Abstract:Particle separation is important in chemical and biomedical analysis. Among all particle separation approaches, microstructure filtration which based particles size difference has turned into one of the most commonly methods. By controlling the movement of particles, dielectrophoresis has also been widely adopted in particle separation. This work presents a microfluidic device which combines the advantages of microfilters and dielectrophoresis to separate micro-particles and cells. A three-dimensional (3D) model was developed to calculate the distributions of the electric field gradient at the two filter stages. Polystyrene particles with three different sizes were separated by micropillar array structure by applying a 35-Vpp AC voltage at 10 KHz. The blocked particles were pushed off the filters under the negative dielectrophoretic force and drag force. A mixture of Haematococcus pluvialis cells and Bracteacoccus engadinensis cells with different sizes were also successfully separated by this device, which proved that the device can separate both biological samples and polystyrene particles.

Abstract:Effective enrichment and separation of particles and mixing of multiphase microflows can be technically challenging in microfluidic sample processing and analysis. Acoustofluidic techniques have advantages such as high biocompatibility, versatility, and simple device design to meet these demands. Here, we present a novel tri-symmetrical acoustofluidic centrifuge based on traveling surface acoustic waves (TSAWs), achieving particle enrichment and bidirectional size-based separation, pushing away the larger particles while enriching the smaller ones, in a rapid time (<6 s). A microfluidic chamber was designed for further extraction of treated particles and cells. Experimental results show that our device can achieve enrichment of particles in a range of sizes and high purity separation (e.g. >90 % in 5 µm and 40 µm sample), demonstrating its sample processing capabilities. The acoustic treatment did not affect cells (consistent with the control group above 80 %). TSAW-induced acoustic streaming enabled efficient mixing (mixing efficiency χ >85 %) of fluorescent multi-phase microflows within a short time (<0.5 s). The underlying physical mechanisms have been verified by numerical simulations.

Abstract:This study introduces an innovative microfluidic particle separation technique that integrates inertial focusing with deterministic lateral displacement (DLD) on a single chip, significantly enhancing the efficiency of particle separation. This new method completes flow rate matching through a designed pre-focusing and pre-separation inertial structure, avoiding the use of sheath flow. The process involves a sequence of channels: a rectangular channel, a contraction-expansion array (CEA), a lateral separation channel, and another rectangular channel. This three-stage inertial method shortens the focusing channel length and reduces the pressure on subsequent separation stages, streamlining the separation process. It has been demonstrated to separate 20 μm particles from a mixture containing both 10 μm and 20 μm particles with remarkable precision. The technique achieves a 100% separation efficiency, ensuring all target particles are correctly isolated, and a 96.1% separation purity, indicating that the isolated particles are almost entirely free from contaminants. By eliminating the need for sheath flow, this method simplifies the apparatus and reduces operational complexity, offering significant advantages over traditional particle separation techniques. The high efficiency and purity levels achieved by this method highlight its potential for a wide range of applications in fields requiring precise particle separation, such as medical diagnostics, environmental monitoring, and industrial processing.

Abstract:The microfluidic passive control of microparticles largely relies on the hydrodynamic effects of the carrier media such as Newtonian fluids and viscoelastic fluids. Yet the viscoelastic/Newtonian interfacial effect has been scarcely investigated, especially for high-resolution particle separation. Here we report a microfluidic co-flow of Newtonian (water or PBS) and viscoelastic fluids (PEO) for the size-dependent separation of microparticles. The co-flow condition generates a stable viscoelastic/Newtonian interface, giving rise to the wall-directed elastic lift forces that compete with the center-directed lift forces, and efficiently hinders the migration of microparticles from the Newtonian to the viscoelastic fluid in a size-dependent manner. An almost complete separation of a binary mixture of 1 μm and 2 μm polystyrene particles is achieved by the co-flow of water and a very dilute PEO solution (100 ppm), whereas the sole use of water or PEO could not lead to an efficient separation. This co-flow microfluidic system is also applied for the separation of Staphylococcus aureus (1 μm) from platelets (2-3 μm) with >90% efficiencies and purities.
A Self-Cleaning Micro-Fluidic Chip Biospired by the Filtering System of Manta Rays

