitrogen extrusion from the palladium ethyl diazoacetate (Pd-EDA) complex is less favorable. The reaction instead proceeds through Ag2CO3 assisted deprotonation and subsequently reductive elimination to afford the products with diazo functionality remained. This C-H functionalization transformation can be further combined with the recently evolved palladium-catalyzed cross-coupling reaction of diazo compounds with aryl iodides to develop a tandem coupling process for the synthesis of α,α-diaryl esters. DFT calculation supports the involvement of Pd-carbene as reactive intermediate in the catalytic cycle, which goes through facile carbene migratory insertion with a low energy barrier (3.8 kcal/mol).

Abstract:In past decades, palladium-catalyzed direct selective C-H bonds functionalization has become a highly attractive strategy in organic synthesis and represents a highly desirable goal. This review introduces three extensively investigated modes for forming C-C bonds from C-H bonds via palladium catalysis; cross-coupling reactions of C-H bonds with aryl or alkyl halides/pseudohalides, cross-dehydrogenative couplings of two C-H bonds, and cross-coupling reactions of C-H bonds with organometallic reagents. A comprehensive summary of recent advances and detailed analysis on the versatility and practicality of these transformations are presented. The research trend for this strategy is also prospected.

Abstract:A novel and efficient palladium‐catalyzed cross‐coupling benzylation reaction of pentafluorobenzene through selective C−H functionalization has been developed. A series of carbonates and polyfluorobenzenes have been examined and found to give moderate to good yields. Abstract:Aromatic functionalizations are of current interest and importance in organic chemistry, recent transition‐metal catalyzed reactions can transform inert C–H bond into C–X (X = C, O, N, etc.) bonds, greatly enriching aromatic chemistry and related fields. Some well known palladium–containing intermediates such as palladacycles and allylpalladium intermediates play an important role in organic conversions. Similar with allyl group, benzyl group also possesses the capability of stabilizing palladium through η3‐binding. In the process of the formation of benzyl palladium intermediate, there is a transition state of dearomatization. Therefore, it is a challenging and meaningful research topic to seize these aromatic destruction intermediates, realize selective functionalization reaction on aromatic ring via them, and inhibit the formation of traditional benzylated products. Herein, we summarized the recent efforts on expanding the synthetic utility, as well as exploring the mechanism in aromatic functionalization via reactive benzyl palladium intermediates. We envision that the transformation via benzyl palladium intermediates will help promote the development in the field of palladium–catalyzed aromatic regioselective functionalization.

Abstract:<p>Transition-metal catalyzed multi-component reactions have captured the attention of researchers in organic synthesis and drug synthesis due to their advantages of simple operation, easy availability of raw materials and without separation of intermediates. Among the multi-component reactions, the three-component processes have been developed into effective organic procedures. This personal account reviews our and other group's studies on the development of three-component coupling reaction for the rapid construction of two new chemical bonds simultaneously via benzylpalladium intermediates. Catalyst-switched three-component reactions of benzyl halides, activated olefins, and allyltributylstannane were successfully conducted to produce the corresponding benzylallylation products. Activation and conversion of carbon monoxide and carbon dioxide via π-benzylpalladium intermediates provide access to a wide range of unsaturated ketones and esters with excellent functional group tolerance. Meanwhile, other methods to produce benzylpalladium intermediates, including Heck insertion of alkenes into arylpalladium complexes, the oxidative addition of benzyl carbonate to palladium complexes and palladium−carbene migratory insertion, were also highlighted.</p>

Abstract:1,4-cis-disubstituted cyclic compounds play a pivotal role in pharmaceutical development, offering enhanced potency and bioavailability. However, their stereoselective and modular synthesis remains a long-standing challenge. Here, we report an innovative strategy for accessing these structures via mild conditions employing cyclic 1,3-dienes/alkyl(aryl)halides and amines. This procedure exhibits a wide substrate scope that tolerates various functional groups. The utility of this method is demonstrated in the efficient synthesis of a TRPV6 inhibitor, CFTR modulator and other bioactive molecules. Combined experimental and computational studies suggest that the hybrid palladium-catalyzed radical-polar crossover mechanism is crucial for achieving the exceptional 1,4-syn-addition selectivity (dr > 20:1).

Abstract:It is well known that benzyl esters are prevalent functional groups in medicinal agents and natural products. The traditional methods of preparing benzyl esters by utilizing benzyl alcohols/bromides/chlorides with carboxylic acids as coupling partners have been reported over the decades (Scheme 1a). Furthermore, Khan's group proposed benzyl esters can also be obtained by the reaction of aromatic carboxylic acids with substituted toluenes in the presence of NaBrO3/NaHSO3 in early 2003. However, some approaches have disadvantages of long reaction time or sensitivity to the environment. With increasing interest in transition metal-catalyzed C–H bonds activation to construct C–O bonds, several types of transition metals, including Cu, Pd, Pt and Rh have been recently revealed admirable catalytic activity in esterication. Among them, the approach of Pd-catalyzed benzylation of toluene with carboxylic acids under 1 atm of oxygen has been disclosed by Zhang's group (Scheme 1b). It did not need to be pre-functionalized and provided a more facile, atom-economic method to prepare benzyl esters. Despite their obvious potential application, these transition metal-catalyzed C–H bond activation methods suffered from a few drawbacks such as expense and toxicity of transition metals, which limit their practicability. Hence, the development of a facile and practical approach for constructing benzyl esters under metal-free conditions is extremely desirable.

Abstract:Herein, we report a palladium/ norbornene/ copper‐catalyzed single‐step approach that merge selective ortho C‐H bond esterification with ipso thiolation for construction of synthetically versatile 2‐arylthio aryl esters under exceptionally mild conditions. Importantly, this process proceeded in a highly efficient manner, allowing alkoxycarbonyl and thio groups to be installed into one aryl iodides simultaneously by harnessing thiocarbonate as bifunctional reagent. The method has been demonstrated to accommodate good functionalities and feature broad substrate scope.