Abstract:Size-based particle filtration has become indispensable in numerous biomedical and environmental applications. In this study, bioinspired by the filter-feeding mechanism (lobe filtration) of manta rays, we designed a U-shaped biomimetic gill rake filter that combined lobe filtration and Dean flow to filter monodisperse suspensions, bi-disperse suspensions and yeast cells. Compared with other equipment using the inertial focusing technology, our equipment can perform high-throughput (up to 8 mL min-1) and high-efficiency filtration of particles (maximum filtration efficiencies of 96.08% and 97.14% for 10 and 15 mu m monodisperse suspensions at the optimum flow rate of 6 mL min-1). The complex velocity field of the micro-fluidic flow within the filter is numerically simulated, and in combination with experiments, a threshold for the flow rate is identified. When the inlet flow rate exceeds the threshold value, the efficiency of particle filtration is increased rapidly. Afterwards, by analysing the filtration mechanism, we develop three novel filtration processes. The equilibrium positions of the particles and yeast cells in the main channel are close to the outer wall at high flow rate, which diminishes the likelihood of particles and yeast cells entering the side channel. This configuration establishes a self-cleaning mechanism, ensuring prolonged and efficient operation of the filter with high-throughput processing. Furthermore, the influence of the filter lobe angle and channel width on the filtration efficiency and outlet flow rate ratio are explored, and an optimisation plan is prepared. Size-based particle filtration has become indispensable in numerous biomedical and environmental applications.
A Simplified Microfluidic Device for Particle Separation with Two Consecutive Steps: Induced Charge Electro-osmotic Prefocusing and Dielectrophoretic Separation.

Abstract:Continuous dielectrophoretic separation is recognized as a powerful technique for a large number of applications including early stage cancer diagnosis, water quality analysis, and stem-cell-based therapy. Generally, the prefocusing of a particle mixture into a stream is an essential process to ensure all particles are subjected to the same electric field geometry in the separation region. However, accomplishing this focusing process either requires hydrodynamic squeezing, which requires an encumbering peripheral system and a complicated operation to drive and control the fluid motion, or depends on dielectrophoretic forces, which are highly sensitive to the dielectric characterization of particles. An alternative focusing technique, induced charge electro-osmosis (ICEO), has been demonstrated to be effective in focusing an incoming mixture into a particle stream as well as nonselective regarding the particles of interest. Encouraged by these aspects, we propose a hybrid method for microparticle separation based on a delicate combination of ICEO focusing and dielectrophoretic deflection. This method involves two steps: focusing the mixture into a thin particle stream via ICEO vortex flow and separating the particles of differing dielectic properties through dielectrophoresis. To demonstrate the feasibility of the method proposed, we designed and fabricated a microfluidic chip and separated a mixture consisting of yeast cells and silica particles with an efficiency exceeding 96%. This method has good potential for flexible integration into other microfluidic chips in the future.

Abstract:Droplet sorting and enrichment, as a prominent field within microfluidic technology, represent a pivotal stage in the manipulation of droplets and particles. In recent times, droplet sorting methods based on lab-on-disk (LOD) have garnered significant interest among researchers for their inherent merits, including high throughput, ease of operation, seamless device integration, and independence from supplementary driving forces. This study introduces a centrifugal force-driven microfluidic chip comprising spiral microchannels. The chip incorporates microhole arrays along the sidewall of the spiral channels, enabling size-based sorting and enrichment of microdroplets under the influence of multiple forces. Firstly, a comparative analysis was performed to assess the influence of the separation port structure and rotational speed on efficiency, and a mechanical modeling approach was employed to conduct kinetic analyses of droplet behavior during instantaneous separation. Those findings demonstrated a good agreement with the experimental results at ω <100 rpm. Subsequently, sorting experiments on homogeneous droplets indicated that repetitive sorting could increase the recovery ratios, RT(α), of high-concentration droplets (20.7%) from 35.3% to over 80%. We also conducted a sorting experiment on three-component homogeneous-phase emulsions using a serially connected chip array, and the sorting throughput was 0.58 mL/min. As a result, the RT(α) for 60 and 160 μm droplets were 99.4% and 88.9%, respectively. Lastly, we conducted elution experiments and dualsample sorting on a single chip, and the fluorescence results demonstrated that this study provided an efficient and non-cross-contaminating sorting method for non-homogenous phase multi-sample microreactor units.
High-throughput and Clogging-Free Microfluidic Filtration Platform for On-Chip Cell Separation from Undiluted Whole Blood.

Abstract:Rapid separation of white blood cells from whole blood sample is often required for their subsequent analyses of functions and phenotypes, and many advances have been made in this field. However, most current microfiltration-based cell separation microfluidic chips still suffer from low-throughput and membrane clogging. This paper reports on a high-throughput and clogging-free microfluidic filtration platform, which features with an integrated bidirectional micropump and commercially available polycarbonate microporous membranes. The integrated bidirectional micropump enables the fluid to flush micropores back and forth, effectively avoiding membrane clogging. The microporous membrane allows red blood cells passing through high-density pores in a cross-flow mixed with dead-end filtration mode. All the separation processes, including blood and buffer loading, separation, and sample collection, are automatically controlled for easy operation and high throughput. Both microbead mixture and undiluted whole blood sample are separated by the platform effectively. In particular, for white blood cell separation, the chip recovered 72.1% white blood cells with an over 232-fold enrichment ratio at a throughput as high as 37.5 μl/min. This high-throughput, clogging-free, and highly integrated platform holds great promise for point-of-care blood pretreatment, analysis, and diagnosis applications.

Abstract:The separation of target microparticles using microfluidic systems owns extensive applications in biomedical, chemical, and materials science fields. Integration of microfluidic sorting systems employing dielectrophoresis (DEP) technology has been widely investigated. However, enhancing separation efficiency, purity, stability, and integration remains a pressing issue. This study proposes a stepwise multi-stage continuous DEP separation microfluidic chip with a microfilter structure. By leveraging a stepwise electrode configuration, a gradient electric field is generated to drive target microparticles along the electric field gradient, thereby enhancing separation efficiency. Innovative integration of a microfilter structure facilitates simultaneous filtration and improves flow field distribution, thus enhancing system stability. Through the synergistic effect of stepwise electrodes and the microfilter structure, superior coupling of electric and flow fields is achieved, consequently improving the sorting purity, separation efficiency, and system stability of the DEP-based microfluidic sorting system. Validation through simulation and separation of polystyrene microspheres demonstrates the excellent particle separation performance of the proposed system. It evidently shows potential for seamless extension to various biological microparticle sorting applications, harboring significant prospects in the biomedical domain field.

Abstract:Controllable manipulation of fluid flow is crucial for efficient particle separation, which is associated with plenty of biomedical and industrial applications. Microfluidic technologies have achieved promising progress in particle positioning depending on inertial force with or without the help of the Dean effect. Herein, we describe an inertial microfluidic system containing a spiral microchannel for various highly efficient particle separations. We demonstrated that Dean-like secondary flow can be regulated by geometric confinement in the microchannel. On the introduction of a library of micro-obstacles into the spiral microchannels, the resulting linear acceleration of secondary flow can be applied to remarkably enhance particle focusing in time and space. Further, multiple separating and sorting manipulations of particles including polymeric particles, circulating tumor cells, and blood cells, can be successfully accomplished in the dimension-confined spiral channels in a sheathless, high-throughput (typically 3 ml min(-1)), long-term (at least 4 h), and highly-efficient (up to 99.8% focusing) manner. The methodological achievement pointing to ease-of-use, effective, and high-throughput particle manipulations is useful for both laboratory and commercial developments of microfluidic systems in life and material sciences.

Abstract:The application of bubbles in lab-on-chip devices has attracted considerable attention and is exerting an increasingly significant role in recent years due to its influential functions for fluid and particle manip-ulation. Herein, we demonstrated a simple air bubble trapping method based on the surface tension of hydrophobic chip material and demonstrated particle manipulation utilizing oscillating bubbles. The mi-crofluidic device has a T-shaped junction structure with triangular obstacles, which allows us to transfer air -pocket generated due to non-synchronized liquid filling in the microchannel to stable bubbles attached to channel wall at the desired location. The trapped bubbles can maintain stable state at a flow rate of less than 50 mu L/min. By activating bubbles with the sound wave at a specific frequency, we can achieve particle focusing, trapping, extraction and enrichment in a contact-free, label-free and continuous manner. The proposed bubble trapping method may serve as a reusable, detachable and biocompatible tool for broader bubble-based applications in chemical, biology, and engineering fields, such as drug delivery, cell focusing, as well as isolation of tumor cell. High-efficiency blood separation utilizing spiral filtration microchannel with gradually varied width

Abstract:A hybrid microfluidic separation device that uses the crossflow and the centrifugation effect to separate human plasma from blood cells has been designed and fabricated. The chip mainly consists of the spiral microchannel, which is divided into inner and outer mcirochannels by micropillar arrays. The slits between the micropillars act as filter. Clogging and jamming in this filtration structure are efficiently alleviated. Remarkably, the width of the inner microchannel is gradually decreasing from inlet to outlet in order to increase separation efficiency. The performances of the separation device were investigated theoretically and experimentally. Due to high separation efficiency and compact structure, it is envisaged that this device can be integrated with other microfluidic device for point-of-care diagnostics in the near future.

Abstract:Recently, the bubble-based systems have offered a new paradigm in microfluidics. Gas bubbles are highly flexible, controllable and barely mix with liquids, and thus can be used for the creation of reconfigurable microfluidic systems. In this work, a hydrodynamically actuated bubble-based microfluidic system is introduced. This system enables the precise movement of air bubbles via axillary feeder channels to alter the geometry of the main channel and consequently the flow characteristics of the system. Mixing of neighbouring streams is demonstrated by oscillating the bubble at desired displacements and frequencies. Flow control is achieved by pushing the bubble to partially or fully close the main channel. Patterning of suspended particles is also demonstrated by creating a large bubble along the sidewalls. Rigorous analytical and numerical calculations are presented to describe the operation of the system. The examples presented in this paper highlight the versatility of the developed bubble-based actuator for a variety of applications; thus providing a vision that can be expanded for future highly reconfigurable microfluidics.

Abstract:For passive droplet generation, multiple parameters such as the fluid viscosities and flow rates of the continuous and discrete phases correlate to each other, raising relevant control difficulties. In the current study, a droplet platform that is capable of handling dissimilar liquids is proposed. Through combining oscillatory flow and electric charge, synchronized generation and forced coalescence of different droplets can be achieved. Its application for the separation of E. coli from blood is tested, which leads to a high capture efficiency with less sample and within a shorter time than usual.

Abstract:Microfluidic techniques have emerged as promising strategies for a wide variety of synthetic or biological sorting. Unfortunately, there is still a lack of sorting with automatic and handy operation. In contrast to passively generated vortices, the thermocapillary vortices produced by temperature gradient have the advantages of flexible manipulation, stable strength, and simple integration. In this Letter, we present a device used for the pump-free separation of particles through vortices interaction without external fluidic control systems required for the majority of existing devices. Specifically, the device induces a different flow type upon the actuation of optical power, and the flow functions, such as simultaneous pumping and sorting, agree with stimulation results very well. More importantly, our developed sorting device can achieve separations by means of tunable cutoff diameter size. Therefore, this versatile device can be utilized to sort complex samples with the advantages of portability, user-friendly control, and automation.

Abstract:A microfluidic separation device that uses the cross flow and the centrifugation effect to separate human plasma from whole blood has been designed, fabricated and evaluated. The chip mainly consists of a spiral channel divided into inner and outer microchannels by micropillar arrays, which are employed to filter blood cells and plasma. The major feature of this chip is that the width of the inner channel decreases gradually from the inlet to the outlet in order to increase the separation efficiency. Clogging and jamming in this filtration structure are efficiently alleviated. The performances of the separation device have been investigated theoretically and experimentally. Due to high purity of plasma and compact structure, this device can be used as either a plasma extraction device solely, or a pretreatment component that is integrated with other microfluidic device for point-of-care diagnostics.

Abstract:In this paper, we report an inertial microfluidic device with simple geometry for continuous extraction of large particles with high size-selectivity (<2 μm), high efficiency (∼90%), and high purity (>90%). The design takes advantage of a high-aspect-ratio microchannel to inertially equilibrate cells and symmetric chambers for microvortex-aided cell extraction. A side outlet in each chamber continuously siphons larger particles, while the smaller particles or cells exit through the main outlet. The design has several advantages, including simple design, small footprint, ease of paralleling and cascading, one-step operation, and continuous separation with ultra-selectivity, high efficiency and purity. The described approach is applied to manipulating cells and particles for ultra-selective separation, quickly and effectively extracting larger sizes from the main flow, with broad applications in cell separations.

Abstract:In this paper, a novel spiral-channel micro-filter with high throughput and compact layout, taking advantages of the centrifugation effect, has been developed. Numerical results indicate that the flux, which penetrates through the filtration wall, in a circle channel device is 2.25 times than that in the straight channel filter at the same conditions. The higher flow velocity allows higher filtration efficiency. The experimental results show that large particles are completely separated from original mixtures, and small particles are remarkably enriched by several filtration cycles.

Abstract:We report on a microfluidic device capable of sorting nanoscale particulates and water-in-oil emulsions at high throughput. The device is passive, relying solely on hydrodynamic forces and the emulsion mass to achieve separation. We use the microfluidic device to deliver surfactants and lipids to the emulsion surface. This is achieved by immersing the emulsions in a fluid stream with a high concentration of the nano-particulates. The particulates assemble on the surface of the emulsions as they are transported along the stream. The emulsions are then transferred (i.e. separated) into a second fluid stream that is devoid of surrounding material. The performance of the device is evaluated for a range of flow rates, nano-particulate concentrations, and emulsion sizes.
